Infections of Larval Stages of Dicrocoelium dendriticum and Brachylaima sp. in Brown Garden Snail, Helix aspersa, in Turkey

  • The aim of this study was to determine the presence and prevalence of larval stages of Dicrocoelium dendriticum and Brachylaima sp. in the first intermediate host, a species of land snail, Helix aspersa, in Turkey. A total of 211 snails were collected in April-May 2014 from pastures in Mersin District. Larval stages of D. dendriticum were identified under a light microscope. Hepatopancreas from naturally infected H. aspersa snails were examined histologically. The prevalence of larval stages of D. dendriticum and Brachylaima sp. in H. aspersa snails was found to be 2.4% and 1.9%, respectively, in Mersin, Turkey. Cercariae were not matured in sporocysts at the beginning of April; however, it was observed that cercariae matured and started to leave sporocysts by early-May. Thus, it was concluded that H. aspersa acts as an intermediate host to D. dendriticumin and Brachylaima sp. in Mersin, Turkey. A digenean trematode Brachylaima sp. was seen for the first time in Turkey.

Cytochemical and Immunocytological Study on the Cellulase Activity in the stomach of the Land Snail Nesiohelix samarangae (동양달팽이 Nesiohelix samarangae 위에서의 Cellulase 활성에 대한 세포화학적 및 면역세포화학적 연구)

  • 동양달팽이의 위 상피에서 섬유소 분해효소의 분비여부를 화확인하기 위해 세포화학적 방법과 immunogold labeoling mithod를 수행되었다. Nesiohelix samarangae의 위에선 3가지 종류의 원주상피가 관찰되었다. 이 중 TYpe1세포는 microvilli가 발달하고 매우 많은 분비과립을 함유하고 있었으며, Type2세포는 위로 치우친 핵과 전자밀도가 높은 세포질에 rER이 매우 발달해 있었다. Type3 세포는 섬모와 미세융모를 함께 가지고 있는 세포로서 분비과립을 약간 함유하는 것으로 관찰되었다. 세포화학적 방법으로 위 상피에서의 cellulase의 활성을 시험해본 결과 상피세포들의 대다수를 차지하는 Type 1세포들과 Type3 세포들의 분비과립들의 주변에서 cellulase활성이 높았고, immunogold staining결과는 황금입자가 위 상피의 분비과립 주위의 세포질과 rER의 막과 주변세포질, 그리고 상피세포들의 상단부 세포질 등에서 표지되었다. 이상의 모든 실험 결과는 동양달팽이의 위 상피세포에서 cellulase가 분비된다는 사실을 알았다.

Study on the Feeding Preferences of Luciola unmunsana Larvae (운문산반딧불이(Luciola unmunsana) 유충의 먹이선호도 연구)

  • The purpose of this study is to investigate the habitat of Luciola unmunsana to understand the status of land snails, which are the larvae prey, and to evaluate the effects of larval feeding preferences and prey size for the restoration of the L. unmunsana habitat as a basis for future restoration projects. The research methods included field surveys of the habitat and laboratory experiments on feeding preferences using three types of land snails (Allopeas clavulinum kyotoense, Euphaedusa fusaniana, Pupinella rufa) observed in the habitat. Field surveys of 13 L. unmunsana habitat sites revealed a total of 24 land snail species. The experimental results regarding the larvae prey were analyzed for statistical significance of feeding preferences using Chi-square tests and independent sample t-tests. Consequently, larvae showed the highest preference for A. c. kyotoense, with statistically significant differences in feeding preferences (p<0.05). Also, statistically significant differences were observed in the size between the land snails that were eaten as prey and those that were not eaten. This research can deepen the understanding of the feeding preferences of L. unmunsana larvae and provide practical guidelines for the effective conservation and restoration strategies of the firefly habitat.

  • A cytochemical study by the Benedict's reaction method was conducted to find out the cellulase activity in the digestive organs of Helix aspersa. Out of the all digestive organs, the stomach, the intestine, and the digestive gland showed cellulase activities under the transmission electron microscopy.

  • The species of land snails (Stylommatophora) that feed on perilla plants were observed under leaf perilla greenhouses cultivated in the Milyang and Busan areas. Four species of land snails were recorded. Among them, three species were slugs, Meghimatium bilineatum in Phlomycidae, Deroceras reticula tum, and Limax marginatus in Limacidae, and one species was the snail Acusta despecta sieboldiana in Bradybaenidae. D. reticulatum occurred most frequently followed by A. despecta sieboldiana, L. marginatus and M bilineatum. Annually, land snails showed two peaks in the middle of April and in late November for D. reticulatum, early May to middle November for A. despecta sieboldiana, and middle April and early November for L. marginatus. M bilineatum, however, did not show any clear peak because of its low population occurrence.

  • An anatomical and ultrastructural study on the dart sac and the dart of a Korean snail Nesiohelix samarangae was conducted to understand their morphological characteristics with the help of the light microscope and electron microscopes, TEM and SEM. Nesiolelix samarangae had two darts which are pure white structures 6-8 mm in length, tapering from 0.6-0.8 mm to 0.15-0.3 mm. The dart sac had a long conic lumen subdivided into two by a septal wall, and the darts were centrally embedded in the thick muscular layers of the sac. The darts occupied each of the two luminal spaces one per each. The convexed surfaces of the darts had many crystal buds in the shape of the petals. Otherwise, the convexed surfaces of the darts had numerous crystal buds in the shape of candle or topaz. The luminal surface of the dart sac was covered with a single columnar epithelium. The epithelial cells possessed microvilli on their free surface.

Molecular Phylogenetic Study of Nesiohelix samarangae Based on CO-I Gene (동양달팽이 (Nesiohelix samarangae)의 CO-I 유전자를 이용한 분자계통학적 연구)

  • Previously, we have reported expressed sequence tags (ESTs) analysis on the land snail, Nesiohelix samarangae (Ns). Of these ESTs, we have identified four partial fragments of N. samarangae cytochrome oxydase I (NsCO-I) gene which lead to obtain an 852 bp partial cDNA. Since NsCO-I is one of the best-known molecular phylogenetic markers, we have attempted to conduct comparative in silico analysis by using the NsCO-I gene. The combined results from BLAST analyses, multiple sequence alignment and molecular phylogenetic study of NsCO-I cDNA indicate that N. samarangae has similarity to three land snails such as Elona quimperiana, Euhadra herklotsi and Euhadra idzumonis.

  • Comparative karyological analyses of the Korean land snail, Diplommatina (Sinica) paxillus and Diplommatina (Sinica) changensis, were peformed by the Giemsa-staining and air-drying method. The karyotypes of both species were the same (2n = 26). However, the chromosome lengths and arm ratios, and relative chromosome lengths of the two species were distinctly different.
