Benzyl Isothiocyanate-Induced Cytotoxicity via the Inhibition of Autophagy and Lysosomal Function in AGS Cells

  • Gastric adenocarcinoma is among the top causes of cancer-related death and is one of the most commonly diagnosed carcinomas worldwide. Benzyl isothiocyanate (BITC) has been reported to inhibit the gastric cancer metastasis. In our previous study, BITC induced apoptosis in AGS cells. The purpose of the present study was to investigate the effect of BITC on autophagy mechanism in AGS cells. First, the AGS cells were treated with 5, 10, or 15 μM BITC for 24 h, followed by an analysis of the autophagy mechanism. The expression level of autophagy proteins involved in different steps of autophagy, such as LC3B, p62/SQSTM1, Atg5-Atg12, Beclin1, p-mTOR/mTOR ratio, and class III PI3K was measured in the BITC-treated cells. Lysosomal function was investigated using cathepsin activity and Bafilomycin A1, an autophagy degradation stage inhibitor. Methods including qPCR, western blotting, and immunocytochemistry were employed to detect the protein expression levels. Acridine orange staining and omnicathepsin assay were conducted to analyze the lysosomal function. siRNA transfection was performed to knock down the LC3B gene. BITC reduced the level of autophagy protein such as Beclin 1, class III PI3K, and Atg5-Atg12. BITC also induced lysosomal dysfunction which was shown as reducing cathepsin activity, protein level of cathepsin, and enlargement of acidic vesicle. Overall, the results showed that the BITC-induced AGS cell death mechanism also comprises the inhibition of the cytoprotective autophagy at both initiation and degradation steps.

Evaluation of Antimicrobial Activity of Allyl Isothiocyanate (AITC) Adsorbed in Oyster Shell on Food-borne Bacteria

  • 굴 패각은 한국 남쪽의 해안의 바다 양식 폐기물로써 처리문제로 대두되고 있다. 폐기물인 굴 패각을 실용화하기 위해서, 현지 회사에서 구입한 소성된 굴 패각(COS)에 AITC (allyl isothiocyanate)를 흡착시킨 후 식품 감염 질병을 일으키는 박테리아에 대해 성장억제능력을 시험하였다. COS 분말은 Escherichia coli, Staphylococcus aureus, Salmonella thyphimurium 균에 대해 1% 정도의 농도에서 세균 발육 억제 효과를 3에서 5 log 10 CFU/mL로 나타냄으로써 세균 발육 억제 효과를 보였으며, 순수 AITC의 MIC 결과는 Escherichia coli, Staphylococcus aureus, Salmonella thyphimurium에 대해 각각 1 mg/mL, 0.8 mg/mL 0.7 mg/mL을 나타내었다. 소성된 굴 패각은 소성과정에서 기공이 생성되어 225 mg/g의 AITC를 흡착하였고, FTIR 결과로 COS에 AITC가 흡착이 되었음을 확인하였다. 입자의 특성은 매우 미세한 입자 크기 및 높은 선상 표면을 나타내었다. AITC가 흡착된 소성된 굴 패각(ACOS)은 1% 농도에서 완전히 세균 세포를 억제함에 따라, ACOS는 COS보다 더 나은 항균활성을 나타냄을 확인하였으며, 이는 박테리아에 대해 AITC와 소성된 굴 패각의 상승효과가 있음을 나타내었다.

Activity of Allyl Isothiocyanate and Its Synergy with Fluconazole against Candida albicans Biofilms

  • Candidiasis involving the biofilms of Candida albicans is a threat to immunocompromised patients. Candida biofilms are intrinsically resistant to the antifungal drugs and hence novel treatment strategies are desired. The study intended to evaluate the anti-Candida activity of allyl isothiocyanate (AITC) alone and with fluconazole (FLC), particularly against the biofilms. Results revealed the concentration-dependent activity of AITC against the planktonic growth and virulence factors of C. albicans. Significant (p <0.05) inhibition of the biofilms was evident at ${\leq}1mg/ml$ concentrations of AITC. Notably, a combination of 0.004 mg/ml of FLC and 0.125 mg/ml of AITC prevented the biofilm formation. Similarly, the preformed biofilms were significantly (p <0.05) inhibited by the AITC-FLC combination. The fractional inhibitory concentration indices ranging from 0.132 to 0.312 indicated the synergistic activity of AITC and FLC against the biofilm formation and the preformed biofilms. No hemolytic activity at the biofilm inhibitory concentrations of AITC and the AITC-FLC combination suggested the absence of cytotoxic effects. The recognizable synergy between AITC and FLC offers a potential therapeutic strategy against biofilm-associated Candida infections.

[1,2,4]-Triazole 유도체의 합성 및 항암활성 (Synthesis of [1,2,4]-Triazole Derivatives and Their Anticancer Activities)

  • 2-Chlorobenzoyl hydrazine refluxed with benzoyl isothiocyanate and phenyl isothiocyanate in ethanol for 3 hours to give benzamide derivative (1) and anilinederivative (2) on yield of 71%and 95%, respectively. Benzamide derivative (1) reacted with ethanolic sodium hydroxide on reflux to afford cyclization product (3), followed by general substitution reaction of two steps to give acetamide (5), and derivatived acetamides 7a-7k, while aniline derivative (2) reacted with ethanolic sodium hydroxide on reflux to afford another cyclization product (4). Thiol (4) reacted with N-phenyl chloroacetamide in the presence of potassim carbonate to give acetamide derivative (6). Compounds 1-7kwere evaluated for their growth inhibition against five cancer cell lines, including human lung carcinoma (A-549), human prostate cancer (DU145), human colon adenocarcinoma (HT-29), human malignant melanoma (SK-MEL-2) and human ovary malignant ascites (SK-OV-3) with sulforhodamine B (SRB) assay. All compounds (1-7k) showed low inhibition activities under 50% on 100M concentration.

Glucosinolates (3-Butenyl Isothiocyanate, Total Glucosinolates) Change in Seeds and Young Seedlings on Chinese Cabbages

  • The main objective of this study was to investigate the patterns of glucosinolates (3-butenyl isothiocyanate, total glucosinolates) in different parts of young seedling and seeds on Korean Chinese cabbages 55 days and Winter pride cultivars. For determination of glucosinolates, two cultivars of Chinese cabbages seeds and different parts of 1-day-old, 3-day-old, 5-day-old, and 7-day-old seedlings were used for analytical sample preparation provided with an anion exchanges column, and measured by GC and UV-Visible Spectrophotometer. 3-Butenyl ITC concentration was the highest in the cotyledon and the hypocotyl parts of 55 days cultivar seedling. In the cotyledon part of Winter pride cultivar seedling, 3-butenyl ITC amount was increase to 3-day-old seedling and then reduced. The cotyledon of 55 days cultivar contained the highest concentration of total glucosinolates while those were increased in the hypocotyl and decreased by degrees in the root. Total glucosinolates amounts were increased to 5-day-old seedling and then decreased in the cotyledon and the hypocotyl parts of seedling on Winter pride cultivar. There was no significant difference in the root part. In the seeds, both of 3-butenyl ITC and total glucosinolates, 55 days cultivar concentration was higher than Winter pride cultivar. The study has shown the variability in 3-butenyl ITC and total glucosinolates concentrations among cultivars, growth stages, and parts.

Sulforaphane is Superior to Glucoraphanin in Modulating Carcinogen-Metabolising Enzymes in Hep G2 Cells

  • Glucoraphanin is the main glucosinolate found in broccoli and other cruciferous vegetables (Brassicaceae). The objective of the study was to evaluate whether glucoraphanin and its breakdown product sulforaphane, are potent modulators of various phase I and phase II enzymes involved in carcinogen-metabolising enzyme systems in vitro. The glucosinolate glucoraphanin was isolated from cruciferous vegetables and exposed to human hepatoma cell line HepG2 at various concentrations (0-25 ${\mu}M$) for 24 hours. Glucoraphanin at higher concentration (25 ${\mu}M$) decreased dealkylation of methoxyresorufin, a marker for cytochrome P4501 activity; supplementation of the incubation medium with myrosinase (0.018 U), the enzyme that converts glucosinolate to its corresponding isothiocyanate, showed minimal induction in this enzyme activity at concentration 10 ${\mu}M$. Quinone reductase and glutathione S-transferase activities were unaffected by this glucosinolate; however, supplementation of the incubation medium with myrosinase elevated quinone reductase activity. It may be inferred that the breakdown product of glucoraphanin, in this case sulforaphane, is superior than its precursor in modulating carcinogen-metabolising enzyme systems in vitro and this is likely to impact on the chemopreventive activity linked to cruciferous vegetable consumption.

  • $Ni(L)_4(NCS)_2$(1) (L=2-ethylimidazole)착물의 합성하고 단결정 구조를 규명하였다. 이 착물은 정방정계, 공간군 P4nc a=10.587(2), $c=12.927(3){\AA}$, $V=1448.9(4){\AA}$, Z=2로 결정화되었으며 676개의독립적인 회절반점에 대한 최종 신뢰도 인자 $R_1$$wR_₂$값은 각각 0.0581 및 0.1675이었다. 이 착물의 결정 구조는 Ni(II) 금속 원자에 4개의 2-ethylimidazole리간드의 질소 원자가 xy 평면에 배위되고 있고. 2개의 isothiocyanate 리간드의 질소원자 z축 방향에서 트란스 형태뢰 배위되어 균일한 팔면체 구조의 단핵 착물을 형성한다.

Liquid Crystal Mixtures of High Response Time and High $T_{NI}$ for LCD-TV Application

  • In In order to develop liquid crystal mixtures of high response time and high nematic-isotropic transition temperature ($T_{NI}$) for LCD-TV application, novel liquid crystal molecules with a fluoro-isothiocyanate moiety were synthesized. They showed remarkably high $T_{NI}$ over 200 $^{\circ}C$, wide mesophase range of 170 $^{\circ}C$, high dielectric anisotropy of 14 and high optical anisotropy of 0.19. New LC Mixtures of the high $T_{NI}$ ($85{\sim}100 ^{\circ}C$) and fast response time ($8{\sim}10ms$) were blended with the novel fluoro-isothiocyanate containing LC molecules, phenylcyclohexanes, bicyclohexanes and ester compounds. It was also studied on optimum pitch of liquid crystal for high speed twisted nematic LCD-TV applicaton. The LC mixtures show a fast speed of the below one frame rate in real 17" TV panel.

  • The thin films were obtained by plasma polymerization of phenyl isothiocyanate. Polymerizations were carried out in rf(13.56[MHz]) glow discharge generated in an inter-electrode capacitively coupled gas flow system. It was found that this monomer produces uniform films with a wide range of thicknesses, from hundreds of nanometers to tens of micrometers. The deposition rate appeared to be dependent on the substrate distance from the monomer inlet. The IR data revealed significant decrease in -NCS groups content in the polymer as compared with the monomer spectrum and indicated for the appearance of new absorption bands corresponding to the -CN and C-H aliphatic groups. The soluble fraction by GC was found to be composed of numerous low molecular-weight compounds.
