Pharmacognostical Studies on the Folk Medicine 'DeungNaMu' (민간약 등나무의 생약학적 연구)

  • Korean folk medicine 'DeungNaMu' has been used mainly to cure apoplexy, pain of the stapes and tonic for weak man. With regard to the botanical origin of DeungNaMu, it has been considered to be Wisteria species of Leguminosae, but there was no pharmacognostical confirmation on it. To clarify the botanical origin of 'DeungNaMu', the anatomical characteristics of the branches of Wisteria species growing wild in Korea, Wisteria japonica and W. japonica forma. alba were studied. As a result, it was clarified that DeungNaMu was the branches of Wisteria japonica.

  • During our research program to find DNA strand-scission agents from higher plants, the MeOH extracts of the stems of Mucuna birdwoodiana Tutcher. (Leguminosae) exhibited the most potent activity with an $IC_{50}$ value of $4.9\;{\mu}g/ml$. Thus, detailed laboratory investigation was performed, and led to the isolation of known compounds, $({\pm})$-catechin (1) and (-)-epicatechin(2) as active principles. Compounds 1 and 2 showed significant activity of DNA strand-scission with $IC_{50}$ values of 10.8 and $7.5\;{\mu}g/ml$, respectively (positive control, bleomycin: $IC_{50}\;3.3\;{\mu}g/ml$.

  • Two pterocarpans [(6aR,11aR)-maackiain, (6aR,11aR)-medicarpin], one flavanone [(2S)-7-hydroxy-G-methoxy-flavanone], one isoflavan (sativan) and two isoflavones (pseudobaptigenin, genistein) were isolated from the Spatholobus suberectus (Leguminosae). Their chemical structures were determined by comparison of their spectroscopic parameters of CD, ElMS, 1D-NMR and 2D-NMR with those reported in the literatures. All of these compounds are reported for the first time from this plant through the present study.

  • The populations of the legume lotus corniculatus var. japonicus regel were polymorphic for the character of leaf cyanogenesis. The distribution of the related phenotypes was investigated at ten different sites scattered in south Korea. The frequencies of leaf-cyanogenic plants were respectively and in percentage (the number of the samples) ; ch'onmasan(2 sits) 98.00%(200), 99.44%(180); ch'onggyesan 92.30%(104) ; myongjisan 97.00%(100); songnisan 98.00%(100); kyeryongsan 92.50%(120) ; chilgapsan 91.82%(110); hwasan 96.00(100); chirisan(2 sites) 46.67%(105),68.59%(156): χ2=309.25, df=9, p 0.001. the factors that may be responsible for the distribution of the phenoytypes are discussed.

Extractives from the Bark of Sophora japonica L

  • This study was carried out to investigate the constituents of Sophora japonica(Leguminosae) bark. To isolate compounds, bark was extracted with ethanol and then partitioned with hexane, dichloromethane, ethylacetate and butanol successively. After partitioned, DCM fraction was subjected to column chromatography with various solvent system in silica gel and/or Sephadex LH-20. Structures were elucidated by spectroscopic methods including MS, 1H, 13C and 2D-NMR experiments. Four compounds were isolated from the bark and identified as 3',7-dihydroxy-4'-methoxyisoflavone (cyclosin), puerol A, maackiain-3-O-𝛽-D-glucopyranoside (trifolirhizin), and 4', 5, 7-trihydroxyisoflavone (genistein). Among these compounds, cyclosin and trifolirhzin were first isolated from S. japonica.

  • The n-butanol soluble fraction of the extract obtained from the whole plants of Ononis serrata afforded the pterocarpan derivative medicarpin-3-O-glucoside and the isoflavone glucoside rothindin. Structures were elucidated by chemical methods, detailed spectral analyses as well as comparison with the literature data.

  • Legumes are important agricultural and commercial crops characterized by root nodules formed as a result of the symbiotic relationship with nitrogen-fixing rhizobia. Due to the economic significance the chemistry of some of these species has been well recognized.
