Effect of Exogenous Estrogen on Growth and Mammary Gland Development in Korean Native Heifers (외인성 Estrogen이 한우 암송아지의 성장과 유선 및 유방의 발달에 미치는 영향)

  • The effects of exogenous estrogen on growth, mammary development, and the age at which the first estrous symptoms are detected were investigated for the first time in Korean native heifers at 6 months of age. At 90 days after treatment, weight gains of heifers implanted with 10 mg of estradiol were not different from those of the control heifers. However, weight gams of heifers implanted with 20 mg of estradiol were significantly increased at 90 days by 18%, relative to control values. At 180 days, cumulative weight gains of heifers implanted with either 10 mg or 20 mg of estradiol averaged 6.7% or 17.8%, respectively, greater than controls. But these values were not statistically significant. Regardless of dosage, teat diameter gains in the treatment groups were significantly increased at 90 days about 159%, relative to controls. Cumulative teat diameter gains in the heifers implanted with either 10 mg or 20 mg of estradiol were also significantly increased at 180 days by 100% or 128%, respectively, compared to controls. Teat length gains in the heifers implanted with either 10 mg or 20 mg of estradiol were significantly increased at 90 days by 200% and 295%, respectively, compared to controls. Cumulative teat length gains in the heifers implanted with either 10 mg or 20 mg of estradiol were significantly 265% and 325%, respectively, higher than controls. The heifers implanted with either 10 mg or 20 mg of estradiol showed a significant increase in teat volume gains at 90 days of 282% or 246%, respectively, over those of the control heifers. Cumulative teat volume gains in the heifers implanted with either 10 mg or 20 mg of estradiol were significantly increased at 180 days by 251% and 244%, respectively, compared to control values. However, there were no significant differences m gams of teat diameter, length and volume between heifers implanted with 10 mg and 20 mg of estradiol regardless of 90 days or 180 days. Heifers implanted with 10 mg or 20 mg of estradiol showed the first estrus 20 or 124 days, respectively, faster than controls. Overall, These data strongly suggest that estradiol implantation into heifers stimulates growth and mammary development and hasten the age at which the first estrus in Korean native heifers is observed.

Effects of Gestation Length and Birth Weight on Survival Rate in Cloned Korean Native Calves (복제 송아지의 임신 기간과 생시체중이 출생 후 생존율에 미치는 영향)

  • This study was performed to investigate the relation between birth weight and survivability on the production of cloned Hanwoo calves. The 580 cloned embryos were transferred into the 293 recipients. The pregnancy rate of the cloned embryos was 72.3% at 50 days after embryo transfer, and then the rate was dramatically decreased. The mean gestation lengths were 287 days in both clone (range of$279{\sim}295$ days) and artificial insemination (AI, range of $255{\sim}293$ days) calves, respectively. The mean birth weight of cloned calves (30.3kg) was significantly higher compared to that of AI calves (23.7kg) (p<0.05). Among the cloned calves, the birth weight was not different in both normal delivery (n=17, 29.9kg) and caesarean section (n=14, 32.3kg). The weight, however, was significantly higher in the clones (n=18, 32.8kg) dead within 175 days than that of the clones (n=11, 28.3kg) alive more than 175 days after birth (p<0.05). Interestingly, all cloned calves weighed <15kg (n=5) or >35kg (n=9) at birth have been dead within 175 days from the date of birth. The causes of death in the cloned calves were premature birth (n=2, 10.0%), abnormal function of lung and liver (n=2, 10.0%), abnormal function of lung (n=4, 20.0%), malformation (n=4, 20.0%), unknown (n=4, 20.0%), and sudden death syndrome (n=4, 20.0%), respectively. Our findings suggest that normal birth weight is one of the most important factors to survive more than 6 months in cloned calves.

A Study on DNA Polymorphism of the Bovine c-KIT Receptor Gene (소 c-KIT Receptor 유전자의 다형성에 관한 연구)

  • We considered KIT gene as a candidate gene for the white-spotting pattern in cattle. This study was carried out to detect genetic variation of c-KIT receptor gene and to investigate association between the mutation and the white-spotting pattern in cattle. PCR-RFLP analysis within intron 6 of c-KIT receptor gene were performed with 8 cattle breeds including Hanwoo, Angus, Brown Swiss, Charolais, Hereford, Holstein, Limousin and Simmental. When PCR product of approximately 2,440 bp including intron 6 of c-KIT receptor gene was sequenced, four nucleotide substitutions were found within intron 6 of the bovine c-KIT receptor gene. In PCR-RFLP analysis, three alleles (A, B and C), two alleles (A and B) and two alleles (A and B) at each locus were identified by MspⅠ, BsrBⅠ and NdeⅠ, respectively. Although frequencies of allele at each locus were different among cattle breeds, we could not get any evidence related with white or white spotting phenotypes in these mutations on intron 6 of c-KIT receptor gene. However, we can not entirely exclude the possibility that c-KIT receptor gene is responsible for white spotting phenotype in cattle. Thus, further studies need to detect other mutations in c-KIT receptor gene and to test association of those mutations and coat color phenotypes in cattle.

Effect of Brown Seaweed Waste Supplementation on Lactational Performance and Endocrine Physiology in Holstein Lactating Cows (미역부산물의 첨가가 홀스타인 비유우의 비유성적과 내분비생리에 미치는 영향)

  • This study was conducted to investigate effects of the brown seaweed waste(BSW) supplementation on milk production and related endocrine response in serum in Holstein dairy cows. A total of 14 Holstein dairy cows(initial mean live weight 625kg, average lactation days 225, Reproduction 2.4) were randomly allocated into control(basal diet) and treatment groups (4% BSW/basal diet) with 7 replications for 90 days. Dry matter intake was not affected by brown seaweed waste supplementation, but daily milk yield(kg) at the last experiment significantly increased (6.25kg) in treatment group compared with control group(p<0.05) at the last experiment. The plasma insulin-like growth factor(IGF)-1, triiodothyronine($T_3$) and thyroxine($T_4$) levels were significantly increased in treatment group compared with control group(p<0.05), although the concentration of plasma growth hormone(GH) was not significantly different. Milk composition was not significantly different between groups. The somatic cell count(SCC) in milk were significantly reduced in treatment group compared with control group(p<0.05), but antibodies(total IgG, G1, G2) were not significantly different between groups. Therefore we strongly believe that the increased milk yield is related to metabolic hormones as IGF-1, $T_3$ and $T_4$ and the mechanism of reducing SCC in milk must do more study related nonspecific immunsystem in the future.

Effects of Insoluble Dietary Fiber Supplementation on Performance and Nutrient Digestibility of Weanling Pigs (난용성 식이섬유의 첨가가 이유자돈의 생산성 및 영양소 소화율에 미치는 영향)

  • An experiment was conducted to determine the effect of supplementation of insoluble dietary fiber (Vitacel®) on growth performance and nutrient digestibility in weanling pigs. A total of 96 pigs that averaged 6.49$\pm$0.52kg BW and 23$\pm$2.1d age were allocated in a randomized block design with two pigs per pen and 12 pens per treatment. Pigs and feeders were weighed 10-days interval for the 40-d trials to determine ADG, ADFI and feed:gain ratio(F:G). Pigs were fed one of four diets:1) Control diet (C) 2) C+0.3% insoluble dietary fiber(IDF) 3) C+0.6% IDF and 4) c+0.9% IDF. For the determination of fecal nutrients digestibility, pigs were fed diets(diet 2) with 1% Celite-545(Fluka) as a marker and feces were collected on $9^{th}$ day and $18^{th}$ day after feeding diet 2. During the whole experimental period, pigs fed diet with 0.3% IDF have significantly higher ADG than other dietary treatment groups(P<0.05). ADG of pigs fed diet with 0.6% IDF was higher than that of pigs fed control diet(P<0.05). However, there was no significant difference in ADG between control group and 0.9% IDF group(P>0.05). ADFI of pigs fed diet with 0.3% IDF was significantly higher than any other dietary treatment groups(P<0.05). There was no significant difference in ADFI among control group, 0.6% and 0.9% IDF supplementation groups (P>0.05). Digestibilities of organic matter, crude protein, crude fiber and energy were significantly higher in 0.3% IDF supplementation group than any other dietary groups. However, there was no difference in over all nutrient digestibilities between 0.6% and 0.9% IDF group. Feeding diets more than 0.6% IDF did not affect the rest of the nutrients digestibilities except for ADF digestibility compared to control diet. Dietary supplementation level of IDF showed a significant quadratic effect on performance improvement of piglets. This response of growth performance to IDF supplementation is, as expected, in agreement with that of nutrient digestibility. Our results showed that IDF supplementation to diet for weaned piglets might be beneficial in terms of growth and nutrient digestibility. However, there should be more study on the relationship between level of IDF supplementation and piglet response as well as the exact mode of action of IDF in weaned piglets.

The Effects of the Transgenic Aspergillus oryzae Supplementation on Performance, Egg Quality and Intestinal Microflora of Layers (형질 전환 Aspergillus oryzae의 첨가가 산란계의 생산성, 계란 품질 및 장내 미생물 변화에 미치는 영향)

  • An experiment was conducted to investigate the dietary effects of a transgenic Aspergillus oryzae(AO) culture on the performance, egg quality and intestinal microflora of layers. A total of 840 Hy-line Brown layers of 39wks old were assigned to one of the following 7 dietary treatments: control(C), C+0.2% AO culture, C+0.5% AO culture, C+0.2% transgenic AO culture, C+0.5% transgenic AO culture, C+0.2% transgenic mutant AO culture, and C+0.5% transgenic mutant AO culture. The transgenic AO was made by inserting Salmonella gallinarum gene to AO. And the transgenic mutant AO was made by inserting Salmonella gallinarum gene to mutant AO which was mutated by UV irradiation. Each treatment was replicated six times with 20 birds housed in 2 bird cage. Twenty birds units were arranged according to completely randomized block design. Feeding trial lasted for 8wks under 16 hour lighting regimen. Laying performance and egg quality were significantly(P<0.05) affected by the treatments. Transgenic AO culture supplementation at the level of 0.2% significantly increased egg production, while its egg weight was significantly decreased compared to that of the control. Feed intake and feed conversion ratio(FCR) were not significantly different among the AO treatments and the control. The eggshell strength of the AO treatments was significantly higher than that of the control. Transgenic mutant AO culture supplemented at the level of 0.5% significantly increased egg yolk color. Intestinal microflora were significantly(P<0.05) affected by the treatments. The cfu of Lactobacilli spp. significantly increased and those of Salmonella species and E. coli decreased in the AO treatments. The transgenic AO and transgenic mutant AO culture were more effective than the AO culture in reducing the cfu of Salmonella species and E. coli. It is concluded that supplementation of the transgenic AO culture at the level of 0.2% could be recommended for the improvement of egg production. Supplementation of transgenic AO or transgenic mutant AO culture at 0.2% level effectively controlled intestinal Salmonella species population.

Seasonal Variations in Chemical Composition of Dried Food Waste in Wonjusi and Its Feeding Effects in Finishing Pigs (원주지역 남은 음식물의 계절별 성분 함량 및 비육돈에 대한 건조 남은 음식물 급여 효과)

  • A study was conducted to evaluate seasonal variations in chemical composition of food waste (FW) and its feeding effects on growth performance and pork quality in finishing pigs. FW was collected for 1 year (6 times a month) to establish a database for use of FW as a feed ingredient. For a feeding trial (8 weeks), a total of 117 pigs ${\times}$D; 54.80$\pm$4.60kg) were used to evaluate the processing effects of FW. Treatments were: Control (a corn-soybean meal diet without FW), simple dried FW (SD) and vacuum fermented FW (VF). The gross energy, crude protein, crude fat, ash, calcium and phosphorus in FW (DM, average of 4 seasons) were 5,111kcal/kg, 22.92%, 14.31%, 15.48%, 2.7% and 1.05%, respectively. Among seasons, the energy and crude protein contents were the highest (p<0.05) in winter and summer, respectively. In lactic acid bacterial counts, there was no difference between SD and VF. Pigs fed the control diet grew faster (p<0.05) than those fed diets containing food wastes, but not feed conversion ratio. There were no differences in production traits between SD and VF. No differences were also found in dressing percentage, backfat thickness, and pork quality (color, drip loss and TBARS) among treatments. The feed cost (₩/kg body weight) was lower in pigs fed FW than those fed a control diet. In conclusion, a pelleted diet containing food waste less than 20% would reduce feed cost in finishing pigs. However, it seems that a vacuum fermentation of food waste is not necessary for diet processing.

Effects of Total Mixed Rations on Ruminal Characteristics, Digestibility and Beef Production of Hanwoo Steers (섬유질배합사료 급여가 비육후기 거세한우의 반추위 발효성상, 소화율 및 산육성에 미치는 영향)

  • This experiment was carried out to compare the effects of feeding commercial formula feed and rice straw separately (control) versus a total mixed ration (TMR) on productivity of Hanwoo steers in late stage of fattening and on ruminal fermentation characteristics and digestibilities. Ruminal digesta from the cannulated cattle were sampled at 0, 1, 2, 3, 5, 8 hour after feeding. The steers fed TMR consumed 7.4kg per day and there was no difference between feeding systems. Daily weight gain was not significantly (P>0.05) between feeding systems, however, TMR group showed lower daily gain than control group. The amount of feed consumption per kg weight gain was higher in TMR group than control group (10.5kg and 9.7kg, respectively), resulting in a greater efficiency of feed utilization for gain. In the result of appearance rates of quality grade A were 33% higher for TMR group than those in control group. Appearance rates of grade 1 showed 56% and 75% when fed the control and TMR, respectively. Digestibilities of dry matter, crude protein, crude fiber and gross energy for TMR treatment were significantly higher (P<0.01) than those of control. Prior to feeding (0 h) and each subsequent hour, the TMR resulted in higher rumen pH (P<0.05) when compared with control ration. The concentration of NH3-N for TMR treatment maintained at higher level up to 8hr after feeding, especially increased up to 28.2mg/$d\ell$ during 1-2 hour which was two times (P<0.05) more than control. The amount of total VFA showed same trends between feeding systems. However, the ratios of branched chained fatty acid such as iso-butyric acid and iso-valeric acid for TMR treatment were significantly(P<0.01) higher than control for 3-5hr. Results showed that TMR in these trials is effective feeding system for fattening Hanwoo steers in the respect of ruminal characteristics, total tract digestibility and productivities.

The Effects of Processing Methods of Corn on In sacco Starch and Protein Degradability in the Rumen (옥수수 가공방법이 In sacco 전분 및 단백질 분해율에 미치는 영향)

  • The objective of this study was to examine the effects of processing methods of corn grains on protein and starch degradability in the rumen by three ruminally cannulated dry Holstein cows. The corns for these experiments were untreated; whole corn L(density; 660 g/$\ell$), whole corn H(density; 740 g/$\ell$), and treated by four different types: Ground corn, 3.8 mm, 2.8 mm, and 1.5 mm flaked corn. The results obtained were summarized as follows: For 48 hrs, the protein degradabilities were high in order, ground corn, 1.5 mm, 2.8 mm, and 3.8 mm flaked corn(82.6, 76.5, 64.5, and 33.9%, respectively). Flaked corn grains were degraded lower than ground corn. However, as increasing the processing degree of flaking, the protein degradabilities, from 4 hrs to 48 hrs, were increased. The starch degradabilities on 48 hrs were higher in 1.5 and 2.8 mm flaked corns, ground corn, 3.8 mm flaked corn(99.1, 91.5, 89.5, and 68.9%, respectively) than whole corn L(32.0%) and whole corn H(20.5%)(P<0.05). By increasing the processing degree of flaking, the protein degradabilty between 2.8 mm and 3.8 mm was increased significantly from 68.9% to 91.5%, however, that of 1.5 mm flaked corn, processed thinner, tended to be increased slightly, but was not significantly different. From 12 hrs to 24 hrs, whole corn L was degraded little more than whole corn H in starch, was not significantly different. However, after 48 hr incubation in the rumen, whole corn L was degraded more 50% than whole corn H(P<0.05). The value of degradation parameter “a” of protein was lower in all flaked corns than in ground corn. In contrast, the value of degradation parameter “a” of starch was significantly higher in all flaked corns than in ground corn(P<0.05). It seemed that by flaking the corn grains, starch particles were gelatinized, and then, starch was degraded more rapidly, while protein was degraded more slowly. Referring to these kinds of physical characteristics of grain sources in ruminal degradabilities, it is possible to synchronize the fermentation of nitrogen and carbohydrate sources, in formulating the cattle diets.

  • The objective of these studies were to examine the effects of processing methods of corn grains on in vitro dry matter digestability and in sacco degradability in the rumen by three ruminally cannulated dry Holstein cows. The results obtained were summarized as follows: The DM degradabilities, after 48 hr incubation by in sacco method, were the highest(94.4, 88.0 and 87.0%, respectively) in 1.5 mm flaked corn, ground corn, and 2.8 mm flaked corn. The 3.8 mm flaked corn was degraded significantly lower than these. Until 12 hr incubation, whole corn L tended to be degraded little more than whole corn H, was not significantly different. However, after 24 hr incubation, the significant differences between whole corn L and whole corn H were shown(P<0.05). The DM digestabilities by in vitro digestion were the highest for 1.5 mm flaked corn and ground corn(92.3 and 91.2%, respectively)(P<0.05), followed by 2.8 mm and 3.8 mm flaked corn(83.9 and 83.4%, respectively), tended to be similar to those by in sacco method. Whole corn L was digested twice more rapidly than whole corn H. Summarizing the experimental data, compared with unprocessed corns, the flaked corns were significantly increased in the degradabilities of dry matter in the rumen. In addition, as increasing the flaking degree of corn, the degradabilities of dry matter were significantly improved. Referring to these kinds of physical characteristics of grain sources in the ruminal degradabilities, it is believed to be possible to optimize the environment of the fermentation in the rumen.
