Isolation and Identification of Lactic Acid Bacteria for Preparation of Probiotics (생균제용 유산균의 분리 및 동정)

  • In order to prepare probiotics, a strain which has resistant against acid, bile salts and antibiotics was isolated from infant and anirnat feces. Among these isolated strains, TK- 19 showed a strong acid tolerance and growth at 500 $\mu g$/ml of Na-deoxycholate. Also strain TK-19 was resistant to kanarnycin (30 $\mu g$/ml), streptomycin (10 $\mu g$/ml), nisin (50 $\mu g$/ml) and tetracycline (10 $\mu g$/ml). The isolated strain TK-19 was identified as Streptococcus faeciam, and was named Streptococcus faeciurn TK-19.

Probiotic Properties of Bifidobacteria Isolated from Feces of Infants (유아 분변에서 분리한 비피도박테리아의 프로바이오틱스 기능성 연구)

  • Bifidobacteria are a prototype probiotic, which normally inhabit the intestinal tract of humans. In the present study, four species of Bifidobacterium isolated from the feces of infants were characterized. The tolerance for acid or bile salt, autoaggregation, and antibiotic resistance of the bacteria were examined. The four species were resistant to low pH, bile salts, and up to 3% bile acid. Autoaggregation rates were as high as 90%. The bacteria were consistently resistant to gentamicin, kanamycin, streptomycin, ciprofloxacin, and nalidixic acid. Due to their tolerance to environmental factors like acid and bile salts, B. longum MG723, B. breve MG729, B. bifidum MG731, and B. animalis subsp. lactis MG741 are potentially valuable as probiotics and may be useful for industrial application.

  • Commercial products with antibiotics like streptomycin as active ingredients have been used to control soft rot disease caused by Pectobacterium species for a long time. In this study, antibiotic resistance of twenty-seven Korean strains of Pectobacterium species including P. carotovorum, P. odoriferum, P. brasiliense, and P. parmenteri, which were previously shown to be susceptible to the bacteriophage phiPccP-1 was surveyed using a disk diffusion assay. While all strains were highly susceptible to ampicillin, kanamycin, chloramphenicol, tetracycline, and rifampicin, some strains showed weak susceptibility to 300 ㎍/ml of streptomycin. Furthermore, some of them are partially or completely resistant to commercial pesticides-Buramycinand streptomycin at the concentration of 250 ㎍/ml that is recommended by the manufacturer for streptomycin-based pesticides. These results indicate the presence of streptomycin-resistant Pectobacterium strains in South Korea, and the development of antibiotic alternatives to control soft rot is needed.

Antimicrobial Drug Resistance and R Plasmids of Serratia marcescens (Serratia marcescens의 항균제 내성 및 R plasmid)

  • Forty clinical isolates of Serratia marcescens were tested for their susceptibility to 19 antimicrobial drugs and studied on the molecular characteristics of R plasmids. Cefotaxime (Ct) was the most effective drug and only 2 (5%) strains were resistant to this drug. Thirteen to 18% of strains were resistant to cefoperazone (Cz), amikacin (Ak). and trimethoprim (Tp), and 28 to 40% were resistant to piperacillin (Pi), nalidixic acid (Na), gentamicin (Gm), and cefoxitin (Cx). A majority of strains were resistant to carbenicillin (Cb), tobramycin (Tp), kanamycin (Km), and cefamandole (Cd), and all to cephalothin. One half of the isolates were resistant to 10 or more drugs. $MIC_{90}$ of Pi to Gm-resistant strains (Gm') were 8 times higher than that to Gm-susceptible strains (Gm'), but $MIC_{90}$ of Ak, Cx, Ct, and Cz were almost the same between both Gm' and Gm' strains. Nine (23.7%) strains among 38 of multiply drug-resistant S. marcescens transferred conjugally their partial patterns of resistance to E. coli or Klebsiella strains, and two S. marcescens strains producing bacteriocin transferred their resistance to Klebsiella only, but not to E. coli. The plasmid profiles of S. marcescens were studied by the methods of SDS lysis and agarose gel electrophoresis. Twenty-four (60%) strains carried one to four plasmids of 1.4. to 144 Mdal, and conjugative R plasmids of 49 to 127 Mdal were noted in transconjugants. MIC levels of drugs in transconjugants were variable by the R plasmids and recipient strains.

Studies on Chronic Mastitis of Dairy Cattle in Kyungnam Province (경남지방(慶南地方)에서 발생(發生)한 젖소의 만성(慢性) 유방염(乳房炎)에 대한 연구(硏究))

  • Some investigations on chronic mastitis in dairy cattle in Kyungnam Province during the year 1982 were conducted with the special reference to the causative agents and their drug resistance. Milk samples from 46 isolated cases of chronic mastitis cattle were investigated bacteriologically and the organisms recovered were examined for their drug susceptibility against the major antibiotics used in this country by the use of disk diffusion susceptibility test. Four major causative agents involved in chronic mastitis were in order of prevalence Staphylococcus aureus (32.6%), Escherichia coli (28.3%), Streptococcus agalactiae (8.7%) and Candida albicans (8.7%). Staph. epidermidis, Streptococcus uberis, Klebsiella pneumoniae and Candida subtropicalis were found to be one of the minor agents. The majority of staphylococcal isolates were highly resistant to the most of antibiotics employed while 8% of them were resistant to gentamicin and 32% to chloramphenicol. The percentages of staphylococcal cultures resistant to penicillin, lincomycin. streptomycin, methicillin, oleandomycin, tetracycline, cephalothin, ampicillin and erythromycin were 100%, 96%, 96%, 92%, 84%, 84%, 80%, 76%, and 64% respectively. Streptococcal isolates were also highly resistant to the majority of the drugs used although 85.7% of them were susceptible to gentamicin. All Escherichia coli isolates were found to be resistant to erythromycin, lincomycin and penicillin while the majority of them were resistant to ampicillin (92.9%), carbenicillin (85.7%), oleandomycin (85.7%), streptomycin(85.7%), kanamycin (78.6%), methicillin (78.6%) and tetracycline (71.4%). The percentages of E. coli cultures resistant to gentamicin, nitrofurantoin, cephalothin and chloramphenicol were 21.4%, 21.4%, 35.7% and 50.0% respectively.

  • Salmonella strains isolated from blood in the Dong-san hospital, Taegu during the period from 1971 to 1986 were studied for species distribution, drug resistance, and R plasmids. The number of strains was 2,527 and all of them were classified into S. typhi and S. paratyphi A. Approximately 300 strains were isolated in the period from 1974 to 1976 and 1978, 268 in 1982, and 204 in 1983, but the numbers isolated in the 1980's have a tendency to decrease as compared with those of the 1970's. S. typhi occupied 85% or more of strains isolated until 1976, but the isolation frequency decreased yearly with some variation, and S. paratyphi A increased gradually from 1974. Only 4 strains of S. paratyphi A were resistant to some drugs, and the resistance was not transferred to E. coli by conjugation. S. typhi resistant to drugs were 15 in 1971 through 1973, 24 in 1974, and 13 in 1975, but afterwards only few resistant strains were isolated. These strains were resistant to two or more drugs; chloramphenicol(Cm), tetracycline(Tc), streptomycin(Sm), sulfisomidine(Su), ampicillin(Ap), and kanamycin(Km) and no strain resistant to other drugs tested was found. Strains resistant to 3 or less drugs didn't transfer the resistance to E. coli by conjugation. There were 15 strains resistant to four or more drugs, and were isolated in years from 1972 to 1976. These strains transferred the resistance to E. coli, and the resistance was considered to be mediated by R plasmids. Transfer frequency was higher at $25^{\circ}C$ than at $37^{\circ}C$ and patterns of transferred resistance were Cm, Tc, Sm, Su; Ap, Km; Cm. R plasmids having markers of Cm, Tc, Sm and Su were classified into Inc H1.

Establishment of an Efficient Agrobacterium Transformation System for Eggplant and Study of a Potential Biotechnologically Useful Promoter

  • An efficient and reliable Agrobacterium transformation procedure based on TDZ (thidiazuron)-induced organogenesis was established and applied to six Brazilian eggp1ant varieties. Optimum transgenic plants recovery was achieved upon the study of the following parameters affecting transformation efficiency, using F-100 variety as a model: i) explant source; ii) pre-culture period; iii) physical state of the pre-culture medium and iv) coculture conditions. The highest frequency of kanamycin-resistant calli derived from leaf explants (5%) was obtained without a pre-culture period and co-cultivation for 24 h in liquid medium followed by five days on solid RM (regeneration medium). For cotyledon explants, best results were achieved upon a pre-culture of 24 h in liquid RM and a co-cultivation period of 24 h in liquid RM followed by three days in solid RM, resulting in a transformation Sequency of 22.7%. Kanamycin-resistant organogenic calli were also obtained from cultivars Emb, Preta Comprida, Round nose Shaded, Campineira and Florida Market. The expression pattern of an epidermis-specific promoter was studied using transformants expressing a chimaeric construct comprised by the promoter Atgrp-5 transcriptionally fused to the coding region of the gus gene. The expression pattern was similar to that previously observed in tobacco and Arabidopsis thaliana, with preferential expression at the epidermis and the stem phloem. These results support the idea that the Atgrp-5 promoter can be used to drive defense genes in these tissues, which are sites of pathogen interaction and spread, in programs for the genetic improvement of eggplant.

  • The susceptibility of 536 isolates of Staphylococcus and 313 isolates of Streptococcus to a number of chemotherapeutics were studied. These organisms were isolated from bovine mastitis during 1973 and 1974. In addition to this, the rate of multiple resistance of 425 isolates of Staphylococcus and 164 isolates of Streptococcus, isolated in 1974, to the chemotherapeutics was analysed. The results obtained in this work were summerized as follows: 1. Staphylococcus and Streptococcus isolated in 1974 showed a higher resistance, with 3 exceptions of chemotherapeutics, than the isolates of 1973. 2. Staphylococcus isolated in 1973 and 1974 showed a higher susceptibility than Streptococcus. 3. The strains of Staphylococcus resistant to colistin were 39 strains (9.2%), to colistin and sulfisoxazole 33 (7.8%), to streptomycin, kanamycin, colistin and sulfisoxazole 20 (4.7%), and to penicillin, colistin and sulfisoxazole 18 (4.2%). 4. The strains of Streptococcus resistant to colistin were 17 strains (10.4%), to streptomycin, kanamycin, colistin and sulfisoxazole 13 (7.9%), to colistin and sulfisoxazole 11 (6.7%) and to penicillin, colistin and sulfisoxazole 11 (6.7%).

Antibacterial Activities of Mushroom Liquid Culture Extracts Against Livestock Disease-Causing Bacteria and Antibiotic Resistant Bacteria (가축질병 세균 및 항생제 내성 세균에 대한 버섯 균사체 배양 추출물의 항균 활성)

  • The ethyl acetate extracts from the liquid cultures of Coriolus versicolor, Phellinus linteus, and Hericium erinaceus showed significant antibacterial activities against Escherichia coli K88, E. coli K99, E. coli 987P, and Salmonella typhimurium 14058 causing bacterial diarrhea in Korean house pigs and chicken. Of these extracts, Coriolus versicolor extract showed the highest antibacterial activity. In addition, these extracts also showed significant growth inhibition against Staphylococcus aureus CARM3230 and E. coli CARM1381 which are known as kanamycin and ampicillin-resistant strains. These results showed that the mushroom extracts could be developed as a livestock feed additives that can replace commercial antibiotics, and also could be good resources for the development of a new antibacterial agent.

  • The rational approach to antimicrobial therapy of infected root canals is based on accurate identification of the infecting organism and on the organism's susceptibility to antimicrobial agents as measured by standardized techniques. In establishing criteria for the selection of antibiotics, a susceptibility test should be performed. The purpose of this study was to investigate the susceptibility of 224 aerobic and anaerobic microbial strains isolated from infected root canals to various antibiotics. This was performed by using 7 antibiotic sensi-disc: Penicillin (10 units), Ampicillin (10 mcg), Tetracycline (30 mcg), Streptomycin (10 mcg), Kanamycin (30 mcg), Lincomycin (2 mcg), and Clindamycin (2 mcg). The results were as follows; 1. Strains isolated from infected root canals was shown to be most susceptible to Clindamycin, while Streptomycin exhibited least antibacterial properties. 2. Anaerobes were found to be susceptible to Penicillin, Ampicillin, and Clindamycin. 3. Streptomycin and Kanamycin were shown to be effective against Staphylococcus aureus and Staphylococcus epidermis, however, unidentified G(+) cocci organisms were found to be resistant to these agents. 4. Bifidobacterium sp. was susceptible to Lincomycin while G(+) rods were resistant to it. 5. Staphylococcus aureus, Micrococcus sp., and anaerobes were highly susceptible to Clindamycin. 6. All of the antibiotics tested were shown to be very effective against Eubacterium sp.
