Background: Cell-free DNA circulating in blood is a candidate biomarker for malignant tumors. Unlike uniformly truncated DNA released from apoptotic non diseased cells, DNA released from necrotic cancer cells varies in size. Objectives: To measure the DNA integrity index in serum and the absolute DNA concentration to assess their clinical utility as potential serum biomarkers for colorectal carcinoma (CRC) compared to CEA and CA19-9. Materials and Methods: Fifty patients with CRC, 10 with benign colonic polyps and 20 healthy sex and age matched volunteers, were investigated by real time PCR of ALU repeats (ALU q-PCR) using two sets of primers (115 and 247 bp) amplifying different lengths of DNA fragments. The DNA integrity index was calculated as the ratio of q-PCR results of ALU 247/ALU 115bp. Results: Serum DNA integrity was statistically significantly higher in CRC patients compared to the benign and control groups (p<0.001). ROC curves for differentiating CRC patients from normal controls and benign groups had areas under curves of 0.90 and 0.85 respectively. Conclusions: The DNA integrity index is superior to the absolute DNA concentration as a potential serum biomarker for screening and diagnosis of CRC. It may also serve as an indicator for monitoring the progression of CRC patients. Combining CEA and CA19-9 with either of the genetic markers studied is better than either of them alone.
Extension plate attached PHC piles and bolts spliced PHC piles were installed in field test site. Pile integrities were checked during installation with dynamic pile test and the actual pile conditions after installation were compared with integrity index (${\beta}$ index) by PDA test. Theoretically the break in near pile end or pile end (especially extension plate itself) was very difficult to access by integrity index (${\beta}$ index) and also require a high level of knowledge and field experience on PDA test. First actual wave equation of bolts spliced PHC piles due to bolts spliced equipment can be different with welding spliced. Second wave length of the stress wave from installing can be longer than the height of bolts spliced equipment (about 100 mm). Third Beta processing in PDA analysis function is very difficult to access without a high level of knowledge and field experience on wave equation. Above-mentioned three reasons can make conclusion that traditional PDA test in domestic site can not access the integrity of bolts spliced equipment in bolts spliced PHC piles.
To evaluate the biological integrity of aquatic ecosystems, we developed Korean multimetric diatom index (KMDI) using metrics that many researchers have been previously described, and compared to single-metric diatom index such as trophic diatom index (TDI) and diatom assemblage index to organic pollution (DAIpo). For the biological and environmental data, we investigated the epilithic diatoms, water quality and the land-use for 923 sites in Korean streams between 2013-2017, and estuaries between 2010-2017, respectively. Five appropriate metrics were selected according to the following steps; 1) extraction of 300 potential metrics (biological, chemical, physical, and geographical) based on previous references, of these, 2) 46 samples having high separation power were selected, 3) the selected metrics were each tested for variability, redundancy, and sensitivity to the environments, finally 4) construction of multi-metric diatom index comprising single type metrics such as TDI, DAIpo, % motile diatoms, % Achnanthes / (Achnanthes+Navicula), and % number of Gomphonema species. The biological integrity of the 233 sites from the Geum River basin were independently investigated using KMDI. Collectively, the new KMDI showed high sensitivity and explanatory power for environmental factors such as land-use, biochemical oxygen demand, total nitrogen, and electric conductivity. However, it had slightly higher biological integrity for the same sites as compared to single type diatom metrics. Finally, more data accumulation from all over Korea and the development of acceptable diatom metrics were required.

The concept of "Allowable Load Set (ALS)" introduced by the author allows an easy understanding of load and strength characteristics of a structure in relation to its integrity under uncertainties. Two criteria of safety are introduced: A relative safety index denotes the distance to the boundary of the ALS and a normalized safety index is a distance in terms of functional value. They have been utilized in several examples, including multi-body mechanical systems such as a biomechanical system. Both formulations amount to robust designs in the sense that designs most insensitive to uncertainties are obtained in the context of newly defined safety indices, without using any input probability information.
We evaluated the aquatic ecosystem of Keum-Ho River through applications of the Index of Biological Integrity (IBI) using fish assemblages and Qualitative Habitat Evaluation Index (QHEI) during June-November 1999. Overall IBI values ranged from 13 to 37 with mean of 23 (n=25, Std. error= 1.16), indicating a "Poor" or "Very Poor" condition according to the criteria of Karr (1981) and U.S. EPA (1993). The values of mean IBI declined at the rate of $0.22km^{-1}$(($r^2$=0.91, p< 0.05) along the longitudinal distance from the headwaters to the down-river. Reduced IBI values at down-river (St. 4 and 5) were attributed to the decreases in riffle benthic species and the relative abundance of insectivore and increases in tolerant species, anormalies and exotic species. Spatial pattern in IBI agreed with QHEI values, which showed a linear relation ($r^2$=0.998, p< 0.001) with mean number of species. Field measurements of conductivity and pH, indicators for variation of conservative ions, showed that the river water was diluted up to 30% by summer precipitation and surface run-off from the watershed, resulting in physical and chemical instability during the monsoon. For these reasons, average IBI values during monsoon and postmonsoon decreased more than 20% compared to pre -monsoon. Before the perturbation of the system (i.e., pre-monsoon), values of QHEI were inversely correlated (r=-0.99, p< 0.0001) with realtive abundance of native omnivore and were positively correlated (r=0.87, p=0.05) with relative abundance of native carnivore. These results indicate that spatial degradation of habitat quality modified the species richness and trophic structure, producing decreased IBI values. Objective: Human sperm nucleus DNA damage may negatively affect pregnancy outcome, and the spermatozoa of infertile men have more DNA damage than that of fertile men. The aim of this study was to evaluate the effect of microsurgical varicocelectomy on human sperm nucleus DNA integrity. Methods: We reviewed the medical records of 18 subfertile male patients who underwent microsurgical varicocelectomy at our hospital from April 2006 to April 2007. Varicocele was diagnosed by physical examination and Doppler ultrasound. Standard semen analysis was performed in 18 patients before and 4 months after microsurgical varicoceletcomy using a computer assisted semen analyzer. Sperm nucleus DNA integrity was assessed by a single-cell gel electrophoresis (comet assay). Results: No recurrence of varicocele was observed after 4 months later. The DNA fragmentation index improved after varicocelectomy compared with pre-operatively (19.3 versus 13.7%, respectively, p<0.05). Semen analysis parameters (total count, concentration, motile sperm, viability, strict morphology) increased after varicocelectomy, but the difference did not reach statistical significance. Conclusion: Our data suggest that microsurgical varicocelectomy can improve semen analysis parameters and human sperm nucleus DNA integrity in infertile men with varicocele.

As digital evidence has a highly influential role in proving the innocence of suspects, methods for integrity verification of such digital evidence have become essential in the digital forensic field. Most surveillance camera systems are not equipped with proper built-in integrity protection functions. Because digital forgery techniques are becoming increasingly sophisticated, manually determining whether digital content has been falsified is becoming extremely difficult for investigators. Hence, systematic approaches to forensic integrity verification are essential for ascertaining truth or falsehood. We propose an integrity determination method that utilizes the structure of the video content in a Video Event Data Recorder (VEDR). The proposed method identifies the difference in frame index fields between a forged file and an original file. Experiments conducted using real VEDRs in the market and video files forged by a video editing tool demonstrate that the proposed integrity verification scheme can detect broken integrity in video content.
We evaluated the health condition of Pyong-Chang river, the tributary of Han- River, using the Index of Biological Integrity (IBI) and Qualitative Habitat Evaluation Index (QHEI) during September 1999${\sim}$August 2000. The annual mean of IBI, which was estimated using eleven metrics, was 49 (range: $45{\sim}51$) and the mean of QHEI, which was estimated using seven parameters, was 88 (range: $76{\sim}94$) during the study. The river health, based on the IBI criteria of Karr (1981), ranged from "excellent" to "good" conditions, while based on the habitat criteria of Plafkin et al. (1989), it ranged from "pristine" (comparable to reference) to "supporting" conditions. Values of IBI showed slight differences between upstream and downstream sites and QHEI values varied weakly depending on characteristics of variables. Regression analyses showed that annual values of QHEI had no functional relations with stream order (p = 1.82; n = 8) but showed some decreases near slight point-sources. This result indicates that conditions of physical habitat did not change highly with increases of the stream order. According to analyses of feed guilds, relative abundance of insectivores, omnivores and carnivores was 85.1%, 3.5% 0.3%, respectively. Also, relative abundance of sensitive and tolerant species was 75% and 4.6%, respectively, while exotic and morphological anomalies were not found in the river. These outcomes indicate that health condition of fish, based on the trophic conditions of U.S. EPA (1993), was excellent in the river. Regression analyses of IBI values against the QHEI showed that the variation of habitat conditions accounted 57% for the variation of the Index of Biological Integrity (p<0.05; $R^2\;=\;0.57$; n = 7).Overall data of IBI and QHEI suggest that the river health in the present is in optimal conditions but may be degradated by acceleration of chemical inputs and physical-habitat disturbance.
Bogie frame of the electric car is an important structural member for the support of vehicle loading. In general, more than 25 years' durability is necessary. Much study has been carried out for the prediction of the structural integrity of the bogie frame in experimental and theoretical domains. One of the useful methods is reliability-based approach. The objective of this paper is to estimate the structural integrity of the bogie frame of an electric car, which is under the running test. We used two approachs. In the first approach probabilistic distribution of S-N curve is used. In the second approach, limit state function is used.
