본 실험은 고온기 파프리카 재배에서 CaCl2 엽면 살포에 의한 배꼽썩음과 발생과 경감 효과를 알아보고자 과실 크기에 따른 살포 시기, 횟수 및 시간에 미치는 영향을 구명하였다. CaCl2(Ca 0.4%) 엽면 살포는 6월 3일 부터 7월 1일 까지 방아다리 위 4-9 마디의 착과 과실과 잎에 식물체 1주당 350mL/회를 처리하였다. CaCl2를 7일 간격으로 4주간 엽면 살포 후 수확한 파프리카의 Ca 함량은 배꼽썩음과 과실의 과폭 11-20mm 처리에서 가장 낮았고, pedicel > stem-end > middle, blossom-end 순으로 낮아졌다. 정상과 과실의 Ca 함량은 과폭 31-40mm 처리에서 뚜렷하게 증가하였고 배꼽썩음과와 비교할 때 78% 높았다. 과폭 21mm 이상의 과실 middle과 blossom-end 부위 Ca 함량은 정상과에서는 19.8%-28.8%였으며, 배꼽썩음과에서는 15.7%-18.5%였다. 과폭 31-40mm 처리에서 배꼽썩음과 발생율은 60% 이상으로 급격히 증가하였다. 과폭 크기별 엽면 살포 처리에 따른 파프리카 과중은 차이가 없었으며, 상품과율은 21-30mm 크기에서 가장 높았고, 당도는 과폭 11-30mm 처리에서 높았다. 과폭 21-30mm 파프리카에 7일 동안CaCl2를 3회 엽면 살포하였을 때 세포벽 결합(cell wall-bound, CWB) Ca 함량은 가장 높았고, 배꼽썩음과 발생율은 6.3%로 가장 낮았다. CaCl2 엽면 살포 처리 10일 후 파프리카의 CWB Ca 함량은 대조구에 비해 모든 처리에서 2.9배-3.5배 증가하였다. 하루 중 1회 시간을 달리하여 CaCl2 엽면 살포하였을 때, 7일째 엽소현상이 오전 9시 30분 처리에서 부터 오후 17시 처리 까지 관찰되었고, proline 함량은 처리 시간이 늦어질수록 증가되었다. 따라서 파프리카 여름재배시 배꼽썩음과 발생 경감을 위한 CaCl2 엽면 살포 방법은 과폭 21-30mm인 시기에 3일 간격으로 2회-3회, 오전 8시 이전에 살포 하는 것이 적합하리라 본다.
조풍해(briny wind)에 대한 과수 과종간 피해 양상을 알아보기 위해 본 실험을 수행하였다. 갈변율의 경시적 변화에서 배와 복숭아는 처리 후 1시간 만에 갈변이 시작되었고, 사과와 포도는 2시간 후부터 갈변증상을 나타냈나. 배와 포도는 농도가 높아질수록 낙엽률도 증가되었다. 염분처리에 대한 사과의 에틸렌 함량은 NaCl 3.0% 처리한 경우 1, 2시간 후까지 무처리와 큰 차이가 없었지만, 24시간 후에는 무처리 보다 약 3배 이상 높은 함량을 나타냈고, 5일 후에는 차이가 없었다. 사과의 염분처리에서 뒷면처리는 앞 뒷면 전체처리한 경우와 동일한 갈변과 낙엽률을 보였다. 엽령에 따른 NaCl의 반응은 사과와 배 모두에서 성엽이 유엽보다는 피해가 크게 나타났고 배보다는 사과가 염에 대해 민감하였다. 처리시기에 따른 염피해 에서 7월 30일 처리는 사과에서 측아가 발아되었으며, 정단은 배 30%를 제외하고 나머지 과종은 100% 발아하였다. 상위엽과 정단의 호르몬 함량을 조사한 결과에서, 잎의 t-zeatin 함량은 대포구가 염처리구 보다 높은 함량을 나타냈다. 하지만 IAA와 ABA는 염농도가 높아질수록 상위엽의 함량이 증가하였다.

Turnip mosaic Potyvirus (TuMV) and Ribgrass mosaic Tobamovirus (RMV) are major viruses infecting crucifer crops in Korea. RMV-FG22 was isolated from oriental cabbage. TuMV isolates were TuMV-CA7 from oriental cabbage, TuMV-TU and TuMV-TU2 from turnip, TuMV-RA from rape, TuMV-ST from stock, and TuMV-R9 from radish. The six isolates of TuMV were classified by symptom expression in inbred lines of crucifers. TuMV-CA7 and TuMV-TU isolates infected mostly oriental cabbages; TuMV-ST, TuMV-TU2, and TuMV-R9 infected radishes; and TuMV-RA infected both oriental cabbages and radishes. Crops used in six combinations of mixed infections were 'Tambok' cultivar resistant to TuMV,'SSD63' susceptible inbred line of oriental cabbage, pure line of leaf mustard, and‘Daeburyungyeorum’cultivar of radish. External symptoms in 'Tambok' and radish by each of the six single infections of TuMV showed similar results by bioassay. Synergistic response of necrotic death occurred within 1 week after inoculation in all combinations mixed with TuMV and RMV-FG22 on leaf mustard. In oriental cabbage 'SSD63' , synergism of necrosis occurred in four TuMV isolates, but not in TuMV-ST and TuMV-R9. In oriental cabbage 'Tambok' , synergism was expressed only in two combinations of RMV-FG22+TuMV-CA7 and RMV-FG22+TuV-TU, but other combinations had the same symptoms produced by RM-FG22. In radish‘Daeburyungyeorum’, only mild mosaic symptoms were induced by combinations of RMV-FG22+TuMV-CA7, RMV-FG22+TuMV-TU, RMV-FG22+TuMV-RA, and RMV-FG22+TuMV-R9. Mosaic and severe mosaic were induced in combinations of RMV-FG22 +TuMV-TU2 and RMV-FG22+TuMV-ST, respectively.
The potential of three plants extracts, to protect potato plants against bacterial wilt caused by Ralstonia solanacearum was determined under greenhouse and field conditions. All soil drenching treatments of aqueous plant extracts of Hibsicus sabdariffa, Punica granatum and Eucalyptus globulus significantly reduced the disease severity compared with inoculated control. Although the applications of all three plant extracts resulted in similar reductions of disease severity in field up 63.23 to 68.39%, treatment of E. globulus leaf extract was found greater in restricting the symptom development than other the two plant extracts in the greenhouse. More than 94% reduction in the bacterial wilt symptom was observed in potato plants. All tested plant extracts were effective in inhibiting the growth of bacterial pathogen, not only in vitro, but also in stem of potato plants as compared with the inoculated control Potato plants treated with extract of H. sabdariffa reduced bacterial growth more effectively than treatment with P. granatum and E. globulus. Activity of defence-related enzymes, including peroxidase, polyphenoloxidase and phenylalanine ammonia lyase, were significantly increased in plants treated with the plant extracts compared to the control during the experimental period. In general, the higher enzymes activities were determined in both inoculated and non-inoculated treated potato plants after 8 days from plant extracts treatment. These results suggested that these plant extracts may be play an important role in controlling the potato bacterial wilt disease, through they have antimicrobial activity and induction of systemic resistance in potato plants.
국내에서 분리 동정한 오이모자이크 바이러스(CMV)로부터 외피단백질(CP) 유전자를 분리하였고 이의 염기서열을 결정한 결과 129번째 아미노산이 proline으로서 mosaic symptom을 보이고 염기 및 아미노산 서열의 유사성을 비교한 결과 subgroup I (type I)에 속한다는 것을 확인하였다. 분리한 CP 유전자를 식물형질전환용 운반체에 삽입하여 담배 재배종인 NC82와 바이러스 증식용인 Samsun에 잎절편을 통하여 형질전환시켰다. 형질전환된 담배의 DNA를 분리하여 Southern blotting과 PCR을 수행한 결과 대부분의 형질전환체에 CP 유전자가 삽입되였음을 확인하였고 T$_1$ 종자를 kanamycin이 첨가된 배지에서 후대 항생제 저항성 검정을 실시한 결과 1개의 CP 유전자가 삽입된 경우가 50%에 달했고 2개와 3개의 유전자가 삽입된 경우도 각각 39%와 11%에 달했다.
From the view point of phytopathological anatomy, the author has tried to study the effect of the shoot cluster disease virus on the internal structure of vascular tissues of chinese date tree (Ziziphus jujuba var. inermis Rehd.) comparing healthy checks and diseased plants. The materials were collected at the several sites, Kumgock-Ri, Masuc-Ri, Kyungi-Do, and near the campus of Korea University and around the area of Chongam-Dong, Seoul City, from August 15th to September 5th 1959. The leaf materials of healthy and diseased plants are fixed and aspirated in two kinds of killing solutions, formalin-acetic acid alcohol solution and Craf III solution. Sections were cut at 5-10$\mu$ thickness and stained with the double staining reagents of safranin and fast green. In this experiment the author has observed that there are marked structural changes in the infected plants in contrast of healthy checks. As figures 3-7 show that the following characteric changes have taken place on infected plants: 1) the arrangement of irregularly developed sieve elements in phloem, 2) the degeneration of phloem elements, 3) the irregular arrangement of epidermis in mid-vein, 4) more necrosis is observed among the parenchymatous cells, 5) abundant accumulatin of starch grains in parenchymatous cells, . In contrast to the above irregularities caused by the virus disease, the healthy checks appear normal structures as shown in figures 1 and 2. In adding to the all features noted above, the author could also observe an interesting feature that the xylem elements in mid-vein vascular bundle tissues are considerably disorganized to show the unspecialized vessel elements, the irregularly arranged xylem elements. However, this kind of irregularities which occur in xylem under the virus infection has not been reported previously. The features noted on the internal structure of vascular bundle under the condition of infection by the shoot cluster disease on chinese date trees appear to be more or less closely similar to the symptoms of the bunchy-top of banana and the yellow dwarf disease of barley in respect to the fact that whether phloem necrosis takes place as a primary symptom or a secondary symptom. In all these disease, primary histological changes of hypoplasia and hypertrophy are preceeded by the necrosis of phloem.
Bacterial blight is one of the most serious diseases of rice (Oryza sativa L.), and it has been known that Xanthomonas oryzae pv. oryzae (Xoo) causes this disease symptom. To develop resistance rice cultivars against Xoo, 3,000 lines of $M_3$ mutants, which were irradiated with gamma ray, were tested by 'scissor-dip method' primarily, and 191 putative resistant lines were selected. In $M_4$ generation, these lines were screened again with various ways such as measuring of symptom of bacterial blight in leaf, number of tiller, fresh weight, and phenotypic segregation ratio in next generation. Finally, six resistance lines were selected. RT-PCR analysis revealed that these lines displayed high level of R-genes such as Xa21, Pi36, and Pi-ta. These results indicate that mutations by gamma ray cause disruptions of regulatory signal transduction systems of these R-genes. Furthermore, these selected mutants could be useful for the development of rice cultivar resistant to Xoo.
세균과 바이러스에 감염된 작물의 생물적 스트레스 탐지를 원격탐지 기술 기반의 열적외 센서를 이용하여 실내 생육 챔버에서 실험하였다. 감염으로 인한 엽온의 증가와 감염 농도에 따른 엽온 차이를 확인했다. 또한 엽온 기반으로 산출한 CWSI 값은 감염 잎에서 증가하였고, 그러한 현상은 육안으로 병징을 발견하기 하루 전에 시작되었다. 따라서 스마트팜 시스템의 작물 모니터링에 열적외 센서를 이용한다면, 병해의 탐지는 물론 피해 등급 평가, 조기 알람 등에 활용될 수 있을 것이다. 하지만 실제 스마트팜 적용을 위해서는 향후 엽온 측정 정확도 향상 기술, 자료 해석 방법, 생물 비생물적 스트레스 구별 알고리즘 연구 등이 추가로 필요할 것이다.
The pathogenicity to pear trees and other experimental hosts of the Apple stem grooving virus Korean isolate (ASGV-K) carried by a fungal vector, Talaromyces flavus was examined. ASGV-harboring T. flavus induced mild symptoms on virus-free pears. Symptom severity was intermediate between pears showing typical PBNLS and virus-free pears. Ten cultivars of Phaseolus vulgaris showed 35%-90% infectivity by direct infiltration into leaves and roots by ASGV-harboring T. flavus. Application of fungal cultures to soils showed 0%-70% infectivity depending on the P. vulgaris cultivar. Sap extracted from ASGV-infected Chenopodium quinoa induced similar symptoms on P. vulgaris at 25 days after inoculation. Similar symptoms were also detected on P. vulgaris which were inoculated with ASGV-harboring T.flavus. When healthy P. vulgaris leaves were challenged with sap extracted from P. vulgaris leaves infected with ASGV-harboring T. flavus, typical symptoms were observed. These data suggest that T. flavus mediates the transfer of ASGV to host plants.
