Cloning of Mouse AQP-CD Gene

  • Water transport in highly-permeable membranes is facilitated by some specialized pathways, which are called aquaporins (AQP). AQP1 (AQP-CHIP) is the first recognized aquaporin identified from red cells and renal proximal tubules. Up until now 4 other aquaporin homologs have been reported. Each aquaporin has its unique tissue distribution and regulatory mechanims. To elucidate molecular mechanisms for their transcription regulation and tissue-specific expression isolation of aquaporin genes is required. To clone promoters of the AQP family mouse genomic library was screened by the 1st exon-specific probe of AQP4, and 5 different plaques were positively hybridized. Phage DNAs were purified and characterized by restriction mapping and sequencing. One of them is the mouse AQP-CD gene. The gene was consisted of 4 exons and the exon-intron boundaries of mouse AQP-CD gene were identified at identical positions in other related genes. The 5'-flanking region of AQP-CD gene contains one classic TATA box, a GATA consensus sequence, an E-box and a cyclic AMP-responsive element. The cloning of the mouse AQP-CD gene, of which product is expressed in the collecting duct and is responsible for antidiuresis by vasopressin, will contribute to understand the molecular mechanisms of tissue-specific expression and regulation of AQP-CD gene under various conditions.

Members of the ran family of stress-inducible small GTP-binding proteins are differentially regulated in sweetpotato plants

  • Ran is a small GTP-binding protein that binds and subsequently hydrolyzes GTP. The functions of Ran in nuclear transport and mitotic progression are well conserved in plants and animals. In animal cells, stress treatments cause Ran relocalization and slowing of nuclear transport, but the role of Ran proteins in plant cells exposed to stress is still unclear. We have therefore compared Ran genes from three EST libraries construed from different cell types of sweetpotato and the distribution pattern of Ran ESTs differed according to cell type. We further characterized two IbRan genes. IbRan1 is a specific EST to the suspension cells and leaf libraries, and IbRan2 is specific EST to the root library. IbRan1 showed 94.6 % identity with IbRan2 at the amino acid level, but the C-terminal region of IbRan1 differed from that of IbRan2. These two genes showed tissue-specific differential regulation in wounded tissues. Chilling stress induced a similar expression pattern in both IbRan genes in the leaves and petioles, but they were differently regulated in the roots. Hydrogen peroxide treatment highly stimulated IbRan2 mRNA expression in the leaves and petioles, but had no significant effect on IbRan1 gene expression. These results showed that the transcription of these two IbRan genes responds differentially to abiotic stresses and that they are subjected to tissue-specific regulation. Plant Ran-type small G-proteins are a multigenic family, and the characterization of each Ran genes under various environmental stresses will contribute toward our understanding of the distinctive function of each plant Ran isoform.

Cloning and Regulation of Schizosaccharomyces pombe Gene Encoding Ribosomal Protein L11

  • The cDNA encoding ribosomal protein was identified from a cDNA library of Schizosaccharomyces pombe. The nucleotide sequence of the 548 by cDNA clone reveals an open reading frame, which encodes a putative protein of 166 amino acids with a molecular mass of 18.3 kDa. The amino acid sequence of the S. pombe L11 protein is highly homologous with those of rat and fruit, while it is clearly less similar to those of prokaryotic counterparts. The 1,044 by upstream sequence, and the region encoding N-terminal 7 amino acids of the genomic DNA were fused into the promoterless $\beta$-galactosidase gene of the shuttle vector YEp357 in order to generate the fusion plasmid pHY L11. Synthesis of $\beta$-galactosidase from the fusion plasmid varied according to the growth curve. It decreased significantly in the growth-arrested yeast cells that were treated with aluminum chloride and mercuric chloride. However, it was enhanced by treatments with cadmium chloride ($2.5\;{\mu}M$), zinc chloride ($2.5\;{\mu}M$), and hydrogen peroxide (0.5 mM). This indicates that the expression of the L,11 gene could be induced by oxidative stress.

Regulation of signal transducer and activator of transcription 3 activation by dual-specificity phosphatase 3

  • Since cancer is the leading cause of death worldwide, there is an urgent need to understand the mechanisms underlying cancer progression and the development of cancer inhibitors. Signal transducer and activator of transcription 3 (STAT3) is a major transcription factor that regulates the proliferation and survival of various cancer cells. Here, dual-specificity phosphatase 3 (DUSP3) was identified as a regulator of STAT3 based on an interaction screening performed using the protein tyrosine phosphatase library. DUSP3 interacted with the C-terminal domain of STAT3 and dephosphorylated p-Y705 of STAT3. In vitro dephosphorylation assay revealed that DUSP3 directly dephosphorylated p-STAT3. The suppressive effects of DUSP3 on STAT3 were evaluated by a decreased STAT3-specific promoter activity, which in turn reduced the expression of the downstream target genes of STAT3. In summary, DUSP3 downregulated the transcriptional activity of STAT3 via dephosphorylation at Y705 and also suppressed the migratory activity of cancer cells. This study demonstrated that DUSP3 inhibits interleukin 6 (IL-6)/STAT3 signaling and is expected to regulate cancer development. Novel functions of DUSP3 discovered in IL-6/STAT3 signaling regulation would help expand the understanding of cancer development mechanisms.

국내외 드론산업 동향 분석을 통한 공공분야에서의 드론 활용방안에 대한 연구 (A Study on Utilization of Drone for Public Sector by Analysis of Drone Industry)

  • The drone is an unmanned aerial vehicle which has no human pilot. Drones can be classified into military drones, commercial drones, and personal drones by usage. Also, drones can be classified from large-sized to nano-sized drone by size and autonomous, remote controlled drone by control type. Especially, military drones can be classified into low-altitude drones, medium-altitude, and high-altitude drones by altitude. Recently, the drone industry is one of the fast growing industries in the world. As drone technologies have become more advanced and cost-effective, Korean government has set its goal to become a top-level country in drone business. However, the government's strict regulation for drone operations is one of the biggest hurdles for the development of the related technologies in Korea and other countries. For example, critical problems for drone delivery can be classified into technical issues and institutional issues. Technical issues include durability, conditional awareness, grasp and release mechanisms, collision avoidance systems, drone operating system. Institutional issues include pilot and operator licensing, privacy rules, noise guidelines, security rules, education for drone police. This study analyzes the trends of the drone industry from the viewpoint of technology and regulation. Also, we define the business areas of drone utilization. Especially, the drone business types or models for public sector are proposed. Drone services or functions promoting public interests need to be aligned with the business reference model of Korean government. To define ten types of drone uses for public sector, we combine the business types of government with the future uses of drones that are proposed by futurists and business analysts. Future uses of drones can be divided into three sectors or services. First, drone services for public or military sectors include early warning systems, emergency services, news reporting, police drones, library drones, healthcare drones, travel drones. Second, drone services for commercial or industrial services include parcel delivery drones, gaming drones, sporting drones, farming and agriculture drones, ranching drones, robotic arm drones. Third, drone services for household sector include smart home drones.

『성호사설(星湖僿說)』의 형성 배경과 인용서목 분석 (An Analysis on the Foundation Background and Cited References of Miscellaneous Explanations of Seongho (星湖僿說))

  • 본 연구는 성호 이익의 대표적인 저술인 "성호사설"에 인용된 문헌 443종을 추출하여 인용문헌의 규모 및 문헌의 성격, 중국본의 비중, 최신본의 비중, 주제별 비중 등을 분석하였다. "성호사설" 인용문헌 443종을 살펴본 결과, 고려 조선본이 전체 23.3%, 중국본이 76.7%를 차지하였고, 중국본의 편찬시기를 살펴본 결과, 송대 이전 편찬본이 70% 이상을 차지한 반면, 명 청대 문헌은 22.7%인 것으로 나타났다. 인용문헌의 주제별 비중을 살펴보면 가장 많은 비중을 차지하는 주제영역은 역사서류(歷史書類), 시문집류(詩文集類), 유교경전류(儒敎經典類), 소설필기류(小說筆記類)의 순으로 나타났으며, 성호 이익은 이외에도 잡가류, 유가류, 지리류, 제도규정류, 서화음악류, 자전류, 전기류, 백과사전류 등 다양한 주제에 관심을 가졌던 것으로 조사되었다.

Nebulin C-terminus Interacts with NCBP51, a New Isoform of RING Finger Protein 125 (RNF125)

  • Nebulin, a giant modular protein from muscle, is thought to act as molecular ruler in sarcomere assembly. In skeletal muscle, the C-terminal ${\sim}50 kDa$ region of nebulin extends into the Z-line lattice. The most recent studies implicated highlighting its extensive isoform diversity and exciting reports revealed its expression in cardiac and non-muscle tissues containing brain. Also these novel findings are indicating that nebulin is actually a multifunctional filament system, perhaps playing roles in signal transduction, contractile regulation, and myofibril force generation, as well as other not yet defined functions. However the binding protein of nebulin and function in brain is still unknown. A novel binding partner of nebulin C-terminal region was identified by screening a human brain cDNA library using yeast two-hybrid system. Nebulin C-terminus binding protein 51 (NCBP51) was contained a RING-finger domain and identified a new isoform of RING finger protein 125 (RNF125). The interaction was confirmed using the GST pull-down assay. NCBP51 belongs to a family of the RING finger proteins and its function remains to be identified in brain. The role of nebulin and NCBP51 will be studied by loss-of-function using siRNA technique in brain.

Identification and Characterization of Genes Differentially Expressed in the Resistance Reaction in Wheat Infected with Tilletia tritici, the Common Bunt Pathogen

  • The differentially virulent race T1 of common bunt (Tilletia tritici) was used to inoculate the wheat lines Neepawa (compatible) and its sib BW553 (incompatible) that are nearly isogenic for the Bt-10 resistance gene. Inoculated crown tissues were used to construct a suppression subtractive hybridization (SSH) cDNA library. Of the 1920 clones arrayed from the SSH cDNA library, approximately 10% were differentially regulated. A total of 168 differentially up-regulated and 25 down-regulated genes were identified and sequenced; 71% sequences had significant homology to genes of known function, of which 59% appeared to have roles in cellular metabolism and development, 24% in abiotic/biotic stress responses, 3% involved in transcription and signal transduction responses. Two putative resistance genes and a transcription factor were identified among the up regulated sequences. The expression of several candidate genes including a lipase, two non-specific lipid transfer proteins (ns-LTPs), and several wheat pathogenesis-related (PR)-proteins, was evaluated following 4 to 32 days post-inoculation in compatible and incompatible interactions. Results confirmed the higher overall expression of these genes in resistant BW553 compared to susceptible Neepawa, and the differential up-regulation of wheat lipase, chitinase and PR-1 proteins in the expression of the incompatible interaction.

붉은 털 원숭이의 뇌조직에서 CCDC94 유전자 대체 전사체의 분자적 분석 (Molecular Analysis of Alternative Transcripts of CCDC94 Gene in the Brain Tissues of Rhesus Monkey)

  • 붉은 털 원숭이의 유전체는 인간의 것과 93% 정도로 동일하여, 진화적 연구 및 생물의학적 연구에 널리 활용되고 있다. 숙주 개체 내로 가동성 유전인자(TEs)의 삽입은 유전자 전사체의 다양성과 발현양상을 다르게 만든다. 본 연구에서는 붉은 털 원숭이의 뇌 조직으로부터 만든 cDNA 라이브러리에서 112개 전사체를 동정하여 분석하였다. 하나의 전사체 R54는 인간과 원숭이의 다양한 조직에서 유전자 발현양상을 비교분석 해 본 결과 서로 다른 패턴을 보여 주었다. 이러한 현상은 가동성 유전인자인 L2A의 삽입으로 인한 스플라이싱 도너 사이트가 변화된 것으로 생각된다. 따라서, 영장류의 진화과정에 있어 유전체 내로 TEs의 삽입은 전사체의 다양성과 유전자 발현 조절에 변화를 주는 것으로 시사된다.

  • 이 논문은 한국기록관리학회지를 오픈액세스 학술지로 전환하는 과정을 기록한 것이다. 이 과정을 이해하기 위하여 상용DB업체와 학회, 공공DB운영기관 사이에서 벌어진 저작권을 중심으로 한 갈등과 업체와 도서관 사이의 학술DB 가격에 대한 갈등문제를 설명하였다. 또한 최근 국내의 문헌정보학 분야와 인문사회분야 학회들의 오픈액세스 선언의 의미를 설명하였다. 한국기록관리학회지를 오픈액세스로 전환하는 과정에서 부딪힌 출판비 문제, 논문 유통의 문제, 학술지 출판과 관련한 제도의 변경 등 학술지 출판사로서 실무적인 사안들을 처리한 방법과 과정을 기록하고 문제점들도 드러내고자 했다. 또한 이 논문은 기록관리학회지의 오픈액세스 전환을 학술논문의 지식커먼즈를 실현하려는 과정으로 설명하였다.
