Paenibacillus sp. DG-22로부터 열에 안정한 β-xylosidase를 암호화하는 유전자의 클로닝, 염기서열결정 및 발현 (Cloning, Sequencing and Expression of the Gene Encoding a Thermostable β-Xylosidase from Paenibacillus sp. DG-22)

  • 세균인 Paenibacillus sp. DG-22의 유전체 DNA library가 제조되었으며, ${\beta}-xylosidase-$양성 클론이 형광기질인 $4-methylumbelliferyl-{\beta}-D-xylopyranoside$ $({\beta}MUX)$를 사용하여 확인되었다. 이 클론으로부터 재조합 플라스미드가 분리되었고 삽입된 4.3-kb 크기 DNA의 염기서열이 결정되었다. ${beta}-xylosidase$ 유전자는 분자량이 78.710 dal-ton이고 pI가 5.0인 701개의 아미노산을 암호화하는 2,106 염기쌍의 열린해독틀(ORF)로 구성되어있었다. xylA 유전자산물의 추론된 아미노산 서열은 과(family) 52에 속하는 클리코실 가수분해효소로 분류된 ${beta}-xylosidase$들과 상당한 유사성을 가지고 있었다. 이 xylA 유전자에 6개의 히스티딘-꼬리표를 붙이기 위해 pQE60 발현벡터에 다시 클로닝하였다. 재조합 ${beta}-xylosidase$ $(XylA-H_6)$가 열처리와 고정화금속친화성 크로마토그래피(IMAC)에 의해 순수하게 정제되었다. $XylA-H_6$ 효소의 최적 pH와 온도는 각각 pH 5.5-6.0과 $60^{\circ}C$이었다.

A Plausible Method for the Diagnosis of Genetic Disorders Using Full Length cDNA

  • A cDNA of coagulation Factor IX gene has been screened from the $\lambda$gt11 human fetal liver cDNA library, and used to construct a 2.8-kb full length cDNA after recombining with the N-terminal fragment from pTZ-FIX. Human genomic DNA was isolated, digested with the restriction endonucleases, TaqI, EcoRI, and HindIII, and Southern hybridization was performed using the full length factor IX cDNA as a probe. The hybridized bands generated by the restriction endonucleases were the followings: TaqI, 0.3, 1.0, 1.6, 1.8, 2.7, 3.7, and 5.3 kb bands; EcoRI, 1.8, 4.8, 4.9, 5.5, 6.8, and 12.6 kb bands; HindIII, 4.1, 4.4, 5.2, 5.8, 7.6, and 12.5 kb bands. When the Southern bands were physically mapped along the genome, about 50-kb continuous region harboring almost all of the genomic region of Factor Ⅸ gene was covered. These results suggest a possibility of using an exonal cDNA probe to diagnose abnormalities including large deletions, insertions, and rearrangements along the genome, if there is any.

  • A Southern-hybridization analysis and size-selected DNA library screening led to the isolation of a 6.3-kbp S. setonii DNA fragment, from which the Cl20-encoding genetic locus was found to be located within a 1.4-kbp DNA fragment. A complete nucleotide sequencing analysis of the 1.4-kbp DNA fragment revealed a 0.84-kbp ORF, which showed a strong overall amino acid similarity to the known high-G+C gram-positive bacterial mesophilic C120s. The heterologous expression of the cloned 1.4-kbp DNA fragment in E. coli demonstrated that this Cl20 possessed a thermophilic activity within a broad temperature range and showed a higher activity against 3-methy1catechol than catechol or 4-methy-catechol, but no activity against protocatecuate.

조합화학 기법을 이용한 Y1-x(P1-y-zNbyVz)O4:Eux 형광체의 합성 및 빛 발광 특성 (Synthesis and Photoluminescence Properties of Y1-x(P1-y-zNbyVz)O4:Eux Phosphors by Modified Combinatorial Chemistry Method)

  • 조합화학 기법을 이용하여 청색 및 적색 발광을 하는 $Y_{1-x}(P_{1-y-z}Nb_yV_z)O_4:Eu_x$ 형광체를 합성하였다. 합성한 $Y_{1-x}(P_{1-y-z}Nb_yV_z)O_4:Eu_x$ 형광체에 대하여 254 nm 및 147 nm 여기 하에서의 발광 세기에 관한 라이브러리를 완성하였고, 또한 형광체의 결정성과 입자 현상은 XRD와 SEM으로 특성을 관찰하였다. 조합검색 결과로부터 $Y_{1-x}(P_{1-y-z}Nb_yV_z)O_4:Eu_x$ 형광체의 발광특성은 P 자리에 치환되는 금속이온의 영향을 많이 받았다. 결과적으로 $Y_{0.88}PO_4:Eu_{0.12}$ 형광체와 비교하여 발광 효율이 우수한 새로운 $Y_{0.88}(P_{0.92}Nb_{0.05}V_{0.03})O_4:Eu_{0.12}$ 형광체를 찾아내었다.

인지행동중재가 암 환자의 심리적 디스트레스, 자기간호 및 삶의 질에 미치는 효과: 메타분석 (Cognitive Behavioral Therapy for Psychological Distress, Self Care and Quality of Life in Patients with Cancer: A Meta-analysis)

  • Purpose: The purpose of this study was to assess the effects of cognitive behavioral therapy (CBT) on depression, anxiety, self care behavior and quality of life in cancer patients. Methods: Two thousand and eighty three abstracts were identified through six electronic databases (1980 to June 2012) in Korea. Seventeen studies involving 679 participants met the inclusion criteria for meta analysis. Two authors independently assessed trial quality by Cochrane's Risk of Bias and Methodological Items for Non Randomized Studies and extracted data. The data were analyzed by the RevMan 5.2 program of Cochrane library. Results: Overall, study quality was moderate to high. CBT was conducted for a mean of 4.2 weeks, 7 sessions and an average of 36.1-minutes per session. CBT was effective for depression (d=-0.85; 95% CI=-1.09, -0.61), anxiety (d=-0.52; 95% CI=-0.75, -0.29), self care behavior (d=-1.34; 95% CI=-1.93, -0.74), and quality of life (d=-0.42; 95% CI=-0.80, -0.04). Publication bias was not detected as evaluated by funnel plot and Egger's test. Conclusion: CBT has small to large effects on depression, anxiety, self care and quality of life. These finding suggests that various CBT interventions can assist cancer patients in reducing emotional distress and improving self care and quality of life.

새로운 알츠하이머 치료제의 안전성 유효성에 관한 체계적 문헌고찰 (Systemic Literature Review Study on the Efficacy and Safety of Novel Alzheimer's Disease Treatments)

  • Background: Innovative Alzheimer's disease drugs received approval in the United States in 2021 and 2023. This study aims to assess the safety and efficacy of these novel treatments, elucidate their mechanisms of action, and compare their impact on cognitive function improvement with approved drugs. Methods: We conducted a comprehensive search of pivotal clinical studies related to Alzheimer's disease treatments in PubMed/Medline, Embase, and the Cochrane Library databases from January 1st, 2020 to December 31st, 2022. Meta-analysis was performed using RevMan 5.4 software. Results: A total of 14 studies were included in this systematic review. When compared to the placebo, the new drugs did not exhibit a statistically significant effect on MMSE (Mini-Mental State Examination) (mean difference= -0.04, 95% confidence intervals [CIs]: -0.31, 0.23, N=3662, I2=0%). However, they demonstrated a significant impact on ADAS-cog (Alzheimer's Disease Assessment Scale-Cognitive Subscale) (standardized mean difference= -0.15, 95% CIs: -0.2, -0.1, N=6710, I2=17%). When compared to the approved drugs, the new drugs showed a statistically significantly lower effect on MMSE (test for subgroup difference Chi2=23.13, N = 5870, p<0.00001) but showed only a trend of decreased efficacy on ADAS-cog (Chi2=1.16, N = 8670, p=0.28). Conclusion: New drugs yielded diverse clinical endpoint results compared to the placebo, and in comparison to existing approved drugs, they exhibited lower efficacy in improving cognitive function. The safety profile of these new drugs, as reported in clinical trials, was generally well-tolerated.

Isolation of Deletion Mutants by Reverse Genetics in Caenorhabditis elegans

  • Obtaining mutant animals is important for studying the function of a particular gene. A chemical mutagenesis was first carried out to generate mutations in C. elegans. In this study, we used ultraviolet-activated 4,5',8-trimethylpsoralen to induce small deletion mutations. A library of mutagenized worms was prepared for recovery of candidate animals and stored at $15^{\circ}C$ during screening instead of being made into a frozen stock library. In order to isolate deletion mutations in target genes, a polymerase chain reaction (PCR)-based screening method was used. As a result, two independent mutants with deletions of approximately 1.0 kb and 1.3 kb were isolated. This modified and improved reverse genetic approach was proven to be effective and practical for isolating mutant animals to study gene function at the organismal level.

한국의 현행색인지에 관한 분석적 연구 (An analytical study on the major Korean periodical indexes)

  • This study attempts to analyze the major periodical indexes and catalogs of 231 titles published in Korea in the form of monographs from 1973 to 1993, in order to identify the trends of their publications and the problems in its compilation and to suggest an idea for the improvements. The results of this can be summarized as follow : 1. The publication of indexes are concentrated on some subject fields including those of social science. And there should be general indexes in the fields of the humanities and pure sciences for the researches in these fields to consult. 2. The titles of the indexes are represented by various forms. 3. The materials included are extended to all types of them including monographs, serials, these and dissertations, and newspapers, so that enhances the usefulness of the subject indexes. 4. Most of indexes are published in quarterly or yearly and seldom publish the cumulative editions. 5. Most of indexes(140 titles, 90%) are classified indexes. 6. The descriptions include basic elements of author, title, translator, contributed title, issue number, date, pages, etc, in the case of most of indexes. 7. Cross reference are seldom used, because most of indexes are classified ones. 8. Auxiliary indexes or a n.0, ppendixes are not included in 34 titles of 21%.

Cloning of the Alkaline Phosphatase Gene from Kluyveromyces fragilis

  • In order to clone the gene coding for alkaline phosphatase in the yeast Kluyveromyces fragilis, a genomic library was constructed using the yeast-E. coli shuttle vector pHN114 as a cloning vector. From the genomic library, a clone carrying the gene was isolated and the plasmid was designated as pSKH101. A restriction enzyme map was made using this plasmid. Subcloning experiments and complementation studies showed that alkaline phosphatase was active only in the original 3.1 kb insert. Southern hybridization analysis confirmed that the cloned DNA fragment was derived from K. fragilis genomic DNA. Using a minicell experiment, the product of the cloned gene was identified as a protein with a molecular weight of 63 KDa. A 0.6 kb HindIII fragment, which showed promoter activity, was isolated using the E. coli promoter-probe vector pKO-1.

온라인 정보검색 시스템의 명령어비교

  • This study is an attempt to furnish some helpful data for the maximum use of online information retrieval systems based on the comparative analysis of commands and operating terms employed by DIALOG, ORBIT and BRS which are three largest information retrieval systems in the world. To begin with, the historical development, service contents and retrieval functions of three systems were overviewed in the second chapter, on the basis of which the concrete functions and contents of the commands and the operating terms of the three systems were compared and analyzed one another. Commands were explained divided into basic and special ones. The basic commands subdivided into 44 items were put into the form of a diagram(Table 1). The special commands were first subdivided into search commnad, output-related commands and su n.0, pplementary commands, and then 14 items of which were comparatively analyzed and were represented as a chart(Table 2). In addition, any stand-alone commands employed by each system were also analyzed as a chart(Table 3). The Operating terms other than commands were subdivided into 16 items and were represented as a chart(Table 4). Password, inverted file, search step number in search session, etc were explained to the very detailed extent. A self-evident, single word is that the through understanding of commands is essential to the maximm use of all the functions of each of three systems.
