IFIT1 (also known as ISG56) is a member of the interferon-inducible protein with tetratricopeptide repeats (IFITs) family. IFITs are strongly induced by type I interferon (IFN), double-stranded RNA and virus infection. Here, we investigated IFIT1 expression in human umbilical vein endothelial cells (HUVECs) and in human bronchus epithelial cells (BEAS-2Bs) induced by the H9N2 virus and inactivated viral particle at different time points. We also investigated the effect of H9N2 virus and viral particle infection on $IFN-{\alpha}/{\beta}$ production, and assessed whether hemagglutinin or neuraminidase protein induced IFIT1 expression. Results showed that both H9N2 virus infection and viral particle inoculation induced the expression of IFIT1 at mRNA and protein levels in the two cell lines. Hemagglutinin or neuraminidase protein binding alone is not sufficient to induce IFIT1 expression. Surprisingly, the expression patterns of IFIT1 in response to H9N2 virus and viral particles in the two cell lines were opposite, and production kinetics of $IFN-{\alpha}/{\beta}$ also differed. An additional finding was that induction of IFIT1 in response to H9N2 virus infection or viral particle inoculation was more sensitive in HUVECs than in BEAS-2Bs. Our data offers new insight into the innate immune response of endothelial cells to H9N2 virus infection.
Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a highly transmissible and potentially fatal virus. So far, most comprehensive analyses encompassing clinical and transcriptional manifestation have concentrated on the lungs. Here, we confirmed evident signs of viral infection in the lungs and spleen of SARS-CoV-2-infected K18-hACE2 mice, which replicate the phenotype and infection symptoms in hospitalized humans. Seven days post viral detection in organs, infected mice showed decreased vital signs, leading to death. Bronchopneumonia due to infiltration of leukocytes in the lungs and reduction in the spleen lymphocyte region were observed. Transcriptome profiling implicated the meticulous regulation of distress and recovery from cytokine-mediated immunity by distinct immune cell types in a time-dependent manner. In lungs, the chemokine-driven response to viral invasion was highly elevated at 2 days post infection (dpi). In late infection, diseased lungs, post the innate immune process, showed recovery signs. The spleen established an even more immediate line of defense than the lungs, and the cytokine expression profile dropped at 7 dpi. At 5 dpi, spleen samples diverged into two distinct groups with different transcriptome profile and pathophysiology. Inhibition of consecutive host cell viral entry and massive immunoglobulin production and proteolysis inhibition seemed that one group endeavored to survive, while the other group struggled with developmental regeneration against consistent viral intrusion through the replication cycle. Our results may contribute to improved understanding of the longitudinal response to viral infection and development of potential therapeutics for hospitalized patients affected by SARS-CoV-2.
Orientia tsutsugamushi, a gram-negative bacterium, causes severe acute febrile illness in humans. Despite this danger, the route of infection, infectivity, and protective mechanisms of the host's immune response to O. tsutsugamushi are unclear. Dendritic cells (DCs) are one of the most important cell types in bridging the innate and adaptive immune responses. In this study, we observed that O. tsutsugamushi infects and replicates in monocyte-derived DCs (MODCs). During infection and replication, the expressions of the cytokines IL-12 and TNF-${\alpha}$, as well as the co-stimulatory molecules CD80, CD83, CD86, and CD40, were increased in MODCs. When O. tsutsugamushi-treated MODCs were co-cultured with autologous $CD4^+$ T cells, they enhanced production of IFN-${\gamma}$, a major Th1 cytokine. Collectively, our results show that O. tsutsugamushi can replicate in MODCs and can simultaneously induce MODC maturation and increase proinflammatory cytokine levels in MODCs that subsequently activate $CD4^+$ T cells.
선천성 면역은 숙주의 물리적 방어벽을 뚫고 침입하는 감염성 질병 원인균에 대항하는 첫 번째 방어로서 아주 중요한 역할을 한다. Mannose-binding lectin (MBL 또는 mannan-binding protein, MBP)은 혈청 내에 존재하는 면역성 단백질로서 감염 후 즉시 유발되는 acute phase response의 특정 단백질이다. MBL 단백질은 세균, 바이러스, 곰팡이, 기생충 등의 탄수화합물 구조에 결합하여 식균 작용을 돕거나 보체경로를 활성화 시킨다. MBL 단백질은 C-말단이 탄수화물을 인식하는 도메인이며, 연결 목 부위와 콜라겐 부위로 구성되어 있다. 혈청 내의 MBL 농도가 낮으면 높은 빈도로 면역결핍현상이 관찰된다고 알려져 있다. MBL 단백질의 기능과 유전에 대해 많은 연구가 되어져 왔으나 아직 MBL 단백질 복합체 등에 대한 연구는 많이 이루어져 있지 않다. 따라서 MBL 연구에 필수적인 MBL cDNA 제조와 재조합 단백질의 합성, 그리고 재조합 단백질을 항원으로 사용하여 polyclonal antibody를 생산한 연구 결과를 보고하는 바이다. 본 연구결과로 획득한 MBL cDNA, 재조합 단백질과 anti-MBL 항체는 앞으로의 MBL 연구에 절대적으로 필요한 도구가 될 것으로 생각된다.
점막면역체계는 병원체에 대항하여 정밀하게 설계된 생물체의 방어체계로 그 중 위장관은 최전방에서 가장 중요한 기능을 하는 기관이다. 숙주 감염을 일으키는 병원성 미생물 가운데 식중독을 유발하는 살모넬라는 구강경로로 체내로 들어와 파이어판을 통해서 침입한다. 다양한 혈청형의 살모넬라 균주는 여러 톨유사수용체의 리간드를 통해서 숙주의 선천면역을 자극하지만, 숙주 특이성과 병원성에 따라 장내 점막에서 다양한 후천면역반응을 유도하기도 한다. 우리는 살모넬라 균주 중 비교적 연구가 덜 되어있는 S. enteritidis에 의해 감염되었을 때 장 내에서 일어나는 면역반응을 연구하였다. 우선 마우스에 처리할 적정 농도의 살모넬라 농도를 정하기 위해 살모넬라의 농도를 다르게 하여 실험을 진행하였고 고농도의 살모넬라 처리군에서 더 낮은 생존율과 높은 몸무게 감소율을 관찰하였다. 반수치사량의 반에 해당하는 농도의 살모넬라를 마우스에 감염시켰을 때 장 내 수지상세포와 T 세포에 유의적인 차이가 없는 것을 확인하였다. 하지만 동일한 조건의 마우스에서 회장조직을 염색했을 때 뮤신 분비의 증가와 술잔세포의 감소를 확인할 수 있었다. 또한 밀착연접단백질 유전자의 상대적인 발현량을 분석하였고 살모넬라 처리군에서 Claudin의 유의적인 감소를 확인할 수 있었다. 이어 살모넬라 감염 후 DSS를 처리한 실험에서 살모넬라 처리군에서 생존율이 낮아짐을 확인하여 S. enteritidis가 염증반응을 악화시킨다는 것을 알 수 있었다.
지구상에 존재하는 전체 동물 중 가장 큰 부분을 차지 하고 있는 곤충은 예로부터 인간의 식품, 농업, 산업, 의약 등의 일상 생활에 이용되어 왔다. 많은 수와 높은 영양학적 가치로 곤충의 생리활성물질이 미개발 생물자원으로 재평가 되고 있다. 곤충은 면역세포, 곤충 혈구세포, 효소들의 연쇄반응 혹은 항균 단백질/펩타이드 같은 방법으로 외부의 감염에 저항성을 가지게 된다. 항균 펩타이드는 곤충의 혈림프의 선천성 면역 시스템 중 주요한 성분중의 하나로 항생제 내성 균주의 출몰이 빈번하게 일어나 해결책으로 새로운 항생제 개발이 시급한 시점에서 유력한 후보물질로 주목 받고 있다. 곤충 유래 항균 펩타이드는 150개가 넘게 분리되었으며 크게 세크로핀, 디펜신, 글라이신/프롤린이 풍부한 펩타이드로 이루어진다. 이 논문에서, 향균 펩타이드를 생산하는 여러 곤충 중에서 벌, 소똥구리, 울도하늘소, 나비 그리고 울도하늘소에서 얻을 수 있는 펩타이드의 종류 그리고 작용 기작에 대해 알아보았다. 이 펩타이드들은 항균효과를 가지고 있으며 멜리틴을 제외하고 적혈구의 용혈 현상이 나타나지 않고 주로 세포막을 붕괴시키거나 세포자살기작을 유도하여 병원성 미생물의 성장을 억제한다. 곤충 유래 펩타이드와 같은 생리활성물질이 그 활용 가능성의 면에서 엄청난 가능성을 가지고 있어 이에 대한 연구는 앞으로 더 주목을 받을 것이다.
본 연구는 인공재배 상황버섯 자실체 추출물인 금사목질진흙버섯 자실체 추출물의 mouse sarcoma cells인 S-180 세포에 대한 정맥 투여 시 항암 효과와 급성독성 및 4주 아 급성 독성에 대한 보고이다. 금사목질진흙버섯 자실체 추출물의 항암효과는 250 mg/kg에서 대조군 대비 42.7%의 크기로 가장 높은 효과를 나타내었으며, 농도 의존적인 경향을 나타내었다. 급성 독성에서는 $LD_{50}$치가 632.84 mg/kg (♂)과 814.48 mg/kg (♀)로 나타났으며, 이자와 장기의 무게비가 증가하는 현상이 나타나, 영향이 있는 것으로 추측되었다. 4주 아 급성 독성 시험에서 $LD_{50}$치는 355.41 mg/kg (♂)과 383.53 mg/kg (♀)로 나타났으며, 특히 250 mg/kg 투여군에서는 간의 무게 비가 2배 정도 증가하는 현상과 황색 반점이 나타났다. 특히 125 mg/kg 이상 투여군에서는 점프 능력과 민첩성 등의 운동 능력이 현저히 증가되는 현상이 관찰되었으며, 이에 대한 정확한 메카니즘은 확인할 수 없었다. 이상의 결과를 바탕으로 금사목질진흙버섯 자실체 추출물에 대한 표준물질 정제 과정을 통한 작용기전과 운동능력 향상에 대한 실험 등이 추가된다면 유효 농도에서 독성이 낮으면서도 암을 제어할 수 있는 새로운 천연물 신약의 후보 물질로 이용될 가능성이 있는 것으로 판단된다.

Airway mucus secretion is an essential innate immune response for host protection. However, overproduction and hypersecretion of mucus, mainly composed of the gel-forming MUC5AC protein, are significant risk factors for patients with asthma and chronic obstructive pulmonary disease (COPD). The transforming growth factor β (TGFβ) signaling pathway negatively regulates MUC5AC expression; however, the underlying molecular mechanism is not fully understood. Here, we showed that TGFβ significantly reduces the expression of MUC5AC mRNA and its protein in NCI-H292 cells, a human mucoepidermoid carcinoma cell line. This reduced MUC5AC expression was restored by a TGFβ receptor inhibitor (SB431542), but not by the inhibition of NF-κB (BAY11-7082 or Triptolide) or PI3K (LY294002) activities. TGFβ-activated Smad3 dose-dependently bound to MUC5AC promoter. Notably, TGFβ-activated Smad3 recruited HDAC2 and facilitated nuclear translocation of HDAC2, thereby inducing the deacetylation of NF-κB at K310, which is essential for a reduction in NF-κB transcriptional activity. Both TGFβ-induced nuclear translocation of Smad3/HDAC2 and deacetylation of NF-κB at K310 were suppressed by a Smad3 inhibitor (SIS3). These results suggest that the TGFβ-activated Smad3/HDAC2 complex is an essential negative regulator for MUC5AC expression and an epigenetic regulator for NF-κB acetylation. Therefore, these results collectively suggest that modulation of the TGFβ1/Smad3/HDAC2/NF-κB pathway axis can be a promising way to improve lung function as a treatment strategy for asthma and COPD.
