치태형성 억제세균의 분리 (ISOLATION OF THE BACTERIA INHIBITING THE FORMATION OF PLAQUE)

  • 치태의 주요성분인 비수용성 글루캔의 형성을 억제시키는 세균을 분리하기 위하여 유치원 원아 만여명으로부터 타액을 채취하였다. 일회용 큐벧을 사용하여 비수용성 글루캔의 형성을 억제시킨 세균을 분리하였다. 분리된 세균을 그람 염색과 API 20S kit와 API 50 CHL kit를 사용하여 당발효 및 생화학적 특성을 검사한 결과, Streptococcus oralis, Streptococcus mitis, Streptococcus mitior, Streptococcus sanguis, Enterococcus durans, Lactococcus lactis로 동정되었다. 비수용성 글루캔 형성 억제 정도를 판정하기 위하여 일회용 큐벧에서 S. mutans 단독 배양시에는 550nm에서의 흡광도가 1.503이었으나, S. mutans와 Streptococcus oralis, Streptococcus mitis, Streptococcus mitior, Streptococcus sanguis, Enterococcus durans, Lactococcus lactis, Lactobacillus acidophilus 혼합 배양시에는 각각의 흡광도가 0.823, 0.912, 0.894, 0.878, 0.753, 0.845, 1.021로 감소되었다.

향부자 메탄올 추출물의 Streptococcus mutans에 대한 성장, 산생성, 부착 및 비수용성 글루칸 합성 억제에 미치는 영향 (Effects of methanol extract of Cyperus rotundus on the growth, acid production, adhesion, and insoluble glucan synthesis of Streptococcus mutans)

  • Streptococcus mutans (S. mutans) is known as the causative bacterial playing the most important role informing plaque and it is being noticed as major causative bacteria of dental caries. Therefore, development of more effective, substantial and safe preventive agent against dental caries and periodontal disease is honestly required. The present study was designed to investigate the effect of Cyperus rotundus (Cyperaceae) methanol extracts on the growth, acid production, adhesion, and insoluble glucan synthesis of S. mutans. The methanol extract of C. rotundus showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.5, 1, 2 and 4 mg/ml compared to the control group. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 50% at the concentration of 0.5 mg/ml and complete inhibition was observed at the concentration of 4 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan from sucrose, methanol extract of C. rotundus showed more than 10% inhibition over the concentration of 2 mg/ml. Thus, the application of C. rotundus can be considered a useful and a practical method for the prevention of dental caries.

삼백초 추출물의 항치아우식 효과 (Anticariogenic Properties of the Extract of Saururus chinensis)

  • It has been well established that S. mutans is the major etiological agent in dental caries, one of the most common oral diseases worldwide. The present study was designed to investigate the effect of Saururus chinensis (S. chinensis) ethanol extracts on the growth, acid production, biofilm formation, adhesion, and insoluble glucan synthesis of S. mutans. The ethanol extracts of S. chinensis showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.025, 0.05, 0.1, 0.2 and 0.4 mg/ml compared to the control group. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 80% at the concentration of 0.05 mg/ml and complete inhibition was observed at the concentration of 0.4 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan form sucrose, ethanol extract of S. chinensis showed more than 10% inhibition over the concentration of 0.025 mg/ml. The synthesis of insoluble glucan was decreased in the presence of 0.025 ${\sim}$ 0.4 mg/ml of the ethanol extract of S. chinensis. Our research strongly suggested S. chinensis was a promising natural product for the prevention of dental caries.

  • Dental plaque is a film of microorganisms on the tooth surface that plays an important part in the development of caries and periodontal diseases. Streptococcus mutans (S. mutans) is present in almost all types of dental plaque. Teeth and their supporting structure, the gums (gingiva) are subjected to infection by S. mutans that causes cavities and pyorrhea which, if left untreated, can eventually lead to gingivitis. Various chemical agents have been evaluated over the years with respect to their antimicrobial effects in the oral cavity; however, all are associated with side effects that prohibit regular long-term use. The present study was designed to investigate the effect of Aralia continentalis (Araliaceae) extracts on the growth, acid production, adhesion, and insoluble glucan synthesis of S. mutans. The methanol extract of A. continentalis showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.25, 0.5, 1, 2 and 4 mg/ml compared to the control group. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 60% at the concentration of 0.25 mg/ml and complete inhibition was observed at the concentration of 4 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan from sucrose, methanol extract of A. continentalis showed more than 10% inhibition over the concentration of 0.25 mg/ml. The synthesis of insoluble glucan was decreased in the presence of 0.25 - 4 mg/ml of the methanol extract of A. continentalis. Hence, we conclude that A. continentalis might be a candidate of anticaries agent.

  • 기능성 식품소재인 ${\beta}-glucan$을 식빵재료의 10, 20 및 30% 첨가하여 제조한 반죽의 이화학적 특성을 분석하였다. ${\beta}-Glucan$ 첨가량이 많을수록 반죽형성시간이 증가한 반면 발효에 걸리는 시간은 단축되었다. 반죽의 발효팽창력은 ${\beta}-glucan$ 20% 첨가까지 ${\beta}-glucan$ 첨가량이 증가할수록 증가하였고 ${\beta}-glucan$ 30% 첨가시에는 유의적으로 감소하였다. 반죽의 pH는 대조군의 경우 발효과정을 거치면서 낮아지는 경향을 나타내었으나 ${\beta}-glucan$ 첨가군은 다소 증가하였다. 반죽의 물성 중 탄력성은 ${\beta}-glucan$의 첨가량이 증가함에 따라 증가하고 경도는 감소하였다. 검성은 ${\beta}-glucan$ 10 및 20% 첨가군의 경우 대조군과 차이가 없었고 ${\beta}-glucan$ 30% 첨가군은 유의적으로 낮았다. 발효과정이 진행됨에 따라 반죽은 탄력성, 검성, 경도 및 응집성이 모두 감소하는 경향을 보였으나, ${\beta}-glucan$ 첨가에 따른 유의적인 변화는 없었다. Amylogram에서 호화개시온도와 최고점도에 달하는 온도가 모두 높아져 ${\beta}-glucan$이 호화를 지연시켰으며, setback과 consistency는 감소하여 ${\beta}-glucan$이 식빵노화를 지연시켜 줄 수 있음을 암시하였다. 반죽의 초미세구조를 관찰한 결과 ${\beta}-glucan$ 30% 첨가시 ${\beta}-glucan$ 입자가 반죽에 흡수되지 못하였음을 알 수 있었다. 따라서 제빵적성에 적합한 ${\beta}-glucan$ 첨가율은 20% 이하가 바람직한 것으로 보인다.

  • 본인 등은 베타-1,3-글루칸 합성효소능이 낮은 Saccharomyces cerevisiae의 조건성 돌연변이주들을 분리하여 그 특성을 조사하였다. 이 돌연변이주들은 비허용온도인 37'C에서 삼투감수성을 나타나며, 세포벽 성분 중 알칼리 비용해성 글루칸의 함량이 낮았다. 베타-1,3-글루칸의 합성효소능의 결함원인을 조사한 결과, 효모의 베타-1,3-글루칸 합성효소를 구성하는 촉매성분(cataytic component)과 GTP-결합성분 두가지 중 촉매 성분의 결함이 이 돌연변이주들의 베타-1,3-글루칸 합성효소능의 손상원인인 것으로 추정되었다.

Purification and characterization of glucosyltransferase and fructosyltransferase from Leuconostoc mesenteroides NRRL B-1149

  • A glucan and a fructan producing enzymes from Leuconostoc mesenteroides NRRL B-1149 were prepared and concentrated from the cu1ture of 1.5% sucrose using polysulfone ultrafiltration hollow fiber in the presence of 0.1% (w/v) Tween 80, 1 mM $CaCl_2$, and 0.02% $NaN_3$. The molecular masses of the enzymes were estimated to be about 213.6 kDa and 180 kDa, respectively, based on the PAS staining for the glucosyltransferase and Mukasa method for fructosyltransferase. Polymers produced by the enzymes showed different solubility; an insoluble glucan and a soluble fructan. The linkages of polymers were determined by methylation using Hakomori reagent and following acid hydrolysis. The glucan was composed of ${\alpha}$-1,6 and 1,3 linkages and the fructan showed similar linkage data of levan.

보리의 발아정도가 맥아의 ${\beta}$-glucan 용해성 및 맥주의 점도에 미치는 영향 (Effects of Malt Modification on ${\beta}$-Glucan Solubility and Beer Viscosity)

  • 발아시간에 따라 덜 변형된 맥아(60시간 발아)와 적절히 변형된 맥아(96시간 발아)의 총, 불용성 및 수용성 ${\beta}-glucan$ 함량을 측정하였다. 보리의 총 ${\beta}-glucan$ 함량은 3.96%였으며 발아 중에 감소하였는데 덜 변형된 맥아에서는 1.02%인 반면에 적절히 변형된 맥아에서는 0.18%로 급격히 감소하였다. 적절히 변형된 맥아는 $21^{\circ}C$$45^{\circ}C$ 추출온도에서 ${\beta}-glucan$ 용해성이 덜 변형된 맥아에 비해 현저하게 증가하였다. 변형정도가 다른 두가지 맥아에 대하여 당화온도별로 당화 후 맥즙과 맥주의 점도를 분석하였다. 당화온도가 $45-75^{\circ}C$로 증가함에 따라 맥즙과 맥주의 점도가 증가하였으며 적절히 변형된 맥아가 덜 변형된 맥아에 비해 점도의 상승이 크지 않았다. 이는 적절히 변형된 맥아는 덜 변형된 맥아에 비해서 당화과정중에 ${\beta}-glucan$ 용해성이 상대적으로 높지만 초기 ${\beta}-glucan$의 함량이 낮을 뿐 만 아니라 맥아에 충분히 합성되어 발달된 ${\beta}-glucanse$의 활성에 의해 당화과정 중에 용출되어지는 ${\beta}-glucan$의 분해가 이루어지기 때문인 것으로 판단되었다.

  • Dextranase와 glucose←oxidase의 향치아우식 인자로서의 효괴와 dentifrice 앙분으로서의 이용 가능성융,경토하였다. W Water-insoluble glucan에 의한 plaque의 형성은 dextranase와 glucose-oxida앤플 사용함으호써 억제할 수 9.]였다. D Dextranase의 경우 낮은 농도에서도 plaque의 형생억저l와 분해에 매우 효과적이였다. Glucose-oxidase의 경우 얄균작용 에 의해 생균수를 줄임으로써 plaque의 형성억제에는 효과적이었으나 분해작용은 미약하였다. Dentifrice의 각 성분에 대한 compatibility test 를 통해 효소의 안쟁화를 위한 Mode] Dentifrice 플 구생하였고. Model Dentifrice 에서의 activity 변 화플 관창한 결과 안정성이 오랫동안 유지될을 확인할 수 있었다.

오배자 Galla Chinensis 추출물이 Streptococcus mutans의 우식활성 억제에 미치는 영향 (Inhibitory Effects of Galla Chinensis Extract on Cariogenic Properties of Streptococcus mutans)

  • Streptococcus mutans (S. mutans) is one of the most important bacteria in the formation of dental plaque and dental caries. S. mutans adheres to an acquired pellicle formed on the tooth surface, and aggregates with many oral bacteria, and initiates plaque formation by synthesizing glucan from sucrose, which is catalyzed by glucosyltransferases. S. mutans metabolizes the dietary sugar to the organic acids. The organic acids demineralize tooth surface and result in dental caries. Galla Chinensis have been traditionally used for stopping bleeding of gingiva, removing edema and halitosis, drainage, fixing the teeth and as an antiphlogistic agent. In previous reports, antibacterial effects of Galla Chinensis have been investigated whereas anti-cariogenic effects is still not examined enough. Therefore we tested effects of ethanol extracts of Galla Chinensis on the cariogenic properties such as the growth, acid production, adhesion, and insoluble glucan synthesis of S. mutans. In the result, ethanol extracts of Galla Chinensis showed the inhibition of S. mutans growth and organic acids production over 0.031 mg/ml concentrations. The adhesion of S. mutans to Saliva-coated Hydroxyapatite beads S-HAs has decreased with the increase of concentration of ethanol extracts of Galla Chinensis. And it seems to have adhesion inhibitory effect in concentration of over 0.25 mg/ml. It gives us the result that Galla Chinensis have anti-caries effects. But ethanol extract of Galla Chinensis didn't have inhibitory effect on insoluble glucan synthesis. Preliminary phytochemical analysis of the ethanol extract of Galla Chinensis showed strong phenolic compounds, medium steroids & terpenoids and glycosides, and weak organic acids and peptides. These results suggest that the ethanol extracts of Galla Chinensis may have anti-cariogenic properties, which may be able to be related with strong phenolic compounds.
