해양환경에서 Pyrene의 생분해 (Biodegradation of Pyrene in Marine Environment)

  • 난분해성 다핵방향족 탄화수소인 pyrene의 생분해를 해수와 해사로 구성된 microcosm에서 조사하였다. 농도 100~1,000 ppm pyrene의 5주동안 자연분해도는 5~$20^{\circ}C$에서 1~11% 이고 이때 전환율은 310~2,200 ng/g(ml).day 이었다. 생분해에 미치는 분해균주 접종량, pyrene 농도, 온도와 계면활성제 첨가의 영향등을 영양염류가 투여된 시료에서 조사하였다. 혼합 분해균주접종시 해수보다는 해사에서 전환율이 전체적으로 높았으며 $10^{7}$cells/ml 접종시 pyrene 200 ppm 농도의 해사에서 대조구보다 분해도가 7.8배 높았다. 가장 높은 전환율 4,860 ng/g/day는 1,000 ppm pyrene이 첨가된 해사에 복합균주를 접종한 시료에서 측정되었다. Glucose 첨가시 해사에서는 배양초기에만 분해도의 증가가 나타났으며, 해수에서는 분해도가 8~10% 증가하였다. 계면활성제와 mineral oil 의 첨가는 오히려 pyrene 분해도를 대부분 감소시켰는데 CMC가 생분해에 큰 영향을 미치는 것으로 판단된다.

Chlorella saccharophila 배양 최적화 및 유용물질의 생산 (Optimization of Chlorella saccharophila Cultivation and Useful Materials Production)

  • 본 연구에서는 Chlorella saccharophila의 배양을 통하여 바이오에너지 자원을 대량으로 확보하고자 배지 최적화 실험을 진행하였다. 최적화 인자로는 배양 형태, 초기 접종량, 탄소원 종류 및 농도, 질소원 종류 및 농도, 배양시간이다. 실험 결과, 배양 형태는 광원과 외부탄소원을 모두 공급하는 mixotrophic 배양이 적절하였다. 초기 접종량은 3% (v/v), 탄소원은 glucose 30 g/L, 질소원은 $NaNO_3$ 0.95 g/L를 첨가하는 것이 우수하였다. 최적 배지 조건으로 배양한 결과, oil의 함량은 12일에서 가장 높았으나, 회수되는 C. saccharophila의 biomass양과 chlorophyll의 양은 10일에서 가장 높았다. 위의 결과는 미세조류의 배지 최적화를 통하여 대량배양을 위한 기초자료로 사용될 수 있으리라 판단된다.

함박꽃나무의 현탁배양세포로부터 [+]-Eudesmin의 생산을 위한 최적화 (Optimization of Culture Conditions for the [+]-Eudesmin Production in Magnolia Sieboldii Cells)

  • 목련과 수종인 함박꽃나무의 현탁배양세포로부터 생리활성을 갖는 리그난화합물인 (+)-eudesmin을 효율적으로 생산하기 위한 연구로써 플라스크배양 단계에서의 다양한 배양조건들 즉, 배지, 초기 당농도, 교반속도, 초기 접종농도, 그리고 elicitation 효과를 확인하고자 하였다. MS배지를 포함한 4종의 배지에서는 물질의 생산성과 생중량 모두에서 MS배지가 적합한 것으로 나타났다. 130 rpm으로 교반되는 항온배양기에서 $3\%$ sucrose와 0.5 mg/L 2,4-D가 첨가된 MS배지에 0.5 mg (DCW)의 농도로 세포를 접종한 실험구에서 8주 후 플라스크 당 3.71 g (DCW)의 생중량을 얻었으며, 지표물질인 (+)-eudesmin의 함량은 $5\%$ sucrose와 200 mg/L chitosan 처리구에서 $3.2{\mu}g/g$ (DCW)으로 대조구에 비해 1.7배의 증가를 나타내었다. 이와 같은 연구결과는 생물반응기를 이용한 목련과 수종에서의 유용물질 생산 연구에 활용될 수 있을 것으로 사료된다.

Production of Tween 80-inducing Esterase by Acinetobacter sp. B1 Using Response Surface Methodology

  • Esterase produced by Acinetobacter sp. B1 (strain B1) was optimized by means of one-variable-at-a-time and response surface methodologies. Results of the one-variable-at-a-time experiment showed that Tween 80 significantly increased esterase production of strain B1. The addition of Tween 80 to the culture medium increased the biomass and esterase activity of strain B1, stimulated content of total extracellular protein, and enhanced the oleic acid (C18:1) composition in the cell membrane of strain B1. The influence of eight culture variables on esterase production was evaluated by Plackett-Burman design. Results showed that Tween 80, pH, and $K_2HPO_4$ significantly affected the esterase production of strain B1. Tween 80, pH, and $K_2HPO_4$ were further optimized by central composite design. Under the optimized conditions (w/v, soluble starch 2.5%, tryptone 1.5%, Tween 80 0.8%, $K_2HPO_4$ 0.5%, NaCl 0.5%, pH 8.0, inoculum size 1%, and inoculum age 24 h), the maximum esterase activity of strain B1 was 152.13 U/ml, which was 10-fold higher than that of non-optimization after 36 h cultivation.

Engineered bioclogging in coarse sands by using fermentation-based bacterial biopolymer formation

  • Sealing of leakage in waterfront or water-retaining structures is one of the major issues in geotechnical engineering practices. With demands for biological methods as sustainable ground improvement techniques, bioclogging, defined as the reduction in hydraulic conductivity of soils caused by microbial activities, has been considered as an alternative to the chemical grout techniques for its economic advantages and eco-friendliness of microbial by-products. This study investigated the feasibility of bioaugmentation and biostimulation methods to induce fermentation-based bioclogging effect in coarse sands. In the bioaugmentation experiments, effects of various parameters and conditions, including grain size, pH, and biogenic gas generation, on hydraulic conductivity reduction were examined through a series of column experiments while Leuconostoc mesenteroides, which produce an insoluble biopolymer called dextran, was used as the model bacteria. The column test results demonstrate that the accumulation of bacterial biopolymer can readily reduce the hydraulic conductivity by three-to-four orders of magnitudes or by 99.9-99.99% in well-controlled environments. In the biostimulation experiments, two inoculums of indigenous soil bacteria sampled from waterfront embankments were prepared and their bioclogging efficiency was examined. With one inoculum containing species capable of fermentation and biopolymer production, the hydraulic conductivity reduction by two orders of magnitude was achieved, however, no clogging was found with the other inoculum. This implies that presence of indigenous species capable of biopolymer production and their population, if any, play a key role in causing bioclogging, because of competition with other indigenous bacteria. The presented results provide fundamental insights into the bacterial biopolymer formation mechanism, its effect on soil permeability, and potential of engineering bacterial clogging in subsurface.

양수의 대장균에 대한 세균증식 억제효과 (Bacterial Growth-inhibiting Activity of Amniotic Fluid Against E. coli)

  • The amniotic fluid provides a medium in which the fetus can readily move, cushions him against possible injury and helps him maintain an even temperature. Besides above mentioned functions, investigators reported that human amniotic fluid contains host-resistance factors which prevent bacteria from producing infectious disease and this activity shows difference among human racial groups or bacterial genera, species and strains. 40 amniotic fluid specimens from Korean women in their second and third trimesters of pregnancy were examined for inhibiting the growth of Escherichia coli. And various factors which might affect bacterial growth inhibiting activity such as pH, initial inoculum size, concentration of amniotic fluid, and heat resistance, were also tested using a strongly inhibitory amniotic fluid specimen. Finally plate diffusion tests were carried out using other strongly inhibitory amniotic fluid. The following results were obtained: 1. Of the 40 fluid samples examined, 18 specimens(45%) had inhibitory activity and samples from women in their second trimester of pregnanancy showed non-inhibitory activity(2 specimens). 2. The pH of the fluids varied between 7.43 and 8.33. There was no correlation between pH and inhibitory activity. 3. No. 19 amniotic fluid showed bacteriostatic activity after 24 hours incubation when an inoculum of $10^2$ organisms per milliliter was used, but non-inhibitory with an inoculum of $10^3$ and $10^4$ bacteria per milliliter. 4. The content of amniotic fluid in culture media influenced E. coli growth. At 90 percent, E. coli was inhibited growth but at 10 percent and 50 percent. 5. Inhibitory activity of No. 19 amniotic fluid was retained after heating to $50^{\circ}C$ for 30 minutes or 100^{\circ}C$ for 30 minutes. 6. Plate diffusion tests with No. 27 amniotic fluid showed that 0.7ml amniotic fluid gave clear zone of growth inhibition around the central well but 0.2ml and 0.1ml amniotic fluids were not.

사과식초 제조를 위한 사과주스의 초산발효 최적화 (Optimization of the Acetic Acid Fermentation Condition of Apple Juice)

  • 초산발효조건에 따른 사과주스의 발효특성을 알아보기 위하여 flask scale에서 초기 알코올 농도, 당농도, 초산농도 및 종초접종량 조건에 따른 초산발효를 실시하였다. 초기 알코올농도 3, 5, 7, 9%를 달리하여 사과주스의 초산발효를 12일간 실시한 결과 알코올 함량 5% 일때 발효 10일 째 산도가 최대치 5.88%로 측정되었으며, 알코올 함량이 9%일 경우 초산발효가 정상적으로 진행되지 않았다. 초기 당 농도 1, 5, 10, 14 $^{\circ}Brix$로 달리하여 초산발효를 실시한 결과 1 $^{\circ}Brix$의 경우 산도가 다른 조건에 비해 서서히 증가하여 발효 12일차에 산도 4.48%로 측정되었으며, 5, 10 $^{\circ}Brix$가 상대적으로 빠른 시간 내에 산도 4% 이상 생성됨을 알수 있었다. 초기 산도를 0.3, 0.5, 1, 1.5 및 2%로 달리하여 초산발효를 실시한 결과 초기 산도가 1% 이상일 때는 발효가 원활히 진행되는 것으로 나타났다. 종초 접종량은 10%, 15% 이상일 때 초산발효가 정상적으로 진행되어 발효 8일차에 산도가 각각 5.60, 6.05%에 도달하였다. 따라서, 사과주스를 이용한 초산발효시에는 알코올농도 5%, 초기 당농도 5 $^{\circ}Brix$ 이상, 초기 산도 1.0% 이상 및 종초 접종량 10% 이상이 적절할 것으로 생각된다. 산업적 규모의 초산발효 전단계로서 Mini-jar fermentor를 이용하여 초산발효를 실시한 결과 14 $^{\circ}Brix$ 사과농축액 2L에 알코올 7%와 $30^{\circ}C$, 300 rpm에서 활성화 된 종초 10%(총산함량: 4.03%)를 접종하여 초기산도를 약 1.0%로 조정한 후 $30^{\circ}C$, 300 rpm, 1.0 vvm으로 통기하여 초산발효를 실시한 후 시간대별로 분취하여 분석한 결과 Flask scale에서 보다 시간이 많이 단축되어 48 hr 발효하였을 때 산도 5% 이상의 식초를 생산할 수 있었다.

Influence of Diet Induced Changes in Rumen Microbial Characteristics on Gas Production Kinetics of Straw Substrates In vitro

  • The effect of diets varying in level and source of nitrogen (N) and fermentable organic matter on dynamic characteristics of microbial populations in rumen liquor and their impact on substrate fermentation in vitro was studied. The diets tested were straw alone, straw+concentrate mixture and straw+urea molasses mineral block (UMMB) lick. The same diets were taken as substrates and tested on each inoculum collected from the diets. Diet had no effect on the amino acid (AA) composition of either bacteria or protozoa. Differences among the diets in intake, source of N and OM affected bacterial and protozoal characteristics in the rumen. Upper asymptote of gas production (Y$\alpha$) had a higher correlation with bacterial pool size and production rate than with protozoal pool size and production rate. Among the parameters of the gas production model, Y$\alpha$ and lag time in total gas has showed significant (p<0.01) correlation with bacterial characteristics. Though the rate constant of gas production significantly differed (p<0.01) between diet and type of straw, it was least influenced by the microbial characteristics. The regression coefficient of diet and type of straw for Y$\alpha$ indicated that the effect of diet on Y$\alpha$ was threefold higher than that of the straw. As microbial characteristics showed higher correlation with Y$\alpha$, and diet had more influence on the microbial characteristics, gas production on a straw diet could be used effectively to understand the microbial characteristics.

Xylanase Production in Solid State Fermentation by Aspergillus niger Mutant using Statistical Experimental Designs

  • The production of xylanase from Aspergillus niger mutant in SSF was optimized by' using statistical experimental designs. An inoculum size of $5{\times}10^5$ spores/g. initial moisture content of 65 %. cultivation time of 5 days and 10 times concentration of basal medium containing 50 times concentration of CSL were optimum for xylanase production ‘ Under the optimized conditions. the activity and productivity of xytanase obtained after 5 days of fermentation were 5.071 IU/gram of rice straw and 14.790 IU/l.h. respectively.

해양세균 Zoogloea sp.로부터 유용 다당류의 생산에 미치는 발효조건의 영향 (Effects of Fermentation Conditions on the Production of the Useful Polysaccharides from Marine Bacterium Zoogloea sp.)

  • The fermentation conditions for the maximal production of the useful polysaccharides(water soluble polysaccharide and cell bound polysaccharide) from marine bacterium Zoogloea sp.(KCCM 10036) were investigated with a 5 L jar fermentor. The maximal production of these polysaccharides was obtained under the conditions of initial pH 7.8, 30$^{\circ}C$, 400 rpm of agitation speed, 2 Wm of aeration rate, 10%(w/v) of inoculum size and 2.5%(w/v) of glucose substrate and 10.38 g/L of total polysaccharide was produced. Apparent viscosity of the culture broth was increased with the production of these polysaccharides and the maximum value was reached to 22,500 cp.
