An ethanol-producing yeast strain, CHFY0201, was isolated from soil in South Korea using an enrichment technique in a yeast peptone dextrose medium supplemented with 5% (w/v) ethanol at $30^{\circ}C$. The phenotypic and physiological characteristics, as well as molecular phylogenetic analysis based on the D1/D2 domains of the large subunit (26S) rDNA gene and the internally transcribed spacer (ITS) 1+2 regions, suggested that the CHFY0201 was a novel strain of Schizosaccharomyces pombe. During shaking flask cultivation, the highest ethanol productivity and theoretical yield of S. pombe CHFY0201 in YPD media containing 9.5% total sugars were $0.59{\pm}0.01$ g/l/h and $88.4{\pm}0.91%$, respectively. Simultaneous saccharification and fermentation for ethanol production was carried out using liquefied cassava (Manihot esculenta) powder in a 5-l lab-scale jar fermenter at $32^{\circ}C$ for 66 h with an agitation speed of 120 rpm. Under these conditions, S. pombe CHFY0201 yielded a final ethanol concentration of $72.1{\pm}0.27$ g/l and a theoretical yield of $82.7{\pm}1.52%$ at a maximum ethanol productivity of $1.16{\pm}0.07$ g/l/h. These results suggest that S. pombe CHFY0201 is a potential producer for industrial bioethanol production.
The optimum conditions for the production of Lentinus edodes mycelia and it's hypoglycemic effect was studied. Optimum pH and temperature for the production of mycelia in shaken flask culture were 5 and $30^{\circ}C$, respectively, for 24 days cultivation. Culture period for maximum production of mycelia (8.13 g/l) in 5-l jar fermenter cultivation was shortened as much as 6 days compared to shaken flask culture. The mycelial dose of 5 % proved almost equally effective in lowering the plasma glucose, total cholesterol, and triglyceride level as much as 23.0, 20.7, and 27.1%, respectively. The values of alanine transaminase and aspartate transaminase were also estimated and exhibited a substantial reduction (29.6 and 34.3%, respectively) in activity after the administration of dried mycelia.
The production of doxorubicin by a mutant of Streptomyces peucetius subsp. caesius was studied. The optimal culture conditions, such as inoculum size and medium composition were established to improve the productivity of doxorubicin. The optimal medium composition was found to be 4% maltose, 0.5% HEPES, 0.02% $K_2HPO_4$, 0.01% $MgSO_4$. As an antiform agent, 0.001% KG(10% Adekanol+10% Silicone) was suitable one among various agents. Culture was carried out in 2.5 L jar-fermenter with different aeration rates of 1.5, 1.0, and 1.5 v/v min. The maximum production of doxorubicin(29 mg/l) was obtained at 1.5 v/v min of aeration rate, and even at 1.0 v/v min, the production of doxorubicin was increased up to 15% compared with that of shake-flask culture.
The mycelial dispersed growth of Cordyceps sinensis was optimized in submerged batch culture at initial pH of 5.0, 150 rpm, and $25^{\circ}C$. The morphological data showed much more dispersed growth of C. sinenesis at initial pH of 5.0. Also, projected area, main hyphal length and number of tips for the mycelial growth of initial pH 5.0 were higher than those of other initial pHs. The industrial medium for mycelial production of C. sinensis was determined to be molasses of 100 g and crushed brewery yeast of 10 g per liter as carbon and nitrogen sources, respectively. With these culture conditions, the maximum production of mycelia was approximately 30.0 g per liter by batch culture in 5-liter jar fermenter with no controlled pH. This result suggests that large-scale mycelia production of C. sinensis may be possible in submerged batch culture. The hot water extract of mycelia from C. sinensis was mainly composed of 83.0% carbohydrate, 11.8% protein, 1.9% lipid, and 2.4% ash and there were present glucose, mannose, galactose, and arabinose as molar ratio of 8.79 : 2.59 : 1.34 : 1.0 in the carbohydrate, respectively. In the experiment using spleen cell and macrophage, the extract showed potent mitogenic and immuno-stimulating activities and among various components, an important factor that contribute to the immunological activities was turned out to be carbohydrate moiety.
Lactococcus lactis SA72 from Jeot-gal (Korean traditional fermented fish foods) produces lacticin SA72. The influence of several parameters on the fermentative production of lacticin SA72 by Lactococcus lactis SA72 was studied. MRS medium among several media was selected for enhanced bacteriocin production. The mean growth rate and bacteriocin productivity of L. lactis SA72 increased as the initial pH of the media increases. The highest lacticin SA72 activity was detected 3,200 AU/ml at pH 6.0, $32^{\circ}C$, and 1% (inoculum size, v/v) in the jar fermenter. Enhanced production of lacticin SA72 was investigated by a fed-batch cultivation with the intermittent feeding of the concentrated glucose solution. Under the optimized conditions, lacticin SA72 activity finally reached to 6,400 AU/ml.
In order to produce alcohol for the alternative energy from dried powder of Jerusalem artichoke was investigated with Kluyveromyces marxianus UCD(FST)55-82, which was reported to assimilate inulin. The optimal condition for the production of ethanol by K. marxianus was elucidated to be incubation temperature of $30^{\circ}C$, initial pH 5.44, agitation of 100 rpm, 1,000 ml of medium in a 2.5l-vessel, anaerobic state, and inoculation of 2.5%(v/v). Addition of antifoam A concentrate(si1icon polymer) of 0.01% and urea of 0.1% increased the concentration of ethanol effectively. The optimized condition showed ethanol concentration of 6.8%(v/v) in Jerusalem artichoke liquid medium, production yield of 91.91% and productivity of 2.71 g/l/hr during the first day and 0.71g ethanol/l/hr during four days of incubation.

In a study of hydrogen-producing bacteria, strain T4384 was isolated from rice field samples in the Republic of Korea. The isolate was identified as Enterobacter sp. T4384 by phylogenetic analysis of 16S rRNA and rpoB gene sequences. Enterobacter sp. T4384 grew at a temperature range of $10-45^{\circ}C$ and at an initial pH range of 4.5-9.5. Strain T4384 produced hydrogen at 0-6% NaCl by using glucose, fructose, and mannose. In serum bottle cultures using a complete medium, Enterobacter sp. T4384 produced 1,098 ml/l $H_2$, 4.0 g/l ethanol, and 1.0 g/l acetic acid. In a pH-regulated jar fermenter culture with the biogas removed, 2,202 ml/l $H_2$, 6.2 g/l ethanol, and 1.0 g/l acetic acid were produced, and the lag-phase time was 4.8 h. Strain T4384 metabolized the hydrolysate of organic waste for the production of hydrogen and volatile fatty acid. The strain T4384 produced 947 ml/l $H_2$, 3.2 g/l ethanol, and 0.2 g/l acetic acid from 6% (w/v) food waste hydrolysate; 738 ml/l $H_2$, 4.2 g/l ethanol, and 0.8 g/l acetic acid from Miscanthus sinensis hydrolysate; and 805 ml/l $H_2$, 5.0 g/l ethanol, and 0.7 g/l acetic acid from Sorghum bicolor hydrolysate.
The ginsenoside Rh2 has strong anti-cancer, anti-inflammatory, and anti-diabetic effects. However, the application of ginsenoside Rh2 is restricted because of the small amounts found in Korean white and red ginsengs. To enhance the production of ginsenoside Rh2-MIX (comprising 20(S)-Rh2, 20(R)-Rh2, Rk2, and Rh3 as a 10-g unit) with high specificity, yield, and purity, a new combination of enzymatic conversion using the commercial enzyme Viscozyme L followed by acid treatment was developed. Viscozyme L treatment at pH 5.0 and $50^{\circ}C$ was used initially to transform the major ginsenosides Rb1, Rb2, Rc, and Rd into ginsenoside F2, followed by acid-heat treatment using citric acid 2% (w/v) at pH 2.0 and $121^{\circ}C$ for 15 min. Scale-up production in a 10-L jar fermenter, using 60 g of the protopanaxadiol-type ginsenoside mixture from ginseng roots, produced 24 g of ginsenoside Rh2-MIX. Using 2 g of Rh2-MIX, 131 mg of 20(S)-Rh2, 58 mg of 20(R)-Rh2, 47 mg of Rk2, and 26 mg of Rh3 were obtained at over 98% chromatographic purity. Then, the anti-cancer effect of the four purified ginsenosides was investigated on B16F10, MDA-MB-231, and HuH-7 cell lines. As a result, these four rare ginsenosides markedly inhibited the growth of the cancer cell lines. These results suggested that rare ginsenoside Rh2-MIX could be exploited to prepare an anti-cancer supplement in the functional food and pharmaceutical industries.
The biosynthesis of lovastatin, a cholesterol lowering agent formed by the filamentous fungus Aspergillus terreus, was examined in a 2.5 L jar fermenter. In batch bioreactor cultures conducted at various agitation rates, 400 rpm showed the best result in terms of lovastatin production. Notably, the effect of pH on lovastatin biosynthesis was found to be significant: when the pH was controlled at around 5.8 during the whole fermentation period, lovastatin concentration reached 598 mg/L, which is much hihger than the amounts obtained by pH-uncontrolled and pH 7.4-controlled fermentations. In addition, both L-histidine and L-tryptophan were observed to be favorable amino acids for the enhancement of lovastatin production when 6 g/L of the respective amino acids were supplemented at the beginning of the fermentation period. By further optimization of the production media and the physical environment, lovastatin production was increased to 836 mg/L (3.5 mg/L/hr) which is approximately 10 times higher than the productivity of the basic control culture.
The aim of this study attempted to verify the possibility of bioethanol production using wasted medium density fiberboard (wMDF). In order to produce bioethanol from wood cellulosic materials must be carried out the process of pretreatment, saccharification, fermentation and distillation. First, the wMDF was pretreated using sodium chlorite and pretreated wMDF was prepared to 8% slurry and then slurry was saccharified with the commercial enzyme (Cellic CTec3). The fermentable sugar and pH of saccharified substrate were about 5.5% glucose and 4.4, respectively. Herein we compared the results of ethanol yield according to the nutrients added or without addition to increase ethanol yield. Ethanol fermentation was finished in about 24 hours, but it was delayed in experimental group without nutrients. Ethanol content and fermentation ratio of the final fermented mash prepared by utilizing jar fermenter was 25.40 g/L and 86.64%, respectively. At this time, the maximum ethanol productivity was confirmed as 1.78 g/Lh (ethanol content 21.38 g/L, 12 h), and the overall ethanol productivity was 1.05 g/Lh (ethanol content 25.27 g/L, 24 h). Using fermented liquid we could produced bioethanol 95.37% by continuous distillator packed with copper element in laboratory scale. These results show that wMDF has a potential valuable for bioethanol production.
