Cytotoxicities of Tumor-specific T Lymphocytes Primed by Glioma Apoptotic Body - or Glioma Cell Lysate-pulsed Dendritic Cells

  • Objective : The choice of tumor antigen for dendritic cell[DC]-loading has still been an unresolved problem in the DC-based vaccine strategies against malignant gliomas that has not been found well-characterized tumor specific antigens. In this study, we compare tumor-specific T cell response induced by glioma apoptotic body[GAB]-pulsed DCs to response induced by glioma cell lysate-pulsed ones quantitatively. Methods : DCs generated in the presence of granulocyte macrophage-colony stimulating factor and interleukin[IL]-4 from peripheral blood mononuclear cells[PBMCs] of HLA-A2 positive healthy donors were cultured. Each GABs and glioma cell lysate generated from HLA-A2 positive T98G glioblastoma cells were co-incubated with DCs. $CD8^+$ T lymphocytes isolated from PBMCs of same donors were cultured in media containing IL-2 and either stimulated by GAB- or lysate-pulsed DCs three times at a weekly interval. The interferon[IFN]-${\gamma}$ concentrations of each cell culture supernate were measured by enzyme immunoassay technique. Cytolytic activity of the generated cytotoxic $CD8^+$ T cells either stimulated with GAB- or lysate-pulsed DCs was determined by a standard 4-h $^{51}Cr$-release assay. Results : IFN-${\gamma}$ production and cytolytic activity of effector T cells stimulated by GAB-pulsed DCs were significantly higher than those of T cells stimulated by lysate-pulsed ones. Conclusion : These results indicate the choice of antigen is a critical determinant in the induction of antitumor immunity against malignant glioma. Antigen preparations from GABs represent a promising alternative to glioma cell lysate in DC-based glioma vaccine strategies.

RAW 264.7 대식세포와 염증유도 동물모델에서 산겨릅나무 추출물의 항산화 및 항염증 효과 (In Vitro and In Vivo Anti-Oxidative and Anti-Inflammatory Activities of Acer tegmentosum Maxim Extracts)

  • 산겨릅나무 $80^{\circ}C$ 열수 및 95% 에탄올 추출물을 제조하여 항산화 및 항염증 효과를 확인한 결과 추출물은 우수한 항산화 효과를 나타내었고 RAW 264.7 세포에서도 항염증 효과를 보였으며, 그중 95% 에탄올 추출물의 효능이 더 큰 것으로 나타났다. 따라서 효능이 더 뛰어난 산겨릅나무 에탄올 추출물을 이용하여 in vivo 실험에서 추출물의 세포 보호효과 및 항염증 효능을 재확인하였다. ICR mouse에 일주일간 100 mg/kg B.W 산겨릅나무 에탄올 추출물을 경구투여한 후 LPS를 처리하여 염증반응 및 산화적 스트레스를 유도시켜 Comet assay, serum tumor necrosis $factor(TNF)-{\alpha}$$interferon-{\gamma}$, 장점막세포에서의 $TNF-{\alpha}$와 interleukin-6에 대한 생성능을 측정한 결과 산겨릅나무 에탄올 추출물은 DNA 손상을 억제하였으며 사이토카인 생성을 유의적으로 억제하여 항산화와 항염증 효과를 보였다. 따라서 본 연구에서 사용한 산겨릅나무의 에탄올 추출물은 산화적 스트레스 및 염증을 억제하는 효과를 나타내는 세포보호 기능적 소재로서 가능성이 있는 것으로 생각한다.

동충하초 추출물이 생쥐 비장세포에 미치는 영향 (Research on Effects of Cordyceps Sinensis in Spleen Cells of Mouse.)

  • Objectives : This study was carried out to know the effect of Cordyceps sinensis(CS) on the immune inflammatory responses of spleen cells and function or immunocytes of the normal mouse. Methodes: We investigated effects of Cordyceps sinensis(CS) on normal immunocytes, gene expression of IL-12, IFN-$\gamma$ and surface-receptor expression of $CD3_{\epsilon}+$, CD4+, CD8+ and CD19+ cells were measured by PCR and FACS. Results : CS activated adhisive splenic cells morphologically as compared with the control group in the normal spleen cells of BALB/C mice. CS enhanced gene expression of interleukin-12 and interferon-gamma in a dose-dependent manner in the normal spleen cells of BALB/C mice. CS reduced the number of activating cells and surface-receptor expression of CD4+, CD8+ and CD19+. Conclusion : Cordyceps sinensis will be used as a stable remedium in the auto-immune diseases.

Anti-inflammatory and radical scavenging properties of Verbena officinalis

  • Verbena officinalis (Verbenaceae) has been used as herbal medicine or health supplement in both Western and Eastern countries for centuries. In the present study, we examined the anti-inflammatory and antioxidant activities of the methylene chloride fraction of V. officinalis (VMC). To elucidate the anti-inflammatory properties of VMC, we investigated the inhibition effects of nitric oxide production in interferon-gamma (IFN-$\gamma$) and lipopolysaccharide-stimulated mouse peritoneal macrophages. VMC suppressed nitric oxide production, inducible nitric oxide synthase and cyclooxygenase-2 expression dose-dependently without notable cytotoxicity. In various radical scavenging assays, VMC exhibited strong scavenging effect on DPPH radical, superoxide radical, nitric oxide radical and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) radical. VMC also showed potent reducing power. These findings strongly suggest that VMC may be beneficial in oxidative stress-mediated inflammatory disorders.

Fiber-Optic Sensor Simultaneously Detecting Localized Surface Plasmon Resonance and Surface-Enhanced Raman Scattering

  • This study reports a fiber-optic sensor detecting biomolecule by simultaneously monitoring localized surface plasmon resonance (LSPR) from gold nanoparticles (Au NPs) of ca. $50{\pm}5$ nm attached on one end of optical fiber and surface enhanced Raman scattering (SERS) of the reporter molecules adsorbed on the gold surfaces as an additional sensing tool. The sensor was fabricated by immobilizing Au NPs on one end of an optical fiber by chemical reaction. LSPR and SERS signals of the sensor were measured using various refractive indices solutions. Finally, the sensor was applied to observe real-time LSPR sensor-gram and SERS spectra of the reporter molecule of 4-aminothiphenol during the antibody-antigen reaction of interferon-gamma (IFN-${\gamma}$) as a proof-concept experiment of biological applications.

Anti-Tumor and Immunoregulatory Effects of Fermented Papaya Preparation (FPP: SAIDO-PS501)

  • Various beneficial effects have been described for fermented papaya preparation (FPP: SAIDO-PS501) based on its anti-oxidative and anti-inflammatory functions. The present study was designed to determine the effects of FPP on carcinogenesis in vivo, and immunomodulatory function in vitro. Mice were injected with RL male 1 cells subcutaneously or 3-methylcholantherene (MCA) intravenously to induce cancer and orally or intraperitoneally treated with FPP solution. Human peripheral blood mononuclear cells (PBMC) were obtained from healthy volunteers and patients with atopic dermatitis, treated with FPP, and subjected to measurement of cytokine production and changes in Foxp3-expressing regulatory T cell (Treg) stimulated with phytohemagglutinin (PHA). Administration of FPP suppressed tumor size and the incidence of malignancy. In vitro, treatment of PBMC with FPP induced IL-$1{\beta}$, $TNF{\alpha}$ and $IFN{\gamma}$ production. Moreover, FPP suppressed proliferation of PHA-stimulated Foxp3-expressing Treg. These results suggest that FPP has chemotherapeutic properties.

인진 추출물의 소염진통작용 (The Anti-inflammatory and Analgesic Activities of Artemisia capillaris Thunberg)

  • Inflammatory mediators such as interleukin-1 (IL-1), tumor necrosis $factor-{\alpha}\;(TNF-{\alpha}),\;interferon-{\gamma}\;(IFN-{\gamma})$ and lipopolysaccharide (LPS) are thought to play major roles in joint diseases such as a rheumatoid arthritis (RA), and there is considerable evidence playing a role for these cytokines in osteoarthritis (OA). Therefore, we have studied the effects on anti-inflammation and analgesic by ethyl acetate fraction from 70% ethanol extract of Artemisia capillaries (EAC). As a positive control, apigenin, which is known as an anti-inflammatory agent as an iNOS inhibitor, was used and showed the dose-dependent inhibitory effect. EAC showed strong inhibitory efficacy against cytokine-induced proteoglycan degradation, $PGE_2$ production, nitric oxide (NO) production, and matrix-matalloproteinases (MMPs) expression in rabbit articular chondrocyte. In the writhing test induced by acetic acid, EAC $(200{\sim}400\;mg/kg)$ exhibited a dose-dependent inhibition of writhing. The results indicate that EAC have anti-inflammatory and analgesic activities, and could be a good herbal medicine candidate for curing of RA and/or OA.

Immunomodulating and Anticoagulant Activity of Glycosaminoglycans Derived from Porcine Testis

  • Glycosaminoglycans (GAGs) were isolated from the porcine testis, and their immuno-modulating and anticoagulant activity was investigated. From anion exchange chromatography (Dowex Macropolous Resin) used for further isolation of porcine testis GAGs (PT-GAGs), two fractions (PT-GAG-1.5 and PT-GAG-16) eluted by different salt concentration were obtained. In immunomodulating activity test, PT-GAG-1.5, but not PT-GAG-16, significantly enhanced the growth of murine peritoneal macrophages. In addition, treatment with PT-GAG-1.5 induced the production of cytokines, interleukin-1$\beta$ (IL-1$\beta$), interferon-${\gamma}$ (IFN-${\gamma}$) and tumor necrosis factor-$\alpha$ (TNF-$\alpha$), from murine microphages. Unexpectedly, both of PT-GAGs had no effect on the growth of murine splenocytes. The anticoagulant activity of PT-GAG-1.5 and PT-GAG-16 was examined by activated partial thromboplastin time (aPTT) assay and thrombin time (TT) assay. Both of PT-kGAGs significantly increased the clotting times of aPTT and TT in a dose-dependent manner. The anticoagulant activity of PT-GAG-16 was found to be higher than that of PT-GAG-1.5. These results suggest that PT-GAGs possess biological activities such as immunomodulating activity and anticoagulant activity.

Analysis of Biological Experiments on the Anti-inflammatory and Antipyretic Effects of Hwangryeonhaedok-tang

  • Objectives: To establish scientific and objective evidence for the use of a Korean medicine, articles regarding Hwangryeonhaedok-tang (HRHDT), a herbal medicine frequently used in Korean medical clinics and hospitals, were gathered and analyzed. Methods: The articles were classified as being from domestic or international journals, and by their year of publication. The mechanisms of the anti-inflammatory and antipyretic effects of HRHDT were investigated. Results: Of the 25 articles analyzed, 7 were published from Korea, 7 were from China, and 11 were from Japan. HRHDT showed anti-inflammatory and antipyretic effects through the regulation of the expression of Th1 cytokines including interleukin-2 (IL-2), IL-8, interferon-${\gamma}$ (IFN-${\gamma}$), and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$); and Th2 cytokines including IL-4, IL-6, and IL-12, which inhibit leukotriene B4 (LTB4), cyclooxygenase-2 (COX-2), prostaglandin E2 (PGE2), and inflammatory cells. It also lowered preprodynorphin (PPD), and corticotropin-releasing factor (CRF) in the peripheral nerve system and hypothalamus. Conclusions: We speculate that the anti-inflammatory and antipyretic effects could be related to the therapeutic efficacy of HRHDT in removing pathogenic fire and heat.

십전대보탕의 면역활성에 관한 기초 실험 연구 문헌 분석 (Analysis of Biological Experiment on Immunoactivity of Sipjeondabo-tang)

  • To investigate scientific evidence for the use of a Korean Medicine (KM), the papers regarding Sipjeondabo-tang (Shiquandabu-tang) frequently used in Korean medical clinics and hospitals were collected and analyzed. The papers were classified as being from domestic or international journals and by the year of publication. The mechanisms of immuno-activity Sipjeondabo-tang (Shiquandabu-tang) were researched. Among 41 papers, 4 were published as Korean papers whereas 7 were as Chinese papers and 40 were as Japanese papers. Sipjeondabo-tang (Shiquandabu-tang) regulated the productions of Th1 cytokines such as interleukin-2 (IL-2) and interferon-${\gamma}$ (IFN-${\gamma}$), and Th2 cytokines such as IL-4, IL-5, IL-6, IL-12 improving the function of immune organ including bone marrow, spleen, liver, thymus and peyer's patch. It also modulated the releases of immunoglobulin-G (IgG) and IgM. Thus, Sipjeondabo-tang (Shiquandabu-tang) balanced the Th1/Th2 immune responses as well as T/B cell. The regulation of immunoactivity could be speculated as an objective and scientific evidence of Sipjeondabo-tang (Shiquandabu-tang).
