배나무잎 이상반점증상에 관한 연구 - 2. 원인구명 - (Studies on the Pear Abnormal Leaf Spot Disease - 2. Identification of Causal Agent -)

  • Alternaria spp.were predominantly isolated from the abnormal leaf spot lesions of pear cultivars Niitaka and Nijiiseiki. Alternaria isolates from the cultivar Niitaka were not pathogenic to both cultivars, but the isolates from the cultivar Nijiiseiki developed typical lesions of black leaf spots and were identified as A. kikuchiana. However, no typical abnormal leaf spot lesions were produced by the Alternaria isolates. Foliar spray of twelve different agrochemicals including lime sulfur, either alone or in combinations, with 7 times applications from April to July failed to reduced the disease development. Application of 17 different pesticides including fungicides, insecticides and herbicides currently used in pear orchards did not cause leaf injury similar to the abnormal leaf spot. Simulated acid rain of as low as pH 3.0 did not incite any leaf lesions alike the abnormal spot lesions. Mineral contents in the leaves of both cultivars did not differ significantly between the healthy leaves and those with abnormal leaf spots. When cuttings of pear tree were obtained in February from newly emerged twigs of the healthy or the diseased trees of Niitaka and planted in sand in the greenhouse, only those from the diseased trees developed typical leaf lesions of the abnormal spot. These results indicate that abnormal leaf spots are caused by unknown systemic agents in pear trees, rather than by Alternaria spp., chemical injury or acid rain.

Different tolerance of zooplankton communities to insecticide application depending on the species composition

  • Natural zooplankton communities are composed of many different species at different trophic levels in the aquatic food web. Several researchers have reported that in mesocosm/enclosure experiments, larger cladocerans tend to be more sensitive to carbamate insecticides than smaller ones (Daphnia > Moina, Diaphanosoma > Bosmina). In contrast, results from individual-level laboratory tests have suggested that large cladoceran species are more tolerant than small species. To clarify this inconsistency, we conducted a microcosm experiment using model zooplankton communities with different species compositions, where animals were exposed to lethal (near to the 24 h LC50, concentration estimated to kill 50% of individuals within 24-h for the small cladoceran Bosmina) and lower, sublethal concentrations of carbaryl. In the experiment, population densities of the small cladocerans (Bosmina and Bosminopsis) decreased subsequent to the applications of chemical, but no impacts were observed on the large cladoceran Daphnia. Our results supported the reports of previous individual level toxicity tests, and indicated that the sensitivity of zooplankton to the insecticide was unchanged by biological interactions but the response of population can be modified by compensation of population through hatching from resting eggs and/or the persistence of insecticide in the systems.

자외선에너지(UV-C)를 이용한 유기인계 화합물의 분해 (Decomposition of Organophosphorous Compounds with Ultraviolet Energy(UV-C))

  • 두 종류의 유기인계 살충제인 디클로로보스와 클로로피리포스를 UV조사, UV 조사와 $TiO_2$분말 그리고 UV조사와 sea sand를 저압수은 램프를 이용하여 분해실험을 하였다. 이 화합물의 확인은 가스크로마토그래프, 총유기탄소 그리고 이온크로마토그래프를 사용하였다. 두 물질 모두, UV조사와 sea sand가 UV 조사와 UV조사와 $TiO_2$분말보다 분해가 더 잘 되었다. 반응 최종생성물은 디클로르보스에서는 $Cl^-$, 클로로피리포스에서는 $Cl^-$ $SO_4{^{2-}}$등이 생성되었다.

  • 전국 621개 양잠시범부락의 84가을 누에병 발생율과 그 원인을 조사한 결과는 다음과 같다. 1. 누에 사육 기간중 누에병 발생율은 약 17%이었다. 2. 누에병 종류별 발생율은 농병(38.5%), 경화병 (20.3%), 연화병(17.9%) 및 쉬파리병 (9.4%)의 순이었다. 3. 누에병은 종잠기보다 장잠기 이후에 주로 발생하였다. 4. 누에병 발생의 주원인은 기상환경불량, 농약피해 및 소독불량이었고, 엽질불량, 사육기술부족 및 공해는 큰 문제가 되지 않았다.

광양만 저질에서의 미량오염물질 분포특성에 관한 연구 (A Study on Distribution Property of Micropollutants in Gwang-Yang Bay Sediment)

  • This study investigated the distributive property of micropollutants in sediment around Gwang-yang bay using simultaneous analytical method of 310 chemicals. In the results, the major micropollutants detected were CH type chemicals such as aliphatic, polycyclic compounds, benzenes, and CHN(O) type chemicals aromatic amines, and pesticides. Insecticides of pesticide type were frequently detected at all sampling site. The total concentration of micropollutants were higher in summer than in winter and measured within the range of $ND{\sim}36.50{\mu}g/L$. Also, because of effect of seomjin river, GY6 of all sampling site detected by the highest concentration, GY10 and 11 located in Gwang-yang outside bay were not detected the micropollutants. From the result of this study, we should estimated that the industrial complex located in Gwang-yang inside bay and an inflow of fresh water through the Seomjin river are major pollution sources of Gwang-yang bay.

Acquiring Vitellogenic Competence in the Rice Pest Nilaparvata lugens Stal: Effects of a Juvenile Hormone Analogue, Hydroprene

  • Though many insecticides are commercially available, development of resistance, pest resurgence and effects on non-target organisms led to the search for alternate insect pest management (IPM) strategy based on larval growth and reproductive fitness. Reproductive potential of insects depends on its acquiring of vitellogenic competence which is under hormonal control. Exogenous application of analogues of JR (JHAs) and ecdysterone could derail normal development and reproduction in insects by manipulating an array of physiological processes. In the rice pest, brown planthopper, Nilaparvata lugens, JHA, hydroprene induced metathetely from the fifth (final) instar nymphs in an age-dependent manner. Day 0 of the final instar showed highest sensitivity to induce this abnormal development. Adults emerged from treated day 3 nymphs looked normal. Both the morphotypes were reproductively incompetent and showed partial to complete sterility. Pre-adult exposure of the ovarian tissue to hydroprene suppressed mitotic division of germinal cells and induced abnormalities in the later s1ages of growth and differentiation of ovary in N. lugens. More over the nymphal exposure to hydroprene inhibited patency changes of follicular epithelium and affected competence of the follicles for yolk sequestration. In the absence of ovarian growth and oocyte differentiation, germarium found disintegrated, trophic core regressed and terminal oocytes resorbed. Hydroprene exposure to newly ecdysed brachypterous females did not affect ovarian development and egg production. Proper larval-adult transition appeared as a. prerequisite for vitellogenic competence in N. lugens for which the ovarian tissues must be exposed to ecdysterone in the internal milieu devoid of JH.

A Spectrophotometric Assay for Cytochrome P450 Monooxygenase Activity

  • An assay for cytochrome P450 monooxygenase activity by determination of the products of organophosphate oxidation via inhibition of acetylcholinesterase was described. Extracts from strains of Oryzaephilus surinamensis selected for resistance to chlorpyrifos-methyl (QVOS 102), fenitrothion (VOS F) and malathion (VOS 3), and a standard susceptible strain VOS 48, were incubated with an organophosphate in the presence of a NADPH-generating system and acetylcholinesterase. The degree of inhibition of the acetylchoinesterase activity was converted to manooxygenase activity using standard curves for the inhibition of acetylcholiesterase by chlorpyrifos-methyl-oxon, fenitrooxon and malaoxan. Activity was detectable in VOS 48 and was present at different increased levels with the different organophosphates in the three resistant strains, suggesting that different forms of P450 might be involved in organophosphate oxidation in these insects. The assays were carried out using crude insect homogenates and much smaller samples of insect material than the standard aldrin to dieldrin assay. It should be possible to use the method for determination of monooxygenase activity in single insert.

Urinary concentration of 3-phenoxybenzoic acid in elementary students in South Korea

  • Objectives Pyrethroid pesticides are among the most commonly using insecticides in South Korean households and have been the subject of considerable interest among public health professionals for their potential health effects. The objective of this study is to examine the level of urinary 3-phenoxybenzoic acid (3-PBA) among elementary students in South Korea. Methods We conducted a cross-sectional study to evaluate pyrethroid pesticide exposure levels by measuring the urinary metabolites of 3-PBA using a gas chromatographymass spectrometry method in March 2011. Study participants were 70 Asan-area and Incheon-area elementary students. Results All respondents had values above the detection limit, and the geometric means of 3-PBA in all children were $1.85{\mu}g/L$ and $1.46{\mu}g/g$ creatinine. Children with the top 10% urinary levels of 3-PBA were more likely to be girls, under nine years of age, living in a rural area, and living in a residential type apartment. Conclusions South Korean children have a higher concentration of urinary 3-PBA compared with those of other countries. Further research identifying exposure pathways and intervention efforts to reduce environmental pesticide use are needed in South Korea.

Salmonella System에 있어서의 농약의 돌연변이 유발성 (Mutagenicity of Pesticides in the Salmonella System)

  • Salmonella typhimurium LT-2의 histididine auxotroph 중 base substitute mutant인 TA 1535와 TA100, 그리고 frame-shiftmutant인 TA 1538과 TA 98을 각각 사용하였고 microsomal enayme activation system을 병용하여 국내 사용농약 20종에 대한 돌연변이 유발성을 검하였다. 이중 다음 4종의 농약이 양성 및 의양성의 유발성을 나타냈다. (1) Captan은 강력한 돌연변이 유발성질로서 TA1535, TA100 및 TA98에 대해 직접적으로 10$\mu\textrm{g}$/plate의 농도에서 plate당 각각 205, 1012, 그리고 297의 돌연변이 균종의 출현을 나타냈으며 microsomal enzyme activation에 의해 그 유발능력이 약간 감소되는 경향을 보였다. (2) Micut는 TA100에 대해 직접적으로 10mg/plate의 농도에서 55 revertant 정도의 미비한 돌연변이 유발성을 나타냈다. (3) Dimethoate는 TA100에서 역시 직접성으로 처리가능 최고농도인 0.2λ/plate에서 55 revertant/plate 정도의 징약한 돌연변이 유발성을 바타냈다. (4) Triforing도 역시 TA 100에서 직접적으로 10mg/plate로 처리된 농도에서 78 revertant/plate정도의 약한 돌연변이 유발성을 나타했다.

  • Biologically, MGK-264 (N-octyl bicycloheptene dicarboximide) acts as a synergists for insecticides mainly pyrethrins and pyrethroids. It's used extensively in combination with pyrethrin and piperonyl butoxide and also with personal insect repellent and cockroach repellents. But the toxic effect of MGK-264 in mamalians was a relatively little known therefore in this studies it was initiated to examine the toxic effect of MGK -264 in rats. For 5 weeks it administrated daily in each 250 mg and 500 mg of MGK-264 per kg of body weight in rats. 1) The body weight gain and the LYMPH (%) value in blood were observed a slight tendency to reduce in accordance with amount of dose and number treatment time. 2) The content of cytochrome P-450 and activity of NADPH-cytochrome c reductase were decreased in liver and those were observed some tendency in the kidney as liver but not significant. 3) The liver cholinesterase activity in the both 250 mg/kg and 500 mg/kg per kg of body weight with treated groups and the liver aniline hydroxylase in 500 mg/kg treated group were gradually decreased from 4 weeks after treated groups. In consequence it would sugested that the toxic effect of MGK-264 was low but in could offer hazard effect in liver and nervers system of rats if it was administrated move dose of MGK-264 and agumented in number of treated time.
