닭의장풀과 자주달개비에서 적색광과 이산화탄소에 의해 유도된 공변세포의 전위차 변화에 미치는 엽육세포의 영향 (Influence of the Mesophyll on the Change of electrical Potential Difference of Guard Cells Induced by Red-light and CO2 in Commelina communis L. and Tradescantia virginiana L.)

  • Intact leaf과 detached epidermis에 있는 공변세포의 전기 생리학적 특성에 대한 빛과 이산화탄소의 효과를 조사하였다. 빛을 intact leaf의 abaxial side에 처리하면 공변세포막이 과분극 (hyperpolarization)되었다. 닭의장풀의 intact leaf에 있는 공변세포들은 빛에 의해 최대 13 mV 그리고 이산화탄소에 의해 42 mV까지 membrane potential difference(MPD)가 negative하게 변했다. 자주달개비에서도 비슷한 결과를 얻었다. 그러나, 빛과 이산화탄소를 detached epidermis에 있는 공변세포에 처리할 경우에는 공변세포의 MPD가 변하지 않았다. 위의 결과로부터, 엽육세포가 공변세포의 MPD 변화에 영향을 주는 것으로 사료되어, 엽육세포들을 광합성 억제제들을 침윤시켜 엽육세포 광합성의 어느 기작이 공변세포 MPD 변화에 영향을 주는지 조사하였다. CCCP로 침윤한 잎의 공변세포막은 적색광에 의해 약간 탈분극(depolarization)되었고, 이산화탄소에 의해 과분극되었다. 반면에, DCCD와 DCMU로 침윤한 경우에는 대조구 잎과 마찬자기로 적색광과 이산화탄소에 의해 과분극되었다. Azide로 침윤한 잎에 적색광을 처리하면 공변세포의MPD는 변하지 않았고, 이산화탄소를 처리하면 다른 처리구들에 비해 훨씬 감소한 막의 과분극을 보였다. 이는 azide가 잎에 손상을 유도하며 세포내 대사활성을 감소시킨 결과 이산화탄소에 의한 MPD 변화가 작았고, 적색광은 아무 효과도 보이지 않은 것으로 사료된다. 따라서, 엽육세포가 적색광을 감지하며 빛에 의해 유도된 공변세포막 과분극은 순환적 광인산화 반응에 의해 생성된 에너지에 의존하나 이산화타소에 의해 유도된 공변세포막 과분극은 광합성가 무관하다고 볼 수 있다. 또한 이산화탄소를 intact leaf에 처리하면 공변세포 액포가 알칼리화되는 것을 관찰하였는데, 이는 막의 과분극이 양성자 이온의 방출에 의해 일어난다는 것을 의미한다.

Bioelectrochemical Denitrification by Pseudomonas sp. or Anaerobic Bacterial Consortium

  • In a bacterial denitrification test with Pseudomonas sp. and anaerobic consortium, more nitrates and less substrate were consumed but less metabolic nitrite was produced under an anaerobic $H_2$ condition rather than under $N_2$ condition. In a bioelectrochemical denitrification test with the same organisms, the electrochemically reduced neutral red was confirmed to be a substitute electron donor and a reducing power like $H_2$. The biocatalytic activity of membrane-free bacterial extract, membrane fraction, and intact cell for bioelectrochemical denitrification was measured using cyclic voltammetry. When neutral red was used as an electron mediator, the electron transfer from electrode to electron acceptor (nitrate) via neutral red was not observed in the cyclic voltammogram with the membrane-free bacterial extract, but it was confirmed to gradually increase in proportion to the concentration of nitrate in that of the membrane fraction and the intact cell of Pseudomonas sp.

The Electrophysiology Application on Guard Cells to See the Influence of Carbon Dioxide

  • The effect of $CO_2$ on the opening of stomata in the intact leaf of Commelina communis has been investigated. Full opening of stomatal apertures(around $18{\mu}m$) was achieved in the intact leaf by addition of $CO_2$($900{\mu}mol\;mol^{-1}$). At 90 minutes, the stomatal apertures of leaves treated with $CO_2$ free air were reduced. In contrast, stomata opened most widely with the treatment of $CO_2$ air at 90 minutes. The effects of light, $CO_2$ air and $CO_2$ free air on the change of membrane potential difference(PD) were measured. Fast hyperpolarization of guard cell membrane PD was recorded reaching up to -12 mV in response to light. If $CO_2$ free air was given firstly, there was no response. When light was given after $CO_2$ free air, the light effect was very clear. At the onset of $CO_2$ air, the PD showed a dramatic hyperpolarization to about -25 mV. Changes in the pH of apoplast in intact leaves in response to $CO_2$ air were observed. $CO_2$ air caused a change of 0.4 pH unit. Therefore, it can be hypothesized that $CO_2$ flowing could stimulate proton efflux which is a necessary precursor of stomatal opening.

한우 무손상 적혈구의 superoxide 및 과산화수소 제거능력 (Scavenge of superoxide and hydrogen peroxide by bovine intact red blood cells)

  • The ability of bovine intact red blood cells to scavenge superoxide and hydrogen peroxide by superoxide dismutase, catalase and glutathione peroxidase was investigated. Intact red cells(up to 0.4%) suspensions did not inhibit ferricytochrome c reduction by superoxide in the superoxide generating system. On the other hand, intact red cell(0.4%) suspensions almost completely inhibit ferrocytochrome c oxidation by hydrogen peroxide. The ability of intact red cells to scavenge hydrogen peroxide was mainly attributed to either membrane bound catalase or glutathione peroxidase. The scavenge of hydrogen peroxide by 0.1~0.2% intact red cells showed a trend of dependence on mainly glutathione peroxidase. However, at blood cell concentration higher than 0.3%, the process depended upon peroxidase-independent scavengers like catalase. Enhancement of ferrocytochrome c oxidation by red cells treated with aminotriazole proved that the protection against hydrogen peroxide was due to catalase, while the protection in the presence of glutathione indicated scavenging effect of glutathione peroxidase against hydrogen peroxide.

  • Ultrathin sections of intact mycelia, released protoplast and fused protoplast of Micromonospora rosaria were observed by electron microscopy Intact mycelia showed a typical gram-positive bacterial cell wall structure and mesosomes. Released protoplasts had no cell wall components and fibrous nuclear region was distinguished from cytoplsmic region clearly. Protoplasts which treated with sucrose supplemented buffer were stable. But those treated with buffer without sucrose were extensively damaged, forming mom braneous vesicles. It was surmised that those vesicles originated from the damaged cytoplasmic membrane. High frequency of fusion was achieved by 50%(w/v). polyethylene-glycol 1,000 Fusion bodies in different stage of fusion were observed. Cell membrane barrier was stepwise relieved.

Effect of Omeprazole on Membrane P-Type ATPase and Peptide Transport in Helicobacter pylori

  • We investigated the growth-inhibitory mechanism of Helicobacter pylori by omeprazole (OMP) and its activated sulfenamide (OAS). Using dithiothreitol (DTT) and 5,5'-dithio-bis[2-nitrobenzoic acid] (DTNB; Ellman's reagent), we first determined the relationship between the binding capacity of these compounds to H. pylori membrane and its significance to membrane P-type ATPase activity. After incubation of the intact H. pylori cells with either OMP or OAS, the residual quantity of free SH-groups on the cell membrane was measured, and, the resulting values were plotted as a function of time. From this experiment, we found that there was a considerable difference in the membrane-binding rates between OMP and OAS. At neutral pH, the disulfide bond formation on H. pylori membrane was completed within 2 min of incubation of the intact cells with OAS. By OMP, however, it was gradually formed, exceeding 10 min of incubation for completion, whereby, the extent of P-type ATPase inhibition appeared to be proportional to the disulfide forming rate. From this data, it was suggested that the disulfide formation might directly affect enzyme activity. Since OMP per se cannot yield a disulfide bond with cysteine, it is predicted that the enzyme inactivation must be caused by the OAS form. Accordingly, we postulated that, under the neutral pH, OMP could be converted to OAS in the course of transport. By extrapolating the inhibitory slopes, we could evaluate K₁ values, relating to their minimal inhibitory concentrations (MICs) for H. pylori growth. In these MIC ranges, H. pylori uptake or vesicular export of nutrients such as peptides were totally prohibited, but their effect in Escherichia coli were negligible. From these observations, we strongly suggest that the P-type ATPase activity is essential for the survival of H. pylori cells in particular.

The Effect of Different Light Quality on the Change of Membrane PD of the Guard Cell in Tradescantia virginiana L.

  • The effects of different light quality on the change of membrane potential difference (PD) of the guard cell in the intact leaf have been investigated. The mombrane PD was about -5.5 mV by white light of 600 $\mu$moles $m^{-2}\; s^{-1}$. The mean PD of change caused by red light was about -5.2 mV at the light intensity of 80 $\mu$moles $m^{-2}\; s^{-1}$. Membrane PD of guard cells in response to blue light was saturated at low light intensity. However, red and green light enhanced the change of membrane PD of guard cells with increasing intensity. In green light the biggest change of memrane PD was around -4 mV, whereas, with blue light the change of of memrane PD was around -2 mV. Accordingly, the membrane PD of guard cell showed the different degree of hyper-polarization by each wavelength.

  • A 4-year-old cat was referred for a suspected pulmonary mass. True diaphragmatic hernia presence was diagnosed via computed tomography (CT). There was a thin membrane covering the diaphragmatic defect. The membrane was thinner than the diaphragm. After contrast injection, the membrane was less enhanced than that of the normal diaphragm. The membrane was identified as a remnant of the parietal pleura. In addition, contrast-enhanced CT images provided clarity in viewing the herniated liver and falciform fat. A thinner membrane, covering the diaphragmatic defect, and attached to the thicker normal diaphragm, is considered a unique CT feature of true diaphragmatic hernia.

Photodegradation stability study of PVDF- and PEI-based membranes for oily wastewater treatment process

  • In this work, an attempt was made to compare the effects of UV irradiation on the intrinsic and separation properties of membranes made of two different polymeric materials, i.e., polyvinylidene fluoride (PVDF) and polyetherimide (PEI). The changes on membrane structural morphologies and chemical characteristics upon UV-A exposure (up to 60 h) were studied by FESEM and FTIR, respectively. It was found that cracks and fractures were detected on the PVDF-based membrane surface when the membrane was exposed directly to UV light for up to 60 h. Furthermore, the mechanical strength and thermal stability of irradiated PVDF-based membrane was reported to decrease with increasing UV exposure time. The PEI membrane surface meanwhile remained almost intact throughout the entire UV irradiation process. Filtration experiments showed that the permeate flux of UV-irradiated PVDF membrane was significantly increased from approximately 11 to $16L/m^2.h$ with increasing UV exposure time from zero to 60 h. Oil rejection meanwhile was decreased from 98 to 85%. For the PEI-based membrane, oil rejection of >97% was recorded and its overall structural integrity was marginally affected throughout the entire UV irradiation process. The findings of this work showed that the PEI-based membrane should be considered as the host for photocatalyts incorporation if the membrane was to be used for UV-assisted wastewater treatment process.

단감 과실의 과육 갈변과 세포막 투과성 및 지방산 조성 변화의 관계 (The Relationship among Flesh Browning, Membrane Permeability, and Fatty Acid Composition in Fuyu Persimmon Fruits)

  • The cell membrane properties in relation to flesh browning of Fuyu persimmon fruits during CA storage were studied. Compared to intact fruits, the flesh tissue of browned fruits showed higher rate of electrolyte leakage, indicating incresed membrane permeability. It could be assumed that the increased membrane permeability results in 1eakage of phenolic compounds from vacuole and their oxidation by contacting with PPO, inducing finally the development of flesh browning. In addition, lower content of fatty acids and higher saturation rate of them were found in browned fruits. In conculusion, it was suggested that the inhibited fatty acid metabolism and fatty acid saturation during CA storage cause membrane Permeability to increase.
