Bacterial diseases of flounder, Paralichthys olivaceus (넙치의 세균성(細菌性) 질병(疾病))

  • Flounder culture has been developed mainly in the western parts of japan, and, to date, following six bacterial diseases have been reported. Bacterial white enteritis occurs in 16 to 30-day-old flounder larvae and often causes mass mortality in seed production. Bacterium named Vibrio sp. INFL invades and multiplies in the mucosae of posterier part of intestine, and causes desquamative enteritis. Gliding bacterial disease occurs mostly in juvenile stage and in spring to summer. Diseased signs are partial discoloration and erosion of skin and fins. Histologically, epidermis are removed, and the causative bacterium, Flexibacter maritimus, multiplies on the surface of demis and invades into the muscular tissue. Vibriosis caused by Vibrio anguillarum and related organisum is one of the well-known diseases among marine fish. Outbreaks of the disease in flounder culture are relatively few, but mass mortalities in fingerlings due to the disease were reported. An outbreak of nocardiosis in the autumn of 1984 has been reported, but since then the disease scarcely occurred. The disease is characterized by formation of abscesses under the skin and white nodes in the gill, heart, spleen and kidney. Streptococcicosis occurs frequently in recent years. Beta-hemolytic streptococcus is the causative bacterium, which possesses the same biochemical and serological characteristics as $\beta$-streptococci isolated from some marine and freshwater fish, and is seemed to related to Streptococcus iniae. Edwardsiellosis is the disease that causes most damage in flounder culture in Japan. Characteristic symptoms are swelling of abdomen and intestinal protrusion from the anus due to accumulation of ascites. Edwardsiella tarda, a well-known pathogen of freshwater fish, is the causative bacterium of the disease.

A Study on the Liu Wansu's "Theory on the three wasting-thirst" (유완소(劉完素)의 "삼소론(三消論)"에 대한 연구(硏究))

  • Wasting-thirst(消渴) occurs due mainly to dryness-heat(燥熱) which by depleting 'Fluid and Humor'(津液) make symptoms of thirst(口渴) swift digestion with rapid hungering(消穀善饑) frequent urination(小便頻數) according to "Internal Classic"(內經). However, the doctors of those days had a tendency to manage wasting-thirst with herbal drugs of dryness-heat or with drugs that tonifies the kidney. Liu Wansu(劉完素) thought that those kinds of drugs intensify gatro-intestinal dryness-heat and consequently could not be suitable for the patients with wasting-thirst. He said that they had mistaken about the relation between five viscera(五臟) and six climate condition(六氣) and that they did not know thoroughly on the theory of branch and root(標本). For preventing the disease of wasting-thirst, Liu Wansu(劉完素) recommended avoiding gatro-intestinal drying caused by the intake of dryness-heat diet, and warned not to deplete 'Fluid and Humor' due to stress-induced fire(火), not to damage yin qi(陰氣) through severe diseases.

Pathogenesis of Perirenal Edema in Pigs and Rabbits Administered with Oxalate, Nitrate and Calcium (Oxalate, Nitrate 및 Calcium 혼합투여(混合投與)에 의(依)한 돈(豚) 및 가토(家兎)의 Perirenal Edema 발병(發病))

  • In order to clarify the pathogenesis of perirenal edema, pigs and rabbits were administered with oxalate and nitrate, with oxalate nitrate and glycolic acid, with oxalate, nitrate and ascorbic acid and with oxalate, nitrate and calcium, respectively. The results obtained are summarized as follows; The pigs and rabbits administered with oxalate and nitrate with oxalate, nitrate and glycolic acid and with oxalate, nitrate and ascorbic acid, respectively, were not showed perirenal edema despite of observing the abundant oxalate crystals in the proximal convoluted tubles. But pigs and rabbits administered with oxalate, nitrate and calcium were histopathologically showed perirenal edema similar to those of pigs fed Amaranthus retroflexus. Therefore, author considered that oxalate, nitrate and calcium are main factors to cause perirenal edema. It was regarded that perirenal edema in pigs was caused by the reciprocal reactions of those materials including oxalate, nitrate and calcium which may produce vascular damage, decreased osmotic pressure by hypoproteinemia and increased vascular permeability in kidney, rather than the mechanical obstructions by the oxalate crystals in the proximal convoluted tubule.

Subtilisin QK, a Fibrinolytic Enzyme, Inhibits the Exogenous Nitrite and Hydrogen Peroxide Induced Protein Nitration, inVitro and inVivo

  • Subtilisin QK, which is newly identified as a fibrinolytic enzyme from Bacillus subtilis QK02, has the ability of preventing nitrotyrosine formation in bovine serum albumin induced by nitrite, hydrogen peroxide and hemoglobin in vitro verified by ELISA, Western-blot and spectrophotometer assay. Subtilisin QK also attenuates the fluorescence emission spectra of bovine serum albumin in the course of oxidation caused by nitrite, hydrogen peroxide and hemoglobin. Furthermore, subtilisin QK could suppress the transformation of oxy-hemoglobin to met-hemoglobin caused by sodium nitrite, but not the heat-treated subtilisn QK. Compared with some other fibrinolytic enzymes and inactivated subtilisin QK treated by phenylmethylsulfonylfluoride, the ability of inhibiting met-hemoglobin formation of subtilisin QK reveals that the anti-oxidative ability of subtilisin QK is not concerned with its fibrinolytic function. Additionally, nitrotyrosine formation in proteins from brain, heart, liver, kidney, and muscle of mice that is intramuscular injected the mixture of nitrite, hydrogen peroxide and hemoglobin is attenuated by subtilisin QK. Subtilisin QK can also protect Human umbilical vein endothelial cell (ECV-304) from the damage caused by nitrite and hydrogen peroxide.

  • Daekumeumja is a herb-medication which has been used for alcoholic damage. This is a report about the effect of Daekumeumja on Ascites in alcoholic liver cirrhosis. The study of abdominal CT and ultrasonography were performed to evaluate the volume change of ascites. And blood tests were done to measure the liver's and kidney's functions. Reduction of ascites is proved by abdominal CT & ultrasonography. And lab findings of liver functions showed improvement. This study suggests that Daekumeumja is probably effective in the treatment of ascites in alcoholic liver cirrhosis.

  • Diabetes mellitus, commonly known as diabetes, is a group of metabolic disorders characterized by high blood sugar levels over a prolonged period. Diabetes has been found to show many acute complications such as cardiovascular disease, stroke, chronic kidney disease, foot ulcer and damage to eyes. Korean ginseng (Panax ginseng) has been traditionally known to normalize the functional deficiencies of the lung, spleen and stomach, and thus improve the secretion of body fluids, thereby quenching thirst, suggesting it to be effective in the treatment of diabetes. Experimental studies (in vitro and in vivo) have recently shown that Korean ginseng and its extracts exhibit antidiabetic effects, and also insulin secretion and sensitizing effects related to blood glucose control. Moreover, clinical trials on antidiabetic effects of Korean ginseng have been reported to show blood glucose control, improvement of insulin resistance, reduction of postprandial blood glucose level and improvement of serum lipids (TG, TC, LDL-C). These will be critically examined by means of in vitro studies, cell experiment, animal models and human trials with a focus on understanding of molecular mechanisms.

Effects of Cervus elaphus extract solution for Aqua-acupuncture on Antioxidation in Rat's Brain induced by ischemia and reflow (허혈(虛血)/재관류(再貫流) 뇌(腦) 조직(組織)에서 녹용(鹿茸) 약침액(藥鍼液)의 항산화작용(抗酸化作用)에 대한 연구(硏究))

  • Cervus elaphus, being known to reinforce Kidney, have tested to study the effects concerning damages of brain tissue induced by lipid peroxidation. In vitro, the level of lipid peroxide in brain tissue was decreased proportinally according to dose by Cervus elaphus extract solution for aqua-acupuncture (CESAA). It was much more decreased, when lipid peroxidation was induced with Fe(II). And, it was seen proportinally decrease according to the dose of CESAA on xanthine oxidase activities and type conversion ratio. However, I can not find special changes about aldehyde oxidase activities. And, I had observed the effects of CESAA on damages of rat's brain following ischemia and reflow. Before ischemia was caused, CESAA was applied 0.2 ml per 250 g through femoral vein in ischemia and reflow group and normal sailine was applied in normal group. Ischemia was caused by cervical artery's clamp for 30 min and reflowed by clamp remove after 15 min. It was increased on the content of lipid peroxidation, activies and type conversion ratio of xanthine oxidase following ischemia and reflow. However, they were decreased when CESAA was pre-appllied. However, it could not seen special changes on aldehyde oxidase activities, either. In conclusion. CESAA recovers the damage of brain due to ischemia and reflow by decreasing the lipid peroxidation through decreasing of xanthine oxidase activies and type conversion ratio.

  • Acute hyperammonemia is a medical emergency in the newborn. Efficient, prompt removal of serum ammonia is essential in preventing irreversible brain damage in order to prevent the profound central nervous system dysfunction due to hyperammonia. We report a case of 2.3 kg, 5-day old girl with methylmalonic acidemia who presented with severe hyperammonemia and was successfully treated with continuous venovenous hemodiafiltration(CVVHDF). CVVHDF is an effective and safe method of ammonia removal in the newborn.

Genetic Toxicity Test of Emodin by Ames, Micronucleus, Comet Assays and Microarray Analysis Showing Differential Result

  • Emodin (1,3,8-trihydroxy-6-methylanthraquinone) is a major constituent of rhubarb. Although it has been claimed to have a wild spectrum of therapeutic value, its side effects, especially in human kidney cells have not been well characterized. In this study, we have carried out in vitro genetic toxicity test of emodin and microarray analysis of differentially expressed genes in response to emodin. The result of Ames test showed mutations with emodin treatment in base substitution strain TA1535 both with and without exogenous metabolic activation. Likewise, emodin showed mutations in frame shift TA98 both with and without exogenous metabolic activation. The result of COMET assay in L5178Y cells with emodin treatment showed DNA damage both with and without exogenous metabolic activation. Emodin did not increase micronuclei in CHO cells both with and without exogenous metabolic activation. 150 Genes were selected as differentially expressed genes in response to emodin by microarray analysis and these genes would be candidate biomarkers of genetic toxic action of emodin.

Effects of formalin on haematology and blood chemistry in crucian carp (Carassius auratus) (포르말린 침지에 의한 붕어(Carassius auratus)의 혈액학 및 혈액화학치에 대한 영향)

  • To determine the effects of exposure to formalin on the secondary stress indices, changes in haematology and blood chemistry were monitored in healthy crucian carps (Carassius auratus). Fishes were separately exposed in a concentration range of 125 to 500 ppm formalin for 60 min. After exposure, red blood cell (RBC) count and packed cell volume (PCV) were elevated in the 500 ppm formalin exposed group. However, mean corpuscular volume (MCV) and mean corpuscular haemoglobin (MCH) were decreased significantly in the 500 ppm formalin exposed group. Total protein, albumin, aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatinine, total bilirubin, inorganic phosphorus (IP) and magnesium were significantly increased at a concentration of 500 ppm. Alkaline phosphatase (ALP) and glucose were increased at a concentration of 500 ppm, but this was not significant. Lactate dehydrogenase (LDH) and calcium were significantly decreased at concentrations of 250 and 500 ppm. AST, ALT, glucose and magnesium were significantly increased in the 250 ppm formalin exposed group. These results suggests that formalin exposure might cause some damage in the liver and kidney of crucian carp.
