Development and Reproduction of Eucyclops serrulatus(Copepoda: Cyclopoida) in the Laboratory Culture (실험실에서 배양한 Eucyclops serrulatus(검물벼룩목 요각류)의 생식과 성장에 관한 연구)

  • Four cyclopoids were tested for laboratory culture. Among these animals, only Eucyclops serrulatus was successfully cultured in the laboratory. Under the laboratory culture condition, nine kinds of diets were tested for the suitability of nauplius development. Development and reproduction of E. serrulatus were also investigated. Chlamydomonas reinhardii was the only one which could induce complete development from nauplii to adults. It was found that E. serrulatus had relatively short generation time, could produce lots of progenies, and could be handled easily, allowing to obtain many individuals during a short period. With these characteristics, this species may be a candidate for a good test organism for evaluating freshwater pollution.

  • In this study, combustion toxicity evaluation for building interior materials and study for toxicity as using FT-IR analysis. this experiment for the calculation of toxicity index, it using a cone calorimeter model in KS F ISO/TR 9122-4 as a fire model. It is following ISO 19702 procedure for assessing fire toxic gas using FT-IR. This experiment used calculation method for toxicity index (FED) among the international standards. $LC_{50}$ is a concentration that it can cause death to 50 % of experimental animal in 30 minutes - exposure gas test. comparison with the three kinds of toxicity fire gas of construction materials using toxicity index.

Prediction of Chemical Organic Composition of Manure by Near Infrared Reflectance Spectroscopy

  • The organic materials included in excreta of livestock are important resources for organic manure and for improving soil quality, although there is still far from effective using. One reason for this is still unclearly standard of quality for evaluation of manure made from excreta of livestock. Therefore, the objective of this study is to develop rapid and accurate analytical method for analyzing organic compositions of manure made from excreta of livestock, and to establish quality evaluation method based on the compositions predicted by near infrared reflectance spectroscopy (NIRS). Sixteen samples of manure, each eight samples prepared from two treatments, were used in this study. The manure samples were prepared by mixing 560 kg feces of swine,60 kg sawdust with moisture content was adjusted to be 65%. The mixture was then keep under two kinds of shelter, black and clear sheets, as a treatment on the effect of sunlight. Samples were taken in every week (form week-0 to 7) during the process of manure making. Samples were analyzed to determine neutral detergent fiber (NDF), acid detergent fiber (ADF) and acid detergent lignin (ADL) by detergent methods, and organic cell wall (OCW) and fibrous content of low digestibility in OCW (Ob) by enzymatic methods. Biological oxygen demand (BOD) was analyzed by coulometric respirometer method. These compositions were carbohydrateds and lignin that were hardly digested. Spectra of samples were scanned by NIR instrument model 6500 (Pacific Scientific) and read over the range of wavelength between 400 and 2500nm. Calibration equations were developed using eight manure samples collected from black sheet shelter, while prediction was conducted to the other eight samples from clear sheet shelter. Accuracy of NTRS prediction was evaluated by correlation coefficients (r), standard error of prediction (SEP) and ration of standard deviation of reference data in prediction sample set to SEP (RPD). The r, SEP and RPD value of forage were 0.99, 0.69 and 7.6 for ADL, 0.96, 1.03 and 4.1 for NDF, 0.98, 0.60 and 4.9 for ADF, 0.92, 1.24 and 2.6 for Ob, and 0.91, 1.02 and 7.3 for BOD, respectively. The results indicated that NIRS could be used to measure the organic composition of forage used in manure samples.

Effects of Replacing Corn with Brown Rice or Brown Rice with Enzyme on Growth Performance and Nutrient Digestibility in Growing Pigs

  • A 4 week experiment was designed to study the effects of replacing corn with Chinese brown rice or adding different amylase in brown rice basal diet on growth performance and apparent fecal digestibilities of nutrients in growing pigs. One hundred and eight cross-bred pigs (Duroc${\times}$Landrace${\times}$Large White), weighing an average of $18.35{\pm}0.12kg$, were randomly assigned to 6 treatments with 6 replications per treatment. Diet in treatment 1 was corn-soybean meal basal diet, and in treatment 2, 3 and 4, corn was replaced by brown rice on rates of 33.3%, 66.7% and 100% respectively on the basis of treatment 1. And diets in treatment 5 and 6 were similar to treatment 4 except two kinds of amylases, glucoamylase and ${\alpha}$-amylase, were added respectively. The brown rice used in this experiment was husked from one kind of early, long grain, non-glutinous rice (ELGNR, indica rice) in southern China. The results indicated that there was a slight improvement in growth performance of pigs in brown rice treatments (p>0.05). The blood urea nitrogen value in treatment 2 was lower than that in treatment 1 (p<0.05). The differences of apparent fecal digestibilities of most nutrients were significant (p<0.05) except CP. Digestibilities of GE, OM and DM in treatment 4 were the best and digestibility of crude fat in treatment 5 appeared best (p<0.05). Contrast results between treatment 1 and treatment 2 to 4 indicated that the digestibility of GE, OM and DM increased significantly with the replacing rates of brown rice (p<0.05). Contrast results between treatment 4 and 5 indicated that adding glucoamylase in brown rice diet increased growth performance slightly (p>0.05) but not for digestibilities. This experiment shows a positive effect of brown rice on growth performance, especially on nutrient digestibility.

The Nutritive Values and Manufacture of Total Mixed Fermentation Feeds using Green Forage Crops and RiCE-straw (청예 사료작물과 볏짚을 이용한 완전배합발효사료의 제조와 영양적가치)

  • Adequate forage amounts in ruminant animal are necessary for proper ruminal function in dairy cow. This study was carried out to determine the effect of total mixed fermentation feeds made by different green forage crops and grain processings on chemical compositions, RFV (relative feed value) and ruminal characteristics in sheep. The experiment was arranged in a split plot design with 4 replications. The main plot consisted of 6 kinds of green forages (corn, grass, rye, rape, alfalfa and oat) and the sub plots three different grain processings such as non-milling, half milling (7mm mesh over), and regular milling (7mm mesh below). And the different TMFFs (total mixed fermentation feeds) were analyzed for chemical composition and fed to 8 ruminally fistulated sheep for ruminal charactics and palatability. RFV, daily feed intake, acetate/propionate ratio of the rape-TMFFs were higher compared with the other treatment. Ruminal content of VFA (volatile fatty acid) of corn-TMFF was highest as 90.19 mmol% and pH of the feed was lowest as 3.82. But, acetic acid, propionic acid and butyric acid were no difference among treatments. In conclusion, the effect of grain proceeding was not appeared but if consider of only RFV, palatability and dry matter disappearance, grade of TMFF was improved in order of rape-, corn-, alfalfa-, grass-, oat- and rye-TMFF.

Effect of Medium and Soil Conditions on Propagation of Gametophyte and Sporophyte in Leptogramma pozoi (Lag.) Ching subsp. mollissima (Fisch. ex Kunze) Nakaike (진퍼리고사리의 전엽체 및 포자체 증식에 영향을 미치는 배지 조성과 배양토의 종류)

  • This study was carried out to select the optimal condition for prothallus propagation and sporophyte formation of Leptogramma pozoi (Lag.) Ching subsp. mollissima (Fisch. ex Kunze) Nakaike and to provide basic data for mass production system. In the propagation, 300 mg of prothallus were inoculated in different kinds of medium [1/4, 1/2, Murashige and Skoog (MS), Knop], sucrose (0, 1, 2, 3, 4%), and cultured for 8 weeks. Sporophyte formation studies were carried out by inoculating blended prothallus into artificial soils. The soils were mixed with horticultural substrate, peatmoss, perlite, and decomposed granite at different ratios or only with the horticultural substrate. Then the prothallus was cultivated for 12 weeks to figure out the formation and growth of the sporophytes. The growth and development of prothalli were excellent on MS medium. According to medium components, prothallus growth was favorable in all treatments except 0% sucrose treatment and the highest in 2% sucrose. In the experiment of soil mixtures, sporophyte formation was the highest in the horticultural substrate:perlite 2:1 (v:v) and horticultural substrate:decomposed granite 2:1 (v:v) treatment, and the overall growth was good in the horticultural substrate:decomposed granite 2:1 (v:v) mixed soil.

Genetic Variation and Relationships of Korean Cattle(Hanwoo) and Foreign Breeds Using Microsatellite Markers (초위성체 유전표지를 이용한 한우와 외래품종간의 유전적 변이와 유연관계 분석)

  • The purpose of this study was to assess the genetic variation and establish the relationship amongst Hanwoo, Angus and Holstein breed. The genetic characteristics and variability within Hanwoo(300), Angus(80) and Holstein(50) were estimated on the basis of relationships determined using the 10 kinds of microsatellite, which is located on different chromosomes. Frequencies of microsatellites markers were used to estimate heterozygosities, polymorphic information content(PIC) and genetic distances. The PICs ranged from 0.604 to 0.872(Hanwoo), from 0.562 to 0.812(Angus) and from 0.471 to 0.828(Holstein). Observed heterozygosity and PIC of Hanwoo are the highest among the analyzed breeds. Additionally, Estimates of genetic distance can be utilized to identify genetic relationships between Hanwoo and the other breed. Genetic distances(0.233) between Hanwoo and Angus was lower than distances between Hanwoo and Holstein(2.283). Also, Genetic distances between Angus and Holstein was shown for(2,400). The other side, each individuals were not ramified to different group and were spread evenly in phylogenetic dendrogram about all the populations.

A Study of the Additional Toxicity of Mixed Food Additives to Rat (혼합(混合)된 식품첨가물(食品添加物)이 흰쥐의 생리(生理)에 미치는 상승적(相乘的) 독성(毒性) 효과(效果)에 관(關)한 연구(硏究))

  • To improve the food qualities in Korea, two hundred and fourtynine kinds of food additives have been allowed in food processing, of which one hundred and nineteen kinds could be used under the limitted conditions. Hence, in practical uses in food processing, many kinds of them are mixed at random within the permitted amounts for their special purposes. For last several years, many kinds of the food additives were prohibited because they have been proved to be toxic even with the single dose. Until recently a few studies on the toxicity in the mixture of food additives were reported, however, they were shown to be no severe additional effect on the animal. This study was performed to see if any elevation of chronic or subacute toxicity of food additives occur especially when they are mixed with each other, using three kinds of food additives (DHA, AF-2, BHT) most widely used as food preservatives, antiseptics and antioxidants. One hundred and fifty young male rats were taken and divided into ten feeding groups, one first control group (food additives blank), three second control groups (DHA 0.1%, AF-2 0.1%, BHT 0.5%), three mixture groups of low level (mixture of each 60% of two second control level) and three mixture groups of high level (mixture of each 90% of two second control level). As the methods of biological and clinical tests, the change of body weight (growth rate), daily intake of diets, organ to body weight ratio, histopathological findings of organs, hematological observation, liver and kidney function tests were checked three times during the periods of 24 weeks. The following results were obtained. 1. The low level group of DHA, AF-2 mixture and DHA, BHT mixture revealed a little retardation in growth rate than the first control group, however, they were similar to the second controls, while all the mixture groups of high level showed a more remarkable retardation than the first and second controls. 2. Average daily intake of the diets was the same in each group, showing a similar decreasing tendency (70-100g/kg of body weight) in accordance with the growth rate. It was observed that there are no differences in the taste and appetite in each group of rats. 3. Abnormal enlargements of kidney and lung were not seen in all the mixture groups compared with the controls, while a slight hepatomegaly was observed in all mixture groups of low level as in the second controls. Significant differences (almost 1% level) were observed between the high level groups and the first control group. 4. Histopathological effects of the food additives on lung, kidney and liver tissues were found in all mixture group of high level. The less frequencies of the same effects were also seen in the low level groups. 5. The esterified cholesterol to total cholesterol ratio in the mixture groups of high level showed a little lower values, and the activities of serum glutamate oxaloacetate transaminase and alkaline phosphatase decreased almost with significance of 5% level compared with the first control group. The serum A/G ratio in the mixture groups also decreased. The results demonstrated that the liver function was decreased in the mixture groups compared with the controls. 6. In all groups throughout the test period, kidney functions (concentration of protein and creatinine excreated per hour in urine and renal filtration rate) were shown almost as normal as the first control. 7. Average values of hematocrit, erythrocytes and leucocytes in the mixture groups were in the normal ranges as in the controls, which denotes that the production of blood cells in bone marrow were also normal in all groups. With the above results, it is concluded that when the food additives (DHA, AF-2, BHT) were given together to the rats in several combinations even in less amount, they showed more toxic signs than the single doses.

Increase of Production Ratio of Pre-selected Superior Dairy Female Offspring by Combination of OPU derived Oocytes and X-bearing Semen

  • This study was designed to evaluate the possibility of increase through dairy female offspring's ratio by transfer of pre-selected transferrable blastocyst that was produced by pre-selected X-bearing semen with OPU derived oocytes. Elite dairy female cow is demanded strongly compared with male, the so called, farmer wants to produce only an elite female dairy offspring as a candidate female dairy cow for producing milk. In our study, we selected 2 elite dairy bull semen from National Agricultural Cooperative Federation to pre-select X-bearing semen and 5 elite dairy female cows as donor for collecting of OPU derived oocytes. OPU derived embryo production system was carried out an aspiration of immature oocytes from 5 donor cows 2 times per week, total 200 times for 2 to 7 months by an ultrasonographic guided follicular aspiration system and then produced in vitro-produced blastocysts by in vitro maturation, fertilization and culture. Dairy donor semen selected H-319, 320 bull in National Agricultural Cooperative federation was sorted X-bearing semen by flow-cytometer and frozen for using IVF with OPU derived oocytes. Donor cows were selected 5 elite dairy cows from Gyeongju Dairy Cow Community and then disease tests such as 4 kinds of disease before selecting was checked. Oocyte proportion of grade 1 to 3 from total collected oocytes was significantly lower in donor A and B than those in donor C, D and E (82.16 and 70.03% vs. 90.0, 91.78 and 93.57%), respectively (p<0.05). However, number of oocytes per session in donor A, C and E was significantly higher than those in donor B and D ($7.77{\pm}3.26$, $5.85{\pm}2.10$ and $7.03{\pm}2.14$ vs. $4.68{\pm}2.61$ and $5.21{\pm}1.97$ oocytes), but donor A was significantly higher than donor C (p<0.05). Development to blastocyst in donor B, C and E was significantly higher than those in donor A and D (31.0, 25.0 and 25.0% vs. 14.3 and 4.5%), but donor A was not different in donor C and E (p<0.05). Nine out of 10 blastocysts (90.0%) derived from OPU blastocysts were confirmed male embryos that was induced with Y-bearing semen to confirm sex ratio only. Total 96 blastocysts derived from female bearing semen were transferred into synchronized recipients and then confirmed 42 recipients (43.8%) pregnancy rate, 36 offspring (37.5%) and 91.7% female sex ratio (33 female vs. 3 male offspring). Taken together all data, elite dairy female offspring could be produced effectively by in vitro production system between pre-selected x-bearing semen and OPU derived oocytes that would be influential breeder in the breeding of dairy farm to increase effectively elite dairy offspring ratio as well as net income in the dairy farmer.

Characteristics of Flowering and Leaf Emergence in Lycoris Species (상사화류의 개화와 출엽 특성)

  • This study was conducted to examine the pattern and characteristics of flowering and leaf emergence for twenty two kinds of the genus of Lycoris sp. One species of Lycoris was flowered on July to August, twelve species on August, and nine species on September. Shape of flowers were as follows : Nine species belong to L. radiara-shaped flower, seven species L. squamigera-shaped flower, and seven species the intermediate type. Flower color of three kind were an order of descent red and pink. seven kind white, four kind yellow, three kind orange, and two kind purple colors. Ten Lycoris species were good fertile, and two kinds were poor fertile. The average length of pollen grain was ranged from $48.53\;to\;88.62{\mu}m$, and the width from $22.87\;to\;33.67{\mu}m$. Eleven kinds were spring-leaf emergence type, and ten kind autumn-leaf emergence types. Hunter values in leaves were as follows : $L^*$ value was ranged from 31.6 to 45.2; $a^*$ values from -6.74 to -17.46; $b^*$ values from 6.19 to 21.89 Leaf width was ranged from 0.90 to 2.35cm, and its shape was oblong. The length of epidermal cells was 0.33 to 0.75mm, and the width was $38.53\;to\;90.00{\mu}m$. Most of stomata were distributed in the back side of leaves. The length of stomata was ranged from $67.12\;to\;104.89{\mu}m$, and its width was from $14.90\;to\;71.52{\mu}m$.
