Syphacia obvelata: A New Hope to Induction of Intestinal Immunological Tolerance in C57BL/6 Mice

  • The ability of nematodes to manipulate the immune system of their host towards a Th2 and T regulatory responses has been proposed to suppress the inflammatory response. Clinical trials have proposed a useful effect of helminth infections on improvement of inflammatory disorders. In this study, we investigated the immunomodulatory effect of Syphacia obvelata infection to induce intestinal tolerance in C57BL/6 mice. Mice were infected through the cagemates with self-infected BALB/c mice. Four weeks post-infection, expression levels of $IFN-{\gamma}$, $TNF-{\alpha}$, IL-17, and IL-10 were assessed in the supernatant of mesenteric lymph node (MLN) culture. $Foxp3^+Treg$ were measured in MLN cells by flow cytometry. In the S. obvelata-infected group, the percentage of Tregs ($5.2{\pm}0.4$) was significantly higher than the control ($3.6{\pm}0.5$) (P<0.05). The levels of IL-10 ($55.3{\pm}2.2$ vs $35.2{\pm}3.2$), IL-17 ($52.9{\pm}3.8$ vs $41{\pm}1.8$), $IFN-{\gamma}$ ($44.8{\pm}4.8$ vs $22.3{\pm}2.3$) and $TNF-{\alpha}$ ($71.1{\pm}5.8$ vs $60.1{\pm}3.3$) were significantly increased in infected mice compared to the control group (P<0.05). The above results showed the potential effects of S. obvelata to induce intestinal tolerance. Therefore, it seems that S. obvelata may increase the immunological suppressive function in the intestinal tract.

  • 정상인에서 장내세균은 숙주의 면역이나 영양 흡수를 돕지만, 때로는 기회감염균으로서 그들을 위협하기도 한다. 그 중 절대 혐기성 세균인 Bacteroides fragilis는 분비되는 장독소(enterotoxin)인 Bacteroides fragilis toxin (BFT)의 유무에 따라 non-enterotoxigenic B. fragilis (NTBF)와 enterotoxigenic B. fragilis (ETBF)로 나뉜다. ETBF는 가축 및 사람에서 설사 질환 및 대장 질환을 유발한다 그러나 때때로 ETBF를 가지고 있으나 증상이 없는 사람도 존재한다. ETBF는 염증성 설사 질환, 여행자 설사 환자의 대변에서 검출되어 주목 받고 있다. 또한, 몇몇 연구를 통해 inflammatory bowel disease (IBD)나 대장염 및 대장암 환자에서 ETBF가 증가한다는 것이 밝혀졌다. 일반 C57BL/6 마우스 및 germ-free 마우스, multiple intestinal neoplasia (Min) 마우스, 토끼, Mongolian gerbil 등 여러 동물 모델에서 ETBF가 IBD나 대장염, 대장암을 유발 또는 촉진한다는 것이 발표되었다. ETBF의 유일한 병원성 인자인 BFT는 E-cadherin의 분절을 유도하여 장상피 세포의 투과성을 높인다. 이어서 ${\beta}$-catenin 신호전달계가 활성화하여 장상피세포의 증식이 증가한다. 또한 ETBF의 감염은 일반 마우스에서 급성이나 만성의 대장염을 일으키고 Min 마우스에서 종양 형성을 촉진한다. 이는 Stat3에 의존한 $T_H17$ 면역반응의 활성화를 통해 일어난다. 현재 ETBF의 검출 방법에는 크게 BFT toxin assay와 몇 가지 PCR 방법이 있다. 최근 real-time PCR과 같은 분자진단학적 기법의 발달로 일반적인 PCR보다 더 정확한 ETBF의 검출이 가능하게 되었다. 이것을 이용하여 앞으로 실제 임상에서 ETBF와 대장염 및 대장암의 발달 관계에 대한 심도 깊은 연구가 이뤄질 것으로 본다.

Lactobacillus brevis FSB - 1의 균체성분에 의한 면역증진 활성 (Immunopotentiating Activities of Cellular Components of Lactobacillus brevis FSB - 1)

  • 새로운 probiotic 유산균으로써의 잠재적 이용가능성을 평가할 목적으로, 김치로부터 분리한 Lactobacillus brevis FSB-1을 대상으로 각종 면역증진활성의 특성이 조사되었다. L. brevis FSB-1을 전균체, 세포벽, 세포질 및 균체외 획분으로 각각 분리하고 장관면역 활성을 측정한 결과, Peyer's patch 세포를 매개로 한 골수세포 증식활성의 경우, 세포벽 및 세포질 획분에서 상대적으로 높은 활성을 농도 의존적으로 보인 반면, 직접적인 골수세포 증식활성은 나타내지 않았다. 마크로파지의 활성화능은 전균체, 세포벽 및 세포질 획분에서 상대적으로 높은 활성을 보였으며, splenocyte mitogen 활성의 경우, 이들 획분에서 공히 대조군의 약 200%이상의 활성 증가가 관찰되었다. 그러나 양성대조군인 LPS의 활성에는 다소 미치지 못하였다. 한편 보체계 활성화능을 검토한 결과, 균체외 획분을 제외한 모든 획분에서 높은 활성을 보였으며, 특히 세포질 획분에서 농도 의존적으로 매우 강력한 활성을 나타냈다. 또한 세포질 획분에 의한 보체계 활성화는 anti-human C3를 이용한 2차원 면역전기영동에 의해 classical 및 alternative pathway 양 경로를 경유함을 확인할 수 있었다.

선천성 면역에 대한 Lactobacillus acidophilus의 효과 (Effects of Lactobacillus acidophilus on innate immunity)

  • Probiotics have many effects such as antihypertensive, prevention of cancer, antioxidation, reduction of dermatitis symptoms, improvement of mineral absorption, reduction of allergic symptoms, and decrease of cholesterol, However, the main role of probiotics is that they balance intestinal microbials proportion. L. acidophilus is one of probiotics and microflora in intestine. It has an acidification activity, aroma production, texture formation and probiotics properties. We studied on the roles of L. acidophilus in mice. In this study, body weights of mice were decreased when administration of L. acidophilus ($1{\times}10^{10}$ CFU) and swimming ability has been raised than a normal group after feeding on L. acidophilus ($1{\times}10^{10}$ CFU). After taking L. acidophilus ($1{\times}10^{10}$ CFU), total white cells were increased than a normal group; hemoglobin and thrombocytes were increased. The level of cholesterol and triglyceride were decreased in blood analysis. We knew L. acidophilus is related to innate immune system. We found out the secretion of cationic peptide was increased in the Lysoplate assays as a result of L. acidophilus ($1{\times}10^{10}$ CFU) administration. Appearance rate of lysozyme was also increased than the normal group on an immunohistochemistry stain. We confirmed L. acidophilus contributes to host health through innate immune system stimulation. L. acidophilus more than $1{\times}10^{10}$ CFU are thought to be beneficial for the host health and prevention of intestinal diseases in field condition.

Effect of stocking density and alpha-lipoic acid on the growth performance, physiological and oxidative stress and immune response of broilers

  • Objective: The study was conducted to evaluate the effect of stocking density and alphalipoic acid (ALA) on the growth performance, feed utilization, carcass traits, antioxidative ability and immune response of broilers. Methods: A total of 1,530 22-day-old male broilers (Arbor Acres) with comparable body weights ($731.92{\pm}5.26$) were placed into 18 cages ($2.46{\times}2.02m$) in groups of 75 birds ($15birds/m^2$, $37.5kg/m^2$; low stocking density [LD]), 90 birds ($18birds/m^2$, $45.0kg/m^2$; high stocking density [HD]) and 90 birds with 300 mg/kg ALA added to the basal diet ($18birds/m^2$, $45.0kg/m^2$; HD+ALA, high stocking density+${\alpha}$-lipoic acid); each treatment was represented by 6 replicates. The experimental period was 3 weeks. Results: The results showed that the high stocking density regimen resulted in a decreased growth, feed conversion ratio, carcass weight, thigh yield and bursa weight relative to body weight (p<0.05) on d 42. The abdominal fat yield in the HD+ALA group was lower (p = 0.031) than that of the LD group at 42 d. The superoxide dismutase and glutathione peroxidase activities in serum were increased, and malondialdehyde content decreased after adding ALA product (p<0.05) on d 42. Additionally, the serum concentrations of immunoglobulin A (IgA) and IgG were decreased (p<0.05) and the level of diamine oxidase was higher (p<0.01) in the HD group on d 42. Conclusion: The high stocking density significantly decreased broiler growth performance, feed utilization and carcass traits, increased physiological and oxidative stress and induced intestinal mucosal injury. The supplementation of ALA product in broiler diet at 300 mg/kg may reduce the adverse effects of high stocking density-mediated stress by maintaining the antioxidant system and humoral immune system.

Distribution and differential expression of microRNAs in the intestinal mucosal layer of necrotic enteritis induced Fayoumi chickens

  • Objective: Despite an increasing number of investigations into the pathophysiology of necrotic enteritis (NE) disease, etiology of NE-associated diseases, and gene expression profiling of NE-affected tissues, the microRNA (miRNA) profiles of NE-affected poultry have been poorly studied. The aim of this study was to induce NE disease in the genetically disparate Fayoumi chicken lines, and to perform non-coding RNA sequencing in the intestinal mucosal layer. Methods: NE disease was induced in the Fayoumi chicken lines (M5.1 and M15.2), and non-coding RNA sequencing was performed in the intestinal mucosal layer of both NE-affected and uninfected chickens to examine the differential expression of miRNAs. Next, quantitative real-time polymerase chain reaction (real-time qPCR) was performed to further examine four miRNAs that showed the highest fold differences. Finally, bioinformatics analyses were performed to examine the four miRNAs target genes involvement in the signaling pathways, and to examine their interaction. Results: According to non-coding RNA sequencing, total 50 upregulated miRNAs and 26 downregulated miRNAs were detected in the NE-induced M5.1 chickens. While 32 upregulated miRNAs and 11 downregulated miRNAs were detected in the NE-induced M15.2 chickens. Results of real-time qPCR analysis on the four miRNAs (gga-miR-9-5p, gga-miR-20b-5p, ggamiR-196-5p, and gga-let-7d) were mostly correlated with the results of RNAseq. Overall, ggamiR-20b-5p was significantly downregulated in the NE-induced M5.1 chickens and this was associated with the upregulation of its top-ranking target gene, mitogen-activated protein kinase, kinase 2. Further bioinformatics analyses revealed that 45 of the gene targets of gga-miR-20b-5p were involved in signal transduction and immune system-related pathways, and 35 of these targets were predicted to interact with each other. Conclusion: Our study is a novel report of miRNA expression in Fayoumi chickens, and could be very useful in understanding the role of differentially expressed miRNAs in a NE disease model.

Identification of Gene Expression Signatures in the Chicken Intestinal Intraepithelial Lymphocytes in Response to Herb Additive Supplementations

  • Anethole and garlic have an immune modulatory effects on avian coccidiosis, and these effects are correlated with gene expression changes in intestinal epithelial lymphocytes (IELs). In this study, we integrated gene expression datasets from two independent experiments and investigated gene expression profile changes by anethole and garlic respectively, and identified gene expression signatures, which are common targets of these herbs as they might be used for the evaluation of the effect of plant herbs on immunity toward avian coccidiosis. We identified 4,382 and 371 genes, which were differentially expressed in IELs of chickens supplemented with garlic and anethole respectively. The gene ontology (GO) term of differentially expressed genes (DEGs) from garlic treatment resulted in the biological processes (BPs) related to proteolysis, e.g., "modification-dependent protein catabolic process", "proteolysis involved in cellular protein catabolic process", "cellular protein catabolic process", "protein catabolic process", and "ubiquitin-dependent protein catabolic process". In GO analysis, one BP term, "Proteolysis", was obtained. Among DEGs, 300 genes were differentially regulated in response to both garlic and anethole, and 234 and 59 genes were either up- or down-regulated in supplementation with both herbs. Pathway analysis resulted in enrichment of the pathways related to digestion such as "Starch and sucrose metabolism" and "Insulin signaling pathway". Taken together, the results obtained in the present study could contribute to the effective development of evaluation system of plant herbs based on molecular signatures related with their immunological functions in chicken IELs.

한국인 신생아의 분변 미생물 균총 (Fecal Microflora of Korean Neonates)

  • Probiotic bacteria have been administered to neonates to serve as maturational stimuli for the developing gut and intestinal immune system, establish and develop the intestinal microbiota, and mediate host-microbe interactions; further, these bacteria have shown beneficial effects In the treatment and reduction of the risk of infectious diseases, necrotizing enterocolitis, and atopic disease. An LAB isolation project to identify effective lactic acid bacteria for Korean people is in progress. The average total counts of lactic acid bacteria, lactobacilli, bifidobacteria, and coliforms in the fecal samples from 2 provinces were estimated as 8.31, 5.98, 8.13, and 3.01 CFU/g. Additional samples from other provinces will be analyzed to examine the changes in the lactic bacterial counts according to the area, sex of the neonate, mode of delivery, and type of feeding. A database containing the 16S rDNA sequences and the ribosomal protein profile of all the lactic acid bacteria isolated from fecal samples will be constructed. For the effective use of probiotics, a number of clinical studies are needed to formulate guidelines for strain, subject, purpose, and dose.

Oral Administration of Mice with Cell Extracts of Recombinant Lactococcus lactis IL1403 Expressing Mouse Receptor Activator of NF-kB Ligand (RANKL)

  • Receptor activator of NF-kB ligand (RANKL) is known to play a major role in bone metabolism and the immune system, and its recombinant form has been expressed in bacterial systems for research since the last two decades. However, most of these recombinant forms are used after purification or directly using living cells. Here, there were cell extracts of recombinant Lactococcus lactis expressing mouse RANKL (mRANKL) used to evaluate its biological activity in mice. Mice were divided into three groups that were fed phosphate-buffered saline (PBS), wild-type L. lactis IL1403 (WT_CE), and recombinant L. lactis expressing mRANKL (mRANKL_CE). The small intestinal transcriptome and fecal microbiome were then profiled. The biological activity of mRANKL_CE was confirmed by studying RANK-RANKL signaling in vitro and in vivo. For small intestinal transcriptome, differentially expressed genes (DEGs) were identified in the mRANKL_CE group, and no DEGs were found in the WT_CE group. In the PBS vs. mRANKL_CE gene enrichment analysis, upregulated genes were enriched for heat shock protein binding, regulation of bone resorption, and calcium ion binding. In the gut microbiome analysis, there were no critical changes among the three groups. However, Lactobacillus and Sphingomonas were more abundant in the mRANKL_CE group than in the other two groups. Our results indicate that cell extracts of mRANKL_CE can play an effective role without a significant impact on the intestine. This strategy may be useful for the development of protein drugs.

Attenuation of Hepatic Graft-versus-host Disease in Allogeneic Recipients of MyD88-deficient Donor Bone Marrow

  • Acute graft-versus-host-disease (GVHD) is characterized by selective damage to the liver, the skin, and the gastrointestinal tract. Following allogeneic hematopoietic stem cell transplantation, donor bone marrow (BM) cells repopulate the immune system of the recipient. We previously demonstrated that the acute intestinal GVHD (iGVHD) mortality rate was higher in MyD88-deficient BM recipients than that in the control BM recipients. In the present study, the role of MyD88 (expressed by donor BM) in the pathophysiology of hepatic GVHD (hGVHD) was examined. Unlike iGVHD, transplantation with MyD88-deficient T-cell depleted (TCD) BM attenuated hGVHD severity and was associated with low infiltration of T cells into the liver of the recipients. Moreover, GVHD hosts, transplanted with MyD88-deficient TCD BM, exhibited markedly reduced expansion of $CD11b^+Gr-1^+$ myeloidderived suppressor cells (MDSC) in the liver. Adoptive injection of the MDSC from wild type mice, but not MyD88-deficient mice, enhanced hepatic T cell infiltration in the MyD88-deficient TCD BM recipients. Pre-treatment of BM donors with LPS increased MDSC levels in the liver of allogeneic wild type BM recipients. In conclusion, hGVHD and iGVHD may occur through various mechanisms based on the presence of MyD88 in the non-T cell compartment of the allograft.
