Plant Community Structure of Chuwang Valley in Chuwangsan National Park (주왕산국립공원 주왕계곡의 식물군집구조)

  • To investigate the forest structure of Chuwang valley of Chuwangsan National Park, thirty plots were set up and surveyed. Importance values, DBH class distribution, species diversity indices, DCA Ordination, CCA ordiantion and TWINSPAN classification were used for vegetational structure analysis. Pinus densiflora Quercus serata, Q. mongolica, Q. variabilis were appeared to be dominant species in thirty plots. According to the analysis of classification by TWINSPAN, the thirty plots divided four groups. Groups were Q. mongolica-P. densiflora-Carpinus laxiflora community(I), P. densiflora-Q. variabilis community(II), Q. serrata-Q. variabilis community(III), broad-leaved mixed community(IV) Species diversity(H') of investigated area was calculated 1.17~l.32. The successional trend was seemed to be from P. densiflora to Q. spp. in the canopy layer.

Species Diversity and Seasonal Changes of Dominant Ulva Species (Ulvales, Ulvophyceae) in Mikawa Bay, Japan, Deduced from ITS2 rDNA Region Sequences

  • Frequent occurrences of green tides caused by Ulva species (Ulvales, Ulvophyceae) associated with eutrophication along enclosed coasts are currently causing environmental problems in coastal ecosystems. In addition, increasing intercontinental introductions of coastal marine organisms, including Ulva, are also a serious issue. However, due to the considerable morphological plasticity of this genus, the taxonomy of Ulva species based on morphological studies is problematic. Therefore, in order to elucidate the species diversity and seasonal changes of the dominant Ulva species in Mikawa Bay, central Honshu, Japan, we made seasonal collections of Ulva species at seven localities, and identified the dominant species using the ITS2 rDNA region sequences. We identified the following nine taxa as common Ulva species in the area: 1) Ulva pertusa Kjellman; 2) U. ohnoi Hiraoka et Shimada; 3) U. linza L.; 4) U. californica Wille; 5) U. flexuosa Wulfen; 6) U. fasciata Delile; 7) U. compressa L.; 8) U. armoricana Dion et al.; 9) U. scandinavica Bliding. Among the species, U. pertusa was most common and dominant from spring to summer, and U. ohnoi from autumn to winter. Ulva californica and U. scandinavica have not been reported before from Japan.

  • Physicochemical properties of soil, mineral cyclings, production of plants, and relationship between sodium(Na) content and progresses of plant communities were studied in a coastal salt marsh in Incheon. Contents of Na, available phosphorus(A-P) and value of electric conductivity of soil decreased in order of Salicornia herbacea, Limonium tetragonum, Phragmites communis and Zoysia sinica communities, but contents of organic matter, total nitrogen(T-N) and calcium(Ca) of soil were vice versa. Specise diversity index decreased with increase of Na content of soil with correlation coefficient of -0.82. The aboveground biomass of plant communities were 2,981 g.dw/$m^2$ in P. communis, 1,471 g.dw/$m^2$ in Z. sinica, 189g.dw/$m^2$ in S. herbacea and 71 g.dw/$m^2$ L. tetragonum, respectively. Seasonal changes of contents of inorganic nutrients per unit land area coincided with those of biomass of plant communities, however, the maximum contents of K occured earlier than the maximum biomass. Amounts of inorganic nutrients absorbed by plant were directly proportion to its biomass and it was true to reverse in restored amounts of them to soil. In turnover times of nutrients of the communities, it took the shortest time for P but the longest for Ca and P. communis community took the shortest but L. tetragonum the longest. For example, in P. communis turover time of P took one year and that of Na 1,440 years. Lack of P element, therefore, was expected in this study area.

Bacterial Regrowth in Water Distribution Systems and Its Relationship to the Water Quality: Case Study of Two Distribution Systems in Korea

  • This study was done to observe the occurrence of heterotrophic bacteria in terms of free chlorine residuals in two different water distribution systems, which belongs to both K and Y water treatment plant of S city in Korea. The data analyzed in the distribution systems show that the free chlorine residuals decreased from 0.10 to 0.56 mg/l for K, and 0.51 to 0.78 mg/l for Y. The decay of free chlorine is clearly higher in both March and August than in January. The HPC in the distribution systems are ranged from 0 to 40 cfu/ml for K, 0 to 270 cfu/ml for Y, on $R_2$A medium. In particular, its level is relatively high at the consumer's ground storage tanks, taps, and the point-of-end area of Y. The predominant genera that were studied in the distribution systems were Acinetobacter, Sphingomonas (branch of Pseudomonas), Micrococcus, Bacillus, Staphylococcus. The diversity of heterotrophic bacteria increases in the end-point area. Most of them are either encapsulated cells or of Gram-positve cocci. In conclusion, the point-of-end area in distribution systems shows the longer flow distance from the water treatment plants, along with a greater diversity and a higher level of heterotrophic bacteria, due to the significant decay of free chlorine residuals.

The complete chloroplast genome of Scrophularia kakudensis and a comparative analysis of S. kakudensis and S. cephalantha

  • The genus Scrophularia L. (Scrophulariaceae) comprises 200-270 species worldwide and is a taxonomically challenging lineage, displaying morphological diversity and hybridization. S. kakudensis is morphologically similar to the closely related taxa S. kakudensis var. microphylla, S. pilosa, and S. cephalantha. Therefore, the purpose of this study was to sequence the chloroplast (cp) genome of S. kakudensis using next-generation sequencing and compare it to those of related taxa. The complete cp genome sequence of Scrophularia kakudensis was found to be 152,355 bp long, consisting of a pair of inverted repeats of 25,485 bp that separate a large single-copy (LSC) of 83,479 bp from small single-copy regions of 17,909 bp. The cp genome contained 78 protein-coding genes, 30 tRNAs, and four rRNAs. A phylogenetic analysis based on 78 protein-coding genes from six Scrophularia species showed S. kakudensis and S. cephalantha formed with 100% bootstrap values. We compared the complete cp genomes of S. kakudensis and S. cephalantha and identified seven sequence divergence regions: matK/rps16, rps16/trnQ, trnS/trnG, rpoB/trnC, trnS/trnG, rpl32/trnL, and ndhD/psaC. These regions may be useful for determining the phylogenetic relationships among S. kakudensis-related species.

In vitro micropropagation of M.26 (Malus pumila Mill) apple rootstock and assessment of the genetic diversity of proliferated plantlets using simple sequence repeat markers (사과 대목 M.26 (Malus pumila Mill)의 기내 대량번식 및 simple sequence repeat 마커를 이용한 증식된 식물체의 유전적 다양성 평가)

  • The objective of this study was to determine the most effective medium condition of shoot proliferation and root formation for the efficient in vitro micropropagation of M.26 (Malus pumila Mill). Simple sequence repeat (SSR) markers were used to analyze the genetic diversity of micro-propagated and greenhouse grown M.26. Shoot proliferation was carried out in MS (Murashige and Skoog) containing benzyladenin (BA, $0.5{\sim}5.0mg{\cdot}L^{-1}$) and thidiazuron (TDZ, $0.01{\sim}0.1mg{\cdot}L^{-1}$). The highest number of shoots (10.67 shoots per explant) was induced by adding BA at a concentration $1.0mg{\cdot}L^{-1}$. TDZ treatments caused higher hyperhydricity rate in cultured explants than in BA treatments. There was no significant effect of both BA and auxin on shoot proliferation, and the optimum proliferation medium for M.26 was MS medium containing $1.0mg{\cdot}L^{-1}$ BA. To find a suitable medium composition for shoot rooting, we tested different concentrations indole-3-butyric acid (IBA) and ${\alpha}$-naphthaleneacetic acid ($0.5{\sim}5.0mg{\cdot}L^{-1}$), MS medium (1/4-1), sucrose ($0{\sim}30g{\cdot}L^{-1}$). The shoots showed good rooting on half-strength MS medium containing $1.0mg{\cdot}L^{-1}$ IBA and $15-20g{\cdot}L^{-1}$ sucrose. The rooting rate (100%), number of roots (10.45 ~ 13.60 roots per explant), root length (7.41 ~ 8.33 cm), and shoot length (4.93 ~ 5.38 cm) were good on this medium. Fifteen SSR primers were detected in a total of 30 alleles in 20 micro-propagated plantlets, all SSR profiles from micro-propagated plantlets were monomorphic and similar to greenhouse grown control plantlet M.26 plant. The results indicated that M.26 micro-propagated plantlets were genetically stable.

Analysis of genetic divergence according to each mitochondrial DNA region of Haliotis discus hannai (북방전복 (Haliotis discus hannai) 의 mitochondrial DNA 영역별 유전적 변이성 분석)

  • The seven mitochondrial DNA regions (ND2, ND5, ND4, ND4L, ND6, ND1 and 12SrRNA) of Haliotis discus hannai were examined to estimate the availability as a genetic marker for the study of population genetic. The region with the highest genetic variation was ND4 (Haplotype diversity = 1.0000, Nucleotide diversity = 0.0108). On the other hand, ND2 and ND1 regions have significantly appeared genetic divergence between clusters (divergence of 90% and 87%). Also, pairwise $F_{ST}$ between clusters within ND2 and ND1 regions showed high values; 0.4061 (P = 0.0000), 0.4805 (P = 0.0000) respectively. Therefore we can infer that it is the most efficient and accurate way to analyze the region of ND4 with the highest variation in addition to the regions of ND2 and ND1, which formed clusters with high bootstrap value, for study of population genetic structure in this species.

Taxonomic Study of the Subfamily Lycorininae (Hymenoptera: Ichneumonidae) from Korea

  • Korean species of the subfamily Lycorininae Cushman & Rohwer are reviewed. This subfamily along with three species, Lycorina triangulifera Holmgren, 1859, L. spilonotae Chao, 1980, and L. ruficornis Kasparyan, 2007, are reported for the first time from Korea. In this genus, five species have been reported from the Eastern Palaearctic region, six species from China and two species from Japan. Some species of this subfamily is known as parasitoids of Crambidae and Tortricidae. We report three newly recorded species from Korea, Lycorina triangulifera, L. spilonotae, and L. ruficornis, and provide diagnoses with photographs of lycorinine species and a key to the Korean species.

  • In September 1990, the species of Thecamoebae (Rhizopodea ; Protozoa) inhabited in Sphagnum at Mt. Taekwanryng were collected . The six species of Thecamoebae belonged to three families were identified : Arcella bathystoma, Trigonopyxis arcular , Heleopera sphangi, H. petricola, Lesquereusia epistoma, L.spiralis, Except L.Spiralis, five species were newly reported in Korea. They were described and figured with photograph.

Spatio-temporal Variability of Phytoplankton Community in the Jeju Marine Ranching Area (JMRA) (제주 바다목장 해역 식물플랑크톤 군집의 시·공간적 변동 특성)

  • This study describes the spatio-temporal distributions in phytoplankton community such as species composition, standing crops and dominant species from April to November 2008 in the Jeju Marine Ranching Area (JMRA). A total of 106 species of phytoplankton belonging to 63 genera was identified. In particular, diatoms and dinoflagellates were occupied more than 61% and 35% of total species, respectively. The annual dominant species were Skeletonema costatum-like species(ls) in April, Torodinium teredo, Cylindrotheca closterium, Scrippsiella trochoidea in June, S. costatum - ls, Thalassionema nitzschioides, Pseudo-nitzschia pungens, Ebria tripartita in September and Corethron pennatum, Dictyocha fibura, Neoceratium teres in November. Phytoplankton cell density ranged between $0.6cells{\cdot}mL^{-1}$ in June and $64.0cells{\cdot}mL^{-1}$ in April. It fluctuated with an annual mean of $11.7cells{\cdot}mL^{-1}$ between the lowest value of $0.9cells{\cdot}mL^{-1}$ in November and the highest value of $37.0cells{\cdot}mL^{-1}$ by S. costatum -ls in April. Diversity index in September was higher than diatom blooming seasons in April. According to the phytoplankton community structure, the biological oceanographic characteristics of the JMRA was characterized by nanoplankton during a year. That is, primary production is deemed to have a higher possibility of being adjusted by a reproduction by material cycle in the ecosystems than nutrients supply from the lands.
