Cationized Lignin Loaded Alginate Beads for Efficient Cr(VI) Removal

  • In this study, lignin, a lignocellulosic biomass, was chemically modified to produce polyethyleneimine-grafted lignin (PKL) with maximum hexavalent chromium [Cr(VI)] adsorption capacity. Changes in the physicochemical properties due to the cationization of lignin were confirmed through elemental analysis, Fourier transform infrared spectroscopy, and moisture stability evaluation. Alginate (Alg) beads containing PKL (Alg/PKL) were prepared by incorporating cationic lignin into the Alg matrix to apply the prepared PKL in a batch-type water treatment process. The optimal Alg/lignin mixing ratio was selected to increase the Cr(VI) adsorption capacity and minimize lignin elution from the aqueous system. The selected Alg/PKL beads exhibited an excellent Cr(VI) removal capacity of 478.98 mg/g. Isothermal adsorption and thermodynamic analysis revealed that the Cr(VI) removal behavior of the Alg/PKL beads was similar to that of heterogeneous chemical adsorption. In addition, the bulk adsorbent material in the form of beads exhibited adsorption behavior in three stages: surface adsorption, diffusion, and equilibrium.

Conversion Characteristics of Chemical Constituents in Liriodendron tulipifera and Their Influences on Biomass Recalcitrance during Acid-Catalyzed Organosolv Pretreatment

  • The conversion characteristics of the major components of Liriodendron tulipifera were investigated during acid-catalyzed organosolv pretreatment. Glucan in L. tulipifera was slowly hydrolyzed, whereas xylan was rapidly hydrolyzed. Simultaneous hydrolysis and degradation of xylan and lignin occurred; however, after complete hydrolysis of xylan at higher temperatures, lignin remained and was not completely degraded or solubilized. These conversion characteristics influence the structural properties of glucan in L. tulipifera. Critical hydrolysis of the crystalline regions in glucan occurred along with rapid hydrolysis of the amorphous regions in xylan and lignin. Breakdown of internal lignin and xylan bonds, along with solubilization of lignin, causes destruction of the lignin-carbohydrate complex. Over a temperature of 160℃, the lignin that remained was coalesced, migrated, and re-deposited on the surface of pretreated solid residue, resulting in a drastic increase in the number and content of lignin droplets. From the results, the characteristic conversions of each constituent and the changes in the structural properties in L. tulipifera effectively improved enzymatic hydrolysis in the range of 140℃-150℃. Therefore, it can be concluded that significant changes in the biomass recalcitrance of L. tulipifera occurred during organosolv pretreatment.

Extraction and characterization of lignin from black liquor and preparation of biomass-based activated carbon there-from

  • In the present study, biomass-based lignin was extracted from industrial waste black liquor and the extracted lignin was characterized by means of attenuated total reflectance-Fourier transform infrared spectroscopy and $^1H-nuclear$ magnetic resonance spectroscopy. The extracted lignin was carbonized at different temperatures and then activated with steam at $850^{\circ}C$. The extracted lignin in powder state was transformed into a bulky carbonized lignin due to possible fusion between the lignin particles occurring upon carbonization. The carbonized and then pulverized lignin exhibits brittle surfaces, the increased thermal stability, and the carbon assay with increasing the carbonization temperature. The scanning electron microscopic images and the Brunauer-Emmett-Teller result indicate that the steam-activated carbon has the specific surface area of $1718m^2/g$, which is markedly greater than the carbonized lignin. This study reveals that biomass-based activated carbon with highly porous structure can be produced from costless black liquor via steam-activation process.

고추 줄기의 화학 조성분 및 알칼리 펄프화 (Chemical Composition and Alkaline Pulping of a Stem of Red Pepper (Capsium annuum L.))

  • 고추줄기의 화학 조성분과 lignin 화학구조를 분석하였고, 알칼리 증해 적성과 pulp의 섬유장을 측정한 후 목재와 비교하였다. 고추줄기의 화학 조성은 목재에 비해 추출성분, 회분함량이 높았으며, Klason lignin은 침·활엽수의 중간 정도의 함량을 나타내었다. 고추줄기 Klason lignin의 원소분석은 자작나무의 Klason lignin과 비교하여 탄소와 수소는 유사한 양으로 나타난 반면 산소의 함량은 낮게 나타나고 질소의 함량은 비교적 높게 나타났다. 니트로벤젠 산화분해는 S/V가 1보다 낮으므로 고추줄기 리그닌은 vanillin의 양이 syringaldehyde보다 상당히 많은 침엽수 리그닌과 유사한 구조를 보이는 것으로 추정되었다. 고추줄기의 알칼리 증해 적성은 활성알칼리 20%, 액비 1:7의 조건하에서 0.2%-Anthraquinone을 첨가하여 증해하는 것이 리그닌 함량과 수율면에서 최적 증해 조건이었다. 고추줄기 pulp의 섬유장은 약 0.47 mm로 보통 목재섬유에 비해서 매우 짧아 고추대 만으로 제지용 pulp를 제조하는 것보다는 목재 pulp와 혼합하여 사용하거나 비교적 단섬유가 많이 필요한 특수한 용도로 사용 가능하다.

미동정 부후균에 의한 소나무재의 Lignin 분해와 주사전자현미경(SEM)을 이용한 관찰 (Lignin Degradation of Pine Wood by Unidentified Decay Fungi and Observation by Scanning Electron Microscope)

  • 목재의 주성분 중 가장 분해가 어려운 리그닌을 분해하는 균주를 선발하기 위해 산림지역에서 채취한 부후목과 자실체로부터 균을 분리하였다. 리그닌 분해능은 활엽수보다 미생물 분해가 어렵다고 알려진 침엽수인 소나무재에 미생물을 처리하여 Klason 리그닌 정량을 통해 조사하였다. 또한 선발균에 의한 소나무재의 분해과정과 부후정도를 주사전자현미경(Scanning Electron Microscope ; SEM)을 이용해 조사하였다. 선발 균주 중 CJ-6에서 소나무 리그닌의 분해율이 49.48%로 가장 높았으며, 이것은 리그닌 분해 우수 균주로 알려진 Trametes versicolor의 40.58%와 비교해 보았을 때 리그닌 분해력이 더 우수하였다. 균주들 중 리그닌 분해력이 좋게 나타난 2개의 균주를 대상으로 부후에 의한 목재조직의 변화를 관찰하였는데 두 균주의 부후형이 비슷한 경향을 나타내었다. 부후 20일 경과에서는 균사의 침입은 있었으나 아직 목재는 건전한 상태를 유지하고 있었으며, 60일 간의 부후에서는 부후가 어느 정도 진행되어 가도관 벽과 방사조직의 세포벽의 일부가 분해되어 있음을 알 수 있었다. 100일간 부후가 진행된 경우에는 부후가 상당히 진행되어 가도관 세포벽 안쪽이 분해가 많이 진행되어 있었으며, 방사조직의 세포벽이 많이 분해되어 있어 세포간의 구별이 어려웠다.

Fungal Metabolism of Environmentally Persistent Compounds: Substrate Recognition and Metabolic Response

  • Mechanism of lignin biodegradation caused by basidiomycetes and the history of lignin biodegradation studies were briefly reviewed. The important roles of fungal extracellular ligninolytic enzymes such as lignin and manganese peroxidases (LiP and MnP) were also summarized. These enzymes were unique in their catalytic mechanisms and substrate specificities. Either LiP or MnP system is capable of oxidizing a variety of aromatic substrates via a one-electron oxidation. Extracellular fungal system for aromatic degradation is non-specific, which recently attracts many people working a bioremediation field. On the other hand, an intracellular degradation system for aromatic compounds is rather specific in the fungal cell. Structurally similar compounds were prepared and metabolized, indicating that an intracellular degradation strategy consisted of the cellular systems for substrate recognition and metabolic response. It was assumed that lignin-degrading fungi might be needed to develop multiple metabolic pathways for a variety of aromatic compounds caused by the action of non-specific ligninolytic enzymes on lignin. Our recent results on chemical stress responsible factors analyzed using mRNA differential display techniques were also mentioned.

Phanerochaete chrysosporium에 의한 Lignin Peroxidase의 생성과 Pentachlorophenol(PCP)의 분해 (A Study on Synthesis of Lignin Peroxidase and Degradation of Pentachlorophenol(PCP) by Phanerochaete chrysosporium)

  • Experiments for lignin peroxidase production have been conducted by aerobic fermentation of Phanerochaete chrysosporium under low shear rate and enriched oxygen environment. The result of flask cultures of white rot fungus indicated that high oxygen concentration and low shear force were essential for enhancement of lignin peroxidase production. Pentachlorophenol was readily degraded by lignin peroxidase produced in nutrient limited flask cultures. Polyurethane foam was fond to be an effective immobilization matrix of P. chrysosporium.

Studies on the Ligninolytic Enzyme Activities During Biological Bleaching of Kraft Pulp with Newly Isolated Lignin-Degrading Fungi

  • A screening has been performed to find hyper-ligninolytic fungi, which degtrade beech and pine lignin extensively in order to broaden the understanding of the ligninolytic enzymes elaborated by various white-rot fungi. One hundred and twenty two ligninolytic strains were selected from decayed woods with a selective medium for screening ligninolytic wood-rotting fungi. Two of them, Phanerochaete sordida YK-624 and YK-472, showed much higher ligninolytic activity and selectivity in beech-wood degradation than typical lignin-degrading fungi, phanerochaete chrysosporium and Coriolus versicolor. They also degraded birch dioxane lignin and residual lignin in unbleached kraft pulp(UKP) much more extensively than P. chrysosporium and C. versicolor. During fungal treatment of beech wood-powder, the fungus strain P. sordida YK-624 showed higher activity of extracellular manganese peroxidase (MnP) in the medium than P. chrysosporium. It also showed MnP activity, which would not be lignin peroxidast during treatment of oxygen-bleached kraft pulp(OKP) and under enzyme-inducing conditin.

Induction and Stabilization of Lignin Peroxidase from Phanerochaete chrysosporium

  • Veratryl alcohol which has been reported as an inducer for lignin peroxidase showed different effects on the enzyme biosynthesis in Phanerochaete chrysosporium depending on the addition time. Enzyme expression was optimally induced by adding veratryl alcohol when the carbon source began to be depleted. Hydrogen peroxide, to some extent, stimulated production of lignin peroxidase, but beyond a certain concentration, inactivated lignin peroxidase. Tween 80 induced the formation of small pellets, which were resistant to the deactivation by shear stress. Lignin peroxidase production was increased twice compared with that of the control by adopting all the optimal factors in the culture system.

종의 포플라수종에서 리그닌생합성에 관계된 OMT유전자의 발현 (Seasonal Expression of OMT Gene in Relation to Lignin biosynthesis in two Poplar Species)

  • 리그닌함량이 감소한 펄프소재의 육성을 위해 OMT유전자와 리그닌생합성 간의 관계를 2종의 포플라 수종을 이용하여 실험하였다. OMT유전자의 발현은 리그닌생합성이 가장 많이 일어나는 developing secondary xylem에서 높앗다. 이 사부조직에서의 리그닌함량의 증가는 봄에 성장을 개시할 때부터 증가하기 시작하여 8월 말 경에 가장 높았다. 이러한 리그닌의 증가는 OMT유전자의 발현 증가와 일치하였으며, 이는 OMT유전자가 리그닌생합성에서 매우 중요한 역할을 함을 나타내는 것이다.
