Characteristics of Meteorological and Marine Environments for the Red Tide Occurrence of Mid-South Sea in Korea (한국 남해중부해역의 적조발생에 관한 기상 및 해양환경 특성)

  • This study deals with the relationship between the red tide occurrence and the meteorological and marine factors, the prediction of areas where the red tide is likely to occur based on the information, and the satellite monitoring for the red tide in mid-South Sea of Korea. From 1990 to 2001, the red tide was observed every year and the number of occurrences increased as well. The red tide mostly occurred in July, August, and September. The most important meteorological factor governing the mechanisms of the increase in the number of red tide occurrences is found to be a heavy precipitation. It was found that the favorable marine environmental conditions for the red tide formation are some of marine factors such as the warm water temperature, the low salinity, the high suspended solid, the low phosphorus, and the low nitrogen. The necessary conditions for the red tide occurrence are found to be the heavy precipitation (23.4-54.5 mm) for 2∼4 days, the warm temperature $(24.6∼25.9^{\circ}C)$, proper sunshine (2∼10.3 h), and light winds (2∼4.6 m/s & SW) for the day in red tide occurrence. It was possible to monitor the spatial distributions and concentration of the red tide using the satellite images. It was found that the likely areas for red tide occurrence in August 2000 were Yosu - Dolsan coast, Gamak bay, Namhae coast, Marado coast, Goheung coast, and Deukryang bay.

  • Direct synthesis of graphene using a chemical vapor deposition (CVD) has been considered a facile way to produce large-area and uniform graphene film, which is an accessible method from an application standpoint. Hence, their fundamental understanding is highly required. Unfortunately, the CVD growth mechanism of graphene on Cu remains elusive and controversial. Here, we present the evidences for two different growth modes of graphene on Cu investigated by varying carbon feedstock (C2H2 and CH4) and working pressure. The number of uniform graphene layer grown by C2H2 increased with increasing its injection time. A combined secondary ion mass spectrometry (SIMS) and X-ray diffraction (XRD) study revealed a carbon-diffused Cu layer created below surface region of Cu substrate with the expansion of Cu lattice. The graphene on Cu was grown by the diffusion and precipitation mode not by the surface adsorption mode, because similar results were observed in graphene/Ni system. The carbon-diffused Cu layer was also observed after graphene growth under high CH4 pressure. Based on various previous results and ours, we have successfully found that there are two selective growth modes for graphene on Cu substrate, and a desired mode can be chosen by tuning working pressure corresponding to the kind of carbon feedstock. We believe that this finding will shed light on high quality graphene growth and its multifaceted applications.

Preparation and Drug Release of All-Trans Retinoic Acid-Loaded Poly(L-lactic acid) Nanoparticles (레티노산 함유 폴리락탄산 나노입자의 제조 및 약물 방출)

  • To develop an intravenous delivery system of all-trans retinoic acid (ATRA) for the cancer therapy, poly(L-lactic acid) nanoparticles were prepared and characterized. Emulsification-solvent evaporation method was chosen to prepare submicron sized nanoparticles. Spherical nanoparticles less than 200 nm in diameter with narrow size distribution were prepared, and the entrapment efficiency of drug was more than 95%. The endothermic peak at $183^{\circ}C$ and X-ray crystallographic peak of ATRA appeared in the nanoparticle system, suggesting the inhibition of crystallization of ATRA by polymer adsorption during the precipitation process. ATRA was released at $37^{\circ}C$ for 60 days and the release rate was dependent on the concentration of drug incorporated in the nanoparticles. While ATRA was unstable in the light, it was very stable at $4^{\circ}C$. These results suggest the usefulness of PLA nanoparticles as a sustained and prolonged release carrier for ATRA.

Development of Competitive Direct Enzyme-linked Immunosorbent Assay for the Detection of Gentamicin Residues in the Plasma of Live Animals

  • Competitive direct ELISA was developed to detect gentamicin residues. Mice immunized with gentamicin-keyhole limpet hemocyanin (KLH) conjugate developed good antiserum titers, which gradually increased with booster injections, indicating immunization was successfully processed. Monoclonal antibody against gentamicin was prepared using hybridoma cells cloned by limit dilution of fused cells. IgG was purified from ascites fluid of hybridoma cell-injected mice through ammonium sulfate precipitation and Sephadex G-25 gel filtration. After the gel filtration, fractions of high antibody titer were further purified through affinity chromatography on protein A/G column. Monoclonal antibody against gentamicin was confirmed as IgG1, which has kappa light chain. Cross-reactivities ($CR_{50}$) of gentamicin monoclonal antibody to other aminoglycosides (kanamycin, neomycin, and streptomycin) were less than 0.005%, indicating the monoclonal antibody was highly specific for gentamicin. Standard curve constructed through competitive direct ELISA showed measurement range (from 80 to 20% of B/$B_0$ ratio) of gentamicin was between 1 and 40 ng/ml, and 50% of B/$B_0$ ratio was about 4 ng/ml. The gentamicin concentration rapidly increased to 1,300 ng/ml after the intramuscular administration up to 2 h, then sharply decreased to less than 300 ng/ml after 4 h of withdrawal, during which the elimination half-life ($t_{1/2}$) of gentamicin in the rabbit plasma was estimated to be 1.8 h. Competitive direct ELISA method developed in this study using the prepared monoclonal antibody is highly sensitive for gentamicin, and could be useful for detecting gentamicin residues in plasma of live animals.

Seasonal Variation of Planktonic Foraminifera Assemblage in response to Seasonal Shift of Inter-Tropical Convergence Zone in the Northeastern Equatorial Pacific (적도수렴대의 위치변화에 따른 북동태평양 적도해역의 부유성 유공충 군집의 계절변동)

  • A time-series sediment trap was operated at a water depth of 4950 m from July 2003 to May 2004 at KOMO station ($10^{\circ}30^{\prime}N$, $131^{\circ}20^{\prime}W$) in the northeastern equatorial Pacific, with the aim of understanding the temporal variation of planktonic foraminifera assemblages in response to the seasonal shift of Inter-Tropical Convergence Zone (ITCZ). A total of 22130 planktonic foraminifera specimens belonging to 30 species and 11 genera were identified, which shows a distinct seasonal variation with high values (125~288 specimens $m^{-2}day^{-1}$) in the winter to spring (December-May) and low values (16~23 specimens $m^{-2}day^{-1}$) in the fall (September-November). In addition, seasonal ecological differences of foraminifera assemblages are distinctly recognizable: omnivorous foraminifera occurred predominantly during the summer season, whereas herbivorous ones were dominant during the winter season. Such seasonal variations correspond to the seasonal shift of the ITCZ. Enhanced occurrence of herbivorous species during the winter-spring season seems a result of surface water mixing generated by the southward shift of the ITCZ. The increase in omnivorous species during the summer season may be due to the northward movement of the ITCZ caused by weakened wind speed, resulting in the intensification of water column stratification and nutrient-poor environment. A significant reduction of planktonic foraminifera specimens during the fall is attributed to heavy precipitation and reduction in light intensity.

  • Considering the fuel consumption of car, a light structure of aluminum alloys is desired for car body nowadays. However, fusion welding of aluminum alloys has some problems of reduction of joint efficiency, porosity formation and hot cracking. ill the present work, investigation to improve the joint performance of laser welded joint has been carried out by addition of Cu, Ni, and Zr to A6N01 alloy welds. Aluminum alloy plate of 2.0mm in thickness with filler metal bar was welded by twin beam Nd:YAG laser facility (total power:5kW). The filler metals were prepared by changing the chemical compositions for adding the elements into the weld metal. Thirteen filler metal bars were prepared and pre-placed into the base metal before welding. Ar gas shielding with a flow rate of 10 l/min was used. The defocusing distance is kept at 0 mm. At travel speeds of 3 to 9 m/min and at laser power of 5kW (front beam 2kW rear beam 3kW), full penetration welds were obtained, whereas at travel speeds of 12 to 18 m/min and same power, partial penetration was observed. The joint efficiency of laser-welded joint was improved by the addition of Cu, Ni, and Zr due to the solid solution hardening, grain refining and precipitation hardening. The type of hardening has been further considered by metallurgical examination.

Characteristics of Ag ions Photoadsorption Using Photocatalytic TiO2 Nanocrystalline Powder (광촉매용 TiO2 나노분말을 이용한 Ag이온의 흡착 특성)

  • In this study photoadsorption propreties of Ag ion in AgNO$_3$ solution by TiO$_2$nanopowder synthesized by homogeneous precipitation process at low temperature were investigated. It was found that the photocatalytic redliction in AgNO$_3$solution was occurred by TiO$_2$nanopowder even under th sun light irradiation, although the reduction of Ag ions was slow with th small adsorption of 9.32 ppm. Notably the Ag adsorption was promoted in th dark condition probably owing to the chestnut bur shape of TiO$_2$ nanopowder itself. In the application of UV the Ag ions were completely adsorbed within 120 min, showing more significant photocatalytic reaction. The measured adsorption reaction rate and adsorption equilibrium rate constants were 0.0004 g/min and 1494.20(120 $m^2$/g), respectively.

Paleo-Tsushima Water influx to the East Sea during the lowest sea level of the late Quaternary

  • The East Sea, a semi-enclosed marginal sea with shallow straits in the northwest Pacific, is marked by the nearly geographic isolation and the low sea surface salinity during the last glacial maximum (LGM). The East Sea might have the only connection to the open ocean through the Korea Strait with a sill depth of 130 m, allowing the paleo-Tsushima Water to enter the sea during the LGM. The low paleosalinity associated with abnormally light $\delta^{18}O$ values of planktonic foraminifera is interpreted to have resulted from river discharge and precipitation. Nevertheless, two LGM features in the East Sea are disputable. This study attempts to estimate volume transport of the paleo-Tsushima Water via the Korea Strait and further examines its effect on the low sea surface salinity (SSS) during the lowest sea level of the LGM. The East Sea was not completely isolated, but partially linked to the northern East China Sea through the Korea Strait during the LGM. The volume transport of the paleo-Tsushima Water during the LGM is calculated approximately$(0.5\~2.1)\times10^{12}m^3/yr$ on the basis of the selected seismic reflection profiles along with bathymetry and current data. The annual influx of the paleo-Tsushima Water is low, compared to the 100 m-thick surface water volume $(about\;79.75\times10^{12}m^3)$ in the East Sea. The paleo-Tsushima Water influx might have changed the surface water properties within a geologically short time, potentially decreasing sea surface salinity. However, the effect of volume transport on the low sea surface salinity essentially depends on freshwater amounts within the paleo-Tsushima Water and excessive evaporation during the glacial lowstands of sea level. Even though the paleo-Tsushima Water is assumed to have been entirely freshwater at that time period, it would annually reduce only about 1‰ of salinity in the surface water of the East Sea. Thus, the paleo-Tsushima Water influx itself might not be large enough to significantly reduce the paleosalinity of about 100 m-thick surface layer during the LGM. This further suggests contribution of additional river discharges from nearby fluvial systems (e.g. the Amur River) to freshen the surface water.

Fabrication and Characterization of PZT Suspensions for Stereolithography based on 3D Printing

  • PZT suspensions for photo-curable 3D printing were fabricated and their characteristics were evaluated. After mixing the PZT, photopolymer, photo-initiator, and dispersant for 10 min by using a high-shear mixer, the viscosity characteristics were investigated based on the powder content. To determine an appropriate dispersant content, the dispersant was mixed at 1, 3, and 5 wt% of the powder and a precipitation test was conducted for two hours. Consequently, it was confirmed that the dispersibility was excellent at 3 wt%. Through thermogravimetric analysis, it was confirmed that weight reduction occurred in the photopolymer between 120? and 500?, thereby providing a debinding heat treatment profile. The fabricated suspensions were cured using UV light, and the polymer was removed through debinding. Subsequently, the density and surface characteristics were analyzed by using the Archimedes method and field-emission scanning electron microscopy. Consequently, compared with the theoretical density, an excellent characteristic of 97% was shown at a powder content of 87 wt%. Through X-ray diffraction analysis, it was confirmed that the crystallizability improved as the solid content increased. At the mixing ratio of 87 wt% powder and 13 wt% photo-curable resin, the viscosity was 3,100 cps, confirming an appropriate viscosity characteristic as a stereolithography suspension for 3D printing.
