Development of an Enzyme-Linked Immunosorbent Assay for the Organophorus Insecticide Bromophos

  • A competitive enzyme-linked immunosorbent assay (ELISA) was developed for the quantitative detection of the organophosphorus insecticide bromophos. Three bromophos analogues (haptens) were synthesized and were coupled to carrier proteins to use as immunogens or coating antigens. Rabbits were immunized with either one of two haptens coupled to bovine serum albumin (BSA) for production of polyclonal antibodies, and the sera were screened against one of the haptens coupled to ovalbumin (OVA). Using the serum with highest specificity and an enzyme tracer, an antibody-coated ELISA was developed, which showed an $IC_{50}$ of 40 ng/mL with a detection limit of 7 ng/mL. The antibodies in this assay showed negligible cross-reactivity with other organophosphorus pesticides except with the insecticides chlorpyrifos and fenitrothion.

Selective Synthesis of 3,4-Dihydrocoumarins and Chalcones from Substituted Aryl Cinnamic Esters

  • Coumarins are ubiquitous in plant kingdom and have been used as antitumor, antifungals, anticoagulants, insecticides. Chalcones are also widespread in plant kingdom and have been known to possess diverse biological activities; antibacterial, antifungal, antitumor and anti-inflammatory, etc. As they are considered as important natural products, numerous synthetic approaches have been reported up to the present. We devise a new selective method of preparing dihydrocoumarins and chalcones from aryl cinnamates by the selection of reagents. Dihydrocoumarin derivatives were prepared selectively by using intramolecular cyclization catalyzed by p-toluene sulfonic acid. Also, chalcones were prepared by Fries-rearrangement catalyzed by $TiCl_4$. This method can be used for preparing various coumarin & chalcone compounds.

  • Understanding of the impinging behavior of an electrically charged spray is essential in determining appropriate operating conditions for electro-spraying of paints, surface coating materials and insecticides. In the present work, the wall impact behavior of an electrically charged drop has been investigated and compared with that of a neutral drop experimentally. The critical Sommerfeld number representing the spread-splash boundary for the charged drop impacting on the dielectric substrate turned out to be larger compared to that for the neutral drop with the same surface condition. The change of the transition boundary is due to the increase in the surface wettability of the drop on the substrate. However, with the electrically conducting substrates, the charging effect on the transition boundary appeared negligible. This is because the electric discharging time is much shorter than the time required for the flattened drop to reach its maximum extent.

버즘나무방패벌레 (Corythucha ciliata Say)의 생태적 특성 및 살충제감수성 (Ecological Characteristics and Insecticidal Susceptibility of Sycamore Lace Bug, Corythucha ciliata Say (Hemiptera: Tingidae))

  • To understand the biological characteristics of Corythucha ciliata occurring in Korea, developmental periods and its susceptibility to several insecticides were examined under growth chamber condition at $25^{\circ}C$. It took 11.1 day from egg to hatch. And duration of each stage up to the 5th nymph after hatching was 4.0, 2.1, 2.0, 2.9, and 4.0 days, respectively. Total duration from egg to adult was 26.1 days, preoviposition period was 12.2 days, and average number of eggs laid by a female was 83.0. Sex ratio of female was 51.6% and lifespan of female adult was 43.2% days. LC50 value of deltamethrin and esfenvalerate were 0.8 and 0.9 ppm, respectively. Insecticidal effects were better in pyrethroids than organophosphates and carbamates.

Thin Layer Chromatography에 의한 유기 카바메이트계 농약의 스크리닝 테스트에 관한 연구(1) (Studies on screening test for carbamate insecticides by thin layer chromatography)

  • 한국에서 사용하고 있는 carbamate계 살충제의 사용율(%)은 1968년경에는 2.1%인데 비해서 현재는 급증되고 있는 실정에 있다. 따라서 이로 인한 자타살 사건, 환경오염, 살포시 급만성 중독 등의 사고가 빈번히 발생하고 있는 동시에 법화학적 분석에도 많은 문제점이 되두되고 있으므로 1차적으로 시판되고 있는 carbamate계 살충제를 thin layer chromatography에 의해서 성분의 분리 확인 시험을 실시한 결과 1) Silica Gel G-60 및 Silica Gel F-254에서 적합하였고 2) 전개용매는 chloroform: benzeneP: carbontetra chloride (60:30:25), cyclohexane: acetone (80:20), cyclohexane:acetone(70:30), ether:hexane(80:20)에서 9종의 살충제가 잘 분리되었으며 3) UV light (2537 ${\AA}$) 및 2.6-dibromoquinone-4-chlorimide는 전개는 Rf치를 육안으로 구분하는데 가장 양호하였고 4) Carbamate계 살충제 중 Bux-2와 Sevin은 Silica Gel G-60에서 UV light (2537 ${\AA}$)에서 특이한 청보라 및 청색형광을 관찰할 수 있었다. 5) 이상의 결과를 보아 법화학적 시료중에 함유된 카바메이트계 농약의 분석은 가능할 것이며 또한 이 방법의 적용이 현재의 실정으로서는 적합한 것으로 사료되는 바이다.

  • The insecticidal compound priduced by Streptomyces SS-4993 strain was indicated to be a relatively stable small compound by pancreatin treatment. The active compound was adsorbed on HP20 and eluted with 70% acetone. Then, the eluate was extracted with ethyl acetate. The content of the extract was determined by thin layer chromatography (Silica gel 60 F254, hexane-ethyl ether-acetic acid,80 : 20 :1). Three active bands (Rf 0.08, 0.07, 0.88) were identified by dermal application using Bombyx mori larvae. Electrophoretic analysis revealed that the injection of strain SS-4993 culture broth into B. mori larvae induce the reduction of the hemolymph proteins level. Especially, the level of apolipophorin-I was decreased drastically.

  • A new sensitive fluorimetric method for the determination of N-methylcarbamates, a class of well known insecticides, based on the derivatization with 7-chloro-4-nitrosobenz-2-oxa-1,3-diazole (NBD-Cl) has been developed. Unreacted NBD-Cl was eluted ahead of derivatized carbamates from C-18 bonded column. An argon ion laser was used as an excitation source of chromatographic eluents and its fluorescence signal was monitored with optical multichannel analyzer. The detection limits of various carbamates were about 100 pg range and the working curves were linear to $10^4-10^5$ nanogram ranges.

  • In order to evaluate the mutagenic potential in animal for these pesticides which were proved to be mutagenic in the bacterial screening system with a metabolic activation in vitro, we have studied in vivo cytogenetic effects on mouse bone marrow by means of the micronucleus test. The clastogenic activity of the chemical is evaluated as the frequency of micronuclei in polychromatic erythrocytes. We have tested six pesticides, insecticides, DDVP and trichlorfon, fungicide, TMTD, herbicides, NIP and MO and growth regula색, maleic hydrazide. It was found that among the tested pesticides only TMTD exhibited minimal activity in inducing micronuclei. Organophosphorus insecticide DDVP that is the most broadly used and economically important chemical, did not increase the micronuclei frequencies in mouse bone marrow cells as with the all other pesticides tested.

Indirect Bacterial Effect Enhanced Less Recovery of Neonicotinoids by Improved Activities of White-Rot Fungus Phlebia brevispora

  • Bacterial strains that improve mycelial morphology and growth of white-rot fungi in liquid medium could enhance the impact of white-rot fungi towards lesser recovery of neonicotinoids when cocultured. This was demonstrated by the recovery of clothianidin and acetamiprid from cocultures of the white-rot fungus Phlebia brevispora strains with two mycelial-growth-promoting bacteria, Enterobacter sp. TN3W-14 and Pseudomonas sp. TN3W-8. Clothianidin recovery from cocultures of white-rot fungi and bacteria was over 40% lower than that from axenic microbial cultures and mixed-bacterial cultures. About 20% less acetamiprid was equally recovered from both TMIC33929+TN3W-14 cocultures and mixed-bacterial cultures than from axenic fungal and bacterial cultures.
