Effects of Zinc on Lipogenesis of Bovine Intramuscular Adipocytes

  • Zinc (Zn) is a micromineral and functions as a cofactor of many enzymes and its deficiency induces retardation of growth and dysfunction of the immune system in animals. This study was conducted to determine lipogenic activity of Zn in bovine intramuscular adipocytes. Preadipocytes were isolated from intramuscular fat depots of 26 month old Korean (Hanwoo) steers and cultured in media containing Zn. At confluence, the cells were treated with insulin, dexamethasone, and 1-methyl-3-isobutyl-xanthine to induce differentiation (accumulation of lipid droplets in cells). The sources of Zn were zinc chloride (${ZnCl}_2$) and zinc sulfate (${ZnSO}_4$), and the final concentrations of both Zn sources were 0, 5, 25, 50 and 100 ${\mu}$M. Glycerol-3-phosphate dehydrogenase (GPDH) activity, an index of adipocyte differentiation, was increased as the concentration of Zn in media increased showing the highest activity (25.74 ng/min/mg protein) at 25 ${\mu}$M of ${ZnSO}_4$. Supplementation of Zn during differentiation of bovine intramuscular adipocytes tended to decrease the production of nitric oxide (NO). Peroxisome proliferator-activated receptor gamma 2(PPAR$\gamma$2) gene expression was increased 10 days after differentiation induction. The current results indicate that Zn has a strong lipogenic activity in cultured bovine intramuscular adipocytes with remarkable suppression of NO production.

  • The development of marbling in cattle is closely associated with an increase in adipocyte size and number within muscle. The adipose precursor cells have the capacity to differentiate into adipocytes within the muscle during the formation of marbling. In this studies, we established the cell culture system for differentiation of intramuscular preadipocyte isolated from the sirloin of Hanwoo aged 12 months. The intramuscular preadipocyte cells exhibited a fibroblastic appearance and differentiated into adipocytes by treating confluent cells with differentiation medium containing insulin, dexamethasone, and troglitazone. When intramuscular preadipocyte cells were differentiated at 18 day, the triglyceride concentration was higher than control cells. Moreover, the thiazolidinedione treatment increased adipogenesis. RT-PCR analysis confirmed the significant expression of PPARγ mRNA during adipocyte differentiation. In conclution, our culture system used in this study allowed intramuscular preadipocyte cells to differentiated into adipocytes and intramuscular preadipocyte cells may be useful in the further study of differentiation mechanism of adipocytes in Hanwoo.

Intramuscular hemangioma formation in the masseter muscle: a case report (교근에 발생한 근육 내 혈관종: 증례보고)

  • Hemangioma is a benign vascular proliferation. Intramuscular hemangiomas are rare, accounting for less than 1% of all hemangiomas, and occur normally in the trunk and extremities. Approximately 10-20% of intramuscular hemangiomas are found in the head and neck region, most often in the masseter muscles. The typical clinical characteristic is a painful soft tissue mass without cutaneous changes. The suggested treatment is a surgical excision. We report a case of an intramuscular hemagnioma of the masseter muscle. The patient was a 56 year old male who visited our clinic complaining of left facial swelling after 2 years of follow up at a different clinic. After magnetic resonance imaging (MRI), the mass was excised under general anesthesia. The biopsy revealed the mass to be an intramuscular hemangioma. We report the clinical and pathological characteristics as well as the treatment of a case of an intramuscular hemangioma of the masseter muscle.

Comparison of crude fat content and intramuscular fat score at different position of loin from Hanwoo (한우 등심의 위치별 조지방 함량 및 근내지방도 비교)

  • This study was conducted to make certain that the intramuscular fat score at 13th rib could represent the fat content of whole loin from Hanwoo in Korea beef grading system. A total 42 loins from 23 carcasses of grade $1^{{+}{+}}$, 16 carcasses of grade $1^+$, and 3 carcasses of grade 1 of Hanwoo were used for this study. The crude fat content and intramuscular fat score at the $1^{st}$, $6^{th}$, and $13^{th}$ rib, and the last lumbar position of loin were measured. The crude fat content was the highest at the $6^{th}$ rib of loin from the carcasses of grade $1^{{+}{+}}$ and $1^+$ (p<0.05). There were no significant difference of the intramuscular fat scores measured by image analysis and beef marbling score (BMS) between the 6th and $13^{th}$ rib of loin from the carcasses of $1^{{+}{+}}$ and $1^+$ and that were significantly higher than intramuscular fat scores at the $1^{st}$ rib and the last lumbar position of the loin. From the results, we conclude that the intramuscular fat score at the $13^{th}$ rib can represent the fat content of loin when meat graders decide the grade of Hanwoo carcass.

Effectiveness of Video-Record Method on Fundamental Nursing Skill Education - Focused on Intramuscular Injection Practice - (기본간호학 실습교육에서 비디오 녹화학습의 효과연구 - 근육주사 실습교육을 중심으로 -)

  • Effectiveness of the videotaped-record learning method in teaching intramuscular injection skill was investigated using an experimental research methodology. Data was collected from 57 female students attending Fundamental nursing class from two depts. of nursing in Chung-ju and Seoul. The subjects were assigned to two groups, the experimental group of 30 and the control group of 27. The independent variable was videotaped-record learning method and the dependent variable were the degree of knowledge achivement, nursing skill achivement, competence on practicing intramuscular injection skill and satisfaction about the learning method. Data was analyzed using descriptive statistics, chi-square test, t-test and Pearson's correlation coefficient with SPSS-PC program. The results were summarized as follows ; 1. There was statistically significant difference between the experimental group and control group in knowledge achievement about intramuscular injections (t=4.721, P=.000). 2. There was statistically significant difference between the experimental group and control group in nursing skill achievement(t=3.542, P=.001). 3. There was no statistically significant difference between the experimental group and control group in competence on practicing intramuscular injection skill (t=.627, P=.533). 4. There was statistically significant difference between the experimental group and control group in satisfaction about learning method(t=4.708, P=.000). 5. There was significant correlation between nursing skill achievement and knowledge achievement(r=.233, p<.05). In conclusion, this study suggests that videotaped-record learning method is an effective learning method for achieving intramuscular injection skill as a basic nursing skills. Therefore, further study with more developed research design and other fundamental nursing skill practice will be needed to investigate the effectiveness of videotaped-recored learning method.

An Intramuscular Neurofibroma Presenting as a Thenar Mass (엄지 두덩 덩이로 발생한 근육내 신경섬유종)

  • Purpose: Neurofibromas may present as multiple or solitary lesions. Although there is no predilection site for solitary lesions, they are rare on the hand. In addition, solitary intramuscular neurofibromas are a very rare pathological type. Here, we report a rare solitary intramuscular neurofibroma in the hand. This paper examines the clinical characteristics of intramuscular neurofibroma arising from the lumbricalis in order to enable a correct diagnosis and treatment. Methods: A 32-year-old male presented with a painless mass on the palm. The physical examination revealed a $3{\times}2$ cm protruding mass that was non-tender to palpation. The vascular and sensory examinations were unremarkable, while the motor examination showed mild difficulty with flexion and extension. Magnetic resonance imaging demonstrated an enhancing solid mass between the thenar eminence and second metacarpophalangeal joint. The diagnosis of an intramuscular neurofibroma was confirmed following surgical excision and histological evaluation. Results: The pathological examination was consistent with a neurofibroma, with delicate fascicles and loose fusiform cells in a fibrous stroma, with oval or spindle-shaped nuclei and scant cytoplasm. The background matrix was pale staining and had focal myxoid stroma. There was no significant nuclear pleomorphism and no mitoses. Immunohistochemistry with S-100 was slightly positive. At the 6-month follow-up, motor and sensory function were intact and the range of motion was full. Conclusion: A neurofibroma is a rare tumor of the hand, especially the intramuscular type. Hand surgeons should consider the diagnosis of this tumor based on the examination and imaging.

Effects of Vitamins on the Differentiation of Preadipocytes from Hanwoo Cattle Adipose Tissues

  • The experiment was conducted to study the effects of water soluble vitamins and retinoic acid on the differentiation of preadipocyte from omental, subcutaneous, intermuscular and intramuscular adipose tissue of Hanwoo. Differentiation was assessed by the change in enzyme activity, glycerol-3 phosphate dehydrogenase in serum free cell culture system. Preadipocytes treated with biotin ($10{\mu}M$) and pantothenic acid ($100{\mu}M$) were significantly (p<0.05) less differentiated than those from the control in all adipose tissue depots except intramuscular tissue. Although there was no significance, vitamin C was shown to stimulate the adipocyte conversion in omental and subcutaneous, but not in intermuscular and intramuscular adipose tissues. Lower values of GPDH activity in intermuscular preadipocyte were interpreted to be caused by relatively higher amounts of protein. In this experiment vitamin C did not stimulate fat deposition in intramuscular adipose tissue but further experiments are needed on the role of vitamin C in preadipocyte differentiation. When treated with different levels of retinoic acids, differentiation of preadipocytes was significantly (p<0.05) reduced from the level of $0.5{\mu}g/ml$ in omental and intermuscular, from $50{\mu}g/ml$ in subcutaneous, and in intramuscular at $500{\mu}g/ml$, thus showing that intramuscular preadipocytes were least responsive to retinoic acid in differentiation. All-trans retinoic acid appeared to inhibit the differentiation in a dose dependent manner, regardless of adipose tissues type.

Effects of Intramuscular Fat on the Sensory Characteristics of M. longissimus dorsi in Japanese Black Steers as Judged by a Trained Analytical Panel

  • The effects of intramuscular fat on the sensory characteristics of M. longissimus dorsi in Japanese Black steers were investigated by a trained analytical panel (average 13.4 panelists). Five sets (10 head) of artificial identical twins were divided into 2 groups, high level of intramuscular fat group (HG) which utilized the fattening method of increasing intramuscular fat and low level of intramuscular fat group (LG) which did not use the above method. Regarding M. longissmus dorsi which was produced for use in the sensory evaluation, crude fat contents of HG and LG was 25.8% and 23.2% respectively (p<0.05). Warner-Bratzler shear force, water holding capacity, cooking loss and fatty acid composition of HG and LG were similar between the two groups. M. Longissimus dorsi taken from HG and LG were tested for their sensory characteristics by a trained panel. HG was given higher points for juiciness than LG (p<0.05). There were no significant differences for tenderness and flavor between the two groups. Overall acceptability which synthesized each of the sensory characteristics of HG and LG were 5.04 and 4.69 points respectively (p = 0.05). These results suggested that juiciness increased with the increase of intramuscular fat, and this raised the overall acceptability of M. longissimus dorsi.

Studies on Intramuscular Fat Percentage in Live Swine Using Real-time Ultrasound to Determine Pork Quality

  • In the modern pork industry, selection of high intramuscular fat (IMF) in pigs is necessary to improve pork quality. Ultrasound has been used previously to predict subcutaneous fat thickness and IMF in the longissimus muscles of line pigs and Real-time ultrasound has also been reported as a reliable method for estimating IMF in live pigs. So we estimate the correlation between meat quality traits and IMF percentage to investigate the possibility of utilizing real-time ultrasound technology for predicting IMF percentage in line pigs to improve pork quality. The genetic and phenotypic correlations for chemical intramuscular fat (CIMF) and ultrasound intramuscular fat (UIMF) were estimated to be 0.75 and 0.76, respectively. These results suggest that genetic factors strongly influence meat quality. The genetic and phenotypic correlation between UIMF and CIMF were 0.75, 0.76, respectively. The heritability of UIMF and CIMF were 0.48 and 0.50, respectively. So we concluded that CIMF can be replaced with UIMF and Ultrasound machines can be used to test IMF in live swine. In future, UIMF can be utilized to improve pork quality as an alternative to CIMF.

A Comparison of Continuous Intravenous Infusion and Traditional Intramuscular Injection for Postoperative Pain following Cesarean Section (제왕절개술후 통증치료로 지속적 정주방법과 고식적 근주방법의 비교)

  • Background: Recently, improvements in drug administration technology have intensified interest in the treatment of postoperative pain. this has resulted in increased use of continuous intravenous infusion of opioid and epidural opioid as alternative to traditional intramuscular administration of opioid. The goal of this study, therefore, was to document the effects of pain control and side effects following continuous intravenous infusion of morphine or meperidine and intramuscular meperidine following cesarean section. Methods: The vital signs, pain score, oxygen saturation and side effects were compared in 150 patients receiving continuous intravenous infusion of morphine, 30 ${\mu}g/kg/hr$ (n=50, group 1); continuous intravenous infusion of meperidine, 150 ${\mu}g/kg/hr$ (n=50, group 2); or intramuscular meperidine, 50mg/every 6hrs (n=50, group 3). Results: VAS (Visual Analogue Scale) was significantly decreased after 30 minutes of administration in all three groups and was significantly lower at 1 hour, but higher at 6 hours in group 3 than two other groups. Severe desaturation episode, defined as $SpO_2$<90%, occurred in the group 3(0.2%). Moderate desaturation episodes, defined as $SpO_2$ 91~95%, occurred more in group 3 than in group 1 and 2 (17.4% vs. 10.4%, 8.2%). The incidence of side effects were similar among three groups. Conclusion: The continuous infusion of opioid was more effective and safe method of postoperative pain control than traditional intramuscular injection.
