Probabilistic behavior of semi-active isolated buildings under pulse-like earthquakes

  • Seismic isolation systems employ structural control that protect both buildings and vibration-sensitive contents from destructive effects of earthquakes. Structural control is divided into three main groups: passive, active, and semi-active. Among them, semi-active isolation systems, which can reduce floor displacements and accelerations concurrently, has gained importance in recent years since they don't require large power or pose stability problems like active ones. However, their seismic performance may vary depending on the variations that may be observed in the mechanical properties of semi-active devices and/or seismic isolators. Uncertainties relating to isolators can arise from variations in geometry, boundary conditions, material behavior, or temperature, or aging whereas those relating to semi-active control devices can be due to thermal changes, inefficiencies in calibrations, manufacturing errors, etc. For a more realistic evaluation of the seismic behavior of semi-active isolated buildings, such uncertainties must be taken into account. Here, the probabilistic behavior of semi-active isolated buildings under historical pulse-like near-fault earthquakes is evaluated in terms of their performance in preserving structural integrity and protecting vibration-sensitive contents considering aforementioned uncertainties via Monte-Carlo simulations of 3-story and 9-story semi-active isolated benchmark buildings. The results are presented in the form of fragility curves and probability of failure profiles.

Isolation of Cold Acclimation-related Genes in Wheat Chromosome Substitution Line 5D

  • To identify low temperature-induced genes of wheat chromosome substitution line 5D, suppression subtractive hybridization (SSH) was performed with mRNAs from leaf samples that treated with low temperature ($4^{\circ}C$). A cDNA library was constructed using mRNA isolated from wheat chromosome substitution line 5D leaves treated with low temperature ($4^{\circ}C$). The nucleotide and deduced amino acid sequences of the putative gene products were compared. wfr-9 and wfr-32 showed identity over 90% related to vernalization gene. Other two genes, wfr-77 and wfr-83 which is related to freezing-resistant gene have also identity over 90%. This result suggest that those genes may be transcribed into antifreeze proteins which are accumulated within leaf apoplasts, when wheat chromosome substitution line 5D is acclimated during low temperature treatment.

Pure Isolation, Identification and Culture Characteristics of Ethylene Glycol Degrading Bacteria (Ethylene Glycol 분해균주의 순수분리.동전 및 배양특성)

  • A large quantity of ethylene glycol(EG) is remained in the effluent after pretreating polyester weight-loss wastewater physicochemically in the fist stage and must be treated biologically in the second stage. Therefore, an excellent EG-utilizing bacteria strain was isolated from the natural system and the optimal culture conditions of the strain were investigated. The optimal culture conditions of temperature, pH, and nitrogen source were found to be 35$^{\circ}C$, 7.5 and ammonium chloride, respectively, when CODCr removal efficiency was more than 90%. The growth of stains and EG removal efficiency was slightly improved by adding elements such as niacin and biotin. With increasing inoculation size in a batch culture, the removal efficiency of EG was conspicuously increased. Growth rate was inhibited when the initial concentration of EG was more then 30g/L. The strain was identified as Pseudomonas sp. based on morphological and biological characteristics and named as Pseudomonas sp. EG1.

  • Interactions between the acetonitrile and other organic molecules such as furan, pyrrole, pyrrolidine, thiophene, tetrahydrothiophene and acetaldehyde was studied with the infrared absorption spectroscopy under matrix isolation conditions. Xe was used as a major matrix material. Acetonitrile showed strong interactions with pyrrole and thiophene, and little interactions with pyrrolidine and acetaldehyde.

  • In order to screen dextranase with high dextranolytic activity from microbial origins, dextranase producing fungal isolates were isolated from soil of the Taeion area. 197 strains with dextranolytic activities were isolated, out of which 3 strains with high dextranolytic activities were selected in the first screening. A strain (GR-98) with a best dextranolytic activity was selected in the second screening. The strain was identified to be similiar Aspergillus ustus by the morphological and cultural characteristics. The optimum culture temperature and initial pH for the dextranase production of the strain was 30$\circ$C and 7.0, respectively. The optimum culture medium was composed of 2% dextran, 0.3% KNO$_{3}$, 0.05% K$_{2}$HPO$_{4}$, 0.02% MgSO$_{4}$-7H$_{2}$O, 0.05% KC1, and 2.5 $\mu$g/ml pyridoxamine, and the enzyme production was maximum when the strain was subcultured at 30$\circ$C for 7 days.

  • A bacterial strain of Brevibaterium sp. CH1 was isolated and used to produce an enzyme (nitrile hydratase) necessary for earring out the bioconversion of acrylonitrile to acrylamide. The culture and reaction conditions, and medium optimization were studied for the strain. The conversion yield was nearly 100% with a trace amount of acrylic acid produced. The strain showed strong activity of nitrile hydratase toward acrylonitrile and extremely low activity of the amidase toward acrylamide. We sought optimum culture conditions for the formation of nitrile hydratase by Brevibacterium sp. CH1. The effects of temperature and pH on the activity of free and immobilized tells were investigated. The nitrite hydratase of Brevibacterium sp. CH1 acted not only on various aliphatic nitrites such as acrylonitrile, propionitrile and acetonitrile, but also on aromatic nitrile as nicotinonitrile.

  • The dependence of the electrical properties in planar MQW - APD & pn, and p - i - n photodiode implanted with oxygen on the annealing emperatures and ion dose has been investigated. The oxygen implantation was performed for inter-device isolation. The leakage current of as-impanted p-i-n photodiode obtained was less than 50 nA. An annelaing temperature dependence study shows an abrupt increase of leakge current at 600.deg.C for all devices under study. This indicates that donor complex centers introduced by the chemical activity of oxygen increase with increasing annelaing temperatures. Furthermore, leakage current was highly correlated with oxygen dose due to th eimplanted related defects.

Isolation of Bacteria from Freshwater Plant Roots (담수식물 뿌리로부터 세균의 분리)

  • The isolation and identification of bacteria from freshwater plant root system were performed. Twenty four different strains were identified by microbiological identification system. Gram negative bacteria were isolated three times more than Gram positive ones. The ratio of rod and coccus was 11:1. The similarity above 0.5 was 37.5% from the tested samples in identification. Among these isolated bacteria, three dominant strains Pseudomonas cepacia JH10, Xanthomonas maltophilia JH12, Aeromonas salmonicida JH13 were selected to investigate biochemical properties and optimal growth conditions. These strains reached maximum growth after 24-36 hr of incubation in peptone broth, and their optimal temperature was 28$^{\circ}C$ and that of pH was between 7 and 8.

  • Heating floor is the most profitable system of air heating at poultry houses. THe tubes with heating elements inside are laid into the layer of concrete. To prevent lossese of heat penetration deep into the ground the layer of isolation material is laid below the tubes . The depth of isolation laying in every point of the heating floor may be calculated according to the author's formula using the data of temperature on the floor surface and the tube, the distance between tow nearest tubes, and the distance between the tube and the floor surface. Technological investigations allow to estimate the optimal density for greese and ducks when they are bred on the heatuing floor.

  • Four bacterial strains able to degrade dichlorobenzene as the sole source of carbon and energy were isolated from soil by selective enrichment culture, and among them, one isolation was the best in the cell growth and identified as Pseudomonas sp. DCB3 by its morphology and physiological properties. Cell growth dramatically increased in a minimal medium containing 500ppm of dichlorobenzene was not detected any more at 72 hours after cultivation. The optimal temperature and initial pH for the growth of the isolated strain were 30$\circ$C and 7.0, respectively. Cell growth was increased by supplementing $(NH_2)_2CO$ and 50 ppm yeast extract as additional nutrients. Therefore, it was suggested that Pseudomonas sp. DCB3 could be effectively used for the biological treatment of wastewater containing dichlorobenzene.
