Based upon the previous experiments showing that kidney and lung tissues of rat had relatively abundant bradykinin binding sites, we tried to characterize and determine the densities of the bradykinin binding sites in the rabbit kidney tissue and proximal tubular cells under different growing conditions. Among the kidney tissue renal medulla segments showed the highest bradykinin binding sites. To determine which growth factors are to add in the serum free culture medium to express selectively the bradykinin binding sites in the rabbit kidney proximal tubular cells, we tried so called hormone-deletion approach and in here insulin, hydrocortisone, transferrin, triiodothyronine and prostaglandin $E_1$ are examined. By performing receptor binding assay and determination of protein concentrations, we may conclude that the most required hormones in the expression for bradykinin binding sites are insulin and transferrin, and fetal bovine serum is shown to be less effective in this regard.
본 연구는 우리나라에서 재배한 삼채 추출물의 주요생리활성 물질을 2형 당뇨 쥐에 투여하였을 때 인슐린 저항성 개선효과와 췌장에서 β-cell의 형태 및 간 조직의 2형 당뇨 관련 단백질의 발현을 확인 함으로써 그 메카니즘을 확인코자 하였다. 특히 TZDS당뇨약 계열로 현재 시판중인 actoz와 활성을 비교 함으로써 그 유효성 정도를 살펴보고자 본 실험을 디자인하였다. 먼저 식이섭취량, 물 섭취량의 경우, Control군은 2형 당뇨 특징인 다식, 다뇨, 다갈의 형태로 증가하였으나, 삼채 추출물 투여로 식이 및 물 섭취량이 감소하는 경향을 보였다. 식이 투여 2시간 후 혈당변화와 OGTT 검사결과, 2형 당뇨인 Control군에서 혈당의 농도가 가장 높았으나 삼채 추출물 8주 경구투여로 인해 안정화되는 것을 확인할 수 있었으며, serum의 glucose농도 또한 삼채 추출물 투여로 2형당뇨 control군에 비해 혈당이 감소한 것을 확인할 수 있었다. HbA1c (당화혈색소)는 2형 당뇨군(control)에서 9% 수치 범위로 고혈당 범위였으나, 삼채 추출물 섭취로 인하여 Positive control군(actoz 투여군) 7% 수준으로 감소하는 경향을 확인할 수 있었으며, insulin농도는 2형 당뇨인 Control군에서 insulin 농도가 1 ng/ml로 가장 낮은 농도였으며, 삼채 추출물 및 SAC 1.5 ng/ml수준으로 insulin수치가 상승하는 것을 확인할 수 있었다. 혈중 triglyceride, total cholesterol 및 LDL-cholesterol 수치 모두 시료 투여군에서 혈중 지질관련 인자의 농도가 감소하는 것을 확인할 수 있었으며, 특히 AH 400군의 경우 triglyceride, total cholesterol 및 LDL-cholesterol 농도가 각각 482.86±13.40 mg/dl, 163.56±13.24 mg/dl, 142.04±3.24 mg/dl로 대조군에 비해 약 50% 감소한 결과를 나타내었다. 간장 조직 중 지질 과산화의 함량의 경우, 2형 당뇨인 Control군에서 467.74±1.12 MDA nmole/g of tissue로 삼채 추출물 투여(AH 400)로 319.61±26.98 MDA nmole/g of tissue 로 Normal군과 비슷한 수준으로 혈중 lipid peroxide의 함량이 감소하는 것을 확인할 수 있었다. 간장조직을 H&E염색한 결과, Normal군에 비하여 2형 당뇨인 Control군의 간 조직에서 많은 지방적이 나타나 지방간이 확인되었으나, Positive control 및 삼채 추출물 투여에 의해 지방적의 크기와 수가 많이 줄어드는 결과를 얻어 조직학적 검사상 지방간의 소견이 현저히 호전되었으며, 췌장에서 insulin 면역화학 염색 결과 2형 당뇨로 췌도 세포가 거의 손상된 것과는 대조적으로, 삼채 추출물을 투여한 결과 정상군과 유사한 형태의 췌도 세포가 확인되었으며 췌도 세포의 형태가 다양한 정도의 과 형성을 보이는 것이 확인되었다. 또한 간장 조직의 PPARγ, PDK4, SREBP-1, FOX0 1의 발현을 확인한 결과, 발현이 2형 당뇨인 Control군에서 모두 발현이 증가하였으나, 삼채 추출물 투여로 인해 발현 정도가 낮게 나타나 앞서 확인하였던 glucose 및 HbA1c 및 insulin농도의 결과와 상통함을 확인하였다. 이들의 결과로 미루어 볼 때, 삼채 메탄올 추출물의 8주간 경구 투여로 인하여 2형 당뇨에 관여하는 PPARγ 단백질 및 비만 관련 단백질 인자를 조절하였으며, insulin농도 및 식후 혈당조절에도 도움을 주는 것을 확인할 수 있었다. 또한, 췌장의 insulin 면역화학염색 결과, 췌도 세포 형태 및 insulin 합성에도 긍정적인 역할을 하는 것으로 미루어 보아 2형 당뇨에 대한 치료효과가 큰 것으로 사료 된다.

The TAZ activator 2-butyl-5-methyl-6-(pyridine-3-yl)-3-[2'-(1H-tetrazole-5-yl)-biphenyl-4-ylmethyl]-3H-imidazo[4,5-b]pyridine] (TM-25659) inhibits adipocyte differentiation by interacting with peroxisome proliferator-activated receptor gamma. 1 TM-25659 was previously shown to decrease weight gain in a high fat (HF) diet-induced obesity (DIO) mouse model. However, the fundamental mechanisms underlying the effects of TM-25659 remain unknown. Therefore, we investigated the effects of TM-25659 on skeletal muscle functions in C2 myotubes and C57BL/6J mice. We studied the molecular mechanisms underlying the contribution of TM-25659 to palmitate (PA)-induced insulin resistance in C2 myotubes. TM-25659 improved PA-induced insulin resistance and inflammation in C2 myotubes. In addition, TM-25659 increased FGF21 mRNA expression, protein levels, and FGF21 secretion in C2 myotubes via activation of GCN2 pathways (GCN2-$phosphoelF2{\alpha}$-ATF4 and FGF21). This beneficial effect of TM-25659 was diminished by FGF21 siRNA. C57BL/6J mice were fed a HF diet for 30 weeks. The HF-diet group was randomly divided into two groups for the next 14 days: the HF-diet and HF-diet + TM-25659 groups. The HF diet + TM-25659-treated mice showed improvements in their fasting blood glucose levels, insulin sensitivity, insulin-stimulated Akt phosphorylation, and inflammation, but neither body weight nor food intake was affected. The HF diet + TM-25659-treated mice also exhibited increased expression of both FGF21 mRNA and protein. These data indicate that TM-25659 may be beneficial for treating insulin resistance by inducing FGF21 in models of PA-induced insulin resistance and HF diet-induced insulin resistance.
Insulin signaling is coordinated by insulin receptor substrates (IRSs). Many insulin responses, especially for blood glucose metabolism, are mediated primarily through Irs-1 and Irs-2. Irs-1 knockout mice show growth retardation and insulin signaling defects, which can be compensated by other IRSs in vivo; however, the underlying mechanism is not clear. Here, we presented an Irs-1 truncated mutated mouse ($Irs-1^{-/-}$) with growth retardation and subcutaneous adipocyte atrophy. $Irs-1^{-/-}$ mice exhibited mild insulin resistance, as demonstrated by the insulin tolerance test. Phosphatidylinositol 3-kinase (PI3K) activity and phosphorylated Protein Kinase B (PKB/AKT) expression were elevated in liver, skeletal muscle, and subcutaneous adipocytes in Irs-1 deficiency. In addition, the expression of IRS-2 and its phosphorylated version were clearly elevated in liver and skeletal muscle. With miRNA microarray analysis, we found miR-33 was down-regulated in bone marrow stromal cells (BMSCs) of $Irs-1^{-/-}$ mice, while its target gene Irs-2 was up-regulated in vitro studies. In addition, miR-33 was down-regulated in the presence of Irs-1 and which was up-regulated in fasting status. What's more, miR-33 restored its expression in re-feeding status. Meanwhile, miR-33 levels decreased and Irs-2 levels increased in liver, skeletal muscle, and subcutaneous adipocytes of $Irs-1^{-/-}$ mice. In primary cultured liver cells transfected with an miR-33 inhibitor, the expression of IRS-2, PI3K, and phosphorylated-AKT (p-AKT) increased while the opposite results were observed in the presence of an miR-33 mimic. Therefore, decreased miR-33 levels can up-regulate IRS-2 expression, which appears to compensate for the defects of the insulin signaling pathway in Irs-1 deficient mice.
이 연구의 목적은 Caulerpa okamurae 에탄올 추출물(COE)이 제2형 당뇨병 치료의 약물 표적 중 하나인 당 대사 및 인슐린 민감성에 미치는 영향을 평가하는 것이다. COE는 in vitro 실험에서 단백질 티로신 포스타제 1B (PTP1B)와 디펩티딜 펩티데이즈-IV (DPP-IV) 효소 활성을 유의하게 억제시켰다. 또한, COE는 3T3-L1 지방세포와 제브라피쉬에서 당 흡수, 인슐린 수용체 기질(IRS-1) 및 당 수송체(GLUT4) 단백의 발현을 대조군에 비해 유의하게 향상시켰다. L6 근육세포의 덱사메타손(dexamethasone)으로 유도된 인슐린 저항성 모델에서도 COE는 인슐린 신호전달 및 당 흡수 단백의 발현을 효과적으로 증가시켰다. 더불어 인슐린 저항성 지표로 알려진 IRS-1 Ser307의 인산화 활성도 COE 첨가에 의해 유의하게 억제되었다. 그러나 COE는 췌장 베타세포의 인슐린 분비에는 아무런 영향을 미치지 않았다. 결론적으로 COE는 인슐린 표적세포와 제브라피쉬에서 인슐린 신호전달과 당 수송체 GLUT4 단백 발현의 조절을 통해 당 대사 및 인슐린 민감성을 개선시키는 것으로 밝혀졌다.

Adipogenic differentiation of mesenchymal stem cells (MSCs) is critical for metabolic homeostasis and nutrient signaling during development. However, limited information is available on the pivotal modulators of adipogenic differentiation of MSCs. Adaptor protein Lnk (Src homology 2B3 [SH2B3]), which belongs to a family of SH2-containing proteins, modulates the bioactivities of different stem cells, including hematopoietic stem cells and endothelial progenitor cells. In this study, we investigated whether an interaction between insulin-like growth factor-1 receptor (IGF-1R) and Lnk regulated IGF-1-induced adipogenic differentiation of MSCs. We found that wild-type MSCs showed greater adipogenic differentiation potential than $Lnk^{-/-}$ MSCs. An ex vivo adipogenic differentiation assay showed that $Lnk^{-/-}$ MSCs had decreased adipogenic differentiation potential compared with wild-type MSCs. Interestingly, we found that Lnk formed a complex with IGF-1R and that IGF-1 induced the dissociation of this complex. In addition, we observed that IGF-1-induced increase in the phosphorylation of Akt and mammalian target of rapamycin was triggered by the dissociation of the IGF-1R-Lnk complex. Expression levels of a pivotal transcription factor peroxisome proliferator-activated receptor gamma ($PPAR-{\gamma}$) and its adipogenic target genes (LPL and FABP4) significantly decreased in $Lnk^{-/-}$ MSCs. These results suggested that Lnk adaptor protein regulated the adipogenesis of MSCs through the $IGF-1/Akt/PPAR-{\gamma}$ pathway.

Objective: Effects of linseed oil (LO) supplementation on the fat content and fatty acid profile of breast meat, and the expression of three genes in the liver, breast muscle and fat tissues of commercial 154-day-old hybrid male turkeys were investigated. Methods: The animals in the control group were fed a commercially available feed and received no LO supplementation (n = 70), whereas animals in the LO group (n = 70) were fed the same basic diet supplemented with LO (day 15 to 21, 0.5%; day 22 to 112, 1%). The effect of dietary LO supplementation on fatty acid composition of breast muscle was examined by gas chromatography, and the expression of fatty acid desaturase 2 (FADS2), peroxisome proliferator activated receptor gamma (PPARγ), and insulin-like growth factor 1 (IGF1) genes was analysed by means of quantitative reverse transcription polymerase chain reaction. Results: The LO supplementation affected the fatty acid composition of breast muscle. Hepatic FADS2 levels were considerably lower (p<0.001), while adipose tissue expression was higher (p<0.05) in the control compared to the LO group. The PPARγ expression was lower (p<0.05), whereas IGF1 was higher (p<0.05) in the fat of control animals. There were no significant (p>0.05) differences in FADS2, PPARγ, and IGF1 gene expressions of breast muscle; however, omega-6/omega-3 ratio of breast muscle substantially decreased (p<0.001) in the LO group compared to control. Conclusion: Fatty acid composition of breast meat was positively influenced by LO supplementation without deterioration of fattening parameters. Remarkably, increased FADS2 expression in the liver of LO supplemented animals was associated with a significantly decreased omega-6/omega-3 ratio, providing a potentially healthier meat product for human consumption. Increased PPARγ expression in fat tissue of the LO group was not associated with fat content of muscle, whereas a decreased IGF1 expression in fat tissue was associated with a trend of decreasing fat content in muscle of the experimental LO group.
Results on localization of growth hormone receptor (GHR), interaction of growth hormone (GH) with receptor in buffalo adipose tissue and identification of activated signaling molecules in the action of GH are presented. Bovine GH (bGH) was labeled with fluorescein or biotin. Fluorescein-labelled bGH was used for localization of GHRs in buffalo adipocytes. The receptors were present on the cell surface. The affinity of binding of GH to its receptor was determined by designing an experiment in which buffalo adipose tissue explants, biotinylated GH and streptavidin-peroxidase conjugate were employed. The affinity constant was calculated to be $2{\times}10^8M^{-1}$. The receptor density on adipose tissue was found to be 1 femto mole per mg of tissue. Signalling molecules generated in the action of GH were tentatively identified by employing Western blot and enhanced chemiluminescence techniques using anti-phosphotyrosine antibody. Based on molecular weights of proteins reactive to anti-phosphotyrosine antibody, three signaling molecules viz. insulin receptor substrate, Janus activated kinase (Jak) and mitogen activated protein were tentatively identified. These signaling molecules appeared in a time (incubation time of explants with growth hormone) dependent way. The activation of Jak2 was confirmed by employing anti-Jak2 antibody in a Western blot. The activation of Jak2 occurred during 5 min incubation of buffalo adipose tissue explants with GH and incubation for an additional period, viz. 30 min. or 60 min., resulted in a drastic reduction in activation. The results suggest that Jak2 activation is an early event in the action of GH in buffalo adipose tissue.
간의 지질 축적과 인슐린 저항성은 비알콜성 지방간 환자에게서 증가한다. Piperine은 후추(Piper nigrum)와 필발(인도산 후추, P. longum)의 주요 성분으로 항암, 항비만, 항 당뇨병, 항염증 및 항산화 등의 생리활성이 보고되었다. 그러나 piperine의 인간 간세포 HepG2 세포에서 지질 축적과 인슐린 저항성의 억제제로서의 연구는 보고된 바가 없다. 본 연구의 목적은 지질 축적 및 인슐린 저항성에 대한 piperine의 효과를 palmitate처리된 HepG2 세포에서 잠재적인 분자 기전을 밝히는 것이다. 그 결과 piperine처리군은 지질 함량을 감소시켰고, 지방 형성 표적 유전자인 SREBP-1c와 FAS의 발현을 억제함으로써 palmitate처리된 세포내 지질 축적을 감소시켰다. 게다가 piperine처리군은 지방산 산화에 관련된 CPT-1과 인산화된 ACC 및 인산화된 IRS-1 (Tyr632)와 Akt의 레벨을 증가시켰다. 또한, piperine처리군은 인산화된 IRS-1 (Ser307)의 레벨을 감소시켰다. 결론적으로 palmitate처리된 HepG2 세포에서 piperine은 SREBP-1와 FAS발현의 감소 및 CPT-1과 ACC 인산화의 증가 및 인산화된 IRS-1(Try632)와 Akt 신호전달 경로를 조절함으로써 지질 축적 및 인슐린 저항성을 개선함을 확인하였다. 따라서 piperine의 지질 축적 및 인슐린 저항성을 예방하는 약물로써 가능성이 제시되었다.

제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
