본 연구는 DNCB로 유도한 아토피 피부염 유사 병변 실험동물 모델을 활용한 in vivo와 피부 각질세포주를 이용한 in vitro 연구를 통하여 아로니아와 모링가 종자 추출물의 아토피 피부염 개선 효능을 분석하였다. 조직학적 검사 결과 아로니아 추출물(AM) 또는 모링가 종자 추출물(MO) 단독 투여 시보다 1:1로 혼합한 복합물(AM-MO)을 투여했을 때 표피 부분의 과증식과 염증세포의 침윤이 감소하였으며, 염증 및 부종을 나타내는 피부 두께를 측정한 결과에서도 추출물 단독으로 투여하였을 때보다 AM-MO 투여 시 DNCB 도포군과 비교하여 유의하게 감소함을 보여주었다. 피부장벽 손상으로 인한 수분 손실에 대해서는 유의한 효과를 나타내지 않았으며, 혈청 IgE 수준은 AM과 MO 각각 단독으로 투여했을 때보다 혼합 투여하였을 때 현저하게 억제되었다. In vitro에서는 피부각질세포주인 HaCaT 세포를 $TNF-{\alpha}$ 및 $IFN-{\gamma}$로 자극하면 발현이 증가하는 Th2 케모카인 TARC 및 MDC에 대하여 mRNA 수준을 분석하였을 때, 이 역시 단독 추출물 처리 시보다 복합물 처리 시 억제 효과가 더 뛰어남을 확인하였다. 종합적으로 DNCB로 유도된 아토피 피부염 in vivo 모델에서 아로니아 및 모링가 종자 추출물은 단독으로 투여하는 것보다 복합물로 투여하였을 때 현저하게 피부 염증 및 혈청 내 IgE 생성을 억제하였으며, 피부각질 세포주를 이용한 in vitro 연구에서도 추출물 단독 처리보다 혼합하여 처리하였을 때 더 유의한 Th2 케모카인 억제 활성을 나타내었다. 이로 인해 아로니아 및 모링가 종자 추출물은 복합적으로 사용하였을 때 상호작용에 의해서 더 효과적인 아토피 피부염 개선 식품 소재로써 활용될 가능성이 높을 것으로 생각된다.
알레르기성 접촉피부염은 T세포와 대식세포가 관여하는 세포매개성 면역반응으로 항원에 노출된 뒤 수 일 후에 증상이 나타나는 지연형 반응이다. 그 과정은 감작기와 유발기로 나뉘는데 감작기에는, 표피장벽을 통해 유입된 항원이 표피기저층에 있는 항원전달세포에 의해 처리된 후 림프절로 이동되어 T세포에 의해 인식되고 그 T세포는 항원특이 T세포로 활성화된다. 유발기는 동일 항원이 재감작될 때 항원특이 T세포의 반응을 활성화시키고 다양한 cytokine 분비를 통해 염증세포를 항원 유입부위로 이동시킨다. 본 연구에서는 DNCB로 알레르기성 접촉피부염을 유발한 개의 모델에 폴리감마글루탐산의 항염증 효과를 평가하였다. 폴리감마글루탐산은 12일간 적용하였고 실험기간 동안 이틀 간격으로 피부 생리학적 지표를 측정하였으며 적용 후 cytokine 측정과 조직병리학적 검사를 실시하였다. DNCB 적용후 피부 생리학적 지표의 변화로 표피경유수분손실, 피부 수화도, 피부 두께 그리고 홍반지수는 증가하였고 피부 산도는 감소하였다(p < 0.05). 조직병리학적 검사결과 염증세포 침윤과 부종성 변화에 의한 상피두께 증가 및 진피 결합조직의 감소가 특징적으로 나타났다. 또한 진피에서 pro-inflammatory cytokine인 TNF-${\alpha}$와 IFN-${\gamma}$ 수치 및 상피에서 apoptotic change의 지표인 caspase-3와 PARP 면역반응세포의 수치가 유의적으로 증가하였다(p < 0.01). 하지만 폴리감마글루탐산 적용으로 피부 생리학적 지표(p < 0.05) 및 조직병리학적 변화가(p < 0.01) 기본 수치로 회복되었다. 따라서 본 연구를 통해 개에서 DNCB에 의한 알레르기성 접촉피부염 유발 및 폴리감마글루탐산의 우수한 항염증 효과를 확인하였고 그 결과 폴리감마글루탐산은 향후 피부염에 대한 치료제로 사용할 수 있을 것으로 기대된다.
In order to investigate anti-inflammatory, anti-allergic and anti-obesity activities of Samchulkunbi-tang (SCT; Shen zhu jian pi-tang) water and 70% ethanol (EtOH) extracts, in vitro inhibitory activities against nitric oxide (NO), prostaglandin $E_2$$PGE_2$), interleukin (IL)-6 and tumor necrosis factor (TNF)-${\alpha}$ production in lipopolysaccharide-stimulated RAW 264.7 cells, and macrophage-derived chemokine (MDC/CCL22) and regulated on activation of normal T-cell-expressed and -secreted (RANTES/CCL5) production in TNF-${\alpha}$/interferon-${\gamma}$-stimulated HaCaT and BEAS-2B cells as well as glycerol-3-phosphate dehydrogenase (GPDH) activity and leptin production in 3T3-L1 cells were determined. A HPLC was used for quantification of the seven marker components (albiflorin, paeoniflorin, liquiritin, naringin, hesperidin, poncirin and glycyrrhizin) of SCT water and 70% EtOH extracts. SCT showed inhibitory effects against MDC and RANTES production in HaCaT cells, as well as RANTES production in BEAS-2B cells. In addition, SCT reduced not only NO, $PGE_2$, IL-6 and TNF-${\alpha}$ production in RAW 264.7 cells, but also GPDH activity and leptin production in 3T3-L1 cells. Furthermore, the biological activities and the contents of six compounds (except paeoniflorin) were higher in 70% EtOH extract than water extract. These results suggest that SCT has anti-inflammatory, anti-allergic and anti-obesity activities. These efficacies of 70% EtOH extract are relatively higher than that of water extract.
Objectives : This study evaluated activities and ingredient contents concerning extracts according to extraction solvents of Insampaedok-san (IS, Renshen bai du-san). Methods : The herbal constituents of IS were extracted with water and 70% ethanol at $100^{\circ}C$ for 2 hr. Using the HPLC system, the six ingredient contents of different solvent extracts of IS were analyzed. The nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$) production and proinflammatory cytokines were measured in RAW264.7 cells stimulated with lipopolysaccharide (LPS). The macrophage-derived chemokine (MDC/CCL22) and regulated on activation normal T-cell expression and secreted (RANTES/CCL5) production were measured in HaCaT and BEAS-2B cells stimulated tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interferon-${\gamma}$ (IFN-${\gamma}$). The activities of glycerol-3-phosphate dehydrogenase (GPDH) and leptin level were measured in 3T3-L1 cells. Results : The calibration curves showed good linearity ($r^2$=1.0000) for different concentration ranges. The contents of liquiritin, naringin, hesperidin, neohesperirin and glycyrrizin in 70% ethanol extracts of IS were relatively higher than that of water extract, however the content of ferulic acid in 70% ethanol and water extract of IS were similar. The extraction solvents of water and 70% ethanol were evaluated inhibitory effect on the production of NO, $PGE_2$, TNF-${\alpha}$ and IL-6 in RAW 264.7 cells. Their extractions were inhibitory effect on production of MDC/CCL22 and RANTES/CCL5 in HaCaT cell and BEAS-2B cell, respectively. In addition, evaluated reduced on GPDH activity and leptin level in 3T3-L1 preadipocyte cell. Conclusions : Our results suggest that IS extracts were inhibitory effects of disease such as inflammation, allergies and obesity.
In order to investigate experimentally that the effect of aqua-acupuncture solution of Paeonia lactiflora on arthritis of mice induced by collagenII, the author performed several experimental items : that is increase, paw thickness, DTH, weight of spleen, hematological change, expression of $CD4^+$, $CD8^+$, $CD19^+$, gene expression of IL-1, IL-6, IL-12, IFN-${\gamma}$, TNF, proliferation of synovial cells and cytotoxicity. The results were obtained as follows; 1. Inhibitory effects of aqua-acupuncture solution of Paeonia lactiflora on arthritis induced by collgenII. 1) In incidence, paw edema, AI and DTH were inhibited as compared with control group. 2) The splenic weight was increased and the number of leukocytes was decreased as compared with control group. 3) The number of $CD4^+$, $CD8^+$ activated cells and surface-receptor expression were increased as compared with control group. 4) In hematological change, total protein, creatinine and LDH were decreased significantiy as compared with control group. 2. FACS analysis on normal BABL/c of spenic cells treated with aqua-acupuncture solution of Paeonia lactiflora. 1) Aqua-acupuncture solution of Paeonia lactiflora activated adhesive splenic cells of mice morphologically in vitro. 2) Aqua-acupuncture solution of Paeonia dactiflora enhanced the gene expression of IL-12 and also enhanced that of interferon-${\gamma}$ remarkably. 3) Aqua-acupuncture solution of Paeonia lactiflora reduced the number of $CD4^+$, $CD8^+$, $CD19^+$ activated cells and their surface-receptor expression as compared with control group. 3. Effects of aqua-acupuncture solution of Paeonia lactiflora on human synovial cells. 1) In cytotoxicity against synovial cells, aqua-acupuncture solution of Paeonia lactiflora didn't show cytotoxicity at concentration of $10-100{\mu}g/m{\ell}$ but showed significantly at concentration of $200-400{\mu}g/m{\ell}$ as compared with control group. 2) Aqua-acupuncture solution of Paeonia lactiflora reduced the gene expression of IL-6, IL-$1{\beta}$ and TNF-${\alpha}$. 3) Aqua-acupuncture solution of Paeonia lactiflora inhibited proliferation of synovial cells at concentration of 100 and $200{\mu}g/m{\ell}$.
Objective: The genetic effects of an individual on the phenotypes of its social partners, such as its pen mates, are known as social genetic effects. This study aims to identify the candidate genes for social (pen-mates') average daily gain (ADG) in pigs by using the genome-wide association approach. Methods: Social ADG (sADG) was the average ADG of unrelated pen-mates (strangers). We used the phenotype data (16,802 records) after correcting for batch (week), sex, pen, number of strangers (1 to 7 pigs) in the pen, full-sib rate (0% to 80%) within pen, and age at the end of the test. A total of 1,041 pigs from Landrace breeds were genotyped using the Illumina PorcineSNP60 v2 BeadChip panel, which comprised 61,565 single nucleotide polymorphism (SNP) markers. After quality control, 909 individuals and 39,837 markers remained for sADG in genome-wide association study. Results: We detected five new SNPs, all on chromosome 6, which have not been associated with social ADG or other growth traits to date. One SNP was inside the prostaglandin $F2{\alpha}$ receptor (PTGFR) gene, another SNP was located 22 kb upstream of gene interferon-induced protein 44 (IFI44), and the last three SNPs were between 161 kb and 191 kb upstream of the EGF latrophilin and seven transmembrane domain-containing protein 1 (ELTD1) gene. PTGFR, IFI44, and ELTD1 were never associated with social interaction and social genetic effects in any of the previous studies. Conclusion: The identification of several genomic regions, and candidate genes associated with social genetic effects reported here, could contribute to a better understanding of the genetic basis of interaction traits for ADG. In conclusion, we suggest that the PTGFR, IFI44, and ELTD1 may be used as a molecular marker for sADG, although their functional effect was not defined yet. Thus, it will be of interest to execute association studies in those genes.

Background: Delayed hypersensitivity plays a large role in the pathogenesis of tuberculous pleural effusion (TPE). Macrophages infected with live Mycobacterium tuberculosis (MTB) increase the levels of adenosine deaminase2 (ADA2) in the pleural fluid of TPE patients. However, it is as yet unclear whether ADA2 can be produced by macrophages when challenged with MTB antigens alone. This study therefore evaluated the levels of ADA2 mRNA expression, using monocyte-derived macrophages (MDMs) stimulated with MTB antigens. Methods: Purified monocytes from the peripheral blood mononuclear cells of healthy volunteers were differentiated into macrophages using granulocyte-macrophage colony-stimulating factor (GM-CSF) or macrophage colony-stimulating factor (M-CSF). The MDMs were stimulated with early secretory antigenic target protein 6 (ESAT6) and culture filtrate protein 10 (CFP10). The mRNA expression levels for the cat eye syndrome chromosome region, candidate 1 (CECR1) gene encoding ADA2 were then measured. Results: CECR1 mRNA expression levels were significantly higher in MDMs stimulated with ESAT6 and CFP10, than in the unstimulated MDMs. When stimulated with ESAT6, M-CSF-treated MDMs showed more pronounced CECR1 mRNA expression than GM-CSF-treated MDMs. Interferon-${\gamma}$ decreased the ESAT6- and CFP10-induced CECR1 mRNA expression in MDMs. CECR1 mRNA expression levels were positively correlated with mRNA expression of tumor necrosis factor ${\alpha}$ and interleukin 10, respectively. Conclusion: ADA2 mRNA expression increased when MDMs were stimulated with MTB antigens alone. This partly indicates that pleural fluid ADA levels could increase in patients with culture-negative TPE. Our results may be helpful in improving the understanding of TPE pathogenesis.
Type of dietary direct-fed microbials (DFMs) or poultry litter could directly influence the composition of gut microbiota. Gut microbiota plays an important role in shaping the developing immune system and maintaining the homeostasis of the mature immune system in mammal and chickens. The present study was carried out to investigate the interaction among litter, DFMs and immunity in broiler chickens exposed to a field-simulated environment. Immune status of broiler chickens was assessed by serum antibodies against Eimeria spp. and Clostridium spp. and intestinal cytokine mRNA expression. The current experimental design had a $3{\times}2$ factorial arrangement of treatments with three types of litter, i.e., fresh litter or used litter that was obtained from a farm with no disease outbreak (used litter) or a farm with history of a gangrenous dermatitis outbreak (GD litter), and two dietary treatments with or without DFMs. It was found that either DFM addition or type of litter significantly affected anticoccidial antibody levels of broiler chickens at d 42. In general, dietary DFMs increased the anticoccidial antibodies in the fresh-litter raised chickens, but lowered the levels in the GD-litter raised chickens. Serum antibodies against Clostridium perfringens ${\alpha}$-toxin were significantly (p<0.05) higher in chickens raised on GD litter compared with those raised on fresh litter. Cytokine mRNA expression was significantly (p<0.05) altered by either the type of litter or DFMs. Of interest, dietary DFMs lowered interferon-${\gamma}$, interleukin 1beta, and CXCLi2 cytokine mRNA expression in chickens raised on fresh litter but increased them in GD-litter raised chickens. In conclusion, dietary DFMs modulate various immune parameters of broiler chickens, but the DFM-mediated effects were dependent upon the type of litter on which chickens were raised.
본 연구는 홍삼추출물(홍삼단)과 생약복합추출물(헤모힘)이 주요 기능성분은 다르지만 면역활성이라는 생리활성 측면에서 동일한 효과를 인정하고 있어서 이들의 병용 처리가 면역세포에 어떠한 영향을 미치는지에 대해 알아보기 위하여 수행되었다. 면역활성능에 관한 평가를 진행하기 위하여 마우스의 골수에서 분리한 미분화 골수세포를 선천면역에서 중요한 역할을 수행하는 대식세포 및 수지상세포로 분화시킨 후 홍삼추출물과 생약복합추출물을 병용 처리하였을 때 대식세포 및 수지상세포의 세포 증식능 및 사이토카인의 분비능이 증가되는 것으로 관찰되었다. 또한 활성화된 탐식(면역)세포의 세포 표면에서 발현되는 CD80과 CD86의 발현과 탐식(면역)세포의 항원제시에 밀접한 관련이 있는 주 조직적합성 복합체(MHC class II)의 발현이 홍삼추출물과 생약복합추출물의 병용 처리구에서 유의적으로 증가되는 것으로 관찰되었다. 또한 후천면역에서 중요한 역할을 수행하는 면역 T 세포가 다량으로 분포하는 비장 조직으로부터 비장세포를 분리하여 홍삼추출물과 생약복합추출물을 병용처리하였을 때 세포 증식능 및 면역활성을 유도하는 Th1 세포가 분비하는 사이토카인의 함량이 증가되는 것으로 나타났다. 이러한 결과로 미루어 볼 때 홍삼추출물과 생약복합 추출물의 병용 처리는 선천면역뿐만 아니라 후천면역에 관여하는 다양한 면역세포의 활성화에 직 간접적으로 다양한 상승작용을 미치는 것으로 사료된다.
만성C형 간염환자 160예를 대상으로 immunoblot방법과 RT-PCR-hybridization법을 실시하여 임상검사의 유용성을 알아보았다. 양성 150예 중 133예만 RT-PCR-hybridization 법에서 양성을 나타내어 immunoblot법의 진양성율은 88.6%를 나타났으며 두 방법 간의 일치율은 89.3%를 나타났다. 한국인의 HCV 아형을 알기 위하여 혈청형과 유전형을 실시하였다. HCV혈청형 1형과 2형 그리고 1b와 2a유전형이 가장 많은 C형간염 감염원으로 나타났다. 49예를 혈청형과 유전형을 서로 비교한 결과, 혈청형 검사에서 28예(57.1%)가 1형, 21예(42.9%)가 2형으로 나타났으며 유전자형 검사에서 1b형이 25예(51.0%), 1b/2b를 나타낸 예가 1예(2.0%), 2a형이 17예(34.7%), 2a/2c를 나타낸 예가 4예(8.2%), 그리고 2b형이 2예(4.1%)로 나타났다. 본 연구에서는 HCV 간이검출법으로는 immunoblot방법이 유용하며, immunoblot방법 양성 확진검사로는 RT-PCR-hybridization법을 실시하였다. 혈청형이 C형간염의 치료 나 진행과정을 관찰하기 위해서는 유전형보다 유용하나 인터페론 치료효과는 1형과 2형 혈청형별에 따라 차이를 보이지 않았다.

③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
