Functional Analysis of a Histidine Auxotrophic Mutation in Gibberella zeae

  • A plant pathogenic fungus, Gibberella zeae (anamorph: Fusarium graminearum), not only generates economic losses by causing disease on cereal grains, but also leads to severe toxicosis in human and animals through the production of mycotoxins in infected plants. Here, we characterized a histidine auxotrophic mutant of G. zeae, designated Z43R1092, which was generated using a restriction enzyme-mediated integration (REMI) procedure. The mutant exhibited pleiotropic phenotypic changes, including a reduction in mycelial growth and virulence and loss of sexual reproduction. Outcrossing analysis confirmed that the histidine auxotrophy is linked to the insertional vector in Z43R1092. Molecular analysis showed that the histidine requirement of Z43R1092 is caused by a disruption of an open reading frame, designated GzHIS7. The deduced product of GzHIS7 encodes a putative enzyme with an N-terminal glutamine amidotransferase and a C-terminal cyclase domain, similar to the Saccharomyces cerevisiae HIS7 required for histidine biosynthesis. The subsequent gene deletion and complementation analyses confirmed the functions of GzHIS7 in G. zeae. This is the first report of the molecular characterization of histidine auxotrophy in G. zeae, and our results demonstrate that correct histidine biosynthesis is essential for virulence, as well as sexual development, in G. zeae. In addition, our results could provide a G. zeae histidine auxotroph as a recipient strain for genetic transformation using this new selectable marker.

Optimization of Agrobacterium-mediated transformation procedure for grapevine 'Kyoho' with carrot antifreeze protein gene

  • We report an Agrobacterium-mediated transformation procedure optimized for 'Kyoho' that is a major table grapevine cultivar in Korea, and its transgenic plants with antifreeze protein gene of carrot (DcAFP). The full length of DcAFP coding region in accordance with the previous report was isolated from young leaves of carrot and recombined into a plant transformation vector. Ethylene inhibitors such as silver nitrate and aminoethoxyvinylglycin (AVG) supplemented in a co-cultivation medium distinctly increased frequency of shoot regeneration when explants were sub-cultured in a selection medium: particularly ten-fold higher in treatment with 0.1 mg/L AVG than one without ethylene inhibitor. Among various antibiotics and their concentrations, the combination of 150 mg/L cefotaxime plus 150 mg/L $Clavamox^{TM}$ was selected for elimination of Agrobacterium cells in addition to minimization of adverse effect on shoot regeneration, while 50 mg/L kanamycin monosulfate effectively suppressed regeneration of non-transgenic shoots. Applying the elucidated culture condition, we finally obtained a total of 5 transgenic 'Kyoho' plantlets with DcAFP, of which integration with the grapevine genome and transcription was confirmed by nucleic acid analyses.

Construction and Characterization of Transformed Insect Cells Expressing Baculovirus Very Late Factor in an Infection-Independent Manner

  • Transformed Spodoptera frugiperda (Sf9) cells expressing baculovirus very late factor (VLF-1) were constructed by using Autograha nuclear polyhedrosis virus (AcNPV) immediate earthy gene (ie1). Neomycin-resistance gene as a selectable marker was introduced under the control of AcNPV ie1 promoter, and Bombyx mori nuclear polyhedrosis (BmNPV-K1) vlf-1 gene was introduced under the control of the Drosophila heat shock protein gene (hspr70) promoter to yield dual expression plasmid with two independent transcription units. It was transfected into Sf9 cells and cell clones expressing vlf-1 were selected by G4l8 treatment. Genomic DNA from transformed cells was isolated and integration of AcNPV iel harboring vlf-1 was confirmed by PCR using AcNPV iel-specific primers and Southern blot analysis. The transformed cells expressing VLF-1 in an infection-independent manner expressed foreign gene product of recombinant baculovirus in the earlier stage of infection compared with control Sf9 cells. These results suggest the possible to develop highly efficient transformed insect cells for baculovirus expression vector system.

Expression of the S glycoprotein of transmissible gastroenteritis virus (TGEV) in transgenic potato and its immunogenicity in mice

  • Transgenic plants have been tested as an alternative host for the production and delivery of experimental oral vaccines. Here, we developed transgenic potatoes that express the major antigenic sites A and D of the glycoprotein S from transmissible gastroenteritis coronavirus (TGEV-$S_{0.7}$) under three expression vector systems. The DNA integration and mRNA expression level of the TGEV-$S_{0.7}$ gene were confirmed in transgenic plants by PCR and northern blot analysis. Antigen protein expression in transgenic potato was determined by western blot analysis. Enzyme-linked immunosorbent assay results revealed that based on a dilution series of Escherichia coli-derived antigen, the transgenic line P-2 had TGEV-$S_{0.7}$ protein at levels that were 0.015% of total soluble proteins. We then examined the immunogenicity of potato-derived TGEV-$S_{0.7}$ antigen in mice. Compared with the wild-type potato treated group and synthetic antigen treated group, mice treated with the potato-derived antigen showed significantly higher levels of immunoglobulin (Ig) G and IgA responses.

  • This paper explains the impact of frequency error on the performance of WCDMA mobile communication systems and what brings about the frequency error between the base station and the mobile station, and then presents automatic frequency error correction method in mobile receiver. On the basis of system requirement related to frequency stability, the integration test between the base station and the mobile station was accomplished. After applying automatic frequency error correction to mobile receiver, 4 Hz of frequency error at transmitting frequency was obtained. The result met frequency error requirement of ${\pm}$0.1 ppm(about ${\pm}$200 ㎐). Performance degradation due to frequency error was measured by means of Error Vector Magnitude(EVM) at transmitter and Ec/Io at receiver, respectively and then the interface requirement between Modem control signal and RF was suggested to improve the correctness of frequency error control.

Developing an Ensemble Classifier for Bankruptcy Prediction (부도 예측을 위한 앙상블 분류기 개발)

  • An ensemble of classifiers is to employ a set of individually trained classifiers and combine their predictions. It has been found that in most cases the ensembles produce more accurate predictions than the base classifiers. Combining outputs from multiple classifiers, known as ensemble learning, is one of the standard and most important techniques for improving classification accuracy in machine learning. An ensemble of classifiers is efficient only if the individual classifiers make decisions as diverse as possible. Bagging is the most popular method of ensemble learning to generate a diverse set of classifiers. Diversity in bagging is obtained by using different training sets. The different training data subsets are randomly drawn with replacement from the entire training dataset. The random subspace method is an ensemble construction technique using different attribute subsets. In the random subspace, the training dataset is also modified as in bagging. However, this modification is performed in the feature space. Bagging and random subspace are quite well known and popular ensemble algorithms. However, few studies have dealt with the integration of bagging and random subspace using SVM Classifiers, though there is a great potential for useful applications in this area. The focus of this paper is to propose methods for improving SVM performance using hybrid ensemble strategy for bankruptcy prediction. This paper applies the proposed ensemble model to the bankruptcy prediction problem using a real data set from Korean companies.

Numerical Study of Unsteady Supersonic Flow over Tandem Cavities (초음속 비정상 직열배치공동 유동에 관한 수치적 연구)

  • The unsteady supersonic flow over tandem cavities has been analyzed by the integration of Navier-Stokes equations with the k-$\varepsilon$ turbulence model. The unsteady flow is characterized by the periodicity due to the mutual relation between the shear layer and the internal flow in cavities. The upwind TVD scheme based on the flux vector split with the van Leer limiters is used. The results show the principal frequency is very reasonable. The principal frequency of the rear cavity due to the front cavity has been analyzed by the combination of the several aspect ratios of cavities. In the case of the front cavity of low aspect ratio, the frequencies of tandem cavities are almost same, because two shear layers developed from each cavity are mixed and developed to one shear layer. However, in the case of the front cavity of high aspect ratio, the characteristis of frequency are very different, because the second shear layer is developed in the diffused first shear layer.

Development and Characterization of Sporulation Mutants for Overexpression of Recombinant Protein of Bacillus subtilis (재조합 단백질 과발현을 위한 Bacillus snbtilis 포자형성 변이주의 개발 및 특성 분석)

  • Sporulation mutants of Bacillus subtilis were developed for overproduction of heterologous proteins. The strains spoOJ spoIIG, and spoOJ spoIIG double mutant were constructed from two pretense-delfted mutant (DB104). The vector containing aprE gene was integrated in the chromosome of each strain, then the morphology of each strain was observed by TEM (trasmission electron microscopy). The morphology of spoOJ mutant and spoIIG mutant coincides with the description of the previous reports, respectively. The sporulating cells of spoOJ SpoIIG double mutation resemble spoIIG mutant more similarly, but with a little rougher cell wall membrane. The spoOJ mutation in B. subtilis gives negative effect on aprE activity with only a decreased sporulation frequency. On the contrary spoIIG mutation increases the aprE activity twice with an undetectable sporulation frequency. In the case of spoOJ and spolIG, i. e. double mutation, the effect of spoOJ on aprE activity seems to be relieved and the double mutant shows more or less the same aprE activity compared to spoIIG mutant.

Transportation Network Data Generation from the Topological Geographic Database (GIS위상구조자료로부터 교통망자료의 추출에 관한 연구)

  • This paper presents three methods of generating the transportation network data out of the topological geographic database in the hope that the conversion of the geographic database file containing the topology to the conventional node-link type trans¬portation network file may facilitate the integration between transportation planning mod¬els and GIS by alleviating the inherent problems of both computing environments. One way of the proposed conversion method is to use the conversion software that allows the bi-directional conversion between the UTPS (Urban Transportation Planning System) type transportation planning model and GIS. The other two methods of data structure conversion approach directly transform the GIS's user-level topology into the transportation network data topology, and have been introduced with codes programmed with FORTRAN and AML (Arc Macro Language) of ARC/INFO. If used successfully, any approach would not only improve the efficiency of transportation planning process and the associated decision-making activities in it, but enhance the productivity of trans¬portation planning agencies.

Development of Bialaphos Resistant Transgenic Tabacco Plants by Pollination and Utilization of Fertilization Cycle (수분ㆍ수정 시기를 이용한 Bialaphos 저항성 형질전환 담배의 개발)

  • The herbicide bialaphos is a potent inhibitor of glutamine synthetase in higher plants. A bialaphos resistance (bar) gene encoding for an acetyltransferase was isolated from genomic DNA of Pseudomonas syringae pv tabaci. The bar gene was ligated to the binary vector pBI121. Pistils of tobacco plane were heated with the bar gene containing plasmid DNA at various times after pollination. When the treatment was applied at 30 and 40 h after pollination, a number of transgenic plants were obtained. Premary transformation (T$_{0}$ generation) and their progenies (T$_1$T$_2$) were resistant to both bialaphos and kanamycin at a dosage lathal to untransformed control plants. Stable integration of bar gene into chromosomal DNA was proven by Southern blot analysis of genomic DNA isolated from T$_1$progenies. These results show that the bialaphos resistant plane could be obtained by treatment to pistils with the exgenous bar gene through the fertilization cycle of tobacco.o.
