This study was performed to investigate the immunogenicity of autogenous E coli vaccines and their preventive effects on diarrhea in suckling piglets. Autogenous E coli live and killed vaccines were made from the E coli strains isolated from piglets showing diarrhea in field. In group I, pregnant sows were administered with live and killed vaccines at 4 and 2 weeks before parturition, respectively, Killed vaccines were administered twice to pregnant sows at 4 and 2 weeks before parturition in group II, and saline instead of autogenous E coli vaccines was administered to pregnant sows in group III for the control. After parturition, antibody titers in colostrum and milk from sows, incidence of diarrhea in suckling piglets, and immunoreactivity in the ileum of piglets from each treatment group were examined. The results were as follows ; 1. Sixty-two strains of E coli were isolated from suckling piglets with diarrhea. Of the strains, K88 pilus and K99 pilus antigens were identified in 6(9.8%) and 4(6.5%), respectively. Molecular weights of K88 and K99 pilus were 27,500 and 18,500 daltons, respectively. 2. Antibody titers in colostrum from sows after parturition were 1 : 512 to 1 : 1,024 in group I, 1.256 to 1.512 in group II, and 1 : 4 to 1 : 16 in group III. 3. The incidences of diarrhea In suckling piglets of group I, II and III were 3.3%, 9.4% and 21.4%, respectively. 4. When the immunoreactivity in the ileum of piglets from each group was examined, the proportion of IgG-immunoreactivity cells in group I or II was higher than that in group III. In conclusion, administration of autogenous E coli vaccines to pregnant sows before parturition can be an effective way to prevent diarrhea in suckling piglets.
Throughout the studies the following experimental results were summarized. 1. It was impossible to infect and kill the mice, weighing 10 to 12 gm, by inoculating 0.2ml of virulent Cl. chauvoei, diluted 1 to 10 with physiological saline, via subcutaneous, intramuscular, intraperitoneal or intraveonus, route. 2. The mice which were inoculated in brain with 0.03ml of Cl. chauvoei diluted 1 : 5120 with physiological saline were resulted in all death after infection, but not in case of attenuated strain even in dilution of one to five. 3. Virulent Cl. chauvoei were diluted with each of those of whole blood, erythrocytes and serum of horse, calf, swine, sheep, rabbit, guinea pig, chicken and duck, human plasma and 2% CaCl solution, and inoculated subcutaneously 0.25 to 0.5ml in mice, weighing 12 to 15gm. It was resulted in significant increase in virulence as comparing with the case of physiological saline solution except when horse and pig sera were used. Such a phenomena were not seen in attenuated strain. 4. Virulence of virulent Cl. Chauvoei could be increased significantly in rat, as the procedures used in mice, by suspending in whole blood, erythrocytes, serum, or plasma of various animals, or 2% $CaCl_2$ solution and by inoculating subcutaneously 0.5 to 10ml in rat, weighing 30 to 60 gm, as compared with those of control group which used physiological saline solutionos diluent. 5. Mice resisted 100 and 80 percent against challenge of $10^3$ and $10^4$ M.L.D.. respectively, 24 hours after inoculation of 0.5ml black leg antiserum. 6. Immune response to the black leg living vaccine in mice could be obtained more favorably in the group of respected vaccination rather than those of single inoculation and the most profitable inoculm size of the vacine was 0.5 to 1.0ml. 7. Challenge for the immunized mice could be carried out effectively 3 weeks after first vaccination. 8. Satisfactory results could be obtained by inoculating subcutaneously for the immunization and intracerebrally or subcutaneously for the challenge. 9. Mice which were inoculated with 0.5ml of black leg living vaccine via subtaneucously two times at seven days interval and 21 days after first inoculation and challenged with 5 and 10 M.L.D. of virulent strain, resited 100 and 70 to 80 percent respectively. Same results were obtainable in black leg killed vaccine as the procedures used in living vaccine. 10. There were significantly different resistances against the definite challenge does between the mice groups which were immnuized with the living vaccine diluted five or 10 times and the undiluted. 11. For the biological assay of black leg living vaccine and antiserum, satisfactory results could be obtained using mice.
The study was carried out to investigate the pathogenicity of EHV isolate to hamsters and mice and immunogenicity of experimentally produced. vaccine were evaluated in the horses. Hamsters infected. intranasally with $LC_1$ isolate showed symptoms of nasal discharge, conjunctivitis and body weight loss during the observation period of 12 days after infection, while only slight depression and body weight loss were noticed with mice infected with $LC_1$ indicating that hamsters are more susceptible to the virus. Antibody titer of mice and hamsters were gradually increased to highest level of 1:2560~10240, 1:640~1280, respectively, at 7~12 days post vaccination. Horses immunized against $LC_1$ killed vaccine reached to maximum antibody titer of 1:20480 around 4 weeks after 1st vaccination and declined after 12 weeks post vaccination. No significant antibody increase were detected after 2nd vaccination. Mean body temperature and mean total leukocyte counts remained within normal range and no adverse reaction were noticed after vaccination.
Objectives: Nowadays cancer treatment is an important challenge in the medical world that needs better therapies. Many active secretions produced by insects such as honey bees used to discover new anticancer drugs. Bee venom (BV) has a potent anti inflammatory, anti cancer and tumor effects. The aim of present study is evaluation of anticancer effects induced by Apis mellifera venom (AmV) on cell Lines. Methods: AmV was selected for study on cancer cell lines. Total protein, molecular weight and LD50 of crude venom were determined. Then, cells were grown in Dulbecco's Modified Eagle medium supplemented with 10% fetal bovine serum and 1% antibiotics. The A549, HeLa and MDA-MB-231 cell Lines were exposed by different concentration of AmV. The morphology of cells was determined and cell viability was studed by MTT assay. Evaluation of cell death was determined by and DNA fragmentation. Results: The results from MTT assay showed that 3.125 ㎍/mL of A549, 12.5 for HeLa and 6.25 ㎍/mL of MDA-MB-231 killed 50% of cells (p < 0.05). Morphological analysis and the results from hoescht staining and DNA fragmentation indicated that cell death induced by AmV was significantly apoptosis. Conclusion: The data showed that using lower dosage of AmV during treatment period cause inhibition of proliferation in time and dose dependant manner. Findings indicated that some ingredients of AmV have anticancer effects and with further investigation it can be used in production of anticancer drugs.
E. tarda의 독성 비교를 위하여 일반 TSB, iron-chelate인 2,2‘-dipyridyl 첨가 TSB 및 Fe 첨가 TSB의 세 가지 조건에서 E. tarda를 배양하였다. 독성시험의 결과는 iron-chelate 첨가 시험구의 E. tarda에서 가장 낮은 독성을 나타내었다. 그리고 각각의 OMPs와 IROMPs을 분리하여 SDS-PAGE로 항원성을 비교한 결과, iron-chelate 첨가 시험구의 E. tarda IROMPs에서 68, 73 kDa 크기의 분자들이 더 많이 발현된다는 것을 확인할 수 있었다. 두 가지 vaccine 투여구 중에서 2,2‘-dipyridyl 첨가 배지로 배양한 균체로 제작한 DP-FKC 항원 투여구가 응집 항체가가 더 높게 나타났다. 면역반응의 변화를 알기 위한 응집 항체가 조사에서는 FKC 항원과 DP-FKC 항원 투여구에서 모두 대조구보다 높은 값을 나타내었고, 3주 째에 최고치를 나타내었다. ELISPOT을 이용한 항체 생성 세포 수의 검출에서도 응집 항체가와 유사한 pattern을 나타내었는데, 두 가지 vaccine 투여구에서 2주 째에 가장 많은 항체 생성 세포 수를 나타내었다. 공격실험에서는 3주 째에 FKC 항원 투여구가 50%, DP-FKC 항원 투여구에서는 80%의 생존율을 나타내었다. 전체적으로 DP-FKC 항원 투여구의 생존율이 높게 나타났다. 이상의 결과로부터 넙치에 대한 E. tarda의 vaccine으로는 현재 연구되고 있는 FKC vaccine보다 철 결핍 조건에서 배양된 균체의 항원인 DP-FKC vaccine이 동일한 조건으로 사용하였을 때 더 나은 방어력을 가진다는 것을 알 수 있었다.
말인플루엔자(EI) 사독백신 투여 후 virus strains 별로 항체역가 변화를 측정하였다. 더러브렛말(1세) 20마리에서 혈구응집억제반응법으로 혈청항체를 41주 동안 관찰하였다. 기초접종 4주 후에 2차접종을 하였다. 그러나 2차 접종 직전의 혈청에서는 대부분 항체역가가 관찰되지 않았다. 역가 최고치는 접종 후 6-10주였고, 16주 동안 유지되었으며 이후 서서히 감소하였다. 백신무반응은 3마리(15%) 말에서 나타났지만 이는 virus strains에 좌우되었다. A/Equine/La Plata/93(H3N8)는 3개의 strain 중에서 높고 안정적인 항체반응을 유도하였다. 결과를 바탕으로 항체역가를 유지하기 위해서는 1세마에 3회 접종이 권고된다. 저자들은 백신에 대한 무반응은 백신브레이크 또는 개체특이성에 기인하였다고 추측하였다. 이 실험은 국내 최초의 EI 백신 strain에 대한 연구였으며 면역학적 무반응에 대해서는 추가 연구가 필요하다.
To investigate the relationship between metabolic changes in $^1H$-nuclear magnetic resonance (NMR) spectra and fish vaccination, serum was collected from olive flounders treated with a formalin-killed Edwardsiella tarda vaccine and used for $^1H$-NMR metabolite profiling. Principal component analysis and partial least squares were applied to the $^1H$-NMR profile to reduce its complexity and establish class-related clusters. Relative lipid regions were distinguished in vaccinated and non-vaccinated serum. Then, the lipids were extracted from the serum and analyzed. Triolein was identified.
본 연구에서는 병원성 Pasteurella multocida D:4 외막 단백질 H의 방어적 면역성과 백신으로서의 가능성을 검정하고자, 외막 단백질 H 유전자를 대장균에서 발현, Trx와 융합된 형태의 재조합 외막 단백질 H를 분리하여 면역화와 백신 실험에 항원으로 사용하였다. 면역 실험에서 재조합 외막 단백질 H는 높은 역가의 항체를 유도하였으며, 불활화한 사균 백신과 유사한 수준의 백신 효과를 나타내었다.
목 적 : 국내에서 사용되고 일본뇌염 바이러스 백신에 대한 야외 실험을 실시하여 국내 백신 제조에 사용되는 Nakayama-NIH주의 국내 접종자에 대한 항체 생성율 및 중화 항체의 지속율을 조사하고, 일본뇌염 접종 스케줄의 재조정 및 국내 면역 집단의 민감도 조사 등을 통하여 방역 대책의 기초 자료로 활용하고자 하였다. 방 법 : 일본뇌염 백신의 면역효과 및 항체지속 기간을 알아보고자 1994년부터 계속 사업으로 강화군에 소재하고 있는 K 초등학교 학생들을 대상으로 조사를 실시하였다. 대상자들의 혈액을 시기에 따라 채혈하여 hemagglutination inhibition test (HI) 및 plaque reduction neutralization test (PRNT)를 실시하여 항체가를 조사하였다. 결 과 : 총 조사 대상자 213명은 이미 기초 접종이 끝난 자들로서 HI 항체가는 '95년 4월 백신 면역전에 이미 63.4%가 1:10 이상의 항체가를 가지고 있었으며 중화항체가는 전원이 1:20 이상의 항체가를 가지고 있었다. '95년 백신 접종 후 12개월이 지난 1996년 4월까지 HI 항체가 1:20 이상이 55.7%이며, 중화항체가는 전원이 1:20 이상을 보유하고 있었다. 이들에게 1996년 4월에 재접종 후 6개월째에는 HI 항체가가 1:10 이상이 69.7%로 증가하였고 G.M.T값은 1:11.6에서 1:13.23, 중화 항체의 G.M.T는 1:275.7에서 1:348.1로 증가하였다. 결 론 :위의 실험 결과에 따르면 현재 국내에서 사용중인 불활화 일본뇌염 백신은 자연계 에서 일본뇌염 바이러스가 공격하는 $10^5$$LD_{50}$를 막을 수 있는 중화항체가 1:20을 기본면역 1회로 1년 이상 유지하고 있었으며, 충분한 boosting 효과를 보이고 있었다. 따라서 일본뇌염 백신의 추가면역은 기본접종후 개인의 상태에 따라 2~3년 간격의 추가접종이 적절하다 고 생각된다.
본 연구는 넙치, Paralichthys olivaceus에서 Edwardsiella tarda와 Streptococcus iniae에 대한 혼합백신의 효능과 안전성에 대해 조사하고자 하였다. 혼합백신을 주사한 넙치에서 2, 4, 6, 8주 째에 채혈하여 E. tarda와 S. iniae에 대한 항체가 를 microtitre 법으로 조사하였다. 백신 주사 후 3주째에 E. tarda와 S. iniae를 각각 인위감염 시켰을 때 백신구에서 81.0-92.8%의 높은 상대생존율을 보여 혼합백신에 의한 항병력의 증가가 확인되었다. 백신 주사 후 초기에 백신구가 대조구에 비해 체중 증가가 느렸으나 사육기간이 길어지면서 회복되어 대조구와 백신구 체중의 차이를 보이지 않았다. 이 결과는 백신 주사가 넙치의 성장에 영향을 주지 않고 에드와드병과 연쇄구균증을 예방할 수 있음을 보여준다.
