돌연변이를 통한 미세조류 Haematococcus pluvialis의 Astaxanthin 생산성의 향상 (Enhancement of Astaxanthin Production of Haematococcus pluvialis by Mutation)

  • Haematococcus pluvialis는 astaxanthin을 많이 생산하는 미세조류로써 astaxanthin은 항산화제로 면역반응을 강화시켜주며 항암효과 등을 가지고 있다. 그런데 야생종의 낮은 생장속도와 astaxanthin의 생산에는 한계가 있기 때문에 이를 극복하기위해 돌연변이 방법을 사용하였다. 돌연변이 방법으로는 자외선 조사와 EMS와 colchicines 처리를 사용하여 야생종보다 colony가 크고 더 붉은 것은 선택하였다. 선별된 돌연변이들은 carotenoid 생합성과정을 억제하는 nicotine 과 diphenylamine을 이용하여 다시 선별하였다. 그때 생존율은 40-50%이었고 여기서 선별된 균주들을 다시 규모를 키워 배양한 결과 자외선 처리한 돌연변이인 U15-5가 야생종보다 세포당 total carotenold 생산량이 1.68배 증가하였고, colchicine 처리한 DS와 M4-3은 생장속도가 $20\sim30%$ 증가하였다.

자외선이 조사된 사람 피부 섬유아세포에서 권백의 항산화와 MMP 발현에 미치는 영향에 대한 연구 (Antioxidation and Inhibition of Matrix Metalloproteinase in UV-irradiated Human Dermal Fibroblast by Selaginella tamariscina)

  • 다양한 약용식물의 피부노화에 미치는 영향을 평가하였으며, 이 중 권백(Selaginella tamariscina)은 동양에서 암환자 치료를 위한 전통약용식물로 알려져 있다. 우리는 피부노화에 대한 화장품 소재로 권백에 대한 다양한 생물학적 평가를 하였다. 권백의 항산화 효과를 알아보기 위해 DPPH radical과 superoxide anion radical 소거효과를 측정하였다. 그 결과 DPPH radical의 $IC_{50}$ 값은 $65.1{\mu}g/mL$이고, xanthine/xanthine oxidase에 의한 superoxide anion radical의 $IC_{50}$ 값은 $40.9 {\mu}g/mL$이었다. 세포내 활성산소 소거평가를 위해 사람 진피 섬유아세포(human dermal fibroblast)를 배양하여 UVB $20 mJ/cm^2$에 의해 증가된 세포내 활성산소(ROS)가 권백을 처리함으로써 활성산소 소거효과가 증가하였다. 사람 진피 섬유아세포에서 UVA에 의해 발현된 MMP-1 단백질과 mRNA가 권백에 의해 농도 의존적으로 감소하였다. 뿐만 아니라, 권백은 zymography와 semi-quantitative RT-PCR을 이용하여 UVA 조사된 사람 진피 섬유아세포에 MMP-2 (gelatinase)의 활성 감소를 확인하였다. 결론적으로 권백은 자외선에 의한 세포손상을 보호하여 항노화 화장품의 새로운 소재로 이용될 것으로 사료된다.

고선량 및 저선량 방사선 조사 마우스에서 누에동충하초(Paecilomyces japonica)의 효과 (Modification of Radiation Response in Mice by Dongchongxiacao(Paecilomyces japonica))

  • Cordyceps has a reputation for its broad biological activities and as a tonic which replenishs vital function in Chinese traditional medicines. As an attempt to obtain fundamental data for the development of new type Cordyceps, the effects of the fruiting bodies of cultivated fungus of Paecilomyces japonica grown on silkworm larvae on radiationinduced damages were investigated. We performed this study to determine the effect of Dongchongxiacao (Paecilomyces japonica) on jejunal crypt survival, endogenous spleen colony formation, and apoptosis in jejunal crypt cells and hair follicles of mice irradiated with high and low dose of gamma-radiation. Treatment with Dongchongxiacao showed no significant modifying effects on the jejunal crypt survival and endogenous spleen colony formation. The frequency of radiationinduced apoptosis was reduced by pretreatment of Dongchongxiacao (i.p.: 50 mg/kg of body weight, at 12 and 36 hours before irradiation, p<0.01). The spontaneous levels of apoptotic cells are $0.082{\pm}0.041$ in intestinal crypts and $0.231{\pm}0.084$ per hair follicle section of skin. Pretreatment of Dongchongxiacao was associated with decreases of 26.86% in intestinal crypt and 66.36% in hair follicle decrease in the number of cells with nuclei positively stained for apoptosis compared with the irradiation control group. We demonstrated for the first time that Dongchongxiacao administration could reduce the extent of apoptosis produced by radiation in the hair follicle. The results presented herein that Dongchongxiacao given before irradiation is capable of reducing the severity of cell loss as a result of apoptosis.

감마선 조사선량에 따른 버찌(Fruit of Prunus serrulata L. var. spontanea Max. wils.) 에탄올 추출물의 항산화활성 (Antioxidative Properties of Ethanolic Extracts from Flowering Cherry (Prunus serrulata L. var. spontanea Max. wils.) Fruit with Various Doses of Gamma Radiation)

  • 본 연구는 폐자원으로 버려지는 버찌의 활용 증대방안 모색을 위한 기초자료 마련을 위해 버찌의 에탄올 추출물에 0~20 kGy 선량으로 감마선 조사를 실시하고, 이에 따른 항산화활성 변화를 측정하였다. 총 폴리페놀 함량 측정결과, 10 kGy 조사구에서 30.35 mg/g으로 가장 높은 값을 나타내었으나 감마선 조사에 따른 일정한 증가나 감소의 경향은 나타내지 않았다. DPPH radical 소거능 측정결과 10 kGy 조사구에서 327.52 ppm으로 가장 낮은 $IC_{50}$값을 나타내 가장 높은 활성을 나타내었으며 대체적으로 비조사구보다는 조사구에서 높은 활성을 나타내었다. 환원력 측정 결과, 2.5 mg/mL 이하 농도에서 비조사구에 비해 조사구에서 높은 활성을 나타내었고 20 kGy 조사구에서 대체적으로 높은 환원력을 나타내었다. 아질산염 소거능을 측정한 결과 pH 4.2 이하의 pH에서 조사구 및 비조사구 모두 73~94% 이내의 높은 소거능을 나타내 pH 의존적인 아질산염 소거능을 보였으나 조사에 따른 활성 증가는 나타내지 않았다. FRAP value를 측정결과 3 kGy 조사구에서 0.38 mM로 가장 높은 활성을 나타내었다. 결론적으로 버찌 에탄올 추출물에 대한 감마선 조사는 일부 선량에서 추출물의 항산화 활성 증가를 나타내며, 사용목적에 따른 적정 선량의 감마선 조사는 생리활성물질의 증가를 유도하여 산업적인 면에서 효율적으로 이용될 수 있을 것으로 사료된다.

Protoberberine의 고급지방산 유도체합성 및 활성연구(V) (Synthesis of Protoberberine Derivatives and Their Biological Activities)

  • Irradiation of phenolbetaine in a stream of nitrogen produced 8,14-cycioberbine[1]. Compound[1] was treated with 10% HCl solution to give the 8-hydroxycycloberbine[2] in 67.7% yield. Subsequently addition of ethylchloroformate to the compound[2] gave rise to the 8-hydroxy-7-ethylcarboxy-9, 10-dimethoxy-2, 3-methylenelioxy-13-oxo-norochotensane[3] in 78% yield. Treatment of the compound[3] with bis-(2-chloroethyl)amine then lead to the 7-bis(2-chloroethyl)carbamyl-norochoteneare[4]. On the other hand the compound[5], which is the 8-methoxynorochotensane, was derived when compound[1] was treated with methanol in a few drops of BF. Treatment of the compound[6], and the compound[7], 7-bis(2-chloroethyl)-carbanyl-8-methoxy-norocheyensane, was then synthesized by reaction of the compound[6] with bis(2-chloroethyl) amine. In the other synthetic pathway when compound[5] was treated with $POCl_3$ in dried benzene, 13-chloro-6-ene-norochetensane[8] with 42% yield was formed. Finally the 13-bis-(2-chloroethyl) amino-8-methoxy-norochotensane[9] was produced when we treated the compound[8] with bis-(2-chloroethyl) amine. In another pathway, reaction between phenolbetaine which is the precursor of the compound[1] and benzoylchloride in dried chloroform gave us the 5,6,7 trihydro-2, 3-methylene-dioxy-9-chloromethyl-10, 11-dimethoxyphenylisoquinoline-8-benzoate[10] in 73% yield. The results of biological activities for these compounds are also presented in Table I and II.

잠란(蠶卵)의 초기발육과정(初期發育過程)에 따르는 RNase활성(活性) 및 핵산량(核酸量)의 변동(變動) 및 그 X선조사(線照射)에 의한 영향(影響)에 관(關)한 연구(硏究) (Variation of RNase activities and nucleic acid content of non-irradiated and irradiated eggs of Bombyx mori during early development of embryo)

  • Previously identified female pupae were X-irradiated with a dose of 1000r one day prior to moth transformation. Female mothes from irradiated and non-irradiated pupae were copulated with normal male ones and allowed to lay eggs. Fertilized eggs were collected at 6 intervals such as 5, 15, 45, 90 minutes, 12 and 40 hours after laying, and deep-freezed immediately after each collection until measurements. RNase activity and nucleic acid content were determined with each sample and following results were obtained. 1) It was proved to exist two RNases in silk worm eggs as in mammalian tissues, one active maximally at pH 5.8 and the other at pH 8.0, and the acid RNase activity was much higher than that of alkaline RNase. 2) The activity of acid and alkaline RNases increased remarkably during early development of the embryo of silk worm eggs, reaching the maximum activity at 45 minutes from laying time in non-irradiated group. There was no appreciable difference in two RNase activities for 45 minutes after laying in both control and irradiated groups, but the activity of acid and alkaline RNases in latter group was three times as much as that in former group, at 90 minutes from laying time and it was also found the acid RNase activity was 1.8 times higher than alkaline one in irradiated group. 3) The RNA-P content of control group increased considerably for initial 45 minutes, followed by a decline 45 minutes later with sight but steady increase thereafter. The RNA-P content of irradiated group, however, increased at initial 5 minutes, followed by a marked fall 90 minutes after laying, with no change thereafter. The DNA-P of control group showed a sharp increase for initial 45 minutes, followed by a decline 45 minutes later with no appreciable change thereafter, whereas that of irradiated group showed an increase at initial 15 minutes, followed by a sharp decline for following 45 minutes with a gradual increase thereafter. It was thus proved that the synthesis of nucleic acid in silk worm eggs was much suppressed by X-irradiation during early development of embryo. 4) The RNase activity varied in parallel with the RNA-P content in control group, but the RNA-P content in irradiated group was shown to be minimum value in concidence with the maximum activity of both RNases.

방사선 조사 및 녹차 폴리페놀을 첨가한 화장품의 항균효과 (Anti-microbial Effect of Irradiated Green Tea Polyphenol Addition into Cosmetic Composition)

  • 방사선 조사한 녹차 폴리페놀을 스킨과 에센스에 첨가하여 방부제를 첨가하지 않았을 때의 항균효과 및 방부효과를 검증하고 무방부제 화장품으로서의 가능성을 검토하였다. 항균력 측정 결과, C. albicans를 제외한 모든 균주에서 항균효과가 나타났으며, 피부상재균인 S. aureus에서 항균효과가 높게 나타났다. MIC 측정 결과, E. coli와 C. albicans의 경우 2,500 ppm에서, S. aureus의 경우 1,000 ppm에서 저해농도를 나타내었으며, MBC 측정 결과, C. albicans를 제외한 모든 균주에서 측정이 가능하였으며, E. coli의 경우 10,000 ppm, S. aureus의 경우 5,000 ppm에서 모든 균이 사멸하였다. 시간에 따른 생균수 측정 결과, 시간이 지남에 따라 균의 증식을 억제하여 항균력이 뛰어남을 알 수 있었다. 이와 같은 결과로 미루어 보아 녹차폴리페놀은 스킨과 에센스에 첨가 시 항균효과가 우수하여 화장품 첨가제로 이용이 가능할 것으로 사료된다.

수치에 의한 권백의 항산화 효과와 MMP 발현 저해 효과 (Effect of the Processed Selaginella tamariscina on Antioxidation and Inhibition of Matrix Metalloproteinase)

  • 권백은 동양에서 암환자 치료를 위한 전통 약용식물로 알려져 있다. 본 연구에서는 포제기술을 이용한 권백을 항노화 화장품 소재로 적용하고자 다양한 생물학적활성을 평가하였다. 열과 모래를 이용한 포제 권백은 기존 권백을 다른 목적으로 이용하고자 비교하였다. 포제 권백은 페놀함량이 증가하였고 DPPH 라디칼 소거활성도 증가하였다. 세포내 활성산소 소거평가를 위해 사람 섬유아세포를 배양하여 UVB($20 mJ/cm^2$)에 의해 증가된 세포내 활성산소가 포제 권백을 처리함으로써 활성 산소 소거효과가 증가하였다. 사람 섬유아세포에서 UVA에 의해 발현되는 MMP-1효소는 포제 권백에 의해 농도 의존적으로 감소하였다. 결론적으로 포제 권백은 페놀함량의 증가와 항산화 효과가 증가하였고, 자외선에 의한 세포손상을 보호하여 항노화 화장품의 새로운 소재로 이용될 것으로 사료된다.

Oral Administration of Lactobacillus plantarum HY7714 Protects Hairless Mouse Against Ultraviolet B-Induced Photoaging

  • Ultraviolet (UV) irradiation alters multiple molecular pathways in the skin, thereby inducing skin damage, including photoaging. In recent years, probiotics have gained interest due to their beneficial effects on skin health, such as inhibiting atopic dermatitis and improving skin immunity or inflammation. However, little is known about the effects of probiotics on UVB-induced photoaging. In this study, we evaluated the effect of Lactobacillus plantarum HY7714 against UVB-induced photoaging in human dermal fibroblasts and hairless mice. The results showed that L. plantarum HY7714 treatment effectively rescued UVB-reduced procollagen expression through the inhibition of UVB-induced matrix metalloproteinase (MMP)-1 expression in human dermal fibroblasts. Data from a western blot showed that L. plantarum HY7714 inhibited the phosphorylation of Jun N-terminal kinase, thereby suppressing the UVB-induced phosphorylation and expression of c-Jun. Oral administration of L. plantarum HY7714 clearly inhibited the number, depth, and area of wrinkles in hairless mouse skin. Histological data showed that L. plantarum HY7714 significantly inhibited UVB-induced epidermal thickness in mice. Western blot and zymography data also revealed that L. plantarum HY7714 effectively inhibited MMP-13 expression as well as MMP-2 and -9 activities in dermal tissue. Collectively, these results provide further insight regarding the skin biological actions of L. plantarum HY7714, a potential skin anti-photoaging agent.

감마선 조사가 결명자의 생육과 항산화 활성에 미치는 영향 (Effect of Gamma Ray on Germination, Growth and Antioxidant Activity of Senna tora)

  • Background: Senna tora is a flowering plant in the legume family Fabaceae. Its seeds are roasted and consumed as tea in Asia, to reduce inflammation in the liver and improve eyesight. Thus, it has been considered as an important medicinal crops in Asia. However, breeding trials to improve its genetic properties are rare. Mutation breeding by gamma ray is known to be an effective and highly successful approach for the generation of agronomically useful cultivars. Here we analyzed the effects of several dosages of gamma ray on the biological conditions of Senna tora seeds. Methods and Results: The germination rate and growth patterns of Senna tora were examined following irradiation with gamma ray at 100, 200, 300 and 400 Gy. The total phenolic compound contents and antioxidant activities of Senna tora were analyzed. Germination increased at 100 and 200 Gy in the M1 and M2 generations compared with that of the control (M0). The total phenolic compound contents and antioxidant activity of the seeds significantly decreased as the radiation dosage increased above 100 Gy in the M1 generation. Conclusions: Senna tora, irradiated with gamma ray at dosages 100, 200, 300, and 400 Gy, showed maximum germination rate at 200 Gy in the M2 generation. Plant height and leaf size gradually decreased with increasing gamma ray intensity in the M2 generation. The total phenolic compound contents decreased significantly at 400 Gy, and the related antioxidant activity was also decreased as the radiation dosage increased.
