Fenitrothion-degrading microorganisms were isolated from 124 sampling sites of paddy, upland, forest and polluted soil, and wastewater. A total of 1,071 strains were isolated from each selective medium supplemented with 50mg/l of fenitrothion - nutrient agar (NA) 601, potato dextrose agar (PDA) 201, Actinomycetes isolation agar (AIA) 168 and basal salt medium (BSM) 101, respectively. Twenty-eight effective strains of them, which showed more than 80% degradation of fenitrothion by the gasliquid chromatography(GLC) analysis. were successfully selected from each liquid culture supplemented with 50mg/l of fenitrothion - NB 12(upland soil 3, paddy soil 3, forest soil 2, polluted soil 4), PDB 8(upland soil 1, paddy soil 2, forest soil 2, polluted soil 3) and PSB 8(upland soil 1, forest soil 1, polluted soil 6), respectively. Four strains - NPal, NFol, PFol and BPol, which have the most powerful degradation activity were finally selected among 28 fenitrothion-degrading microorganisms based on the degradation rate at the concentration of 100mg/l fenitrothion in enrichment media.
Dormancy-associated protein (DRM) is involved in the dormancy physiology of plants and is conserved in almost all plant species. Recent studies found that DRM genes are involved in the abiotic stress response, and characterization studies of these genes have been conducted in several plants. However, few studies have focused on DRM genes in woody plants. Therefore, in this study, cDNA coding for DRM (PagDRM1) was isolated from poplar (Populus alba ${\times}$ P. glandulosa), and its structure and expression characteristics were investigated. PagDRM1 encodes a putative protein composed of 123 amino acids, and the protein contains two conserved domains (Domain I and Domain II). PagDRM1 is present as one or two copies in the poplar genome. Its expression level was highest in the stem, followed by mature leaves, roots, and flowers. During the growth of cultured cells in suspension, PagDRM1 was highly expressed from the late-exponential phase to the stationary phase. In addition, PagDRM1 expression increased in response to drought, salt stress, and treatment with plant hormones (e.g., abscisic acid and gibberellic acid). Therefore, we suggested that PagDRM1 not only plays an important role in the induction of dormancy, but also contributes to stress tolerance in plants.

It is important to measure the height of trees as an essential element for assessing the forest health in urban areas. Therefore, an automated method that can measure the height of individual tree as a three-dimensional forest information is needed in an extensive and dense forest. Since airborne LiDAR dataset is easy to analyze the tree height(z-coordinate) of forests, studies on individual tree height measurement could be performed as an assessment forest health. Especially in urban forests, that adversely affected by habitat fragmentation and isolation. So this study was analyzed to measure the height of individual trees for assessing the urban forests health, Furthermore to identify environmental factors that affect forest growth. The survey was conducted in the Mt. Bongseo located in Seobuk-gu. Cheonan-si(Middle Chungcheong Province). We segment the individual trees on coniferous by automatic method using the airborne LiDAR dataset of the two periods (year of 2016 and 2017) and to find out individual tree growth. Segmentation of individual trees was performed by using the watershed algorithm and the local maximum, and the tree growth was determined by the difference of the tree height according to the two periods. After we clarify the relationship between the environmental factors affecting the tree growth. The tree growth of Mt. Bongseo was about 20cm for a year, and it was analyzed to be lower than 23.9cm/year of the growth of the dominant species, Pinus rigida. This may have an adverse effect on the growth of isolated urban forests. It also determined different trees growth according to age, diameter and density class in the stock map, effective soil depth and drainage grade in the soil map. There was a statistically significant positive correlation between the distance to the road and the solar radiation as an environmental factor affecting the tree growth. Since there is less correlation, it is necessary to determine other influencing factors affecting tree growth in urban forests besides anthropogenic influences. This study is the first data for the analysis of segmentation and the growth of the individual tree, and it can be used as a scientific data of the urban forest health assessment and management.
Out of 826 isolates of Streptomyces isolated from different soils, their distribution and antibiotic productivity were investigated. Distribution of the organism in the soil was affected by the soil conditions and plants. The highest isolation frequency was occurred from Quercus forest, Robinia forest and grass field, while soils from orchards and cultivating fields showed low density of Streptomyces. More than 49% of the isolates showed antibacterial activity against Bacillus subtilis, Erwinia carotovora subsp. carotovora and Xantomonas campestris pv. oryzae and about 40% of the isolates showed antiyeasty activity against Saccharomyces cerevisiae but only a few isolates showed antibiotic activity against E. coli and Pseudomonas solanacearum. Forty isolates of the Streptomyces showed strong antifungal activity against Pyricularia oryzae. Rate of isolation of Streptomyces was the highest on starch agar among the eight media tested. Antibiotic productivity of the isolates was the highest on potato sucrose agar medium among the 5 media tested.
This experiment was undertaken to investigate proper conditions for protoplast isolation and genetic transformation of the white rot fungi, Polyporus brumalis. The protoplasts were formed from mycelia at a frequency of $1{\times}10^7/ml$ with 0.5% Usukizyme. The transformation vector (pHYgpt) was constructed using hygromycin resistance gene (hph) for the selectable maker. The yield was 100-160 transformants/${\mu}g$ DNA in a transformation mediated by 40% polyethylene glycol solution with aurintricarboxylic acid, heparin and supermidine. The genomic integration of the pHYgpt was confirmed by hph-specific PCR and the expected amplified band appeared only in the transformants. These results could be an efficient tool in gene engineering of the genus polyporus.
Korea Atomic Energy Research Institute (KAERI) constructed the KURT (KAERI Underground Research Tunnel) to analyze the hydrogeological/geochemical characteristics of deep rock mass. Numerous boreholes have been drilled to conduct various field tests. The selection of suitable investigation intervals within a borehole is of great importance. When objectives are centered around hydraulic flow and groundwater sampling, intervals with sufficient groundwater flow are the most suitable. This study defines such points as hydraulic outliers and aimed to detect them using borehole geophysical logging data (temperature and EC) from a 1 km depth borehole. For systematic and efficient outlier detection, machine learning algorithms, such as DBSCAN, OCSVM, kNN, and isolation forest, were applied and their applicability was assessed. Following data preprocessing and algorithm optimization, the four algorithms detected 55, 12, 52, and 68 outliers, respectively. Though this study confirms applicability of the machine learning algorithms, it is suggested that further verification and supplements are desirable since the input data were relatively limited.

For the control of oak wilt caused by Raffaelea quercus-mongolicae, an antifungal microorganism, Streptomyces blastmyceticus, was used as a potential agent. Culture suspension of S. blastmyceticus was injected into Quercus mongolicae in the research forest of Kangwon National University by $ChemJet^{(R)}$ trunk injection and Macro-infusion at root flare injection. $Alamo^{(R)}$ (a.i., propiconazole 14.5%), a fungicide currently used for the control of oak wilt in USA, was also treated by both methods to compare the efficacy. For preventive efficacy, culture suspension of the pathogen was inoculated at 1 month after injection of either agent. Tested trees were cut at 3 months after treatment, stained with 1% Fuchsin acid, and then non-conductive area (NCA) and re-isolation frequency (RIF) of oak wilt fungus were compared among treatments. While NCA was the highest as 47.3% in pathogen only treatment, it was the lowest as 16.0% in sterilized water treatment by Macro-infusion. NCAs of Alamo treatment by Macro-infusion and ChemJet injection were 25.3% and 32.1%, respectively. NCA of S.blastmyceticus treatment by ChemJet injection was 32.3%, similar with Alamo treatment's by ChemJet injection. All treatments by either injection method showed significantly lower NCA compared to the pathogen only treatment. These results indicate that S. blastmyceticus injection shows the preventive efficacy against oak wilt disease by suppressing the growth of pathogen injected. NCA of Macro-infusion injection of sterilized water was lower as 16.0%, compared to 21.3% of ChemJet injection. It means that Macro-infusion is more effective in translocation of sterilized water than ChemJet injection by even distribution. RIF from wood discs of treated trees showed high in pathogen only treatment, but relatively low in S. blastmyceticus treatment. RIF results were correlated with NCA results. From the above results, it was confirmed that S. blastmyceticus showed preventive efficacy against oak wilt disease by ChemJet trunk or Macro-infusion at root flare injection.

Zoysia japonica Steud.(Zj) is a typical warm-season Korean lawn grass, which is used in many places such as river banks, roadside and soccer fields in Korea. Recently, it has also been used in school yards and the Saemangeum reclaimed land to reduce water pollution. Although the cultivated area of turfgrass is steadily increasing worldwide, it grows fast requiring frequent mowing and is difficult to grow in shady areas and the cold region. Therefore this study aims searching for useful gene(s) to develop abiotic stress tolerant and dwarf zoysiagrass. We isolated Ycf4 gene based on the sequence from Oryza sativa Japonica through RT-PCR and RACE PCR. Ultimately, open reading frame (ORF) of ZjYcf4 was 558bp long, encoding a protein of 186 amino acid residues. NCBI blast results showed that the ZjYcf4 protein is evolutionarily closely related to Ycf4 protein from Zoysia macrantha and Setaria italica (100% and 98%, respectively). To determine whether ZjYcf4 was involved in environmental stress in wild-type zoysiagrass, expression patterns of the gene were analyzed by real-time PCR under salt, cold and dark conditions. They were analyzed after each stress treatment for 3 hours. In salt and cold stresses, the expression was higher compared to control (3-fold and 1.5-fold, respectively), although there was a 1.6-fold decrease in expression under dark stress treatment. As reported previously, we suggest that ZjYcf4 gene affects abiotic stress such as salt, cold and dark.

Auxin plays a crucial regulatory role in plant growth and development processes. Three major classes of auxin-responsive transcription factors controlled by the Auxin/indole-3-acetic acid (Aux/IAA), Gretchen Hagen 3 (GH3), and small auxin up RNA (SAUR) genes regulate auxin signaling. Aux/IAA, in particular, encodes short-lived nuclear proteins that accumulate rapidly in response to auxin signaling. In this study, we isolated a PagAux/IAA1 gene from poplar (Populus alba ${\times}$ P. glandulosa) and investigated its expression characteristics. The PagAux/IAA1 cDNA codes for putative 200 amino acids polypeptide containing four conserved domains and two nuclear localization signals (NLSs). Utilizing Southern blot analysis, we confirmed that a single copy of the PagAux/IAA1 gene was present in the poplar genome. The expression of this gene is specific to leaves and flowers of the poplar. PagAux/IAA1 expressed in the early exponential growth phase of cell-cultured in suspension. PagAux/IAA1 expression level reduced in drought and salt stress conditions, and the presence of plant hormones such as abscisic acid. However, expression enhanced in cold stress, cambial cell division, and presence of plant hormones such as gibberellic acid and jasmonic acid. Thus, these results suggest that PagAux/IAA1 participates in cold stress response as well as developmental processes in the poplar.
