형질전환 상추에서 배추 Glutathione Reductase 유전자의 발현 (Expression of Chinese Cabbage Glutathione Reductase Gene in Lettuce (Lactuca sativa L.))

  • 상추의 자엽 조직을 GR 유전자가 도입된 A. tumefaciens LBA 4404와 2일간 공동배양후, carbenicillin 500mg/L, kanamycin 50mg/L NAA 0.1mg/L와 2iP 1.0 mg/L가 함유된 MS 재분화배지에 옳겨 약 4주후에 kanamycin 저항성 개체를 얻었다. 형질전환된 것으로 추정되는 식물체는 kanamycin 100mg/L가 함유된 MS선발배지에서 생존하였다. PCR 분석결과, GR 유전자가 형질전환체의 게놈상에 삽입되어 있음을 확인하였다. 형질전환체의 Southern blot 분석을 통하여 ECL-labelling된 GR 유전자와 동일한 것으로 판단되는 약 1.8 kb 위치에서 밴드를 확인할 수 있었다. RT-PCR 분석으로 GR 유전자가 전사됨을 확인할 수 있었다. 개화후 이들 개체의 종자를 받아 NPTII 유전자의 발현여부를 조사한 결과 R$_1$ 세대에서도 NPTII 유전자가 발현됨을 확인하였다.

채소작물의 형질전환을 위한 재분화체계 확립 및 항생제 검정 (Establishment of Regeneration System and Antibiotic Sensitivity Test for Transformation of Various Vegetable Crops)

  • This study was conducted to determine the concentrations of plant growth regulators required for regeneration and the concentrations of antibiotics for the selection of transformed regenerants from lettuce, musk melon and tomato. The optimal concentrations of plant growth regulators for shoot formation were NAA 0.1 mg/$\ell$ +BA 0.1 mg/$\ell$ for lettuce, NAA 0.01 mg/$\ell$ +BA 2.0 mg/$\ell$ for melon and NAA 0.1 mg/$\ell$ +BA 0.5 mg/$\ell$for musk melon. Shoot induction from tomato, lettuce and melon was completely inhibited by 30 mg/$\ell$ or higher concentrations of kanamycin. Shoot formation from mu나 melon was not affected by kanamycin up to 40 mg/$\ell$, but was reduced in the presence of 50 mg/$\ell$ and completely inhibited by 100 mg/$\ell$. Shoot formation of all four crops was completely inhibited by higromycin at 10 mg/$\ell$. Both carbenicillin and cefatoxinme did not show any negative effects on shoot formation.

  • 상추에 제초제 저항성을 도입하기 위해 상추 자엽 조직을 NPTII-GUS 및 제초제 저항성 유전자 (PAT)가 삽입된 A. tumefaciens MP 90과 2일간 공동배양한 다음 0.1 mg.L$^{-1}$ NAA, 1.0 mg.L$^{-1}$ 2ip, 50 mg.L$^{-1}$ kanamycin, 500 mg.L$^{-1}$ carbenicillin을 첨가한 MS배지에 배양하여 식물체를 재분화시켰다. 재분화 식물체는 kanamycin 내성 검정을 통해 NPTII 유전자의 도입과, PCR, Northern blot 분석을 통해 제초제 저항성 유전자가 식물체의 게놈상에 삽입된 형질전환체임이 확인되었다. 또한 1500배액의 농도로 Basta를 살포한 결과 살포 10일 후 대조 식물체는 완전히 고사하는 데 반해 형질전환체는 지속적인 생육을 보여 얻어진 형질전환 상추가 제초제 저항성 개체임이 확인되었다.

  • Atrophic rhinitis (AR) is the one of important respiratory diseases and causes severe economic losses in pig industry. Severe attempts have been made to reduce the economic losses by preventing the disease. One of the methods is the spraying of antibiotics into nasal cavity of piglets. Recently, the efficacy of the spraying with kanamycin and gentamicin was reduced in the Korean swine industry. Therefore, the preventive methods have been required to be changed based on the antimicrobial susceptibility profiles of causative agents of swine AR. Based on the current situations of this disease, Bordetella (B.) bronchiseptica and Pasteurella (P.) multocida 4D were isolated from pigs with clinical signs of AR. The isolation rates of B. bronchiseptica and P. multocida 4D were 58.5% and 32.9%, respectively. In the antimicrobial susceptibility test, the bacteria were resistant to kanamycin and gentamicin which have been used as the spraying agents, but they were susceptible to amikacin. A new spraying agent was developed using amikacin using ${\beta}$-glucan and yakbaltag as supplementary agents. Field efficacy of the agent was carried out with different schedule. The results from this study suggested that the newly developed spraying agents might be helpful to prevent AR in swine.

형질전환된 꽃양배추에서 Proteinase Inhibitor II 유전자의 발현 (Expression of Proteinase Inhibitor II gene in Transgenic Flowering Cabbage, Brassica oleracea var. acephala DC.)

  • PI-II cDNA가 도입된 식물발현 벡터인 pGA875를 가진 A.tumefaciens LBA4404를 이용하여 꽃양배추의 하배축 조직에 형질전환하여 식물체를 재분화 시켰다. Dot blot 분석으로 PI-II 유전자가 전사됨을 확인할 수 있었다. 또한 이들 개체를 담배거세미나방 유충을 이용하여 생물검정한 결과 대조구에 비해 형질전환체 잎의 섭식정도가 현저히 떨어지는 것을 알 수 있었다. 개화후 이들 개체의 종자를 받아 후대검정을 실시하였을 때 27,4%가 kanamycin내성을 가진 꽃양배추로 확인되었다.

  • 우리나라의 주요 개량 포플러의 하나인 현사시 3호를 대상으로 Arobacterium tumefaciens MP 90/PAT을 이용하여 비선택성 제초제인 Basta에 내성을 갖는 형질전환체를 획득하고자 본 실험을 수행하였다. 기내 배양된 엽절편을 재료로 균주에 10분간 감염, 2일간 공조 배양 및 3주간 선발배지에서 배양으로 kanamycin에 내성을 갖는 잠정적인 형질전환 캘러스를 유도하였다. 선발된 캘러스로부터 배양 4주 후 줄기를 유도하였으며, 증식된 줄기는 발근시켜 완전한 형질전환체를 얻었다. 형질전환체는 PCR기법 및 Southern blot 분석으로 PAT 유전자 전이를 확인하고. 유전자의 발현은 GUS 유전자의 발색 반응으로 확인하였다. 형질전환체는 폿트로 이식하여 4주간 생장시킨 후 제초제 Basta로 살포한 결과 대조구 식물체는 고사되었으나, 형질전환체는 정상적인 생육이 가능하였다

  • 항균제내성(抗菌劑耐性)인 Salmonella typhi를 공시(供試)하여 chloramphenicol(Cm), tetracycline(Tc), ampicillin(Ap), kanamycin(Km) 및 rifampicin(Rif)의 항균작용(抗菌作用)을 한천평판희석법(寒天平板稀釋法)(평판법(平板法))과 broth 희석법(稀釋法)(broth법(法))으로 비교(比較)하였다. 공시균(供試菌)의 최소발육저지농도(最少發育沮止濃度)(MIC)를 보면 Cm, Tc 및 Rif에 있어서는 평판법(平板法)과 broth법(法) 사이에 유의(留意)할만한 차이(差異)는 없었으나 Km에 있어서는 broth법(法)의 MIC가 평판법(平板法)보다 높은 것이 약(約) 반수(半數)있었고 Ap에 있어서도 소수(少數) 있었다. Cm과 Rif는 대부분(大部分)의 공시균(供試菌)에 대(對)하여 정균적(靜菌的)으로 작용(作用)하였으나 Tc, Ap 및 Km은 살균적(殺菌的)으로 작용(作用)하였고 그 작용(作用)은 균주(菌株)에 따라 차이(差異)가 있었다.

한우(韓牛) 및 유우(乳牛)의 생식기내(生殖器內) 세균분리(細菌分離) 동정(同定) 및 약제감수성(藥劑感受性) (Isolation, Identification and Drug Susceptibility of Bacteria from Cow Genital Organs)

  • This study was conducted to determine the microorganisms inhabitating in cow genitalia and their antimicrobial drug susceptibility. During the period between July, 1985 and February, 1986, a total of 111 cow genitalia, 58 from Korean native and 53 from dairy cow, were sampled at three abattoirs. Gross pathological examination and bacterial isolation and identification were performed from the genital samples. In addition antimicrobial drug susceptibility test for the microorganisms isolated, some synergistic activity among drugs were examined on the major organism isolated from the cases of endometritis and pyometra. The results are summerized as follows: 1. Among the bacteria isolated from the genitalia, Staphylococcus spp., C. pyogenes, E. coli, Proteus spp., Streptococcus spp., Bacillus spp. were most frequently isolated whereas the genera of Pasteurella, Pseudomonas, Klebsiella and Yersina were detected far less frequently. 2. In Korean native cow the genera of Straphylococcus and Steptococcus were more isolated than dairy cow while in dairy cow the genera of Corynebacterium, Proteus, Escherichia were more of ten isolated than Korean native cow. 3. From cow genital organs showing lesions of endometritis and prometra, C. pyogenes was most frequently isolated, the isolation rate being 60 percent, and follow by Staphylococcus spp., Proteus spp., E. coli and Pasteurella spp. in the order. 4. Antimicrobial drug susceptibility test conducted on the major organisms isolated showed that all the isolates were susceptible to gentamicin, cephalosporin and sulfisoxazole, but resistant to tetracycline and penicillin. 5. Twenty-nine isolates of C. pyogenes were submitted to the synergistic activity test of cephalosporin, kanamycin and streptomycin with penicillin. Synergists were demonstrated in 90 percent, 31 percent and 27 percent of isolates examined by the combined use of penicillin and cephalosporin, penicillin and kanamycin, penicillin and streptomycin, respectively. About 10 percent of the isolates were found to be indifferent by the synergism test.

돼지 호흡기질병 병인체의 항균제 감수성 조사 (Antimicrobial susceptibility features of porcine respiratory bacterial pathogens by modified broth dilution method)

  • There are several main antibacterial susceptibility tests, such as agar dilution method, broth dilution method and disk diffusion technique. Especially, for minimal inhibitory concentration (MIC) test, agar dilution method has been widely used. But that method is so complicated and bothering that it's difficult to treat a large amount of strains. On the other hand, modified broth dilution method(add 1% glucose and 0.018% phenol red as a pH indicator to broth) is fast and easy to perform. Most of all, it can visualize the result by color. The MICs of 22 antibiotics Including penicillins, aminoglycosides, cephalothin, chloramphenicol, lincomycin, ceftiofur, vancomycin and quinolones, erythromycin, colistin. sul-fadimethoxine, trimethoprim for arcanobacterium pyogenes 14 strains, actinobacillus pleuropneu-moniae 41 strains and pasteurella multocida 37 strains, which were collected from porcine during 1996 ∼ 1999, were determined by modified broth dilution method. Actinobacillus pleuropneumoniae was highly susceptible to all kinds of quinolones such as ciprofloxacin, enrofloxacin and norfloxacin and to all aminoglycosides, like gentamicin, apramycin, kanamycin and ampicillin, cephalothin and ceftiofur. But It was quite resistant to solfadimethoxin, colistin and vancomycin. Pasteurella multocida was found to have high susceptibility to ampicillin, cephalothin, chlorampenicol and gentamicin but had mid-degree susceptibility to other aminoglycosides. In addition, it was susceptible to norfloxacin and nalidixic acid, but not to newer fluoroquinolone like ciprofloxacin and enrofloxacin and it was resistant to colistin and kanamycin. Arcanobacterium pyogenes was highly susceptible to most of quinolones such as cipoofloxacin, enrofloxacin and norfloxacin and gentamicin and penicillin G. But it also obtained high resistance against the early quinolone, nalidixic acid and aminoglycosides such as amikacin, apramycin and kanamycin and erythromycin, chlorampenicol, tetracyclin and vancomycin.

항균제내성의 검사요령과 임상적응용 (Technique of Drug Tolerance Test for Selection of Antibacterial Agents and It′s Clinical Value)

  • The author carried out bovine mastitis test by California Mastitis test(CMT) for the milk from dairy cows in the surburbs of Taejeon. In order to select prefer commercial therapeutic antibacterial agents for mastitis treatment, Staphylococcus aureus were isolated from the CMT positive milk and the strains were tested for the tolerance test to the agents. The results obtained were summarized as follows : 1. The tolerance test appeared graduate tolerance in cases with continuously repeat of therapeutic agents. 2. The antibacterial agents revealed strong tolerance were D-and E-ointments. 3. The antibacterial agents revealed suspect tolerance were A-and F-ointments and ampicillin, penicillin, kanamycin and streptomycin. 4. The antibacterial agents revealed non or rare tolerance were B-and G-ointments and chloramphenicol, erythromycin and oxytetracycline. 5, It is concluded that the use of 3-day-interval in turn with antibacterial agents selected by tolerance test may be beneficial.
