콩과식물화분의 Leucine Aminopeptidase 검출과 그 Isozyme에 대하여 (Identification of Leucine Aminopeptidase in Legume-Pollen Extracts and the Isozymes)

  • Identification and observations of leucine aminopeptidase (LAP) and multiple molecular forms of the enzyme, isozymes, were made with a technique of starch-gel electrophoresis for various legume pollen. Plants tested other than Leguminosae demonstrated either no indication of the presence or at least tract of enzymes and the isozymes, although all legume pollen tested showed strong LAP patterns. The electrophoretic patterns of LAP failed to be shown if the extracts were heated or otherwise denatured. Extent of zymogrammatic appearance of LAP and the isozymes were characteristic of a species.

초지개량에 미치는 가리시비의 효과에 관한 연구 (Sudies on the of $K_2O$Fertilization for the Pasturu Improvement)

  • 초지개량(草地改良)에 있어서 가리(加里)의 시용수준(施用水準) 및 질소(窒素)와 인산(燐酸)의 시비량(施肥量) 관계(關係)가 목초(牧草)의 생산성(生産性)에 미치는 영향(影響)을 구명(究明)하기 위해서 화본과(禾本科) 단파구(單播區), 두과(荳科) 단파구(單播區) 및 혼파구(混播區)로 구분(區分)하여 가리(加里)의 시비수준(施肥水準)을 달리하면서 1977년(年)부터 1980년(年)까지 3년간(年間)에 걸쳐 실험(實驗)한 결과(結果)를 요약(要約)하면 다음과 같다. 1. 가리(加里)의 시비수준(施肥水準)이 목초(牧草)의 정착(定着)과는 영향(影響)이 없으나 화본과(禾本科) 단파구(單播區)에서는 월동성(越冬性)에 효과가 있는 경향(傾向)을 보였다. 2. 목초(牧草)의 수량(收量)은 화본과(禾本科) 단파구(單播區)에서 가리(加里)의 시용(施用) 효과가 높게 나타나는 경향(傾向)이지만 두과(荳科) 단파구(單播區)와 혼파구(混播區)에서는 큰 차이(差異)가 없었다. 3. 고형물(固形物) 수량(收量)은 두과구(荳科區)의 4처리구(處理區)가 10a 1,152kg으로 가장 많고 혼파구(混播區)에서는 3처리구(處理區) 1,093kg, 화본과(禾本科) 단파구(單播區)는 5처리구(處理區)의 834kg이 가장 많았다. 4. 식생상태(植生狀態)의 변화(變化)는 화본과(禾本科) 단파구(單播區)에서 orchard grass와 tall fescue가 해를 경과할수록 증가(增加)하였으며 두과단파구(荳科單播區)와 혼파구(混播區)에서 alfalfa가 90% 이상(以上)의 우점초(優占草)가 되었는데 특(特)히 orchard grass는 가리(加里)의 증비(增肥) 효과가 큰 것 같다. 5. 초지개량(草地改良)에 있어서 본(本) 실험(實驗)의 결과(結果)로 나타난 적정시비수준(適正施肥水準)은 10a 당(當) 질소(窒素), 인산(燐酸), 가리(加里)를 화본과(禾本科) 단파구(單播區)에서 20-20-30kg을 시용(施用)할 때 최고(最高)의 수량(收量)을 나타냈으며 두과(荳科) 단파구(單播區)에서는 8-20-20kg을 시용(施用)하였을 때 최고수량(最高收量)을 얻을 수 있었다. 6. 시비수준(施肥水準)에 따르는 비료가격(肥料價格)과 목초(牧草)의 고형물(固形物) 수량(收量)에 의(衣)해서 1kg 생산(生産)에 소요(所要)된 비료가격(肥料價格)을 비교(比較)하면 혼파구(混播區)의 1처리구(處理구) 수익(收益)이 높은데 그 이유(理由)는 인산(燐酸)의 비료가격(肥料價格)에 의(依)한 것이다. 7. 본(本) 실험(實驗)은 종합적(綜合的)으로 고찰(考察)할 때 목초(牧草)의 고형물(固形物) 수량(收量)과 시비가격(施肥價格)으로 보아 화본과(禾本科)는 10a 당(當) 20-20-20kg, 두과(荳科)와 혼파구(混播區)는 8-20-20kg의 시비수준(施肥水準)이 경제적으로 가장 유리(有利)한 것 같다.

두류의 첨가가 당뇨병 쥐의 장기능과 당뇨증세에 미치는 영향 (Effects of Legume Supplementation on the Gastrointestinal Function and Diabetic Symptoms in Streptozotocin-Induced Diabetic Rats)

  • The present study was conducted to evaluate the usefulness of four kinds of legumes(black soybean: BS, yellow soybean: YS, green peas: GP, soybean curd residue: SCR) as a high-fiber supplement in the therapeutic diet for diabetic patients. Seven groups of normal and streptozotocin-induced diabetic rats were fed isocaloric experimental diets containing 8% dietary fiber from one of legumes for 6 weeks. The effects of legumes on the gastrointestinal function and diabetic symptoms in diabetic rats were also examined and compared with the effects of cellulose or pectin diet. Legume supplementations had no significant effects on alleviating typical diabetic symptoms such as polydipsia, polyphasia, polyuria, and urinary glucose excretion when compared with cellulose or pectin supplementations. The supplementations of legumes all resulted in remarkable changes in gastrointestinal functions: shortening of GI transit time and increases in fecal volume, fecal water and crude fat contents. Legume supplementations appeared to supress the damages of the small intestine villi and the enteritis symptoms in diabetic rats. GP led to regular villus patterns closer to normal villus. BS, YS and GP led to the enlargement of cecum in diabetic rats. In conclusion, the present studies have demonstrated that legumes may possess a significant physiological actions in the control of various diabetic conditions.

두과 작물의 질소고정과 유기조사료생산을 위한 작부체계 (Nitrogen Fixation of legumes and Cropping System for Organic Forage Production)

  • In order to supply 85% of total organic feed in ruminants and 80% in non-ruminants for organic animal production, nitrogen fixation ability of legume should be used in domestic roughages production. 50% of Europe organic farmer use intercropping legume in as green manure. This article is dealing with amount of biologically fixed nitrogen used by legumes and methods for estimating the transfer of biologically fixed N in rotation and separating the N benefit into fixed N and non-fixed N components are reviewed. Available data indicate that transfer amount of N to non legumes was from 50∼9.6(kg/ha) in legume-cereal rotations and proportion of legume N varied with seasons, 90% in summer, 50% in autumn. The important point in cropping system for legumes have to be included for organic forage production 6 year rotation is based on pasture system of 3 year pasture + 2 year annual(com, sudangrass), again pasture. Rye, barly and Italian ryegrass+legumes(vetch, crimson and pea) can be one of option in spring, com, soybean, sudangrass and Japanese bamyard millet would be seeded after spring harvest in the field. Farmer can make good use of rice paddy field as forge production potential area after harvesting rice. Italian, burly and rye+vetch and crimsonclover may be grown in autumn or spring time at the rice field.

Probing Equivocal Effects of Heat Processing of Legume Seeds on Performance of Ruminants - A Review -

  • Published studies show that effects of heat processing of legume seeds on animal performance are equivocal. In this article, we used a nutrition model - the DVE/OEB system to re-analyze nutrient supply (such as truly absorbed intestinal protein DVE value and protein degradation balance OEB value) to ruminants from published studies to probe reasons for such equivocal effects and provided some explanation why equivocal effects occurred. The analysis results showed that an unsuitable supply of nutrients in terms of DVE and OEB intakes (negative total OEB intake, oversupply of total DVE values) resulted in an inability to detect the effectiveness of heat processing in altering bypassing protein (BCP) and/or starch (BST) and their effects. The overall nutrient supply to animal in an experiment should be the context in which any animal performance study is developed. The information described in this article may give better understanding of animal performance in relation to nutritive changes occurring upon processing of legume seeds.

Organogenesis and Production of Some Transgenic Legume Plants by Agrobacterium tumefaciens-mediated Herbicide Resistance Gene Transformation

  • Development of herbicide resistant transgenic legume plants through Agrobacterium-mediated transformation has been worked in many previous studied. Plant regeneration after infection is the important step to obtain successful transgenic plants. Many attempts try to find the optimum media condition for plant regeneration after infection. However, the transformation efficiency of legume plants is still low. In this study, regeneration of some Korean legume species including two soybean cultivars (Dawon and Pungsan) and pea have been done with organogenesis which is used various kind of explants such as cotyledonary-nodes in soybean and bud-containing tissue in pea. We developed the optimum media condition for plant regeneration regulators under Agrobacterium-mediated transformation using different kind and various concentration of plant growth. As the results, B5 medium containing 2 mg/L of 6-benzylaminopurine was selected in this study for the optimum plant regeneration media. The segments were inoculated with Agrobacterium suspension harbored an IG2 vector containing bar gene which confers resistance to phosphinotricin (PPT) in 3, 5 and 7 days. The transformation efficiency was achieved in Dawon 3.03 % and pea 1.46 % with co-cultivation period of 7 days which is showed a high number of GUS positive expression period.

Studies on 5 Protein Fractions Prediction of Forage Legume Mixture by NIRS

  • This study was conducted to assess the feasibility of near-infrared reflectance spectroscopy (NIRS) as a rapid and reliable method for the estimation of crude protein (CP) fractions in forage legume mixtures (sudangrass and pea mixture, and kidney bean and potato mixture). A total of 178 samples were collected and their spectral reflectance obtained in the range of 400~2,500 nm. Of these, 50 samples were selected for calibration and validation, and 35 samples were used for calibration of the data set, and the modified partial least square regression (MPLSR) analysis was performed. The correlation coefficient ($r^2$) and the standard error of cross-validation (SECV) of the calibration models in the CP fractions, A, B1, B2, B3, and C, were 0.94 (1.05), 0.92 (0.74), 0.96 (0.95), 0.91 (0.42), and 0.83 (0.38), respectively. Fifteen samples were used for equation validation, and the $r^2$ and the standard error of prediction (SEP) were 0.87 (1.45), 0.91 (0.49), 0.94 (1.13), 0.36 (0.96), and 0.74 (0.67), respectively. This study showed that NIRS could be an effective tool for the rapid and precise estimation of CP fractions in forage legume mixtures.

두류 전분의 호화와 겔화 성질 (Gelatinization and Gelling Properties of Legume Starches)

  • 묵은 전분의 겔화 성질을 이용한 우리나라 고유의 식품으로서 묵의 재료로는, 녹두, 메밀과 도토리가 이용되어 왔으나 최근 동부가 녹두의 대용으로 이용되고 있다. 묵에 대하여는 많은 연구가 이루어졌으나, 연구자들 사이에 농도와 가열호화방법의 차이로 아직도 묵에 대한 체계적인 결과가 없는 실정이다. 묵의 성질 규명을 위하여 여러 두류의 전분 또는 앙금을 이용한 겔의 성질에 대한 연구가 최근 활발하게 이루어지고 있다. 그러나 아직도 묵에 대한 용어의 정의가 식품학적으로 통일되어있지 못한 실정이므로 앞으로 묵에 대한 체계적인 연구가 요망된다. 이러한 관점에서 우리나라의 주요 두류를 대상으로 전분 또는 앙금의 성질을 종합 정리하였다.
