Isolation and Morphological Characterization of Monilinia sp. KV-27 Associated with Apple Anthracnose of Fuji Apples in Korea

  • This study was undertaken to isolate and to identify a fungal pathogen Monilinia sp. KV-27 associated with apple anthracnose. Rotted Fuji apples were used for the isolation of the fungus. The infected tissues were sterilized with 70% ethanol, washed with sterilized distilled water and were transferred to 50 ml containing potato dextrose broth (PDB) flasks. The peripheral hyphae of the fungal colony which developed from the infected tissues were isolated on to potato dextrose agar (PDA). On PDA plates the fungus grew well at $25^{\circ}C$ and occupied more than half of a 9 cm petri dish within 5 days. The fungal cultures on PDA were used for morphological observation and identification of the fungus. Conidiophores were produced on the gray to whitish sporodochial structures scattered on PDA plates. These conidiophores gave rise to chains of conidia, which were branched and easily detached in water. These structures were dark brown to black and consisted of hyphal masses. Conidia produced on PDA plates were hyline or light colored, lemon shaped or ellipsoidal ($10-13{\times}8.5-11{\mu}m$) in size.

  • A survey was conducted to determine the prevalance of infectious pancreatic necrosis virus (IPNV) on fish farms in Korea and the epidemiology of IPNV infection in the farmed rainbow trout. In total, 43 pools of rainbow trout with apparent signs of viral infection from five provinces were obtained and analyzed. Evident cytopathic effects, including karyopycnosis and cell destruction, were observed in CHSE (chinook samlmon embyro)-214 cells infected with the virus isolates. Of these, ten viral isolates were assumed to be IPNV based on biophysical properties. RNA analysis revealed that the isolates contained two-segmented RNA genomes, further indicating that the viral isolates are IPNV. Antigenic comparison of the IPNV isolates identified three distinct serological groups separable by the cross-neutralization test. Of the ten IPNV isolates, six could be classified as strain DRT, two as strain Ab, and two as strain VR299. We were not able to isolate new strain of IPNV or any isolate serologically similar to the standard strain Sp.poraceae and families of the Agaricales, they are genetically more related to the Polyporaceae. These results are consistent with morphological characters observed in those mushrooms. However, it is premature to conclude taxonomic status Ganoderma species in the present study employing small sample size.

  • As a basic study for quality improvement of Korean soybean paste and soybean sauce, we investigated on microflora of soybean paste and soybean sauce fermentation. Major Microorganisms were isolated from the sample pastes and sauces, and identified systematically. Selected Microorganisms were identified by MIS whole cell fatty acid analysis by gas chromatography. Identification results showed that Bacillus licheniformis, bacillus pumilis and Bacillus subtilis were dominant in soybean paste and Staphylococcus vitulus, Bacillus subtilis, Bacillus pumilus, and Lactobacillus fermentum were dominant in soybean sauce. It seemed that these Microorganisms played an important role in soybean paste and soybean sauce fermentation and could be used for the further studies such as protease and amylase activities.

  • The aim of the present research program was to develop a strain of actinomycetes producing extracellular cathepsin B inhibitor. Soil samples were collected from various sites in Korea and a number of actinomycetes were isolated from the soil samples by applying various physical and chemical pretreatments. An economical and effective method was developed for the screening of strains producing low molecular weight cathepsin B inhibitor, and consequently a strain (SMF28) among over 700 isolates was selected. Chemotaxonomic and numerical identification were carried out for the isolate. Fifty taxonomic unit characters were tested and the data were analyzed numerically using TAXON program. The isolate was identified as a strain of Streptomyces chromofuscus.

  • An anti-fungal material produced by actinomycetes was isolated from domestic soil. This actinomycetes was identified as Streptomyces albogriseus by 16S rDNA sequence. YEME (yeast extract 4 g, malt extract 10 g, glucose 4 g, D.W 1l, pH 7.00.2) medium was used for production of anti-fungal materials. S. albogriseus was cultured in a shaking incubator for 2 weeks at 150 rpm and $25^{\circ}C$. An anti-fungal material produced by S. albogriseus was identified at 340 nm by uv/vis- spectrometer and it showed powerful anti-fungal activity. This is the first report that secondary metabolite produced by S. albogriseus showed an activity against phytopathogenic fungi such as Collectrichum coccodes, Botrytis cinerea, Cladosporium cucumerinum, Didymella bryoniae.
