Installing vibration isolation in structures, such as structures adjacent to subways, may be delicatebecause of the proximity with the vibration source or because of the wave propagation path. This paper discusses on method that install isolation Pads on underground walls as a part of the vibration mitigation system, and also on its efficiency, The proposed method is proven to affect significantly the distribution of acceleration in the neighborhood of the structure and to reduce efficiently the maximum amplitude of the vibration. It is also seen that installing isolating pads until the depth of the foundations and deeper is more efficient than installing such device separately from the structure. This Study being limited to the comparison of installation methods, further Studies considering the thickness, stiffness and other parameters should be required.
Trichoderma viride QM 9414의 생육을 저해하여 colony를 형성 시키고 배지에 첨가된 cellulose가 균이 분비한 cellulase에 의해서 분해되어 생긴 clear zone으로서 육안으로 cellulase 역가를 쉽게 측정할 수 있는 방법과 repressor를 첨가하여 constitutive mutant를 선별할 수 있는 방법에 관한 실험을 하였든 바 그 결과를 요약하면 다음과 같다. 1) Colony를 형성시키고 그 size를 제한 하는데는 deoxycholate를 배지에 0.15% 첨가 하였을 때 가 가장 좋았다. 2) Saponin을 배지에 0.l% 첨가하면 뚜렷한 clear zone을 형성하였다. 3) Constitutive mutant를 선별하기 위해서는 기질로 pulp를 사용하였을 경우 glucsoe를 1% 첨가하거나 glycerol이나 cellobiose를 0.5% 첨가하면 가능하다고 생각되며 기질로 pure cellulose를 사용하였을 경우는 그대로 배지에 spore를 plating하면 가능하다고 생각된다. 4) Cellulase 역가가 높은 균주는 clear zone의 크기로서 쉽게 분리할 수 있다.
Chelating sepharose fast flow gel에 $Cu^{2+}$을 고정화시키고 이들과 단백질과의 친화력 정도에 의해서 용출용매를 통해 단백질을 분획하는 방식의 IMAC법을 이용하여 소의 혈장단백질로부터 IgG의 분리를 시도하였다. 대부분의 혈장단백질은 1차(0.01 M $Na_2HPO_4$, 0.5 M NaCl, pH 4.0)와 2차 용출용매(0.01 M imidazol)에 의하여 용출되었으며 역상 chromatography를 이용하여 각 분획의 단백질조성을 조사한 결과, 1차 용출용매에 의해서는 주로 albumin이, 그리고 2차 용출용매에 의해서는 IgG와 transferrin 등이 IMAC column으로부터 용출되었다. 2차 용출용매에 의해 얻어진 분획으로부터 분자량 100 kD이상의 단백질을 한외여과장치를 이용하여 농축하였을 때, IgG가 효과적으로 분리 정제 되었다.
In this paper, we present an adaptive formant tracking algorithm for speech using closed phase WRLS-VFF-VT method. The pitch synchronous closed phase methods is known to give more accurate estimates of the vocal tract parameters than the pitch asynchronous method. However the use of a pitch-synchronous closed phase analysis method has been limited due to difficulties associated with the task of accurately isolating the closed phase region in successive periods of speech. Therefore we have implemented the pitch synchronous closed phase WRLS-VFF-VT algorithm for speech analysis, especially for formant tracking. The proposed algorithm with the variable threshold(VT) can provide a superior performance in the boundary of phone and voiced/unvoiced sound. The proposed method is experimentally compared with the other method such as two channel CPC method by using synthetic waveform and real speech data. From the experimental results, we found that the block data processing techniques, such as the two-channel CPC, gave reasonable estimates of the formant/antiformant. However, the data windows used by these methods included the effects of the periodic excitation pulses, which affected the accuracy of the estimated formants. On the other hand the proposed WRLS-VFF-VT method, which eliminated the influence of the pulse excitation by using an input estimation as part of the algorithm, gave very accurate formant/bandwidth estimates and good spectral matching.

Vibration-based structural health monitoring (SHM) is crucial for the dynamic maintenance of civil building structures to protect property security and the lives of the public. Analyzing these vibrations with modern artificial intelligence and deep learning (DL) methods is a new trend. This paper proposed an unsupervised deep learning method based on a convolutional autoencoder (CAE), which can overcome the limitations of conventional supervised deep learning. With the convolutional core applied to the DL network, the method can extract features self-adaptively and efficiently. The effectiveness of the method in detecting damage is then tested using a benchmark model. Thereafter, this method is used to detect damage and instant disaster events in a rubber bearing-isolated gymnasium structure. The results indicate that the method enables the CAE network to learn the intact vibrations, so as to distinguish between different damage states of the benchmark model, and the outcome meets the high-dimensional data distribution characteristics visualized by the t-SNE method. Besides, the CAE-based network trained with daily vibrations of the isolating layer in the gymnasium can precisely recover newly collected vibration and detect the occurrence of the ground motion. The proposed method is effective at identifying nonlinear variations in the dynamic responses and has the potential to be used for structural condition assessment and safety warning.

Identification of differently expressed genes under specific tissues and/or environments provides insights into the nature and underlying mechanisms of cellular processes. Although cDNA-AFLP (Amplified Fragment Length Polymorphism) is a powerful method for analyzing differentially expressed genes, its use has been limited to the requirement of radioactive isotope use and the difficulty of isolating the bands of interest from a gel. Here, we describe a modified method for cDNA-AFLP that uses a fluorescence dye for detection and isolation of bands directly from a small size polyacrylamide gel. This method involves three steps: (i) preparation of cDNA templates, (ii) PCR amplification and differential display, and (iii) identification of differentially expressed genes. To demonstrate its utility and efficiency, differentially expressed genes during vegetative growth and appressorial development of Magnaporthe oryzae were analyzed. This method could be applied to compare gene expression profiles in a diverse array of organisms.
In this paper, we propose a novel robust fault isolation filter design method using the left eigenstructure assignment scheme proposed by the authors. The proposed method guarantees that the ${\gamma}$ simultaneous faults can be isolated when the number of available outpur measurements is ${\gamma}$. Moreover, if there exist redundant output measurements, the eigenvaluses of te filter system can be assigned to the desired position or the filter can be designed robustly to, the system parameter variation. Liu & Si developed a filter design method which has the same purpose, fault isolation. However their method cannot use the redundant freedom of the output matrix C. The proposed filter can use the redundant freedom of the matrix C effectively. Beside this in this paper, an eigenstructure assignment methodology that satisfies the required fault isolation conditions is also proposed. The proposed fault isolation filter was applied for isolating the simultaneous faults to a VTOL aircraft in order to verify the fault isolation performance.

The EF-18 agar/hydrophobic grid membrane filter (EF18/HGMF) method was evaluated for the isolation of Salmonella in naturally contaminated chicken carcasses, chicken parts (legs, wings and giblets) and processed chicken products (sausages and hamburgers). Percentages of false positive results for Salmonella (colonies with a similar morphology to those of Salmonella) were 78.75, 81.67 and 80% for carcasses, chicken parts and processed chicken products, respectively. The bacterial isolates that caused false positive reactions using this method were identified as Proteus mirabilis (70.85%), Citrobacter freundii (15.25%), Klebsiella ozaenae (5.83%), Hafnia alvei (4.48%), Escherichia coli (2.69%) and Enterobacter aerogenes (0.90%). The data obtained in this study suggest that the EF-18/HGMF method is not sufficiently selective or specific far isolating Salmonella from meat and chicken products.

The aims of this study were to establish a simple and effective method for isolating male germline stem cells (GSCs), and to test the possibility of using these cells as a new approach for male infertility treatment. Testes obtained from neonatal and adult mice were manually decapsulated. GSCs were collected from seminiferous tubules by a two-step enzyme digestion method and plated on gelatin-coated dishes. Over 5-7 days of culture, GSCs obtained from neonates and adults gave rise to large multicellular colonies that were subsequently grown for 10 passages. During in vitro proliferation, oct-4 and two immunological markers (Integrin ${\beta}1,\;{\alpha}6$) for GSCs were highly expressed in the cell colonies. During another culture period of 6 weeks to differentiate to later stage germ cells, the expression of oct-4 mRNA decreased in GSCs and Sertoli cells encapsulated with calcium alginate, but the expression of c-kit and testis-specific histone protein 2B(TH2B) mRNA as well as the localization of c-kit protein was increased. Expression of transition protein (TP-l) and localization of peanut agglutinin were not seen until 3 weeks after culturing, and appeared by 6 weeks of culture. The putative spermatids derived from GSCs supported embryonic development up to the blastocyst stage with normal chromosomal ploidy after chemical activation. Thus, GSCs isolated from neonatal and adult mouse testes were able to be maintained and proliferated in our simple culture conditions. These GSCs have the potential to differentiate into haploid germ cells during another long-term culture.
Insoluble phosphate-solubilizing bacteria are routinely screened by a plate assay method using Pikovskaya agar and a modified Pikovskaya medium. A modified Pikovskaya medium to improve the clarity of the yellow-colored halo has not necessarity improved the plate assay. Colonies of phosphate-solubilizing bacteria tested could be redily selected after 48 h of incubation by green-colored colony formation on plate in which bromcresol green(BCG) was included. Among them, two bacterial strains did not produce distinct yellow halos after 48 h of incubation. We suggest that the green colony formation on plate medium containing BCG is advantageous ofr rapid isolating phosphate-solubilizing bacteria directly from soil.
