Histological and Morphological Characteristics of New and Latent Bud Formation in Panax ginseng C.A. Meyer (인삼의 신아 및 잠아발생의 조직 형태적 특성)

  • This experiment was conducted to obtain the basic information on new- and latent-bud formation, and stem vestige arrangement on the rhizome of Panax ginseng C.A. Meyer. Latent buds emerged from meristematic region between shoot and root of the embryo, and new buds for the next year were distributed both at the bottom portion of the stem and the rhizome. In the new buds, organs such as leaf, stem, and flower bud were already completely differentiated, while the latent bud had an undifferentiated meristematic tissue arranged linearly in a vertical line, indicating that each year new- and latent-buds are formed successively. This result suggests that the number of stem vestige may be used for the determination of ginseng age. Developmental Pathway of Main Bud in Panax ginseng C.A. Meyer (고려인삼의 주아 발달경로)

  • The researchers studied the composition of latent buds by surveying the embryo of harvested seed, and the developmental pathway of latent buds by analyzing the characteristics of local ginseng. One-year-old ginseng seedlings were transplanted into the field and harvested two years later. The developmental pathway of the main bud, which would be the shoot of ginseng in the fourth year, was also investigated. The main bud of the seedling was formed from the region between root and shoot of the germinating seed. Primary and axillary latent buds protruded in a dome-shape from the cortex and separated from the main bud. Ninety percent of the single main bud was derived from the primary latent bud. Twin main buds were derived from a primary latent bud and one axillary bud, and triple main buds were derived from primary latent bud and two axillary latent buds. In the field, the researchers could not find 2-stem plants in 2-3 years old plants. However, the researchers found a 2-stem plant in a 4-year-old plant because its two main buds developed from a 3-year-old plant. We can conclude that a 2-stem plant was observed in the plant that was at least 4 years old. The main buds of the 4-year-old plant were formed at the primary and axillary latent bud of seed and cortex, the latent bud of rhizome in a 2-3 year old plant. In older plants, twin and triple main buds were derived mostly from the cortex latent bud than the primary latent bud.

Growth Characteristics and Demography of Polygonatum involucratum and Polygonatum humile Ramet Population (용동굴레와 각시둥글레의 생장특성과 라메트 개체군의 동태)

  • Growth hadit and demograph in a mature population of polygonatum and polygonatum humile in kanghwa island were studied for two growing seasons. Shoots of two species emerged early spring from the growing apices of the underdground rhizomes which had persisted for up to 1~5 year.after flowering, the ramets produced two rhizome buds at the shoot base. Bacause of the apical dominance in the rhizome system, a new rhizome was developed from only one bud, an actual bud,and the other latent buds were suppressed sothat remained dormant. The latent dud produced a new rhizome only when the actual dud was severed by the herbivores or by the physical obstacles. Therefore, the ramet number is not increased by the new rhizome from the latent bud. however, new ramets dould sometimes grow from latent buds which had been produced more than a year ago. Production of these ramets was main means increasing the ramet numbew and widening the potential zone of exploitation. Changes in size class of each ramet were noyiceable after a tear in small size-classes. Small ramets replaced themselves with larger-sized ramet, while large ramets with similar-sized or smallar-sized ramets. ramet numbers were average 0.82and 1.14 times of those fromthe previous year inp. involucratum and p. humile, though there was between-site variation. Almost all the ramets in the quadrats were alive during the growing season. when the entire rhizome systems were excavater next spring, there were many rhizomesegments without shoots, especially in p. involucratum. therefore, the drcrease of ramet number in p. involucratum in probably due to the climatic factors of winter.

Effect of $GA_3$ Before Planting on Growth and Yield of Atractylodes japonica Koidz (삽주의 정식전 $GA_3$ 처리가 생육에 미치는 영향)

  • This experiment was carried out to determine the effects of $GA_3$ treatment (concentration: period) to rhizomes before planting on their growth, morphology and yield. $GA_3$ treatment increased the number of shoots, leaves or latent buds, plant height, and leaf length per plant compared with non-treatment. The best treatment concentrations were 50 ppm in stem per plant, 100 ppm in latent bud and rhizome fresh weight per plant and 250 ppm in shoot fresh weight per plant. The number of leaves and roots per plant, plant height and fresh weights of shoot and root were the greatest in one day imbibition of $GA_3$. Numbers of stems and latent buds per plant and rhizome fresh weight, however, were the highest in two day imbibition. In addition, the most rhizomes per land unit were produced in one day imbibition treatment of 100 ppm $GA_3$.

  • Stem and petiol explants of peony culture turned to brownish black soon after placing onto medium and degenerated to death. Disroloration was caused mainly by ferrous and calcium cloride. Nitrate was a main factor for the death of culture. The culture damage was increased with the increment of the medium salt strength. A few latent axillary buds were elongated to shoots without forming callus.

  • This experiment was conducted to investigate the effects of length of storage period under low temperature, $CO_2$ enrichment and addition of plant growth regulators in Murashige and Skoog medium on the plant regeneration of Korean ginseng (Panax ginseng C. A. Meyer). Seeds were treated for 60 and 80 days respectively under $5^{\circ}C$ environment. 2500ppm of $CO_2$ was enriched by ventilation. Three plant growth regulators added to the medium were Indolbutyric acid, Benzyladenin and Gibberellic acid (GA3). The result indicated that : The capacity of differentiation was higher in the aged cotyledons from the seeds treated for 80 days under low temperature condition than in those treated for 60 days. $CO_2$ enrichment had stimulating effects on the growth and development of shoot primordium significantly but less effects on the formation of adventitious buds. From one zygotic embryo hundreds of plantlets were differentiated. $CO_2$ enrichment had effects on the formation of both indirect somatic embryo and direct somatic embryo. Indirect somatic embryo showed little growth and differentiation, being undifferentiated vascular stele and epicotyl. Direct somatic embryos were formed on the epidermis of backside basal part of cotyledon. Those embryos developed to whole plant having latent bud.

Studies on Biology and Control of the Mulberry Small Weevil, Baris deplanata ROELOFS (Coleoptera: Curculionidae) (뽕나무애바구미의 생태 및 방제에 관한 연구)

  • The mulberry small weevil, Baris deplanata ROELOFS, has highly infested mulberry trees in Korea. As the damage caused by the mulberry small weevil in mulberry fields has been increased over the country since 1969, the authors has carried out a series of biological and controlling studies on the pest from 1971 to 1972. The results obtained are summarized as follows. 1. The adult weevil is elongate oval in shape with black in color and the probocis is long as usual in curculionidae. The size of adult female is 3.30${\pm}$0.04mm in length, 1.47${\pm}$0.04mm in width, and the length of proboscis is 1.25${\pm}$0.014mm, while adult male is 3.28${\pm}$0.06mm in length, 1.40${\pm}$0.04mm in width, and the length of proboscis is 1.30${\pm}$0.02mm. The antenna is geniculate consisting of 12 segments. The terminal sternite of the abdomen has a pointed tip in male but not in female. 2. The egg is long oval in shape, milky white in color, 0.51${\pm}$0.05mm in length and 0.32${\pm}$0.02mm in width. 3. The mature larva is cylindrical and light yellowow in color except the head of dark brown, and legless, 3.88${\pm}$0.06mm in length, 1.40${\pm}$0.02mm width, each segment bearing many wrinkless and short setae. 4. The pupa is long oval, milky white and exarate, 3.53${\pm}$0.09 in length, 1.40${\pm}$0.03mm in width. 5. Majority of the species has one generation through a year and overwinters as adult in xylem of withered branch and come out again from late April to early May in next year. But some of the female oviposit in the same year and the offsprings overwinter as larva (0.4%) or pupa (0.1%) 6. The eggs are mostly laid under the cork layer of withered branch and the number of eggs deposited by an adult female is 73.44${\pm}$8.74, the average egg-laying period is 33.88${\pm}$6.04 days. The incubation period is 11.69${\pm}$0.39 days, the larval period 45.04${\pm}$1.63 and the pupal period 11.05${\pm}$0.49 days. The period of adult's activity is 46.7${\pm}$5.90 days. 7. The larvae feed on the cambium under the bark and adults feed on the winter bud, the latent bud, the leaf stalk and the base of newly shoot. 8. An active period of adults was observed during the period of 4 months from April to July. However, the peak of adult-density occurred in the early May (in the fields of spring-prunning) and early to middle June(in the fields of summer-prunning). 9. There is a positive correlation between the density of larvae and diameter and length of the branches. 10. The pattern of distributions of the adult of mulberry small weevil is negative binomial distribution. 11. The chalcid fly was disclosed to be a natural enemy which was parasite on the larvae of mulberry small weevil and its parasitic ratio was 11.9%. 12. Phosvel D, Malix D, Salithion EC, DDVP EC, and Phosvel EC were effective for the control of adults and Satchukoto-S EC, and Salithio EC were effective for the control of larvae.
