벼 도정 부산물을 이용한 탁주 제조에 관한 연구 (Studies on Korean Takju using the By-Product of Rice Milling)

  • 파쇄미, 색채불량미와 일반미에 효모 Saccharomyces cerevisiae, 혼합 밀 누룩으로 Aspergillus oryzae와 Aspergillus niger mut. kawachii를 접종하여 만든 개량누룩으로 탁주를 2단 담금한 후 발효과정 중의 품질을 검토한 결과는 다음과 같다. pH는 3.5 정도로서 잡균에 의한 오염이 방지되면서 곰팡이가 생성한 당화 효소에 의해 2일에 당화되었고, 환원당이 알코올 발효되면서 ethanol 18%로 정상적으로 알코올 발효가 진행되었다. 유기산 함량은 일반미에서 citric acid가 매우 높게 나왔다. 탁주 술덧에 함유하는 미량의 알코올로서 n-propanol, 2-methyl-1-propanol, 1-butanol, 3-methyl-1-butanol이 검출되었다. 일반미에서만 1-butanol이 정량되지 않았다. 술덧의 유리당은 파쇄미, 색채불량미에 비해 일반미에서 sucrose함량이 현저히 낮게 나왔다. 유리아미노산 함량은 일반미 술덧이 단맛을 나타내는 serine, glycine, alanine과 쓴맛을 나타내는 leucine, isoleucine, phenylalanine의 함량이 낮게 나왔으며, 색채불량미와 파쇄미술덧은 이와 반대의 결과를 나타내었다. 색차기에 의한 색의 구분에서 뚜렷한 차이가 없었고, 발효후 5단계 평점법에 의해 맛, 향, 색등을 관능검사한 결과 모두 4.3 이상의 $\ulcorner$좋다$\lrcorner$로 평가되어 전통탁주의 이용이 가능할 것으로 사료된다.

Purification, Characterization, and Partial Primary Sequence of a Major-Maltotriose-producing $\alpha$-Amylase, ScAmy43, from Sclerotinia sclerotiorum

  • A novel $\alpha$-amylase ($\alpha$-1,4-$\alpha$-D-glucan glucanohydrolase, E.C. 3.2.1.1), ScAmy43, was found in the culture medium of the phytopathogenic fungus Sclerotinia sclerotiorum grown on oats flour. Purified to homogeneity, ScAmy43 appeared as a 43 kDa monomeric enzyme, as estimated by SDS-PAGE and Superdex 75 gel filtration. The MALDI peptide mass fingerprint of ScAmy43 tryptic digest as well as internal sequence analyses indicate that the enzyme has an original primary structure when compared with other fungal a-amylases. However, the sequence of the 12 N-terminal residues is homologous with those of Aspergillus awamori and Aspergillus kawachii amylases, suggesting that the new enzyme belongs to the same GH13 glycosyl hydrolase family. Assayed with soluble starch as substrate, this enzyme displayed optimal activity at pH 4 and $55^{\circ}C$ with an apparent $K_m$ value of 1.66 mg/ml and $V_{max}$ of 0.1${\mu}mol$glucose $min^{-1}$ $ml^{-1}$. ScAmy43 activity was strongly inhibited by $Cu^{2+}$, $Mn^{2+}$, and $Ba^{2+}$, moderately by $Fe^{2+}$, and was only weakly affected by $Ca^{2+}$ addition. However, since EDTA and EGTA did not inhibit ScAmy43 activity, this enzyme is probably not a metalloprotein. DTT and $\beta$-mercaptoethanol strongly increased the enzyme activity. Starting with soluble starch as substrate, the end products were mainly maltotriose, suggesting for this enzyme an endo action.

  • Calcium and magnesium in polished rice, polished barley, what flour, and corn which were used for Yakju fermentation were determined by atomic absorption spectrophotometry and their quantitative changes in the filtrates of mashes were checked at 24 hours intervals dueing the whole brewing period. The results obtained were as follows. 1) Mineral contens of raw materials(mg% of each anhydrous materials). Ca : polished rice 8.21, polished barley 26.11, wheat flour 66.10, corn 86.63 2) Mineral contents of kojies made from raw materials (mg %of each anhydrous materials). Ca : rice koji 26.36, barley koji 97.61, what flour koji 87.69, corn koji 16.13, seeding koji 28.76 Mg: rice koji 29.29, barley koji 39.84, what flour koji 244.50, corn koji 102.64, seeding koji 143.79 3) Quantitative changes of minerals in the fitrates of mashes. Calcium contents in the filtrates of mashes were increased gradually after mashing in the first stage but unchanged till the mashes were ripened after mashing in the second stage. On the other hand, magnesium cotents were decreased gradually after increase in the the other hand, magnesium contens were decreased gradually after increase in the first stage but showed a tendency to increase gradually in the second stage. 4)Mineral contents of Yakjues produced, marketing Yakju, and natural water for brewing (g/ml). 주정발효용(酒精醱酵用) 효소제(酵素劑) 배양제조(培養製造) 조건(條件)이 Amylase 활성(活性)에 미치는 영향(影響) (The Effect on the Culture Condition on the Activity of Amylase used for Alcohol Fermentation)

  • The culture used wheat bran as media for four kind of mold strains such as Aspergillus oryzae, Aspergillus kawachii, Aspergillus usamii and Rhizopus javanicus to examine which strain could higher the activity of amylase most which is used for alcohol fermentation. It also provided three different kind of wheat bran media containing starch of 47%, 51% and 55% respectively for each strain. For each media it also added three different kind of nitrogen sources; ammonium sulfate, casein, and ammonium sulfate and casein equally mixed. Each nitrogen source added was subordinately differentiated into three different percentages, 2%, 4% and 6% respectively, except the 2% for the ammonium sulfate. The results obtained were summarized as follows (1) The activity of ${\alpha}-amylase$ was highest in the media of starch value 47% of wheat bran with 6% of casein added. (2) The activity of ${\beta}-amylase$ was highest in the media of starch value 51% of wheat bran with 2% of the equal mixture of ammonium sulfate and casein added. (3) The activities of both ${\alpha}-amylase$ and ${\beta}-amylase$ of Aspergillus usamii were highest in the media of starch value 47% wheat bran with no addition of nitrogen source. (4) Of the four strains examined, the activities of ${\alpha}-amylase$ and ${\beta}-amylase$ cultured in Rhizopus javanicus were both relatively higher. (5) The activities of ${\alpha}-amylase$ and ${\beta}-amylase$ of the strains examined became lower as the percentage of starch contents increased except in Rhizopus javanicus.

The anticancer effect of Bioconverted Danggui Liuhuang Decoction EtOH extracts in human colorectal cancer cell lines

  • Objective: The objective of our study was to investigate anti-cancer effects of Danggui Liuhuang Decoction extract bioconverted by protease liquid coenzyme of Aspergillus kawachii (DLD-BE), compared to a non-bioconverted DLD extract (DLD-E) and determine the underlying mechanisms. Methods: DLD-E and DLD-BE were evaluated for their ability to modulate these signaling pathways and suppress the proliferation of human colorectal cancer (CRC) cells, HCT-116, LoVo, and HT-29. The anti-cancer effects of DLD-E and DLD-BE were measured by using proliferation and migration assays, cell cycle analysis, Western blots, and real-time PCR. Results: In this study, treatment with DLD-E and DLD-BE at concentrations of 25-100 ㎍/mL inhibited proliferation and migration in human CRC cells. DLD-BE induced apoptotic cell death and decreased COX-2 expression in HT-29 cells. The mechanisms of action included modulation of the AKT and extracellular-signal-regulated kinase signaling cascades along with inhibition of COX-2 expression. The results demonstrate novel anti-cancer mechanisms of DLD-BE against the growth of human CRC cells. Thus, we propose that DLD-BE can be developed as a more potent supplement to inhibit colorectal tumor growth and intestinal inflammation than DLD-E.

포도 종류를 달리한 포도 약주의 발효 특성 (Fermentation Characteristics of Grape Yakju made with Three Kinds of Grape)

  • Three kinds of grape yakju (Kyoho, Campbell and Moru) and grape-free control were fermented with rice using koji of Aspergillus kawachii as a fermenting agent, and their fermentation characteristics were investigated. For the three yakju, reducing sugar content decreased slowly over 2 days fermentation after an initial rapid decrease. In case of Brix, the reducing sugar content increased gradually after increasing rapidly during the first 2 days of fermentation. The Kyoho and Moru groups displayed a gradual increase in reducing sugar content after an initial 2-day decrease, and the sugar content of the Campbell group increased gradually during fermentation. Uniformly, pH increased gradually after an initial 2-day decrease, however total acid increased continuously throughout fermentation. Alcohol content of yakju showed almost no change after increasing until day 6 (Kyoho) of day 4 (others), with the control displaying the highest content. Control $\alpha$-amylase activity decreased gradually after an initial 2-day increase, while that of the three yakju decreased throughout fermentation. Activities of $\beta$-amylase activities paralleled the results of reducing sugar. Concerning total anthocyanin, the Moru and Campbell groups displayed a rapid increase until day 2, a decrease until day 12, and a gradual increase thereafter. While the Kyoho and control groups displayed virtually no change after an initial 2-day increase until day 12, after which a gradual increase also occurred. In concerning sensory evaluation, Moru yakju scored highest for violetness, flavor of grape, and sweet taste, and also displayed a low alcohol content. These attributed Moru yakju the preferred choice.

Bioconversion enhances anti-oxidant and anti-inflammation activities of different parts of the Mulberry Tree (Morus alba L.), especially the leaf (Mori Folium)

  • The mulberry tree (Morus alba L.) has been traditionally used in Chinese medicine to treat inflammatory diseases. We investigated the effects of bioconversion on different components of the mulberry tree, and determined changes in the physiological activities. Ethyl acetate-soluble fractions of five different segments (fruit, Mori Fructus; leaf, Mori Folium; twig, Mori Ramulus; root, Mori Cortex; and mistletoe, Loranthi Ramulus) of the mulberry tree show enhanced anti-oxidant effects in the 2,2-diphenyl-1-picrylhydrazyl, and 2,2'-azinobis-(3-ethylvenzothiazoline-6-sulfonic acid) assays, and enhanced anti-inflammatory effects of lipopolysaccharide (LPS)-stimulated nitric oxide (NO) production in RAW 264.7 macrophages, after being treated with a crude enzyme extract from Aspergillus kawachii, in the following order of activity: Mori Folium>Mori Cortex>Mori Ramulus>Mori Fructus>Loranthi Ramulus. Ethyl acetate- soluble fraction of mulberry leaves (Mori Folium) that underwent bioconversion was most effective, and was devoid of any cytotoxicity. The fraction was also effective against mRNA expression of LPS-induced pro-inflammatory cytokines, such as inducible nitric oxide synthase, cyclooxygenase-2, tumor necrosis $factor-{\alpha}$, $interleukin-1{\beta}$, and interleukin-6. In addition, the fraction was effective in LPS-induced phosphorylation of mitogen-activated protein kinases and IKK, and $I{\kappa}B$ degradation, followed by translocation of the nuclear $factor-{\kappa}B$ from the cytoplasm to the nucleus. Thus, bioconversion increased the anti-oxidative and anti-inflammatory activities of the mulberry leaf.

국내에 유통되는 종국 곰팡이의 분류학적 특성 및 안전성 (Taxonomic Characterization and Safety of Nuruk Molds Used Industrially in Korea)

  • 장류와 주류 제조를 위하여 국내에서 유통되는 하경발효의 황국균(HK1), 수원발효의 황국균(SW101), 백국균(SW201), 충무발효의 된장용 황국균(CF1001), 간장용 황국균(CF1002), 청주용 황국균(CF1003), 주류용 백국균(CF1005)과 전통 메주에서 분리하여 산업화 준비를 하고 있는 황국균(KACC 93210)의 분류학적 특성과 안전성을 조사하였다. 공시한 6균주의 황국균은 모두 A. oryzae로 동정되었으며 이중 HK1, SW101, CF1001, CF1003은 분생포자를 생성하는 대(stipe)의 길이가 중간크기($711{\sim}1,121{\mu}m$, 중모(中毛))로써 서로 유사한 형태적 및 분자계통학적 특성을 나타내었으나 청주용인 CF1003이 다른 3균주에 비하여 포자를 다소 적게 그리고 균사를 다소 많이 생성하였다. 간장제조용인 CF1002는 대의 길이가 평균 $543{\mu}m$로서 짧았으며(단모(短毛)) omtA 유전자 분석에서 다른 황국균이 ICAo 그룹에 위치하는 반면에 IBLB-그룹에 소속되어 명쾌히 구분되었다. 전통메주에서 분리되어 산업화 과정에 있는 황국균 KACC93210 균주는 대(stipe)가 평균 $270{\mu}m$로 매우 짧았으며(초단모(超短毛)) 다른 황국균이 양털모양의 집락을 형성하는 반면 벨벳모양의 집락을 형성하여 형태적으로 쉽게 구분되었고 omtA 유전자에서도 공시한 황국균은 물론 세계적으로 보고된 어떤 황국균과도 염기서열이 서로 달랐다. 수원발효의 백국균(SW201)과 충무발효의 백국균(CF1005)은 모두 A. luchuensis (또는 A. luchuensis mut. kawachii)로 동정되었으며 집락 형태가 다소 상이하였으나 기타의 형태적 특징과 분자계통학적 특징에서 서로 구분되지 않았다. 황국균 6균주는 aflatoxin, cyclopiazonic acid, sterigmatocystin의 곰팡이 독소를 생성하지 않았고 백국균(A. luchuensis)은 유해한 독소를 생성하지 않는 것으로 이미 보고되었으므로 국내에 유통되는 황국균과 백국균은 모두 곰팡이독소에서 안전한 곰팡이임이 입증되었다.

막걸리의 제조를 위한 효소제의 개발연구 1 (Studies on improvement of manufacturing method of enzymic source for Maggerley(Korea wine) brewing(I))

  • It is necessry to develop and strengthen the activity of enzymic source which in low applied for maggerley brewing as an amylolytic and proteolytic starter, recently in this country the active and strong enzymic starter is required for the better brewing and to substitute another starch material for the present wheat flour. In this study, manufacturing method the strong enzymic source have been developed and established with use of raw wheat bran plus fungal strains of Rhizopus sp. and Aspergillus usamii the culture of starter. The results on experimental the activities of enzymic sources (stater) are as following ; 1. Method of making the enzymic source (starter) is to cultivate the strains of Asperguillus orzyae, Asp. kawachii, Asp usamii and Rhizopus sp. in the acid treated raw or heatboiled wheat bran. 2. The saccharogenic pwoer (S.P.) of enzymic source which consisted of raw bran plus fungi and cultured in it is generally stronger than those of heat-boiled bran plus fungi, the strongest power was shown in the culture of Rhizopus plus raw bran, and the next other is in mixture of Asp.usamii and Rhizopus on raw wheat bran. 3. The most strong alpha amylase activity was expressed in the plot of Asp.oryzae on heat-boiled wheat bran, the next was in the culture of Rhizopus nad Aspergillus usamii on raw wheatbran. 4. The most vigourous acidic proteinase activity was expressed in the micture of raw bran plus Asp. usamii and Rhizopus those were independentlu cu;tured before mixing for neutral proteinase activity, it was shown in the mixed culture of Asp. usamii and Rhizopus on raw wheat bran, the msot active alkaline proteinase activity of enzymic source was found in the plot of raw bran material. 5. For poly-preptidase activity in pH 6.5 it is found that the culture of Rhizopus and Asp.usamii on raw bran was most active among them of enzymic sources. 6. Generally, it is concluded that culture of fungi on acid treated raw wheat bran is stronger in its activity than those of heat boiled wheat bran, especially the culture of Rhizopus nad Asp.usamii on raw bran exhibited the most vigorous and non-polarized activity for all aspects, so it is considered to be most desirable enzymic stater in Korean Maggerley brewing and this would be able to substitute brewing material for the present wheat flour because of its strong and wide hand activity of amylolytic and proteolytic action.

벌꿀을 이용한 고액분리 숙성 탁주의 주질 향상과 고형분의 조절에 따른 품질유지기간 증대 (Improvement of Takju Quality by a Ripening-Fermentation Process Using Honey and Extension of Shelf Life by Control of Takju Mash Sediment)

  • 우리 나라 전통술인 탁주의 주질 개선을 위한 목적으로 천연성과 기능성을 가진 벌꿀을 사용하였고 저장기간의 연장을 위하여 술액의 침전물 양을 조절할 목적으로 본 연구를 진행하였다. 발효방식은 주모발효 후 3단담금 방식으로 하였고, 입국균은 Aspergillus awamori var. kawachi, 원료는 100% 백미, 알코올 발효 효모는 Saccharomyces cerevisiae를 사용하였다. 제성액의 고형분과 술액을 분리하여 벌꿀을 최종농도 5%(w/v) 첨가하여 5, 10, $15^{\circ}C$에서 23일간 숙성하였을 때 온도에 따른 유의한 차이는 보이지 않았으며 숙성기간 동안 알코올이 미소하게 감소했을 뿐 pH, 산도 및 총당량은 거의 일정하였다. 이와 같이 특징적인 술액의 차이는 없었지만 벌꿀숙성으로 제조한 탁주가 벌꿀 첨가 직후의 미숙성 탁주와 비교할 때 우수한 관능 테스트 결과를 보여주었다. 추가적으로 냉동 보관한 고형분을 100%, 50% 및 25% 비율로 벌꿀첨가 숙성된 술액과 혼합한 후 물을 보충하여 알코올 8%의 탁주를 제조하고 37일간 $10^{\circ}C$에서 날짜 별로 분석하였다. pH는 모든 탁주에서 4.5~4.8 사이를 큰 차이 없이 유지하였으며 고형분 첨가량이 많을수록 효모 세포수, 산도 및 알코올 함량은 상대적으로 증가하였고 총당량은 감소함을 보여 주었다. 즉 고형분의 양이 소량일수록 탁주의 주질이 안정하였다. 결론적으로 발효 후 고액 분리하여 상층액에 벌꿀을 첨가함으로써 저온 숙성기간 동안 주질의 큰 변화를 주지 않고 관능적으로 우수한 술액을 만들 수 있었으며, 고형분의 재혼합 비율을 조절할 경우 저장기간이 증대된 탁주를 제조할 수 있을 것이다.
