Planar microchip-based lactate biosensor (평면 소자형 락테이트 바이오센서)

  • Two electrode-based lactate biosensor was prepared by immobilizing lactate oxidase (LOD) obtained from pediococcus species in a poly(vinyl alcohol). Hydrogen peroxide ($H_2O_2$) produced by the reaction of lactate and LOD was detected on the Pt-black that was electrochemically deposited on the Au electrode. Sensors fabricated with Pt-black deposited Au electrode provided a high current of $H_2O_2$ oxidation at a substantially lowered applied potential (+300 mV vs. Ag/AgCl), resulting in reduced interferences from easily oxidizable species such as ascorbic acid, acetaminophen, and uric acid. An outer membrane is formulated by adjusting water uptake of hydrophilic polyurethane (HPU). The sensor performance was evaluated in vitro with both flow-through arrangement and static mode. The sensor showed a linear range from 0.1 mM to about 9.0 mM in 0.05 M phosphate buffer (pH 7.6) containing 0.05 M NaCl. Storing the sensors prepared in this work at $4^{\circ}C$ buffer solution while not in use, they provided same electrochemical performance for more than 25 days.

  • In this work immobilization technique of enzymes onto the magnetic nanoparticles has been developed for biochip applications. Glucose oxidase and lactate dehydrogenase were immobilized on magnetic nanoparticles via cyanamide and glutaraldehyde. Immobilized enzymes had good operational and storage stability The immobilized glucose oxidase and lactate dehydrogenase were characterized by some factors(pH, temperature, and components of buffer solution etc) which affect the activity, In order to characterize the magnetic nanoparticles, we have used transmission electron microscopy(TEM), X-ray diffraction(XRD) and Fourier transform infrared(FTIR).

A Comparative Study on the Effects of Polygoni Radix and Cynanchi Radix on Rat Livers Intoxicated with Carbon Tetrachloride (백서(白鼠) 간조직(肝組織)에 미치는 적하수오(赤何首烏)와 백하수오(白何首烏)의 효능에 관한 비교(比較) 연구(硏究))

  • The Polygoni Radix and Cynanchi Radix have been used to potentiate the liver functions in clinic of Oriental Medicine. The water extracts of Polygoni Radix and Cynanchi Radix were administered orally to rats intoxicated with carbon tetrachloride and then this experiment have been performed by observing liver fatty degeneration and activities of enzymes such as cytochrome oxidase (CYO), adenosine triphosphatase (ATP), acid phosphatase (ACP), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP). By oral administration of water extracts of the radices between 1 and 10 days, the following results were obtained. 1. The group given Cynanchi Radix extract showed recovery of the fat liver in 4 days, whereas that given Polygoni Radix extract did the recovery in 8 days. 2. In cytochrome oxidase activity, the group given Cynanchi extract showed normal activity in 6 days, whereas that given Polygoni Radix extract did the activity in 8 days. 3. In adenosine triphosphatase activity, the groups given Cynanchi Radix and Polygoni Radix extracts showed normal activities in 2 and 8 days, respectively. 4. In acid phosphatase activity, the groups given Cynanchi Radix and Polygoni Radix extracts showed recovery of the activities in 2 and 4 days, respectively. 5. In lactate dehydrogenase activity, the group given Cynanchi Radix and Polygoni Radix extracts showed recovery of the activities in 6 and 10 days, respectively. 6. In alkaline phosphatase activity, the group given Cynanchi Radix extract showed normal activity in 2 days, whereas that given Polygoni Radix extract showed slight recovery between 4 and 6 days followed by decrease of the activity in 8 and 10 days. From the above-mentioned results, it was found that both of the water extracts of Polygoni and Cynanchi Radix possessed the recovery action of liver function as intoxicated with carbon tetrachloride in rats. It is also noted that the extract of Cynanchi Radix showed more potent activity than that of Polygoni Radix.

The change of ascorbate oxidase isozyme pattern during mycelial development of streptomyces lavendulae (Streptomyces lavendulae의 균사체 발달에 따른 ascorbate oxidase 동위효소 양상 변화)

  • pH decreased as the substrate mycelium developed, $\Delta$pH was 1.05-1.15, but increased after the aerial mycelium formation. The lactic acid content in culture solution showed no difference between 0.2% and 5% glucose, at which the aerial mycelium formation was repressed. The growth and development of mycelium was delayed by the lactate treatment. The activity of catalase was maximum in 24 hours after inoculation, and the wuperoxide dismutase activity showed a constant level during the developmental phases. The ascorbic acid accumulated after the aerial mycelium formation. The ascorbate oxidase isozyme of Rf 0.44 appeared, while the isozyme of Rf 0.36 desappeared during the development.

  • On-line monitoring technique for the concentration of lactate by a FIA(Flow Injection Analysis) system was studied. The lactate oxidase(LOD) was immboilized on VA-Epoxy carrier and integrated into the FIA system. The pH, buffer flow rate and temperature for the LOD-FIA were optimized, and the effects of salts and metabolites dissolved in the sample on the activity of immobilized enzyme were investigated. The LOD-FIA has been applied to monitor the concentrations of lactate in a simulated bioprocess. The on-line monitoring data by the LOD-FIA agreed with the off-line data measured by a fluorescence spectroscopy well.

Redistribution of Lactate Dehydrogenase Isozymes and Morphology of Tissues in Mus musculus after Irradiation (방사선 조사 후 생쥐(Mus musculus)조직의 형태와 젖산탈수소효소 동위효소의 재분포)

  • In order to identify the morphological changes of tissues in mouse after irradiation. We have observed the redistribution of LDH isozymes and the morphological changes of skeletal muscle, heart, kidney, liver and testis in mouse according to variation amount with the time after the 1 Gray and 3 Gray irradiation each. As a result of H-E (hematoxylin-eosin) stain, the apoptotic bodies were more easily observed in the liver than the other tissues and the quantity of the apoptotic bodies was proportionated to radiation amount. The number of apoptotic bodies was shown the highest at 1 day in most tissues and at 7 day in testis after irradiation. TUNEL (terminal deoxyribonucleodtidyl transferase-mediated dUTP-digoxigenin nick end labeling) staining was shown the same results as H-E staining. After the irradiation, the protein content was reduced in tissues except kidney. And protein content was reduced in all tissues at the initial period of 2 hours after 3 Gy irradiation. But it increased at 7 days after irradiation. LDH (EC 1.1.1.27, lactate dehydrogenase) activity was increased mostly in tissues at the early stage after 1 Gy irradiation. The maximum activity was detected earlier stage after 1 Gy irradiation than 3 Gy irradiation. The activity of LDH $A_4$ isozyme was decreased in the skeletal muscle, heart, kidney, and testis. The activity of $B_4$ and $A_2$$B_2$ sozyme was increased in the skeletal muscle and heart, and the activity of heterotetramer isozyme was increased in kidney The activity of $A_4$ isozyme in liver was detected high level and the activity of isozyme including subunit C elevated in testis. Therefore, LDH isozyme seems to play a role of lactate oxidase in most tissues except liver after irradiation. These data support that LDH isozyme is predomintly involved in the aerobic metabolism.

Changes of Activities and Isozymes of Lactate Dehydrogenase in Coreoperca herzi and Pseudogobio esocinus Acclimated to Rapid Increase of Dissolved Oxygen (급격한 용존산소량 증가에 순응한 꺽지(Coreoperca herzi)와 모래무지(Pseudogobioesocinus) 젖산탈수소효소 활성과 동위효소의 변화)

  • The metabolism of lactate dehydrogenase (EC 1.1.1.27, LDH) and $C_4$ isozyme were studied in tissues of Coreoperca herzi and Pseudogobio esocinus acclimated to rapid increase of dissolved oxygen (DO). In C. herzi the LDH activity was changed $35-39\%$ in brain and liver tissues, and within $20\%$ in other tissues. The $B_4$ isozyme was increased and isozyme containing subunit C was decreased in muscle tissue. The $B_4$ isozyme was increased in heart and kidney. In P. esocinus, the LDH activity in liver tissues was largely increased to $150\%$ for 30 minute and $70\%$ in other tissues. The $A_4$ isozyme was increased in muscle and $B_4$ isozyme was increased in other tissues. Especially, the metabolism of liver tissue in P. esocinus was regulated by increasing liver-specific $C_4$ and decreasing $A_4$ isozyme. But the metabolism of eye tissue in C. herzi was regulated by decreasing LDH activity and eye-specific $C_4$ isozyme. The LDH activity and LDH isozyme in P. esocinus were largely increased than C. herzi acclimated to rapid increase of DO. And eye-specific $C_4$ and liver-specific $C_4$ isozymes played role as lactate oxidase. Therefore, the response of species acclimated to rapid increase of DO seems to be variable, perhaps due to prior exposure to environmental conditions.

Effects of Myrrha Water Extract on Rat Myocardial Cells in Cultures (몰약 전탕액이 배양 심근세포에 미치는 영향)

  • Objectives and Methods : In order to elucidate toxic mechanism of myocardial damage and protective effect of myrrha water extract against cytotoxic effect of xanthine oxidase/hypoxanthine(XO/HX), cardioprotective effect of myrrha water extract was examined by MTT assay, LDH (Lactate Dehydrogenase) activity and heart beating rate after cultured myocardial cells derived from neonatal mouse were treated with various concentration of XO/HX, a free radical. Results : XO/HX induced a decrease of cell viability, an increase in the amount of LDH, and a decrease of heart beating rate on cultured myocardial cells in a dose-dependent manner. In cardioprotective effect of myrrha water extract, it showed a decrease in the amount of LDH and an increase of heart beating rate on cultured myocardial cells damaged by XO/HX. Conclusions : From the above results, it is suggested that XO/HX showed toxic effect in cultured myocardial cells derived from neonatal mouse and that myrrha water extract is very effective in the prevention of XO/HX-induced cardiotoxicity.

The Effects of Creatine Oral Delivery on the Muscular Activity and Blood Lactate Density of Rowing Athletes (크레아틴 구강투여가 조정선수들의 근활성도와 혈중젖산농도에 미치는 영향)

  • The purpose of this study is to investigate how effects creatine dosage has on the improvement of rowing athletes' performance ability. Rowing athletes were administered with creatine, through which to examine the change of athletic performance ability, blood fatigue substances, and muscular activity. The subjects (participants) of this Study consisted of 12 male rowing athletes at P University, with at least 5 years of rowing experiences, which divided into two groups - creatine dosing group of 6 persons and control group of 6 persons - for random sampling measurement. Enzymatic-colorimetric method using lacrate oxidase and 4-aminoantipyrine was performed for blood lactate level analysis, and wireless EMG system (QEMG-4: Lxtha Korea) for muscular activity analysis, with 4 channels set for data analysis. As body parts to be measured, two muscular parts - latissimus dorsi and lumbar spinel - were chosen. Then, on the 5th day from the date of administering them with creatine (that is, 4 days after dosing them with creatine), rowing movement with the highest level of activity was calculated as peak value, which was measured twice. The test data used for this Study were SPSS/PC 18.0, pre-movement and post-movement two-way ANOVA for repeated measurement for comparative analysis of each muscle, with significant level at .05. As a result, the change of blood lactate level was significantly higher in creatine dosing group than in non-dosing group (p<.05). As for the change of muscular activity, both latissimus dorsi and lumbar spinel showed a significantly higher change of muscle in creatine dosing group than in non-dosing group (p<.05 and p<.05, respectively).

Effects of Sophorae Radix Extract in Rat Cardiac Endothelial Cells (고삼 추출물이 배양 심장내피세포에 미치는 영향)

  • To test the protective effect of Sophorae Radix (SR) on the damage of cardiac endothelial cells by xanthine oxidase (XO)/hypoxanthine (HX)-induced oxygen tree radical, Neutral Red (NR), lactate dyhydrogenase (LDH), and c-fos immunopositive cells assay were used in the presence of SR extract. The results of these experiments were obtained as follows ; Cardiac endothelial cells treated with XO/HX showed the cytotoxicity such as a decrease in viability, and increases in LDH activity and c-fos immunopositive cells. Cardiac endothelial cells pretreated with SR extract protected the increase of LDH activity. Alos, cardiac endothelial cells pretreated with SR extract inhibited the increase of c-fos immunopositive cells. These results show that XO/HX induces toxic effects in cultured cardiac endothelial cells derived from neonatal rat, and suggest that SR extract is very effective in the prevention of XO/HX-induced toxicity.
