A review of the immunomodulatory activities of polysaccharides isolated from Panax species

  • Panax polysaccharides are biopolymers that are isolated and purified from the roots, stems, leaves, flowers, and fruits of Panax L. plants, which have attracted considerable attention because of their immunomodulatory activities. In this paper, the composition and structural characteristics of purified polysaccharides are reviewed. Moreover, the immunomodulatory activities of polysaccharides are described both in vivo and in vitro. In vitro, Panax polysaccharides exert immunomodulatory functions mainly by activating macrophages, dendritic cells, and the complement system. In vivo, Panax polysaccharides can increase the immune organ indices and stimulate lymphocytes. In addition, this paper also discusses the membrane receptors and various signalling pathways of immune cells. Panax polysaccharides have many beneficial therapeutic effects, including enhancing or activating the immune response, and may be helpful in treating cancer, sepsis, osteoporosis, and other conditions. Panax polysaccharides have the potential for use in the development of novel therapeutic agents or adjuvants with beneficial immunomodulatory properties.

Mitochondrial OXPHOS genes provides insights into genetics basis of hypoxia adaptation in anchialine cave shrimps

  • Cave shrimps from the genera Typhlatya, Stygiocaris and Typhlopatsa (TST complex) comprises twenty cave-adapted taxa, which mainly occur in the anchialine environment. Anchialine habitats may undergo drastic environmental fluctuations, including spatial and temporal changes in salinity, temperature, and dissolved oxygen content. Previous studies of crustaceans from anchialine caves suggest that they have possessed morphological, behavioral, and physiological adaptations to cope with the extreme conditions, similar to other cave-dwelling crustaceans. However, the genetic basis has not been thoroughly explored in crustaceans from anchialine habitats, which can experience hypoxic regimes. To test whether the TST shrimp-complex hypoxia adaptations matched adaptive evolution of mitochondrial OXPHOS genes. The 13 OXPHOS genes from mitochondrial genomes of 98 shrimps and 1 outgroup were examined. For each of these genes was investigated and compared to orthologous sequences using both gene (i.e. branch-site and Datamonkey) and protein (i.e. TreeSAAP) level approaches. Positive selection was detected in 11 of the 13 candidate genes, and the radical amino acid changes sites scattered throughout the entire TST complex phylogeny. Additionally, a series of parallel/convergent amino acid substitutions were identified in mitochondrial OXPHOS genes of TST complex shrimps, which reflect functional convergence or similar genetic mechanisms of cave adaptation. The extensive occurrence of positive selection is suggestive of their essential role in adaptation to hypoxic anchialine environment, and further implying that TST complex shrimps might have acquired a finely capacity for energy metabolism. These results provided some new insights into the genetic basis of anchialine hypoxia adaptation.

Rapid Detection of SdhBP225F and SdhBH272R Mutations in Boscalid Resistant Botrytis cinerea Strains by ARMS-PCR

  • $SdhB^{P225F}$ and $SdhB^{H272R}$ mutations have been found associated with boscalid resistance in Botrytis cinerea from strawberry in Shanghai, China. For rapid detection of two mutations, tetra-primers were designed and optimized to gain the relatively high accuracy and specificity based on the ARMS-PCR technique, by which resistance can be identified with different lengths of products on agarose gels. The tetra-primer ARMS-PCR systems for $SdhB^{P225F}$ and $SdhB^{H272R}$ were validated by 9 SdhB-squenced strains repeatedly. Then, sensitivity of 30 more strains were also tested by the methods, which were accordant with genotypes by sequencing and the sensitivity of conidial germination to boscalid by 100%. Thus, the methods developed in this study are proved to be rapid, inexpensive, accurate and practical for resistance detection of Botrytis cinerea caused by $SdhB^{P225F}$ and $SdhB^{H272R}$ mutations.

A key-insulated CP-ABE with key exposure accountability for secure data sharing in the cloud

  • ABE has become an effective tool for data protection in cloud computing. However, since users possessing the same attributes share the same private keys, there exist some malicious users exposing their private keys deliberately for illegal data sharing without being detected, which will threaten the security of the cloud system. Such issues remain in many current ABE schemes since the private keys are rarely associated with any user specific identifiers. In order to achieve user accountability as well as provide key exposure protection, in this paper, we propose a key-insulated ciphertext policy attribute based encryption with key exposure accountability (KI-CPABE-KEA). In our scheme, data receiver can decrypt the ciphertext if the attributes he owns match with the self-centric policy which is set by the data owner. Besides, a unique identifier is embedded into each user's private key. If a malicious user exposes his private key for illegal data sharing, his identity can be exactly pinpointed by system manager. The key-insulation mechanism guarantees forward and backward security when key exposure happens as well as provides efficient key updating for users in the cloud system. The higher efficiency with proved security make our KI-CPABE-KEA more appropriate for secure data sharing in cloud computing.

  • In the paper, designed digital contents security module to check unlawfulness reproduction and distribution of digital contents. This paper applied Diffie-Hellman algorithm that use discrete logarithm and random number as primary for public key application to create encryption key that agree each other through communication channel between DCPS and HOST, and applied Triple DES repeat DES 3 times through 2 different encryption key that is selecting ANSI X9.17 that is key management standard, ISO 8732 and PEM(Privacy-Enhanced Mail) etc. by secondary protection for safe transmission of digital contents in transmission line. Designed security module consist of key exchange module, key derivation module and copy protection processing module. Digital contents security module that design in this thesis checks reproduction and distribution of digital contents by unauthenticated user through user certification function and digital contents encryption function, and protect digital contents transmission line.

Characterization of the brittleness of hard rock at different temperatures using uniaxial compression tests

  • The failure mechanism of a deep hard rock tunnel under high geostress and high geothermalactivity is extremely complex. Uniaxial compression tests of granite at different temperatures were conducted. The complete stress-strain curves, mechanical parameters and macroscopic failure types of the rock were analyzed in detail. The brittleness index, which represents the possibility of a severe brittleness hazard, is proposed in this paperby comparing the peak stress and the expansion stress. The results show that the temperature range from 20 to $60^{\circ}C$ is able to aggravate the brittle failure of hard rock based on the brittleness index. The closure of internal micro cracks by thermal stress can improve the strength of hard rock and the storage capacity of elastic strain energy. The failure mode ofthe samples changes from shear failure to tensile failure as the temperature increases. In conclusion, the brittle failure mechanism of hard rock under the action of thermal coupling is revealed, and the analysis result offers significant guidance for deep buried tunnels at high temperatures and under high geostress.

  • Artemisinin, the effective ingredient of Chinese herb Artemisia annua L (Qinghao in Chinese), has been proved to be effective to antimalarial. Herein, a reliable, sensitive and convenient electrochemical method was developed for the determination of artemisinin utilizing the excellent properties of multi-wall carbon nanotube (MWNT). The electrochemical behavior of artemisinin was investigated. It is found that the reduction peak current of artemisinin remarkably increases and the peak potential shifts positively by 240 mV at the MWNT film-modified electrode. These phenomena indicate that the MWNT film exhibits efficient catalytic activity to the electrochemical reduction of artemisinin. The effects of pH value, amount of MWNT, scan rate and accumulation time were examined. The limit of detection (S/N = 3) is as low as 10 $\mu$ g $L^{-1}$. Finally, this newly developed method was used to determine the content of artemisinin in Artemisia annua L.

Probabilistic and spectral modelling of dynamic wind effects of quayside container cranes

  • Quayside container cranes are important delivery machineries located in the most frontiers of container terminals, where strong wind attacks happen occasionally. Since the previous researches on quayside container cranes mainly focused on the mean wind load and static response characteristics, the fluctuating wind load and dynamic response characteristics require further investigations. In the present study, the aerodynamic wind loads on quayside container cranes were obtained from wind tunnel tests. The probabilistic and spectral models of the fluctuating aerodynamic loads were established. Then the joint probabilistic distributions of dynamic wind-induced responses were derived theoretically based on a series of Gaussian and independent assumption of resonant components. Finally, the results were validated by time domain analysis using wind tunnel data. It is concluded that the assumptions are acceptable. And the presented approach can estimate peak dynamic sliding force, overturning moments and leg uplifts of quayside container cranes effectively and efficiently.

Synthesis of Polyamine Grafted Chitosan Copolymer and Evaluation of Its Corrosion Inhibition Performance

  • Two new chitosan derivatives, polyamine grafted chitosan copolymers have been synthesized for corrosion protection of carbon steel in acidic medium. First, methyl acrylate graft chitosan copolymer (CS-MAA) was prepared by the reaction of chitosan (CS) and methyl acrylate (MAA) via the Michael addition reaction. Then, CS-MAA was reacted with ethylene diamine (EN) and triethylene tetramine (TN) respectively to synthesize ethylene diamine grafted chitosan copolymer (CS-MAA-EN) and triethylene tetramine grafted chitosan copolymer (CS-MAA-TN), and the structures were characterized by Fourier-transform infrared spectroscopy (FT-IR). At last, the corrosion inhibition activities on Q235 carbon steel were investigated by using gravimetric measurements, metallographic microscope, potentiodynamic polarization and electrochemical impedance spectroscopy (EIS) measurements. The compounds CS-MAA-EN and CS-MAA-TN show an appreciable corrosion inhibition property against corrosion of Q235 carbon steel in 5% HCl solution at $25^{\circ}C$. It has been observed that CS-MAA-EN shows greater corrosion inhibition efficiency than CS-MAA-TN. The inhibition efficiency of CS-MAA-EN was close to 90% when the mass fraction concentration was 0.2%~0.3%; the inhibition efficiency of CS-MAA-TN was close to 85% when the mass fraction concentration was 0.02%. The present work provided very promising results in the preparation of green corrosion inhibitors.

Nigrospora Species Associated with Various Hosts from Shandong Peninsula, China

  • Nigrospora is a monophyletic genus belonging to Apiosporaceae. Species in this genus are phytopathogenic, endophytic, and saprobic on different hosts. In this study, leaf specimens with disease symptoms were collected from host plants from the Shandong Peninsula, China. The fungal taxa associated with these leaf spots were studied using morphology and phylogeny based on ITS, TEF1, and TUB2 gene regions. In this article, we report on the genus Nigrospora with N. gorlenkoana, N. oryzae, N. osmanthi, N. rubi, and N. sphaerica identified with 13 novel host associations including crops with economic importance such as bamboo and Chinese rose.
