  • The preparation of the fine BN powders by ammonia-nitridation of ammonia-decaborane derivate was attempted at temperature between 300 to 150$0^{\circ}C$. The formation mechanism of BN was examined and the resulant BN powder was characterized by means of IR, XRD, SEM and PSA method. In the nitridation below 80$0^{\circ}C$ bonding materials were identified with mainly BH and NH but above 80$0^{\circ}C$ with BN by IR spectra and X-ray patterns. Crystalite size, lattice constant and particle size distribution of hexagonal BN prepared at 1, 50$0^{\circ}C$ were La=470$\AA$, Lc=180$\AA$, a=2.5062$\pm$1$\AA$, c=6.8285$\pm$2$\AA$ and 2.0-5.4${\mu}{\textrm}{m}$, respectively.

  • Polycrystalline $Sr_{2}Nb_{2}O_{7}$ and $La_{2}Ti_{2}O_{7}$ ceramics with very high Curie temperatures were synthesized by the chemical coprecipitation method (CCP). The powders synthesized were identified by XRD and their sintering behavior and physical properties were studied. The grain-orientation and electrical properties of sintered ceramics were investigated as a function of firing temperature. Single phase could be obtained by CCP method at temperature lower than that of the conventional method by 100 - $150^{\circ}C$. Strontium niobate, $Sr_{2}Nb_{2}O_{7}$, powder was Prepared by CCP method at temperatures as low as $800^{\circ}C$ via intermediate phase of $Sr_{5}Nb_{4}O_{15}$ formed at $700^{\circ}C$. The resulting CCP-derived powder was observed to have finer and more uniform particle size distribution than those obtained through the conventional or the molten salt synthesis method. Sintering of CCP-derived $Sr_{2}Nb_{2}O_{7}$ powder at $1500^{\circ}C$ yielded a highly dense ceramics with 97% theoretical density. Very high grain-orientation developed along the (0k0) direction was observed by SEM, which resulted in anisotropic dielectric properties of the sintered samples, with the dielectric constant values approaching those for single crystal.

  • Polycrystalline $Sr_{2}(Ta_{1-x}Nbx)_{2}O_{7}$ and $La_{2}Ti_{2}O_{7}$ ceramics having very high Curie temperatures were synthesized by the conventional oxide mixing method (CON) and the molten salt synthesis method (MSS). Physical characteristics and phase relationship of calcined powders, as well as the sintering behavior, the grain-orientation and dielectric properties of sintered ceramics were investigated as a function of composition and firing temperature. The single $A_{2}B_{2}O_{7}$ phase was synthesized by using the MSS method at 100 - $150^{\circ}C$ lower temperature compared to the CON method. As Nb content increased in $Sr_{2}(Ta_{1-x}Nbx)_{2}O_{7}$ ceramics, the Curie temperature and the dielectric constant at Curie temperature were raised, and the sintering behavior and the degree of grain-orientation were also improved at the same time. The use of MSS-derived powders made it possible to lower the sintering temperature and to improve the dielectric properties of the sintering samples. However, the piezoelectric properties as well as the grain-orientation were not improved any further by the MSS route.

Optimization of Culture Conditions for Xylitol Production by A Mutant of Candida parapsilosis (Candida parapsilosis 돌연변이주에 의한 Xylitol 생산조건의 최적화)

  • Effect of culture conditions such as pH, temperature, agitation speed and oxygen transfer rate on xylitol production from xylose by Candide parapsilosis ATCC 21019 mutant was investigated in a jar fermentor. The initial concentration of xylosr was fixed at 50 g/l in this experiment. When pH was increased, cell growth and xylose consumption rate were increased, but maximum xylitol production was shown in the range of pH 4.5 and 5.5 with a yield of 0.68 g/g-xylose. The optimal temperature for xylitol production was determined to be $30^{\circ}C$. Considering the importance of dissolved oxygen tension, for xylitol production, the effect of oxygen transfer rate coefficient $(k_La)$ on fermentation parameters was carefully evaluated in the range of $20{\sim}85\;hr{-1}\;of\;k_La$ (corresponding to $100{\sim}300$rpm of agitation speed). The xylitol production was maximized at $30\;hr^{-1}\;of\;k_La$(150 rpm). A higher oxygen transfer rate supported better cell growth with lower xylitol yield. It was determined that maximum xylitol concentration, xylitol yield and productivity was 35.8 g/l, 71.6% and $0.58\;g/l{\sim}hr^{-1}$, respectively, at $30\;hr^{-1}\;of\;k_La$ In order to further increase xylitol productivity, ferementation using the concentrated biomass(20 g/l) was carried out at the conditions of pH 4.5, $30^{\circ}C$ and $30\;hr\;1$ of oxygen transfer rate. The final xylitol concentration of 40 g/l was obtained at 18 hours of culture time. From this result, it was calculated that xylitol yield was 80ft on the basis of xylose consumption and volumetric productivity was $2.22\;g/l{\sim}hr$ which was increased by $3{\sim}4$ fold compared with $0.5{\sim}0.7\;g/l-hr$ obtained in a normal fermentation condition.

  • The purpose of this study was to investigate the effect of exercise and/or L-arginine on abdominal fat, IGF-1 on GH/IGF-1 axis, fibrinogen, and PAI-1 in aged and obese rats. Male Sprague-Dawley rats were treated with a D-galactose aging inducing agent (50 mg/kg) given intraperitoneally for 12 weeks. Thirty-two male Sprague-Dawley rats were treated and divided into four groups: aging-high fat diet group (AG+HF), AG+HF with L-arginine intake group (AG+LA), AG+HF with exercise group (AG+EX), and AG+EX with L-arginine intake group (AG+LA+EX). The experimental rats underwent treadmill training (60 min/day, 6 days/week at 0% gradient) for 12 weeks. L-arginine was given orally (150 mg/kg/day) for 12 weeks. After the experiment, blood was collected from the left ventricle and abdominal fat was extracted. The results showed that GH was significantly increased in AG+EX and AG+AL+EX. IGF-1 was significantly increased in both the AG+AL+EX and AG+EX group ($p$<0.05), while fibrinogen and PAI-1 were not significantly different among the groups. Abdominal fat was significantly decreased in the AG+LA, AG+EX, and AG+LA+EX groups ($p$<0.05) compared with the AG+HF group. In conclusion, this study suggests that exercise alone or L-arginine alone or a combination not only increases the GH and IGF-1 concentration, but also decreases the abdominal fat mass.

Cytotoxicity and Antigenotoxic Effects of Cordyceps militaris Extracts (번데기동충하조(Cordyceps militaris) 추출물의 세포독성 및 유전독성 억제효과)

  • This study was performed to investigated the effects on the cytotoxicity and antigenotoxicity of Cordyceps militaris extracts on the human cancer cell lines. The ethanol extract and five fractions which were hexane, chloroform, ethylacetate, butanol and aqueous were screened for crytotoxicity on human lung carcinoma(A549). human breast adenocarcinoma (MCF-7) human epitheloid carcinoma(HeLa), human fibrosarcoma(HT1080) human hepatocellular carcinoma(Hep3B), human gastric carcinoma(KATOIII) and chronic myelogenous leukemia(K562) cell by SRB and MTT assays. The results showed that growth inhibition rates of the human cancer cell in the presence of Cordyceps militaris were inhibited with increasing concentration of the extract. The ethanol extract from Cordyceps militaris had strong inhibitory effects in1 mg/mL treatment by SRB assay , showing 89.4%, 85.7%, 72.9% and 65.5% inhibition in HT1080, HeLa, Hep3B and A549, respectively. The treatment of 1 mg/mL hexane fraction by SRB assay had the strongest cytotoxicity with 97.0% on HT1080 followed by MCF-7(92.9%) and HeLA(90.3%). The inhibition ration on KATOIII by MTT assay was much higher in the butanol (83.7%) and aqueous (80.4%) than in the ethanol extract (61.5%) And also, K562 showed similar tendency with KATOIII. The effects of Cordyceps militaris extracts on the frequencies of micronucleated polychromatic erythrocytes (MNPCEs) induced by N-methyl-N-nitro-N-nitrosoguanidime(MNNG) were investigated in the bone-marrow cells of ICR male mice. The amount of 10, 20, 40 and 80 mg/kg of each extract were administered to animals immediately after injection of MNNG, and the exposure time was 36 hours. Significant reductions(p<0.05) with 39.7%, 52.7%, 71.4% and 83.9% were observed in the frequencies of MNPCE when 10, 20, 40 and 80 mg/kg of the hexane fraction of Coryceps militarus extracts were given to the mice.

  • Background : Thyroid Transcription Factor-1(TTF-1) acts as a tissue specific transcription factor in the regulation of lung specific gene expression and as morphogenic protein during lung organogenesis. Currently, there is very little information on the cis-acting sequences and transcription factors that direct the TTF-1 gene expression. DNAse 1 hypersensitive (DH) sites represent a marker for active or potentially active chromatin and are likely to be especially important in gene regulation, being associated with many DNA sequences that regulate gene expression. It is clear that DH regions correlate with genetic regulatory loci and binding for sequence-specific DNA-binding proteins. Methods : We have used DH site assays to identify putative distal regulatory elements in H441 lung adenocarcinoma cells, which express the TTF-1 gene and HeLa cells. Results : There are four DH sites 5' of the TTF-1 gene. These sites are located at base pair approximately +150, -450, -800, and -1500 from the start of transcription. Conclusion : These data suggest that there may be at least one intragenic site and regulatory region 5' prime to the promotor region.

  • The compositional and structural properties of alkali metal ion exchanged Y-zeolites have been investigated by la number of analytical techniques and their catalytic activities were tested for NOx reduction in combination with a non-thermal plasma. The NOx conversion data for LiY, NaY, KY and CsY were measured by chemiluminiscent NOx meter in the temperature range of 100 to $350^{\circ}C$. The initial activities of the catalyst at $150^{\circ}C$ increased in the order LiY < KY < NaY < CsY in alkali series. The activity of CsY and NaY were increased and showed maximum at $200^{\circ}C$ and then decreased in the plasma reactor, as the temperature increased. The activity of KY maintained same by $200^{\circ}C$ and then decreased, whereas the activity of LiY decreased with the increasing temperature. The CsY catalyst, which showed the highest activity in alkali metal series, exhibits a NOx conversion efficiency of $80\%$ between $170{\~}220^{\circ}C$.

  • Casein is a milk protein and one of the most important nutrients in milk. The composition is over 80% in cow's milk and about 20~45% in human's milk. Casein is highly biocompatible and biodegradable, so it has been studied for various biomedical materials applications as well as drug delivery systems. It is widely known that casein can be prepared as nanoparticles in the presence of the Ca2+ metal ion. Because casein is amphiphilic, hydrophobic drugs could be loaded inside to form a protein-based drug delivery system. In this study, we studied the optimum conditions for casein nanoparticle formation using natural metal ions present in the body, such as calcium, magnesium, zinc, and iron. It was confirmed that nanoparticles have a uniform size of around 150 nm and negative zeta potential values. In addition, it was demonstrated that casein nanoparticles have a cell viability of more than 80% and efficient intracellular uptake properties using confocal microscopy. From the results, it was also shown that the casein nanoparticles prepared using various metal ions have the potential to be biocompatible drug delivery carriers.
