This study is to investigate for obtaining the operating conditions of continuous vinegar production using fluidized bed reactor by Acetobacter aceti cell immobilized in Ca-alginate gel. The optimum conditions obtaining by batch fermentation using fluidized bed reactor were as follows; The fermentation temperature and aeration rate were 3$0^{\circ}C$ and 1.0VVM and the initial concentration of ethanol and acetic acid in medium were 33g/l and 27g/l respectively. The amount of bead used was 25%(w/v). The overall acetic acid productivities of batch fermentations by free cell and immobilized cell were 0.31g/l-hr and 0.48g/l-hr, respectively, at the final acetic acid concentration of 50g/l. In the continuous vinegar production using fluidized bed reactor by immobilized cell under optimum conditions, it was possible to produce 23g/l acetic acid continuously up to 90 days with maximum acetic acid productivity of 2.76g/l-hr at dilution rate 0.12hr-1.
Plant-derived natural products have been and will continue to be valuable sources. Elicitors have been employed to modify cell metabolism in order to enhance the productivity of useful metabolites in plant cell/tissue cultures. In this study, several elicitors were used to improve the productivity of useful metabolites and to reduce culture time for archiving high concentration in P. ginseng hairy root cultures. The addition of chitosan, chitosan oligosaccharide and alginate oligosaccharide to the culture of P. ginseng hairy roots caused growth to be inhibited with the increase in elicitor concentration. The usage of the chitosan elicitor and D-glucosamine caused a slight decrease in hairy root growth, whereas total ginseng saponin accumulated slightly with the increase in elicitor concentration. When gel beads were added to the culture medium at the initial period, hairy root growth was enhanced. The maximum growth was 1.35 times higher than that of the control at $1\%$ (w/v). Total ginseng saponin content decreased due to the addition of alginate beads. This would result in consistent diffusion of lower levels of calcium ions during the culture period that promotes biomass growth.

Effect of the two-stage operation and cell concentration on indirubin production was investigated using bioreactor culture of Polygonum tinctorium. Two-stage culture was operated successfully for 110 days without any adverse effects on continuous indirubin production. Maximum indirubin concentration was found to be at 80 mg/bioreactor. Initial cell concentration significantly affected indirubin production. The indirubin production at 29.2% PCV was improved by 845%, compared to that at 5% PCV. For high-density bioreactor culture of P. tinctorium, a maximum production rate of 10.2 mg indirubin/L day was obtained. Indirubin recovery for bioreactor operation was also examined using XAD-2, XAD-4, XAD-7, and solid silicon. XAD-4 was 1.6-fold more effective than that for solid silicon in indirubin recovery.
Photobiological $H_2$ production was compared using purple non-sulfur bacteria Rhodobacter sphaeroides KD131 in the medium containing various organic acids as the carbon source and electron doner under illumination of $110\;W/m^2$ using halogen lamp at $30^{\circ}C$. The organic acids used were 0~120 mM acetate, butyrate, lactate and malate. Initial pH 7.0 and cell concentration 1.0 at 660nm were increased to pH 8 and 4.4~5.1, respectively during 24hrs of photo-fermentation when lactate and malate were used. However, acetate and butyrate increased pH to 9 and cell concentration to 3.2~3.9 of malate at the same experimental conditions. Optimum ranges of organic acids concentration and carbon/nitrogen ratio were 30~60 mM and 10~20, respectively. When malate was used as the substrate, maximum $H_2$ production 1.1 ml $H_2$/ml broth, which is equivalent to 1.97 mol $H_2$/mol malate was observed.
Batch suspension cultures of hybridoma cell were performed with various initial glutamine concentrations to investigate the effects of glutamine on cell growth and death, monoclonal antibody production, glucose and glutamine consumption, and the production of lactate and ammonium ion. An mathematical kinetic model was formulated to describe the kinetics of cell growth, the consumption of nutrients (glucose and glutamine), and the production of monoclonal antibody and waste metabolites (lactate and ammonium ion) based on experimental data. An equation for the specific growth rate was developed such that superimposed Monod equation in glucose and glutamine, with non-competitive type inhibition relations in ammonium ion and lactate. The inhibition constant for lactate was inversely proportional to the lactate concentration. The specific death rate was considered to be a function of glucose, glutamine, ammonium ion and lactate concentration.
A tower fermentor equipped with a modified settler was used for ethanol fermentation using highly flocculating yeast, Saccharomyces uvarum. The settler was constructed of glass column divided into two chambers by a funnel shaped divider. Gas was allowed to escape from lower chamber of the settler through a small tube. This design significantly reduced the turbulence in upper chamber of the settler and made it possible to operate at high dilution rate. Using the tower fermentor, the effects of operating conditions such as initial glucose concentration, dilution rate and cell recycle ratio were studied. The maximum ethanol productivity, 64.0 g/l' h was obtained at a dilution rate 1.1 h -1 and a cell recycle ratio 5 with the corresponding ethanol concentration of 58.8 g/l, and cell mass of 88 g/l.
The addition of a limited concentration of yeast extract to a minimal salt medium (MSM) enhanced cell growth and increased the production of curdlan whereas nitrogen-limitation was found to be essential for the higher production of curdlan by Agrobacterium sp. ATCC 31749. As the amount of the inoculum increased, the cell growth as well as the production of curdlan also increased in the MSM without a nitrogen source. The cell growth and production of curdlan increased as the initial pH of the medium decreased as low as 5.0. The conversion rate and concentration of curdlan from 2% (w/v) glucose in the MSM with concentrated cells under nitrogen deletion was 67% and 13.4 g/L, respectively. The highest conversion rate of curdlan under the conditions optimized in this study was 71% when the glucose concentrations was 1% (w/v).
The effect of nitrogen source on production of a high viscosity exopolysaccharide, methylan, from methanol by Mentylobacterium organophilum was investigated in fed-batch culture. During the fermentation, cells continued to grow even after the nitrogen source added to the medium was depleted and methylan production was stimulated under the condition which ammonium ion was depleted. Cell growth increased proportionally to the initial concentration of ammonium ion in the medium, but methylan production was significantly inhibited at the high concentration of ammonium ion. As the initial concentration of ammonium ion increased, the specific growth rate, the specific product formation rate and the specific substrate consumption rate decreased due to the inhibitory effect of excess ammonium ions. In order to reduce the inhibitory effect by high concentration of ammonium ion. The control of ammonium ion concentration within the desired level(usually $0.45g/\ell$) was necessary. When ammonium ion concentration was maintained below $0.15g/\ell$ by exponential feeding, methylan production could be increased up to $12.5g/\ell$.
The conditions for production of high cell density of Bifidobacterium longum were investigated and the cross-flow filtration system was used to remove the inhibitory metabolites, lactic acid and acetic acid. The maximum cell growth was observed with glucose as carbon source at the concentration of 50 g/l at $37^{\circ}C$ with the initial pH 6.5. When B. longum was cultured in a cross-flow filtration system, the maximum cell growth was observed at a dilution rate(D) of $0.31\;h^{-1}$ and the dry cell weight was 16.4 g/l($3.5{\times}10^{10}\;cell/ml$), which was about four times higher than that obtained in the batch culture with pH control.
The optimal initial pH for the ethanol production by Saccharomyces K35 was found to be 5.0, and about 80% of yield was obtained when 200g/$\ell$ of glucose was used as a substrate, which showed sugar tolerant. As the additives and cross-linking agent, the addition of 1.67%(w/v) Celite R-634 together with 0.33%(v/v) of glutaraldehyde(ACG bead) resulted in better stability, ethanol productivity and cell viability than Ca-alginate bead. Also, ACG bead seemed to be more resistant to phosphate ion than Ca-alginate bead, considering outgrowing cell concentration in the media. Scanning electron microscopic observation depicted that the surface of ACG bead was almost similar to the original state but not for Ca-alginate bead. When repealpd-batch culture was performed with Ca-alginate bead for 60 days in a 500m1 Erlenmeyer flask, ethanol and cell concentration were maintained about 138g/$\ell$-gel and 29~30g/$\ell$-gel, respectively, up to 40 days(7th run number), and then both were rapidly decreased. In the case of ACG bead, ethanol and cell concentration were maintained about 130~150g/$\ell$-gel and 32~35g/$\ell$-gel, respectively, up to 60days(10th run number). Cell viability was maintained about 70%, and outgrowing cell concentration was below 5.8% of total cell concentration.
