표면개질 알루미나막의 증기투과에 의한 알코올의 분리 (Separation of Alcohol/water Mixtures with Surface-modified Alumina Membrane in Vapor Permeation)

  • 높은 선택도와 투과도를 가진 분리막이 요구되지만 선택도가 높은 막은 투과도가 낮은 것이 일반적이다. 본 연구에서는 적절한 선택도를 유지하면서 높은 투과도를 갖는 분리막을 제조하기 위하여 다공성 알루미나막을 실란커플링제로 코팅하였다. 코팅된막에 대한 물의 접촉각은 90$^{\circ}$ 이상이었으며, 큰 소수성을 가짐을 의미하기 때문에 에탄올, 이소프로판올, 부탄을 수용액의 농축을 위한 증기투과실험을 수행하였다. 공급액중의 알코을 농도가 증가함에 따라 투과도가 증가하였으며, 이는 물에 비하여 코팅된 막에 대한 에탄올, 이소프로판올, 부탄올의 친화도가 크기 때문으로 판단된다. 코팅된 알루미나막의 투과도는 고분자막의 투과도에 비하여 20~1000배로 크게 나타났다.

Aspergillus niger SFN-416으로부터 생산한 $\beta$-Glucosidase의 정제 및 특성

  • $\beta $-Glucosidase (EC 3.2.1.21) was purified from Aspergillus niger SFN-416 by a sequential process of ammonium sulfate precipitation, Sepadex G-100 and DEAE-Sephacel column chromatography. Molecular weight of the enzyme was 46, 000 daltons. The K$_{m}$ and V$_{max}$ values for PNPG were 0.67 mM and 25 moles/ml $\cdot $min., respectively. The optimum pH and temperature of the enzyme activity were 3.5 and 58$\circ $C, respectively. The enzyme activity was decreased by addition of metal ions, and increased by addition of metanol, ethanol, isopropanol and 1-butanol at a concentration of 10% (v/v). Stability of the enzyme was increased by addition of isopropanol and 1-butanol at a concentration of 10% (v/v).

홍조류인 한국산 김종에서의 염색체 DNA 분리방법 (Rapid and Efficient Purification of Nucleic Acids from the Macroalga Porphyra(Rhodophyta))

  • A method for the isolation and purification of DNA from a red algae, Porphyra was innovated. The innovation of the method consists mainly of three steps that include sodium acetate treatment, chloroform extraction, and 0.2 volume isopropanol precipitation step. The sodium acetate treatment was designed to remove polysaccharide contamination, and the isopropanol step to remove proteins and salts contaminents. Genomic DNA,s of several species(for example, P. tenera, P. yezoensis, P. seriata, and P. pseudolinearis) was successfully isolated by the innovated method. The amount of DNA purified from one g of sample material with the innovated method was 53 g in average. The resulting DNA was characterized to include high molecular weight and showed no nuclease activity. The DNA was pure enough to be digested directly by various restriction enzymes without any difficulties. Porphyra DNA was pure enough and adequate for amplification reaction through the polymerase chain reaction (small nuclear rDNA PCR amplification).

플라스틱 필름에 대한 유기 용매의 전이 특성에 관한 연구 (Studies on the Migration behavior of various Printing Ink solvents of plastic films)

  • The increasing use of plastics in food packaging materials has led to the issue of food-packaging mutual interactions from residues in the plastics. Plastic films are commonly printed by using solvent-based ink to decorate the packaged food for consumer attention. However, the residual solvents can not be completely removed and they can migrate into the contained food which lead to undesirable off-flavors. Partitioning (Kp) of printing ink solvents was studied in two types of plastic films with different chemical structure and polarity. At $25^{\circ}C$, Kp of toluene is higher than that of isopropanol in PP, but isopropanol showed higher Kp value than toluene in EVOH. This showed that polarity had a significant effect on the partitioning of printing ink solvents into the plastic films. Printing solvents had a higher affinity to the polymer with similar polarity than it did to the different one.

Removal of acetic acid from wastewater by esterification in the membrane reactor

  • Acetic acid can be removed from wastewater by esterification in a membrane reactor. Pervaporation membrane reactor (PVMR) is an alternative process to conventional separation processes. It is an environmentally friendly process. The main advantages of the PVMR are simultaneous water removal and production of an ester economically. In this study, the synthetic wastewater has been used. Esterification reaction of acetic acid with isopropanol has been studied in the presence of tungstosilicic acid hydrate as a catalyst in a batch reactor and in a PVMR. The effects of important operating parameters such as reaction temperature, initial molar ratio of isopropanol to acetic acid and catalyst concentration has been examined. Removal of acetic acid (conversion of acetic acid) was obtained as 85% using a PVMR by removal of water from the reaction mixture.

  • Sustained-release dosage form of ferruginous hematinics which can be absorbed effectively without forming "iron block" that caused by high concentration of Iron in the gastrointestinal tract, was studied. Gelatinized micropellets containing medicament were prepared according to the method of N. Tanaka et al. and hardened in 10% formalin-isopropanol in various time. Gelatinized micropellets were digested with artificial gastric juice and prepared a graph of iron concentration determined by Hong's method. As a result, it is assumed that gelatinized sustained-release dosage form can be a good ferruginous hematinics and it is shown that the preparation of 72-hour-hardening in 10% formalin-isopropanol has a suitable sustained release.d release.

  • 여러 조성의 2-프로판올-물 혼합용액중의 anilinium chloride 용액의 상대점도를 $20^{\circ}$, $30^{\circ}$$40^{\circ}$에서 각각 측정하고, Jones-Dole식, ${\eta}{\tau}=1+AC^{1/2}+BC$에서 결정되는 점도 A- 및 B- 계수와, 절대반응속도 이론으로 구해지는 점성흐름의 활성화 에너지로 부터, 용매구조에 미치는 용질의 효과를 논의하였다. 2-프로판올의 몰분율이 $0.10{\sim}0.15$인 조성에서 점도 A- 및 B- 계수가 최소인 현상은 이 조성에서 물의 구조가 가장 강화되기 때문인 것으로 해석하였다. 또 anilinium chloride는 물에 대한 structure-maker 또는 structure-breaker의 입장에서 borderline case에 속한다고 결론지을 수 있었다.

GC를 이용한 극성용매의 분석방법 개발 연구 (Study on Development of Analytical Method for Polar Solvents by GC)

  • Objectives: The purpose of this study is to simplify and standardize analytical method of polar solvents(methanol, isopropanol, n-butanol, acetone, methylene chloride and MIBK) in the working environmental by GC. Because NIOSH methods are various and complicated. Methods: The method is using the same stationary phase(5% phenyl 95% dimethylpolysiloxane), absorbent(silica gel) and desorption solvent(DMSO) for above 6 solvents. For the 6 solvents desorption efficiency, calibration curve, and limit of detection were studied Results: As the results, 6 solvents{2 groups ; first group(methanol/isopropanol/butanol) and second group(acetone/methylene chloride/MIBK)} could be separated and quantified within 10 minutes. Desorption efficiency from silica gell of 6 solvents using dimethylsulfoxide(DMSO) was methanol 86.2%, isopropanol 103.2%, n-butanol 101.8%, acetone 98.2%, methylene chloride 103.9% and MIBK 106.2% in the range of 0.2, 0.5, 2.0 times of TWA, consequently, satisfied NIOSH estimation level(beyond 75%). Correlation coefficient(r)in the range of 0.2~2.0 times of TWA, was above 0.999 for 6 solvent. LOD(mg/DMSO ml) using calibration curve in the range of 0.2~2 times of TWA was methanol 0.11, isopropyl alcohol 0.20, n-butanol 0.03, acetone 0.50, methylene chloride 0.05, MIBK 0.04 respectably. Conclusions: This method can be used at the sampling and analytical method for polar solvents by GC. Also, will be able to be applied with NIOSH methods.

졸-겔법에 의한 TiO2 미분말 합성과 반응메카니즘(I): Titanium isopropoxide의 가수분해 (Synthesis of TiO2 Fine Powder by Sol-Gel Process and Reaction Mechanism(I): Hydrolysis of Titanium Isopropoxide)

  • Isopropanol 용매 내에서 titanium isopropoxide($Ti(O-^iPr)_4$)의 가수분해반응에 의하여 $TiO_2$ 미분말을 합성하였고 가수분해 반응 속도를 자외선분광법에 의하여 측정하였다. 반응은 $Ti(O-^iPr)_4$의 농도에 비하여 물 농도를 크게 하여 유사일차반응으로 진행시켰고, 물 농도 및 온도의 변화에 따른 반응속도상수를 Guggenheim method로 계산하였다. 또한 물의 동위원소 효과를 측정하여 반응에 참여하는 물분자의 촉매성을 확인하였다. 입도분석과 미세구조 관찰 결과, 얻어진 $TiO_2$ 미분말은 $0.3{\mu}m$정도의 입자크기를 갖는 구형의 입자로 확인되었다. 또한 반응속도로부터 전이상태에 참여하는 n value와 열역학적 파라메타를 계산한 결과, $Ti(O-^iPr)_4$의 가수분해반응은 이분자 반응인 $S_N2$ mechanism으로 진행하는 것으로 추정되었다.
