한국산 바구미과(科)(딱정벌레목(目)) 9미기록종(種)에 대한 보고 (Newly Recorded Species of the Family Curculionidae in Korea (Coleoptera; Curculrionidae))

  • 본 연구를 통해 바구미과의 Pimelocerus elongates (Roelofs, 1873), Phylaitis maculiventris Voss, 1958, Egiona picta (Roelofs, 1875), Cryptorhynchus electus (Roelofs, 1875), Rhadinopus confinis Voss, 1958, R. sulcatostriatus (Roelofs, 1875), Deiradocranus setosus (Morimoto, 1962), Rhadinomerus annulipes (Roelofs, 1875), R. maebarai Chujo et Voss, 1960 등 9종을 한국에서 처음 보고하며, 분포가 불분명했던 Shirahoshizo hiurai Morimoto는 한국에 분포함을 확인하였다. 이들의 특징에 대한 기술과 형태 사진을 제시하였다.

Alien Hitchhiker Insect Species Detected from International Vessels Entering Korea in 2022

  • Hitchhiker insect species from international vessels entering Korea in 2022 were monitored. A total of 947 samples of hitchhiker insects were collected using a simple collection method by hand. Among them, 856 individuals were classified as 374 species of 86 families in 10 orders through integrative analysis with DNA barcoding and morphological examination. The rest 91 individuals were identified only to the family level. As a result of examining the distribution of the 374 species (856 individuals), 38 species (71 individuals) were confirmed as not-distributed species in Korea, including six species (11 individuals) as 'regulated species' listed by the Korean Animal and Plant Quarantine Agency. Of 38 not-distributed species, 10 species were detected multiple times (at least twice). Accordingly, it is necessary to strengthen monitoring of the area around the port of entry along with continuous surveillance to prevent invasion of species detected multiple times. For monitoring alien hitchhiker insect species, this study provided detection information and biological data for alien species.

榮山江 水系의 水棲混蟲의 分布와 季節的 變動 (Distribution and Seasonal Fluctuation of Aquatic Insects in the Yongsan River System)

  • Aquatic insects were collected from 18 sites in the Yongsan River System from August, 1988 to July, 1989. The monthly fluctuation and distributional pattern of the aquatic insects along the environmental factors were investigated. Quantitatively collected aquatic insects were identified, and numbers, standing biomass, dominance indices and diversity indices in each site and month were determined. The aquatic insect faun of the Yongsan River System was composed of 146 species, 82 genera, 38 families in 7 orders including 44 species of mayflies, 32 species of caddisflies, 24 species of stoneflies, 21 species of flies, 16 species of beetles, 6 species of dragonflies and 3 species of dobsonflies. Seasonal occurrence of species were more diversified in winter and spring than in summer. Also, the number of individuals occurred was higher in winter and spring than in summer while it was the highest in upper stream and the lowest in Kwang-ju stream. Component ratio of the number of individuals according to the insect orders was as follows: Mayflies 38.71%, Flies 36.97%, Stoneflies 10.21%, Caddisflies 6.80%, Beetles 2.96%, Dobbsonflies 2.74%, Dragonflies 0.95%. Percentage of standing biomass according to insect orders were as follows: Mayfiles 32.81%, Stoneflies 26.51%, Caddisflies 17.58%, Dobbsonflies 14.84%, Flies 3.74%, Beetles 3.08%, Dragonflies 1.45%, The highest standing biomass occurred in spring and the lowest in summer.

Subcellular Location of Spodpotera Cell-expressed Human HepG2-type Glucose Transport Protein

  • The baculovirus/insect cell expression system is of great value for the large-scale production of normal and mutant mammalian passive glucose-transport proteins heterologously for structural and functional studies. In most mammalian cells that express HepG2, this transporter isoform is predominantly located at the cell surface. However, it had been reported that heterologous expression of other membrane proteins using the baculovirus system induced highly vacuolated cytoplasmic membranes. Therefore, how a cell responds to the synthesis of large amounts of a glycoprotein could be an interesting area for investigation. In order to examine the subcellular location of the human HepG2 transport proteins when expressed in insect cells, immunofluorescence studies were carried out. Insect cells were infected with the recombinant baculovirus AcNPVHIS-GT or with wild-type virus at a MOI of 5, or were not exposed to viral infection. A high level of fluorescence displayed in cells infected with the recombinant virus indicated that transporters are expressed abundantly and present on the surface of infected Sf21 cells. The evidence for the specificity of the immunostaining was strengthened by the negative results shown in the negative controls. Distribution of the transporter protein expressed in insect cells was further revealed by making a series of optical sections through an AcNPVHIS-GT-infected cell using a confocal microscope, which permits optical sectioning of cell sample. These sections displayed intense cytoplasmic immunofluorecence surrounding the region occupied by the enlarged nucleus, indicating that the expressed protein was present not only at the cell surface but also throughout the cytoplasmic membranous structures.

Study on the Plants Planted in Rooftop and Their Damage by Insect Pests

  • Plants planted in the green-roofed areas in Busan and Jinju were surveyed. The woody plants investigated in this study were classified into 52 families and 156 species, and the herbaceous plants were classified into 30 families and 97 species. Woody plants mainly planted were Rhododendron yedoense var. poukhanense, R. indicum, C. kousa, P. mume, and E. alatus. However, Pinus spp. were planted in all areas. The main herbaceous species planted were Sedum kamtschaticum, S. takesimense, S. middendorffianum, T. quinquecostatus var. japonica, and A. spathulifolius Maxim. According to surveying the distribution of woody plant pests, they could be classified into six orders, 24 families, and 46 species that usually appeared from April to October but especially between June and September. We investigated 39 insect species in relation to pest damage to leaves, 21 insect species in relation to that of branches, and 39 insect species in relation to that of stems of woody plants.

Flow Cytometric Analysis of the Effect of Silkworm Hemolymph on the Baculovirus-Induced Insect Cell Apoptosis

  • The effect of silkworm hemolymph on the inhibition of baculovirus-induced insect cell apoptosis was quantitatively investigated using a flow cytometric analysis. Spodoptera frugiperda (Sf9) cell and Autographa californica nuclear polyhedrous virus (AcNPV) were used as a model for insect cell and baculovirus in this study, respectively. Compared with a mammalian cell cycle, the fraction of G1 cells was relatively small in the Sf9 cell cycle. Silkworm hemolymph did not affect the Sf9 cell-cycle distribution before the baculovirus infection. However, the fraction of cells which are not in the sub-G1 phase remained at a high level for 3 days after the infection in the medium without silkworm hemolymph, while it remained at a high level for 7 days after the infection in the medium supplemented with silkworm hemolymph. The fractions of apoptotic cells in the sub-G1 phase were $4.7\%$, and 4 days after infection, $22.7\%$, in the media with and without silkworm hemolymph, respectively.

지리산 중산리골, 대원사골 및 거림골에서 산림해충의 분포조사 (Survey on the Forest Insect Pests in Junsanri-gol, Daewonsa-gol and Georim-gol of Mt. Jiri)

  • 지리산의 중산리골, 대원사골과 거림골을 잇는 지역에서 수목에 피해를 주는 산림해충은 7목 44과 140종으로 조사되었다. 이 중 나비목은 16과 59종, 딱정벌레목 9과 35종, 매미목 10과 33종, 노린재목 3과 4종, 벌목은 3과 6종, 메뚜기목 1과 1종, 대벌레목 2과 2종으로 조사되었다. 조사지 역별 산림해충은 대원사골 6목 36과 85종, 중산리골 6목 31과 67종, 거림골 7목 27과 56종이었다.

  • 기후변화는 곤충의 밀도와 분포, 몸의 형태와 개체의 크기 등 생물학적인 형질 변화, 생식 및 유전적 특성, 그리고 멸종 등에 영향을 미칠 것으로 예상되고 있다. 기후변화의 영향을 예측하여 피해를 줄이기 위해서는 분산되어 있는 곤충 종별 기본적인 생물학적/생태학적 정보들을 종합하여 검토할 필요가 있다. 따라서 본 연구는 기후 및 환경의 변화에 대한 곤충, 특히 해충의 발생 변화 예측에 필요한 생물학적 정보를 정리하여 이를 활용한 미래 피해 예측을 위한 기초 자료를 제공하고자 수행하였다. 또한 국내외 문헌을 비교 분석하여 국내에서 기후변화 연구를 수행하는데 있어 제한 요인들을 확인하고 향후 필요한 연구소요를 제시하고자 하였다. 국내의 연구들은 단기 모니터링 자료를 이용하여 환경 요인과의 관계를 분석하는 수준에 그치고 있는 반면, 국외 연구들은 장기 모니터링 자료를 이용한 분포 변화 분석이나 기 개발된 생물 종의 파라미터를 이용한 발생 및 분포 변화 예측 그리고 곤충 군집의 구성 변화를 모니터링하는 등 다양한 내용을 주제로 기후변화의 영향을 연구하고 있었다. 결론적으로 기후변화에 대응하기 위해서는 체계적인 모니터링 기술 개발을 통해 곤충의 계절발생, 분포, 월동 특성 및 유전적 구조 변화에 대한 연구가 필수적이며, 주요 해충 및 잠재적인 해충에 대한 기후변화의 영향을 예측하기 위해 곤충 개체군 모델의 개발 역시 중요한 부분이 될 것이다.

국내 서남 지역의 산림 토양 특성과 곤충병원성 진균과의 상관관계 (Correlation between Isolated Entomopathogenic Fungi and Soil Characteristics from Forest Areas of the Southwest Region in Korea)

  • 본 논문에서는 국내 산림 지역의 토양에서 서식하는 곤충병원성 진균을 insect-baiting 방법을 이용하여 분리하고자 하였으며, 곤충병원성 진균과 토양의 물리적, 화학적 특성과의 관계를 분석하고자 하였다. 국내 산림 지역 5곳을 선정하여 토양을 채집하였으며, 채집된 토양에서 총 42 분리주를 분리하였다. 그 중 Beauveria bassiana (42.9%)와 Metarhizium anisopliae (45.2%)가 주로 분리되었으며, 특히 M. anisopliae의 분포는 토양의 총 질소량(g/kg)과 유기물 함량(%)에 따라 달라지는 것이 확인되었다. 국내 산림 지역의 토양은 낮은 pH를 보였으며(pH 4-5), 전체 분리주 중에서 미사질 양토(silt loam)에서 50% 이상이 분리되었다. 본 연구의 결과는 통하여 토양의 물리적, 화학적 특성과 곤충병원성 진균의 분포 및 다양성과의 관계를 이해하는데 도움이 될 것으로 판단된다.

Cutaneous Microflora from Geographically Isolated Groups of Bradysia agrestis, an Insect Vector of Diverse Plant Pathogens

  • Larvae of Bradysia agrestis, an insect vector that transports plant pathogens, were sampled from geographically isolated regions in Korea to identify their cutaneous fungal and bacterial flora. Sampled areas were chosen within the distribution range of B. agrestis; each site was more than 91 km apart to ensure geographical segregation. We isolated 76 microbial (fungi and bacteria) strains (site 1, 29; site 2, 29; site 3, 18 strains) that were identified on the basis of morphological differences. Species identification was molecularly confirmed by determination of universal fungal internal transcribed spacer and bacterial 16S rRNA gene sequences in comparison to sequences in the EzTaxon database and the NCBI GenBank database, and their phylogenetic relationships were determined. The fungal isolates belonged to 2 phyla, 5 classes, and 7 genera; bacterial species belonged to 23 genera and 32 species. Microbial diversity differed significantly among the geographical groups with respect to Margalef's richness (3.9, 3.6, and 4.5), Menhinick's index (2.65, 2.46, and 3.30), Simpson's index (0.06, 0.12, and 0.01), and Shannon's index (2.50, 2.17, and 2.58). Although the microbial genera distribution or diversity values clearly varied among geographical groups, common genera were identified in all groups, including the fungal genus Cladosporium, and the bacterial genera Bacillus and Rhodococcus. According to classic principles of co-evolutionary relationship, these genera might have a closer association with their host insect vector B. agrestis than other genera identified. Some cutaneous bacterial genera (e.g., Pseudomonas) displaying weak interdependency with insect vectors may be hazardous to agricultural environments via mechanical transmission via B. agrestis. This study provides comprehensive information regarding the cutaneous microflora of B. agrestis, which can help in the control of such pests for crop management.
