  • Effects of the ultrasonic treatment on plant represents antifeeding activity and chitin synthesis inhibition activity against diamondback moth (Plutella xylostella L.) by a leaf-dipping method at a concentration of 5,000ppm. Methanol extracts of leaves of the ultrasonic treatment on plant was sequentially partitioned with n-hexane, chloroform, ethlyacetate, butanol and water fractions, and bioassayed. Antifeeding activities were observed in n-hexane, chloroform and ethlyacetate fractions. N-hexane fraction has 94.6% of strongest antifeeding activity against P. xylostella. From the n-hexane sequentially partitioned showed H6 and H8 fractions of antifeeding activities against P. xylostella. And chitin synthesis inhibition activity against P. xylostella was obtained the methaol, n-hexane and water fractions.

Seed propagation and softwood cutting of native styrax japonicus for landscape tree uses (한국 자생 때죽나무의 조경수 이용을 위한 번식방법에 관한 연구)

  • These studies were carried out to investigate seed propagation method and conducted using stem cuttings of Styrax japonicus for using as a woody landscape plant. The results are obtained as follows. After the H\sub2\SO\sub 4\ and GA\sub 3\ treatment to the seeds of Styrax japonicus, three months of warm treatment followed by cold stratification for four months increased seed germination rate. The rooting rate of Styrax japonicus was increased by the treatment with concentrated IBA, especially highest at 7,000ppm. Rooting percentages were highest for cuttings taken on July 15, 1995. Therefore, the most effective method for rooting of Styrax japonicus was treatment with 7,000ppm IBA on July 15 cuttings, which showed rooting rate of over 90%. Half-leaf-remained cuttings were more effective than those with two leaves on rooting of Styrax japonicus. Dipping for 10 seconds in IBA solution resulted in rooting rate of over 70%. The longer the treatment time the worse the rooting.

  • The insecticidal toxicities of seven essential oils against Ricania sp. nymph and adults, newly recorded pest, were evaluated using leaf-dipping and spray methods. At 72 h exposure time, the $LC_{50}$ values of the essential oils of Artemisia vulgaris, Anethum graveolens, Citrus paradisi, Melaleuca alternifolia, Melaleuca leucadendron, Neroli bigarade and Tagetes erecta were 251.02, 387.31, 286.21, 353.54, 275.64, 424.55, $165.68mg/cm^2$ when using the leaf dipping method against Ricania sp. nymphs, and 278.31, 402.11, 306.11, 379.64, 298.22, 459.56, $183.21mg/cm^2$ when using the spray method against Ricania sp. adults, respectively. These results indicate that the essential oil of the Tagetes erecta can be potentially used as new natural insecticidal agents against Ricania sp. nymph and adults.

  • Gerber (Gerbera hybrida) is a valuable ornamental species grown as a potted plant and cut flowers. However, genetic variability within the gerbera genus is very limited. So it is absolutely needed to introduce and widen genetic resources into gerbera lines by genetic transformation. For the purpose, 18 Korean gerbera lines were screened to establish Agrobacterium-mediated genetic transformation procedure. In an experiment to select Korean gerbera lines which are amenable to Agrobacterium-inoculation, 12 lines turned out to be positive in Agrobacterium-inoculation. More callus were produced from BA 2ppm, Zeatin 2ppm, IAA 0.2ppm in pre-culture and regeneration medium (2X media) but there was no difference in the frequency of GUS expression rate. In another experiment to find out optimal condition for highly efficient Agrobacterium-inoculation, petiole and leaf explants have been treated with four different pre-culture periods, two different co-culture periods and two different Agrobacterium strains. As a result, high GUS expression has been showed from petiole and leaf explants treated no pre-culture period with LBA4404 Agrobacterium tumerfaciens, 5 day co-culture period and dipping treatment.

  • Differences in susceptibility of several organophosphate, carbamate and pyrethroid insecticides to three species of aphids were determined by leaf-dipping and spray methods. The insects tested were the apterous adults of the green peach aphid (Myzus persicae), cotton aphid (Aphis gossypii) and soybean aphid (Aphis glycines). The comparison of $LC_{50}$ levels was indicated as follows; (1) The susceptibility to insecticides tested were greatly varied with the test methods. (2) The leaf dipping method is more recommendable than the spray method for insecticide screening with aphids. It was easier, more accurate and less variable than the latter. (3) The susceptibility to insecticides was greatly varied between the aphid species. Soybean aphid was more susceptible to the insecticides than green peach aphid and cotton aphid. Exceptionally, pirimicarb was not effective on the cotton aphid.

  • This experiment was conducted to determine selective mechanism of cyhalofop-butyl ester ((((R-butyl 2-(4-(4-cyano-2-fluorophenoxy) phenoxy) propionate)) between rice and Echinochloa crus-galli. 100ppm of cyhalofop-butyl ester inhibited over 90% of seedling growth of E. crus-galli when applied at 3 leaf stage and complete inhibition was observed at 180ppm applied at the 4 leaf stage, but rice(Chucheongbyeo) was not inhibited by cyhalofop-butyl ester even at 230ppm, regardless of its growth stages(3, 4, 5 and 6 leaf stages). Cyhalofop-butyl ester applied through stem at 10 and 50ppm moved most rapidly to the meristem and resulted in the highest injury on plant height, root length and fresh weight of E. crus-galli. compared with root or leaf application. Seedlings of rice and E. crus-galli at 3 or 4 leaf stage were dipped in 180ppm of cyhalofop-butyl ester solution for 1 minute and aboveground parts of E. crus-galli and rice were removed immediataly after dipping treatment. Regrowth of E. crus-galli was inhibited by the herbicide by 41.7%, but no inhibition was observed in rice. Further, content of chlorophyll reduced to 18.7% of the untreated control, showing appearence of almost being killed, but no effect on chlorophyll content of rice was observed.

  • Transformation system of rolC gene, dwarf gene in Lycium chinenese Mill. established by using system. Pin-punctured leaves induced numerous adventious buds in abaxial side when cultured on 3/2 MS medium containing 2.0 mg/L zeatin. Survival rate and shoot regeneration frequency of leaf explants decreased as kanamycin sulfate level increased. Shoot buds were not regenerated on 3/2 MS medium containing 10 mg/L kanamycin sulfate and 2.0 mg/L zeaein. Of the level tested, 10 mg/L of kanamycin sulfate was optimum in selection of kanamycin sulfate resistant plant. Co-culture time of bacteria and leaf explants was affected at the frequency of shoot regeneration and survival of leaf explants. Leaf explants co-cultivated during above 48hr severely decreased survival rate and shooting rate. Best result on survival rate and shooting rate were obtained when exposed for 24 h. 80 explants of 105 leaf explants survived on 3/2 MS medium containing 2.0 mg/L zeatin and 10 mg/L kanamycin sulfate, and 15 shoots was regenerated on the same medium. To select kanamycin sulfate resistant plant, regenerate as cultured on 3/2 MS medium containing 10 mg/L kanamycin sulfate, and obtained 5 kanamycin resistant plants. Southern blot analysis conformed that the rolC gene was incorporated into the genomic DNA of kanamycin resistant plants.

  • ALA (5-aminolevulinic acid) has been proposed as a tetrapyrrole-dependent photodynamic herbicide and insecticide by the action of the protoporphyrinogen IX oxidase (Protox IX). The present study was conducted to determine growth responses of plant and insects to ALA, biodegradable biopesticidal substance. In the paddy condition experiment, plant height and shoot fresh weight of barnyardgrass (Echinochloa crus-galli) was more reduced by ALA than rice plants, even though both plant species show great phytotoxicity. Hairy crabgrass (Digitaria sanguinalis), a monocot weed, was more sensitive to ALA at 5mM under upland condition when ALA applied on the foliage, compared with soybean (Glycine max) as a dicot crop. ALA solutions were tested for their insecticidal and larvicidal activities against Spodaptera exigua (Hubner) and Tetranychus urticae Koch. by foliar application and leaf-dipping method. The result showed higher insecticidal activity of ALA at 10mM and its mixture with insecticide luferon against S. exigua. Strongest insecticidal activity against T. urticae was observed from the ALA solution at 10mM 72 days after application. This results show that ALA solution had potent herbicidal and insecticidal activities against agricultural pests even though their activities were lower than those of synthetic pesticides.

  • The methanol extracts from 30 species of oriental medicinal plants belonging to 24 families were tested for their lhicidal and antifeeding activit~es against diamondback moth (Plutello xylostella L) and tobacco cutworn (Spodoptera litura F.) by a leaf-dipping method at a concentration of 5, 000 ppm. The methanol extract from Copti chinensis only showed a potent larvicidal activity against P xylostello. Strong antifeeding activity against P. xylostello was observed from the extmds from Platycodon grandiflorurn, Codonopsis pilosula, Asomm sieboldii, Rhus chinensis and Uthospermum erythrorhizon And a potent antifeeding activity against S. liturn was obtained from Akebia quinata and Equlsetum hyemale extracts. A significant antifeeding activity against both species was obtained from R chinensis and C. chinensis extracts.

  • Colorado potato beetle is a most destructive insect pest of potato throughout the world. Although utilization of chemical insecticides is a main method for management of this pest, their negative side-effects such as threat to humans and the environmental pollution prompted researchers to search for natural alternatives. Recently plant essential oils with low or without side-effects against noun-targeted organisms and with high availability were considered as safe bio-pesticides. In the present study, toxicity of essential oil of Iranian lemongrass, Cymbopogon citratus Stapf, was evaluated against 3th instar larvae and adults of Colorado potato beetle by a leaf dipping method. Results displayed essential oil had notable toxicity against both larvae and adults after 24 and 48 h exposure times. Probit analysis revealed $LC_{50}$ values (lethal concentration to kill 50% of population) with 95% confidence limits were 10.32 (9.17 - 11.72) and 7.76 (6.80 - 8.74) ${\mu}l/ml$ for larvae and 6.27 (4.82 - 8.15) and 4.35 (3.24 - 5.62) ${\mu}l/ml$ for adults after 24 and 48 h, respectively. Based on regression analysis, a positive correlation between log concentration of essential oil and insect mortality was achieved. Results indicated C. citratus essential oil can be candidate as a natural alternative to the harmful chemical insecticides in the management of Colorado potato beetle.
