Heterologous Expression and Characterization of a Novel Exo-Polygalacturonase from Aspergillus fumigatus Af293 and Its Application in Juice Extraction

  • Exo-polygalacturonase (exo-PG) hydrolyzes pectin acids and liberates mono-galacturonate, which plays an important role in juice extraction, and has rarely been reported. Exo-PG (AfumExoPG28A) from Aspergillus fumigatus belongs to the glycoside hydrolase 28 family. In this study, its gene was cloned and the protein was expressed and secreted in Pichia pastoris with a maximal activity of 4.44 U/ml. The optimal temperature and pH of AfumExoPG28A were 55℃ and 4.0, respectively. The enzyme exhibited activity over almost the entire acidic pH range (>20.0% activity at pH 2.5-6.5) and remained stable at pH 2.5-10.0 for 24 h. The Km and Vmax values of AfumExoPG28A were calculated by the substrate of polygalacturonic acid as 25.4 mg/ml and 23.6 U/mg, respectively. Addition of AfumExoPG28A (0.8 U/mg) increased the light transmittance and juice yield of plantain pulp by 11.7% and 9%, respectively. Combining AfumExoPG28A (0.8 U/mg) with an endo-PG (0.8 U/mg) from our laboratory, the enzymes increased the light transmittance and juice yield of plantain pulp by 45.7% and 10%, respectively. Thus, the enzyme's potential value in juice production was revealed by the remarkable acidic properties and catalytic activity in fruit pulp.

Histologic evaluation of $Regenafil^{TM}$ on defects of extraction socket (발치와의 결손부위에 $Regenafil^{TM}$의 임상적 적용에 관한 조직학적 연구)

  • The purpose of this study is to evaluate histologic result of bone substituting material on defects followed tooth extraction. We compare the histologic findings control, DFDBA, Bio-Oss(R), and $Regenafil^{TM}$, Briefly, mandibular premolar teeth were extracted available for bone filling. All alveolar sites were checked after extraction and thoroughly debrided with a dental curet to remove the periodontal ligament. Extraction sites were prepared dehiscence on buccal side 7mm height from alveolar crest. The graft materials were filled into the extraction socket and dehiscenc defects. The animals were sacrificed 12 weeks after implantation. Both treated and control mandibular sites were histologically evaluated with light microscopy. Histologic observation at 12 weeks revealed that control and experimental sites were healed uneventfully and directly apposed to new bone without any adverse tissue reaction. DFDBA and Bio-Ossn(R)sites maintain width of alveolar crest but were not fully resorbed. $Regenafil^{TM}$ sites also maintain width and particles were resorbed more than other graft materials. From this results, it was suggested that $Regenafil^{TM}$ is promising boen substituting materials maintaining the width of alveolar crest and height follewed tooth extraction.

Effect of High Voltage Pulsed Electric Fields on Extraction of Purple Sweet Potato Pigment (자색 고구마 색소의 추출에 미치는 고전압 펄스 전기장의 영향)

  • High voltage pulsed electric fields (PEF) is a promising technology for the nonthermal extraction of effective components from biological materials. Plant cells were ruptured with PEF at ambient or refrigerated temperature for a short treatment time of second or microsecond. Treatments of coarsely ground purple sweet potato (PSP) with PEF(30 kV/cm, 500 Hz) resulted in maximum extraction yield of 65% as compared with 45-50% for control. An increase in electric field strength (from 10kV/cm to 35kV/cm) and frequency (from 100Hz to 500Hz) resulted in increased amount of extracted pigments, but treatment time is not affected on pigment extraction. Starch granules were not detected and large intracellular spaces were visible between the cells on light and scanning election microscopy of PEF treated PSP. This result suggests that PEF have potential to use on extraction of pigments from plant cells.

EFFECT OF ELECTRICAL STIMULATION ON BONE FORMATION IN THE EXTRACTION SOCKET OF RAT (전기자극이 치조골 치유과정에 미치는 영향)

  • On the basis of the evidence that electrical stimulation could promote healing and regeneration of bone, this study was performed to investigate the effects of electrical stimulation on rat extraction socket, and to evaluate the potential of clinical application of electrical stimulation. Forty rats were used and divided into control groups(l0)and the experimental groups(30) in this study. The maxillary 1st molar were extracted in both groups. In experimental group, electrical stimulation was given at the current intensity of lmA(Test-1), l0mA(Test-2), 25mA(Test-3) each day. At 1,3,5,7 days after the tooth extraction, rats in both groups were serially sacrificed. And the specimens were prepared with Hematoxylin-Eosin stain for the light microscopic evaluation. The results of this study were as follows; 1. At 1 day after the extraction, the periodontal ligament was found in the extraction socket wall. The formation of blood clot with dense infiltration of inflammatory cells in control group and there were less inflammatory cells in test group. 2. At 3 day after the extraction, the cells and collagen of the periodontal ligament were so actively proliferated and synthesized that invaded into the connective tissue of the extraction sockets in the control group. There were the formation of new bone in the basal & lateral portion of socket wall in test -2 and -3. 3. At 5 days after the extraction, there were no formation of new bone in control group. But the more electrical stimulation was applied, the more formation of new bone in test group. 4. At 7 days after the extraction, the extraction sockets were almost filled with trabecular bone in each group. Bone maturarity was remarkable in test-3. 5. The electrical stimulation at l0mA and 25mA was more effective in the bone formation at 5 and 7 days after the extraction. From the above results, electrical stimulation could promote the extraction socket wound healing, and be utilized in the clinical application of the residual ridge expansion.

  • Many of the natural dyes used for natural dyeing are difficult to maintain colorfastness due to their complex structure and specific properties. Therefore, there is a need for developing of color sustainable ability for use as an advanced coloring agent for fabrics, which would eco benign or not. In this study, the natural dye extracted from the waste of grape fruits was used to dye cotton fabric. Thus, the present study aims at extraction of color from grape seeds, skin, and stem through fermentation and then employing the same in dyeing and mordanting of cotton. Dyeing experiments were done under different conditions of fermentation and protein type mordants which were treated before and after dyeing. Experimental fabrics were used with cotton after scouring. Color value of dyed fabrics and color fastness of cotton dyed fabrics to washing and light were measured. The fastness of dyed experimental fabrics was increased by mordanting of protein fermentation and the color of dyed cotton was light red purple. The color of dyed fabric found with the optimum mordant treatment when treated with pre milk-mordant at $40^{\circ}C$ for 30min and 3% grape seed extract. On the whole, reddish tone very slightly increased with the milk pre-mordant. The color fastness of dyed cotton fabrics to light and washing was increased after fermentation.

Extraction Characteristics of Red Flower Cabbage Pigment (꽃양배추 색소의 추출특성)

  • Extraction characteristics of anthocyanin pigment from red flower cabbage(Brassica oleracea L. var. acephala) as a new source of natural food colorant were investigated. The pigment extracted from red flower cabbage showed the characteristic bathochromic shift of the maximum wavelength of light absorption(${\lambda}_{max}$) as pH of the solution changed from pH 1 to 12. As the concentration of citric acid in the extraction solvent increased, extraction rate and total optical density(TOD) of the extract increased. Maximum TOD was obtained by using the extracting solvent including $0.8{\sim}1.0%$ citric acid and stable pigment solution was obtained by using the extracting solvent including $10{\sim}20%$ ethanol in distilled water. As a result, 10% ethanolic solution with 0.8% citric acid was decided as the optimum extraction solvent for the anthocyanin pigment from red flower cabbage. Within the experimental ranges, the extraction rate increased and therefore extraction time decreased as the extraction temperature increased. The times to reach a certain value of TOD i.e., 2.1 were 24, 8, 4 and 2 hours at extraction temperature of 5, 20, 40 and $60^{\circ}C$, respectively.

Extraction and Stabilization of Anthocyanin Pigments from Morus alba Fruits (오디 anthocyanin 색소의 추출 및 저장 안정성)

  • This study was initiated to search for the most effective method for extraction of anthocyanins from Morus alba Fruits using organic acids and to evaluate the stability of the pigments at various storage environments. The anthocyanins were effectively extracted by addition of organic acids such as citric acid, malic acid and fumaric acid. The anthocyanins were not degraded at $25^{\circ}C$ and under a fluorescent light with 183 lux, but sharply degraded at $60^{\circ}C$.

  • Shoreline changes its shapes and attribution dynamically by natural, unnatural acts and is the most information for country. These shorelines can apply to framework data of MGIS (Marine Geographic Information System), and they are getting important to implement a phase of monitoring around coastal areas. This study proposed an algorithm automatically extracting shorelines to use a new developed LiDAR (Light Detection And Ranging) data which is applying in ocean and coastal areas. Then, in result, it was compared to shorelines which is derived from ground survey. In result, it shows stable shorelines in various coast areas such as nature, artificial coast. Additionally, and a possibility of shoreline extraction through LiDAR data.

  • The purpose of this study is to investigate optical behaviour of coloring matter of gromwell -extracted by water, acetic acid/methanol, and the other solvents. The results were as following: UV, VIS spectra of the solution of coloring matter of gromwell extracted by water at below $80^{\circ}C$ did not reveal in the range of the visible light wavelength and coloring matter of gromwell decomposed to blue particle at $95^{\circ}C$. The solution extracted by acetic acid/methanol was tinged with reddish purple and its spectra shifted to the longer wavelength according to increase of the solvent temperature. The color of the solution extracted by water became extinct as time went by, but that extracted by acetic acid/methanol kept up original color. In case of dried gromwell, extraction by acetic acid/methanol was easier than by water, and variation of spectra did not appear but showed hypochromic shift.

Characteristics of Greenish Pigments from Silkwarm Excrement by Ethanol Extraction (누에 배설물에서 Ethanol 추출한 녹색색소의 일반적 성질)

  • A study of characteristics of greenish pigments from silkwarm excrement by ethanol extraction. Through visible absorption scanning, it showed two absorption peaks at 415 and 657nm, and it was shown to be greenish color. In the presence of light, the stability of pigments rapidly degraded, but in the presence of Al-foil, green and blue cover were very stable. It was shown to be stabilized at the temperature of 7$0^{\circ}C$ until 20days and presence of metal ions, such as Na+, K+, Ca2+, Mg2+ and Zn2+. The pigments was shown to be stabilized in 5% acetic acid solution, but in the presence of lactic acid, citric acid and tartaric acid solution were unstable. On the result of TLC analysis, pigments were shown to be composed of eight color fractions, and main color fractions were F-1, F-2 and F-3. In F-1, F-2 fractions were revealed green color and F-3 fraction were revealed yellow color.
