이용도가 낮은 어패류의 가수분해물을 이용한 속성액젓의 제조 (Rapid Processing of Hydrolyzed Sauce Using Low-Usefulness Fish and Shellfish)

  • A rapid processing method for fermented sauce of favorable flavor was investigated with low-usefulness marine resources. Hydrolyzed at optimal conditions for 6 hours usuing alcalase, and separated by molecularporous membrane. It's very effective for remove bitter taste at below M.W. 100 dalton, and effective at below M.W.500 dalton. Added 2% invert sugar in fermented sauce at below M.W.500 dalton, and thermal treatment at 100$^{\circ}C$ for 30 minutes were improved flavor. Chemical composition of fermented sauce using hair tail were 80.7% of moisture, 2.2% of carbohydrate, 1.8% of total nitrogen, 1.6% of amino nitrogen and pH was 6.5. The ratio of amino nitrogen in total nitrogen was 77.8%. And chemical composition of fermented sauce used gizzard shad, kangdale, pen shell and oyster were similar to fermented sauce used hair tail. Total nitrogen were above 1.8% and the ratio of amino nitrogen in total nitrogen was 77.7∼84.2% in all fermented sauce. Amino acid contents in fermented sauce used hair tail, gizzard shad, kangdale, pen shell and oyster were 4,318.1mg%, 4,681.3mg%, 3,156.2mg% and 4,175.0mg%, respectively. And the predominant free amino acid were glutamic acid, lysine and glycine in all fermented sauce.

천연감미료(天然甘味料) Stevioside의 감미(甘味)에 관한 수응력(受應力) 시험(試驗) (Acceptability on the Sweetness of Stevioside as a Natural Sweetner)

  • 우리나라 1978년산(産) stevia 건옆중의 stevioside를 정량하였고, 정제(精製) stevioside의 단맛에 관한 경향을 알아 보기 위하여 이것과 설탕, 포도탕, 과당, 전화당, saccharin-Na, sorbitol 등과의 혼합감미, 이것과 짠맛과의 조화에 대해서 관능적으로 수응력을 시험해 본 결과를 요약하면 다음과 같다. 1. Stevia 건옆중의 total stevioside 함량은 $7.30{\sim}17.8%$이다. 2. 정제(精製) stevioside의 감미도(甘味度)는 $150{\sim}100$배 정도이다. 3. 감미 물질을 혼합해서 stevioside의 감미를 개선한 순서는 다음과 같다. 설탕>포도당>과당, 전화당>saccharin-Na 4. Stevioside에 2종의 감미 물질을 혼합하더라도 각각의 혼합효과 이상으로 개선되지 못하였다. 5. Stevioside의 단맛은 짠맛(소금)과 조화될 수 있다.

Occurrence of an invertase producing strain of Aspergillus niger LP5 isolated from longan pollen and its application in longan syrup production to feed honey bees (Apis mellifera L.)

  • Background: In northern Thailand, the longan flower is the principal nectar source for honey production. Microorganisms play a critical function in the agricultural ecology. The morphological characteristics of fungal species found in longan pollen were studied. Aspergillus spp. were found to be invertase-producing strains and were employed in the longan syrup production process. The purpose of this study was to evaluate the effects of invertase-added longan syrup on the adult honey bee population numbers that were fed by this syrup for 16 weeks. Results: Different fungal species were found in longan pollen samples. Aspergillus was the main genus, with three predominant sections: Nigri, Flavi, and Terrei. Other isolated species were Trichoderma spp., Rhizopus spp., Neurospora spp., Chaetomium spp., Fusarium spp. and Penicillium spp. However, Aspergillus spp. is the only fungal species that produces the enzyme invertase. The invertase-producing strains belonging to the Aspergillus section Nigri were found to be A. niger LP5 with an optimum activity at pH 6.0 and 60℃. When A. niger LP5 invertase was used for longan syrup processing, the highest levels of glucose (3.45%) and fructose (2.08%) were found in invertase added longan syrup (C), while fresh (A) and boiled longan syrup (B) had lower contents of both sugars. The sucrose content was detected in (A) at 4.25%, while (B) and (C) were at 4.02% and 3.08%, respectively. An appropriate amount of sugar to feed and maintain the honey bee population was considered. The data showed no statistically significant differences between the two selected forms of longan syrup compared to the sugar syrup examined by the adult honey bee population. Conclusions: The main species of isolated fungi from longan pollen were Aspergillus spp. The discovery of an invertase-producing strain of A. niger LP5 has enabled its application for enzyme utilization in the invert sugar preparation process. The adult worker bee populations fed by longan syrup from both boiled and invertase-added sources showed an increasing trend. Artificial syrup made from longan fruit to feed honey bees when natural food sources are limited can be applied.

근적외 분광법을 응용한 휴대용 벌꿀 품질 평가 장치 개발 (Development of a Portable Quality Evaluation System for Bee-honeys by Using Near Infrared Spectroscopy)

  • This study was conducted to develop a portable quality evaluation system of bee-honey by near infrared spectroscopic technique. Two kinds of bee-honeys from acacia and polyflower sources were tested in this study. The system consists of power supply, tungsten-halogen lamp, detector, and optical fiber probe. Performance of the system was analyzed by comparing the prediction accuracy of the laboratory spectrophotometer. Total of 346 spectra was divided into a calibration set and a prediction set. The PLS (Partial Least Squares) models were developed to predict the quality parameters of bee-honeys. Reflectance spectra, moisture contents, ash, invert sugar, sucrose, F/G ratio, HMF(hydroxy methyl furfural), and $C^{12}/C^{13}$ ratio of honeys were measured. The PLS models of the laboratory spectrophotometer showed good relationships between predicted and measured quality parameters of honeys in the wavelength range of 1.100~2.200 nm. The PLS analysis of the portable quality evaluation system showed good relationships between predicted and measured quality parameters of honeys in the wavelength range of 1.100~1.300 nm and 1.400~1.700 nm. The results showed the feasibility of the portable quality evaluation system to determine the quality parameters of bee-honey in the field during harvesting.

토양에서 분리한 Bacilus flexus로부터 Invertase의 생산 (Production of Invertase from Newly Isolated Strain Bacilus flexus)

  • In the present study, we isolated a new bacterial strain producing invertase (EC 3.2.1.26) and determined optimized culture condition in flask culture. The strain was identified as Bacilus flexus determined by the 16S rDNA sequencing method. The invertase was produced only in the sucrose medium as the sole carbon source. Potassium nitrate was an adequate nitrogen source for enzyme production, whereas meat peptone showed the highest bacterial growth. Enzyme production was increased about 2-fold when $MgSO_4\cdot7H_2O$ was supplemented to the growth media. The optimum temperature was found to be $30^{\circ}C$ for both enzyme production and bacterial growth. Invertase exhibited pH optima in the range 5.0-6.0 and have a temperature optimum at $40^{\circ}C$, similarly to other invertases found from different microbial sources. Several mineral ions (K and Fe) stimulated the invertase activity, whereas some bioelements (Ag, Mg, and Mn) inhibited enzyme activity. Under the optimized culture condition, the maximum enzyme production (over 250 units/mL) was achieved at 20 h. To the best of our knowledge, this is the first time to report on invertase production by Bacilus flexus.

  • 식용곤충(食用昆蟲)으로서 누에와 번데기를 분석(分析)하여 단백질(蛋白質)과 amino acid 함량(含量)을 알았고 이를 이용(利用)할 수 있는 가능성(可能性)을 알아보았다. (1) 누에와 번데기에 대(對)하여 단백질(蛋白質)을 정량(定量)한 결과(結果), $54.9{\sim}56%$ 정도의 단백질(蛋白質)이 함유(含有)되어 있음을 알았다. (2) 유리아미노산의 정량(定量)과 조성(組成)을 조사하였더니 tryptophan 을 제외(除外)한 대부분(大部分)의 아미노산이 함유되어 있으며 성장하여감에 따라 함량(含量)도 크게 변(變)하고 있음을 알게 되었다. (3) 탄수화물(炭水化物)로서 sucrose, glucose, fructose가 함유(含有)되어 있음을 확인하였다. (4) milk와의 반응(反應)을 $40^{\circ}C$ 항온조에서 방치하면서 pH와 산도변화(酸度變化)를 보았는데, 약 8시간(時間)동안 $40^{\circ}C$ 항온조에 놓아두는 경우 산도(酸度)는 약 8시간(時間)동안에는 별 변동이 없고, 그 후(後)에 변화(變化)가 심했다.

  • Processing conditions of whole sardine into modified fish sauce were investigated. Thawed and chopped sardine was homogenized and hydrolyzed using commercial proteolytic enzymes such as complex enzyme-2000($2.18{\cdot}10^4U/g solid$) and alcalase($1.94{\cdot}10^4\;U/g solid$) in a cylindrical vessel with 4 baffles and 6-bladed impeller. Optimal pH, enzyme concentration and temperature for the hydrolysis with complex enzyme-2000 were 7.0, $7\%$ (W/W) and $52^{\circ}C$, and-those with alcalase were 8.0, $6\%$ (W/W) and $60^{\circ}C$. In both cases, the reasonable amount of water for homogenization, agitation speed and hydrolyzing time were $100\%$ (W/W), 100 rpm and 210 minutes. Thermal treatment of the filtered hydrolysate at $90^{\circ}C$ for 2 hours with $6\%$ of invert sugar was adequated to inactivation of the enzymes and pasteurization of the hydrolysate. Flavor, taste and color of the hydrolysate were improved during the heating process in which the browning products might participate. The content of free amino nitrogen in the fish sauce seasoned with $15\%$ of table salt was ca. $1,640 mg\%$. Yield of the fish sauce based on the contents of proteinous and free amino nitrogen in the raw whole sardine was ca. $86\%$, and ca. $96\%$ of these compounds of the fish sauce was in the form of free amino nitrogen. The pH, salinity and histamine content of the fish sauce were $6.1\~6.3,\;14.2\~14.3\%$ and less than $10\;mg\%$.

포도주에서 분리한 Saccharomyces cerevisiae JS59가 생성하는 Invertase의 정제 및 특성 (Purification and Characterization of an Invertase Produced with Saccharomyces cerevisiae JS59 Isolated from Home-made Wine)

  • 전화당(invert sugar)을 생산하기 위하여 포도주로부터 분리한 효모가 생산하는 invertase의 특성을 조사한 결과는 다음과 같다. 포도주로부터 분리한 효모는 지방산 분석을 통하여 Saccharomyces cerevisiae JS59로 잠정 동정되었다. 본 균주가 생성하는 invertase를 ammonium sulfate 침전, DEAE-Sephadex A-50, Sephadex G-200 column chromatography 법으로 정제하였을 때 단일성을 보였으며, specific activity가 7620.9 unit/mg, 최종 회수율은 13.9로 약 14배 정제된 효소를 얻었다. 본 효소의 $K_m$ 값은 11.5 mM이었다. SDS-PAGE로부터 분자량은 38.5 kDa으로 나타났다. 정제된 invertase의 최적 pH는 5였고, pH 4에서도 94%의 높은 효소활성을 나타냈으며 4에서 6까지의 pH영역에서 안정하였고, $55^{\circ}C$에서 최적 활성을 나타내었으며 $50^{\circ}C$까지는 안정하였다. $Ag^{2+}$$Hg^{2+}$에 의해서 저해를 받았고, $Co^{2+}$, $Mn^{2+}$에 의해서는 효소활성이 증가되었으며, 기질과 효소 반응물을 thin layer chromatography로 분석한 결과, 본 효소는 기질인 sucrose를 완전히 분해하여 환원당을 생성함이 확인되었다.

한국산 벌꿀의 밀원별 단당, 이당 및 삼당류의 정량 특성 (Characterization of Traditional Korean Unifloral Honey Based on the Mono-, Di-, and Trisaccharides)

  • 한국에서 생산 및 유통되고 있는 서로 다른 밀원의 45개 벌꿀(아카시아꿀 15, 잡화꿀 15, 밤꿀 10, 사양꿀 5)을 대상으로, 단당류, 이당류 그리고 삼당류를 TMSO 및 TMSMe 유도체화를 만들어 GC/MS를 이용하여 분석하였다. 아카시아꿀, 잡화꿀, 밤꿀과 사양꿀에서 전화당의 총 함량은 각각 $69.3{\pm}2.86$, $66.4{\pm}3.28$, $62.4{\pm}2.24$$68.0{\pm}2.32%$였고, F/G 비는 1.61, 1.46, 1.90와 1.13이었다. 벌꿀시료에서 이당류는 약14종을 분리하였으며, 이전의 연구에서 보고된 cellobisoe와 melebiose는 모든 벌꿀시료에서 검출되지 않았다. 주요 이당류로는 turanose, maltulose, maltose, trehalulose, kojibiose, isomaltose, nigerose였으며, 꽃꿀에서 미량으로 존재하는 이당류로는 sucrose, ${\alpha}$-trehalose, ${\alpha},{\beta}$-trehalose, laminaribiose, palatinose, gentibiose 등으로 나타났다. 이당류의 총 함량은 maltulose, turanose, trehalulose의 함량이 높은 밤꿀에서 $14.2{\pm}2.43%$로 가장 높았으며, 아카시아 꿀에서 상대적으로 낮은 $8.79{\pm}1.76%$였다. 밀원별 벌꿀에서 7종류의 삼당류를 정량분석하였으며, 그 중 erlose는 사양꿀에서 $4.59{\pm}1.28%$로 가장 높았으며, 꽃꿀에서는 0.79-1.75%의 함량분포를 나타내었다.. 이당류와 삼당류의 총 함량은 maltulose와 turanose, trehalulose, isomaltose 등의 함량이 높게 나타난 밤꿀에서 $16.0{\pm}2.03%$로 가장 높았으며, 아카시아, 잡화꿀과 사양꿀에서는 각각 $9.70{\pm}1.75%$, $11.5{\pm}3.07%$$15.1{\pm}3.19%$의 평균함량 분포를 보였다.
