This study was carried out to investigate the changes in yeast cell number, organic acid and total acid during the fermentation and distribution processes for enhancement of preservation of Makgeolli. Organic acids, including lactic acid, succinic acid, malic acid and citric acid, were increased with fermentation time, while oxalic acid, phosphoric acid and acetic acid were not detected, respectively. Production of organic acids leaded to pH reduction in Makgeolli. In case of Makgeolli kept at $4^{\circ}C$, there was no change in organic acids until 20 days. On the other hand, when observing the change in organic acid of Makgeolli kept at $25^{\circ}C$, concentration of lactic acid was decreased, while citric acid was not detected from the beginning of storage. However, acetic acid was detected from 10th day and rapidly increased at the 25th day. Therefore, it is suggested that the current expiration date (10 days in a cooler) could be extended.
The effects of salting methods on sensory and microbiological properties of Kakdugi were evaluated during fermentation at 10$\^{C}$ for up to 52 days. Kakdugi samples were prepared by 4 different salting methods at the final salt concentration about 1.5%, which was appropriate for organoleptic quality. The salting methods for radish cubes(2cm size) of Kakdugi included; 1) Treatment S-1: spraying dry salt uniformly onto the radish cubes, at the concentration of about 1.5%(w/w) and cured for 1 hr, 2) Treatment S-5: spraying dry salt uniformly onto the radish cubes, at the concentration of about 1.2%(w/w) and cured for 5 hr, 3) Treatment B-1: brining radish cubes in a 8.5%(w/v) salt solution and cured for 1 hr, 4) Treatment B-5: brining radish cubes in a 4.0%(w/v) salt solution and cured for 5 hr. During the early stage of fermentation, sensory test showed higher scores in the overall acceptability of Kakdugi prepared with salting methods S-1 and S-5 than those with B-1 and B-5. However, the trend of acceptability has been reversed by the treatments B-1 and, more notably, by B-5 at the later stage of fermentation. The counts of lactic acid bacteria increased remarkably and then decreased gradually after the optimum ripening period. The major lactic acid bacteria isolated and identified from Kakdugi were Pediococcus, Streptococcus, Leuconostoc, and Lactobacillus.
This study was conducted to compare the effects of lactic acid bacteria (LAB) or LAB+cellulases on the cell wall compositions and the in vitro dry matter digestibility (IVDMD) of Rhodesgrass (RG) and Italian ryegrass (IRG) silages. LAB (Lactobacillus cassei) at a concentration of $10{\times}10^5\;cfu.g^{-1}$ fresh forage was added to all ensiling samples (except the untreated control) of RG and IRG. The cellulases used were Acremoniumcellulase (A), Meicelase (M) or a mixture of both (AM). Each cellulase was applied at levels of 0.005, 0.01 and 0.02 % fresh sample. The samples were incubated at 20, 30 and $40^{\circ}C$ for about 2 months of storage. LAB inoculation did not affect cell wall components or IVDMD of both the RG and IRG silages, but LAB+cellulase treatments did. Increasing the amount of cellulase addition resulted in further decreases of cell wall concentrations. This reduction more markedly occurred with cellulases A and AM than it did with cellulase M. Cell wall components losses were higher in the IRG silages than in the RG silages. LAB+cellulase treatments decreased IVDMD of the RG silages, but had no effect on the IRG silages. The different effect of LAB+cellulase treatments on cell wall degradation and IVDMD of the RG and IRG silages suggested that RG contains more structural carbohydrates, which were difficult to degrade with cellulase, than did IRG.
Growth curves of seven strains of lactic acid bacteria isolated from Kimchi were graphed, when they were cultivated at 3$0^{\circ}C$ in filter sterilized Chinese cabbage juice containing 0, 2, 4 and 6% salt, and then lag time and generation time were calculated. The shapes of growth curves were changed differently among strains, as salt concentrations were increased. The addition of 2~4% salt resulted in prolongation of lag time were most omspicuous in Leu. Paramesenteroides and Leu. Mesenteroides subsp. Dextranicum, and the next in Leu. Mesenteroides subsp. Mesenteroides,m Lac. Bavaricus and Lac. Gomohiochii, and the least in Lac. Plantarum and Lac. Brevis. And then the prolongations of generation time were most remarkable in Lac. bavaricus and Lac. Homohiochii, and the next in Leu. Mesenteroides subsp. Mesenteroides, Leu. Mesenteroides subsp. Dextranicum and Leu. Paramesenteroides, and the least in Lac. Plantarum and Lac. brevis. By increasing salt concentrations from 0 to 2%, the generation times of Leu. Cesenteroides subsp. Dextranicum and Leu. Mesenteriodes subsp. Mesenteroides were prolonged slightly, while those of Lac. Homohiochii and Lac. Brevis were not changed, and those of Lac. Plantarum, Lac. Bavaricus and Leu. Paramesenteroides were shortened slightly. As salt concentrations were increased from 2% to 4%, inversions in the order of generation time occurred among strains. As a whole, lower salt concentrations were more favorable for the growth of Leuconostoc strains, while higher salt concentrations were for Lactobacillus strains.
This experiment was carried out to observe a biological effect of ginseng on the weight gain, physical performance and lactic acid production after exercise in mice. A group of mice weighing about 19 gm was divided into ginseng and cotrol group. on the treadmill (Exp. I & II) and LDH activity of liver and heart homogenates (Exp. II) were determined. Results are summerized as follows; 1. Body weight gain was greater id ginseng group than in control and the difference was statistically significant at 9th and 16th days of experimental period. 2. Maximal running time of ginseng was found to be longer than that of control (p<0.05) in experiment I and the experiment II also revealed the significant increase in maximal running time in ginseng group. 3. Bloo lactate concentration of 48 hour-rest from physical exertion was lower in ginseng group than in control (p<0.05). 4. LDH activity in liver homogenate was lower compared to control group, but in heart homogenate, it was greater in ginseng group. Above findings may be concluded tat the range of biological dose (20 mg/mice/day) of ginseng powder stimulated the body weight gain and increase of physical performance and its mechanism might be attributable to a lower level of blood lactic acid. The adaptive change of LDH activity also contributed to the change in lactate level in blood and tissue.
For development of an on-line monitoring unit of fermentation process, an auto dilution unit based on traditional chemical and biological analytical method was developed and the performance was evaluated. The dilution unit was constructed with two syringe pumps and flow direction change valves and fully automated. Total delivery volume of two pumps using distilled water was measured to confirm the operating stability And diluted concentrations of three substrate solutions (glucose, lactic acid, ethanol) were compared with a standard method with a high performance liquid chromatograph (glucose, lactic acid) and gas chromatograph (ethanol). Relative error values of total delivery volume of the pumps were below 3% and standard deviation values were 0.003 (n=5). Relative error values of diluted concentration of the dilution unit measurements were below 2% with 1/10 of dilution ratio and 70, $80{\mu}{\ell}$ of sample volume conditions for glucose and lactic acid, 1/30 of dilution ratio and $70{\mu}{\ell}$ of sample volume conditions for ethanol, respectively. In case of the ethanol, cause of the evaporative characteristics, the relative error values showed over 5% whole experimental conditions.
The aim of this study was to evaluate the effect of Lactobacillus acidophilus probiotic (LAP) product supplementation on the blood profile, fecal noxious gas emission, and fecal shedding of lactic acid bacteria and coliform bacteria in healthy adult Beagle dogs. In total, 14 Beagle dogs with an average initial body weight of 10.19 ± 0.61 kg were randomly assigned into two dietary treatments,with and without LAP supplementation, for a 28-day feeding trial. At the end of the experiment, there was no significant (p > 0.05) difference in the concentration of serum total cholesterol, high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), white blood cell (WBC), red blood cell (RBC), blood lymphocyte percentage, fecal hydrogen sulfide (H2S) and total mercaptans (R.SH) emission, and fecal coliforms counts. However, the serum concentrations of the triglyceride and fecal ammonia (NH3) emission of the LAP treatment were significantly (p < 0.05) decreased in the group compared with the CON dogs. Fecal total lactic acid bacteria counts were significantly (p < 0.05) increased in the LAP treatment. In conclusion, the supplementation of LAP in Beagle dog diets could decrease the blood triglyceride level and enhance the gut Lactobacillus count which may have positive effects on dogs.

The discarding of wastes into the environment is a significant problem for many communities. Still, food waste can be used for lactic acid bacteria (LAB) growth. Here, we evaluated three growth media equivalent to de Mann Rogosa Sharpe (MRS), using apple bagasse, yeast waste, fish flour, forage oats, and cheese whey. Cell-free supernatants of eight LAB strains were tested for antimicrobial activity against nine indicator microorganisms. The supernatants were also evaluated for protein content, reducing sugars, pH, and lactic acid concentration. Cell-free supernatants from fish flour broth (FFB) LAB growth were the most effective. The strain Leuconostoc mesenteroides PIM5 presented the best activity in all media. L. mesenteroides CAL14 completely inhibited L. monocytogenes and strongly inhibited Bacillus cereus (91.1%). The strain L. mesenteroides PIM5 consumed more proteins (77.42%) and reducing sugars (56.08%) in FFB than in MRS broth (51.78% and 30.58%, respectively). Culture media formulated with agroindustrial wastes positively improved the antimicrobial activity of selected LAB, probably due to the production of antimicrobial peptides or bacteriocins.
This study was conducted to determine the acid and phenol compounds in smoke total particulate matter(TPM) by the different tobacco variety, and grade of tobacco leaves. Sixteen kinds of tobacco leaves which were flue-curd, burley, orient, reconstituted tobacco, expanded stem, and expanded cut tobacco, were selected for this study. After collecting a TPM by using smoking machine, the concentration of TPM components was analyzed by GC. Acid components of TPM of mainstream smoke were different from the variety and grade. The order of the highest concentration of acid compounds in TPM was flue-cured > orient> burley> expanded cut tobacco> reconstituted tobacco> expanded stem. Though lactic acid and glycolic acid concentrations in flue-cured tobacco were twice higher than those in burley tobacco, the contents of 2-furoic acid and 3,4-dihydroxy butanoic acid in burley tobacco were higher than those in flue-cured tobacco. The content of phenolic compounds in the high grade and thick leaves was higher than that in other tobacco leaves. Phenol and catechol compounds in burley CD3W-2 revealed the least value in concentration among the samples tested. Pyrocatechol and hydroquinone concentrations in flue-cured tobacco were 2-3 times higher than those in burley and orient tobacco.
Some strains of Escherichia coli are categorized as pathogenic bacteria and alternative antimicrobials including bacteriophages for controlling these bacteria have been studied. In this study we screened antimicrobial candidates that present synergistic inhibition of the growth of E. coli DH5α as a model when co-treated with the bacteriophage ECP27 to target the bacteria. As candidates, CaCl2, lactic acid, and citric acid were tested. CaCl2 showed a synergistic inhibition against the strain by dose-dependent manner at 6 h of incubation but the viable cell count was recovered at 12 h. However, lactic acid and citric acid at 30 mM concentration showed synergistic inhibitions at 6 h of incubation and cleared the viable cells of E. coli DH5α at 12 h when co-treated with the bacteriophage even though lactic acid or citric acid alone was effective. Therefore, co-treatment using the bacteriophage and organic acids such as lactic acid and citric acid can be a solution for synergistic inhibition of the growth of E. coli.
