인삼의 효과에 관한 세포생리학적 연구 -제 II 편 Saccharomyces의 분열에 미치는 Saponin과 Histamine의 영향- (A Cellular Physiological Study on the Effects of Korean Ginseng -Part II Effects of Saponin and Histamine on the Division of Saccharomyces-)

  • The effects of saponin and histamine solution on the division of Saccharomyces cerevisiae were compared with the effects of ginseng on the division. 1. $10^{-2},\;10^{-3}%$ saponines were inhibitory on the division but $10^{-5}%$ saponin increased slightly the division, however its increasing rate was much less than that of ginseng. 2. 0.5% histamine was inhibitory on the division but $10^{-2}%$ histamine increased the division, however its increasing rate was higher than that of saponin and lower than that of ginseng. 3. Although some doses of saponin and histamine were inhibitory on the division, the increasing dose of saponin or histamine on the division could be recognized as shown in the case of the optimal dose of ginseng.

葛根湯과 加味葛根湯의 抗알레르기 및 消炎, 解熱, 鎭痛作用에 대한 實驗的 硏究 (An Experimental study on the anti-allergic effects, anti-inflammatory action, anti-pyretic action and analgesic action of Galgeun-tang, Gamigalgeun-tang and Geomahwanggalgeun-tang)

  • Allergic rhinitis and allergic dermatitis are common allergic diseases. Galgeun-tang has been used for treating various diseases, which include allergic rhinitis and allergic dermatitis. Experimental sutdies have been done to research the anti-allergic effects of Galgeun-tang, Gamigalgeun-tang and Geomahwanggalgeun-tang. We have observed: the vascular permeability response induced by serotonin and histamine, the contact dermatitis response induced by picryl chloride, the delayed type hypersensitivity response to SRC, the mice paw edema induced by carrageenin, and the rectal temperature in febrile rats induced by endotoxin, as well as the writhing syndrome induced by $0.7{\%}$ acetic acid. The results were as follows: 1. In the vascular permeability response to intradermal serotonin and histamine, Galgeun-tang and Gamigalgeun-tang showed a significant inhibitory effect. However Geomahwanggalgeun-tang showed an insignificant inhibitory effect. 2. In the contact dermatitis response induced by picryl chloride, Galgeun-tang and Gamigalgeun-tang showed a signigicant inhibitory effect. However Geomahwanggalgeun-tang showed an insignificant inhibitory effect. 3. In the delayed type hypersensitivity response to SRC, Galgeun-tang, Gamigalgeun-tang and Geomahwanggalgeun-tang showed a significant inhibitory effect. 4. In the mice paw edema induced by carrageenin, Galgeun-tang and Gamigalgeun-tang showed a significant inhibitory effect. However Geomahwanggalgeun-tang showed an insignificant inhibitory effect. 5. In the rectal temperature in febrile rats induced by endotoxin, Galgeun-tang, Gamigalgeun-tang and Geomahwanggalgeun-tang showed a significant inhibitory effect. 6. In the writhing syndrome induced by a $0.7{\%}$ acetic acid solution. Galgeun-tang and Gamigalgeun-tang showed a significant inhibitory effect. However Geomahwanggalgeun-tang showed an insignificant inhibitory effect.

Inhibitory Effects of Herbal Extracts on Dopa Oxidase Activity of Tyrosinase

  • Tyrosinase catalyzes the rate-limiting steps in melanin biosynthesis which is involved in skin-coloring and local hyperpigmentation of human beings, and unfavorable darkening of food products, Inhibitory effects on 3,4-dihydroxy-phenylalanine (dopa) oxidase activity of tyrosinase by 594 kinds of herbal extracts prepared from herbal medicines and wild plants in Korea were estimated. Two herbal extracts prepared from radicis cortex of Morus alba and rhizoma of Curcuma longa were selected as those exhibiting potent inhibitory effects on the enzyme activity. These herbal extracts were subjected to sequential fractionations with methylene chloride, ethyl acetate, n-butanol, and polar residue. The inhibitory effects on dopa oxidase activity of tyrosinase were shown in ethyl acetate fraction of Morus alba, and in methylene chloride fraction of Curcuma langa. The ethyl acetate fraction of Marus alba exhibited 50% of inhibition on dopa oxidase activity of tyrosinase at the concentration of 12 ${\mu}g/ml$, and methylene chloride fraction of Curcuma langa at 51 ${\mu}g/ml$.

각종 변이원들에 의해 유도된 돌연변이원성에 대한 수리취 추출물의 억제작용 (Inhibitory Effects of Synurus deltoides Extracts on the Mutagenesis Induced by Various Mutagens)

  • This study was undertaken to determine the antimutagenic effects of Synurus deltoides extracts on the mutagenesis induced by 3-amino-1, 4-dimethyl-5-H-pyrido[4, 3-blindol(Trp-P-1), 2-amitnofluorene (2-AF) and 4-nitroquinolin-1-oxide(4NQO) using Ames test. Raw juice, heated juice, and ethanol extract from Synurus deltoides itself did not induce mutagenesis. The raw juice, heated juice and ethanol extract of 50${mu}ell$/plate showed approximately 90%, 37% and 28% inhibitory effect on the mutagenesis induced by Trp-P-1 against TA98 strain, while 80%, 60% and 58% inhibition was observed on the mutagenesis induced by 2-AF at the concentration of 200${mu}ell$/plate, respectively. TA100 strain was more sensitive than TA98 strain by raw juice, heated juice and ethanol extract on the mutagenesis induced by Trp-P-1 and 2-AF. Meanwhile, the raw juice, heated juice, and ethanol extract showed very limited inhibitory effects on the mutagenesis induced by 4-NQO against TA98 and TA100 strain. These results indicate that raw juice had the strongest inhibitory effect on the Trp-P-1 or 2-AF induced mutagenesis, but all of the extracts had a little antimutagenc effects on the 4-NQO induced mutagenesis.

Structural Changes on the HL-60 Cells of TPA-induced Adherence by Asadisulphide

  • Asadisulphide were purified from Ferrula assafoetida by organic solvent extraction and chromatography. Since ethyl acetate extracts of F. assafoetida has the strongest inhibitory effects on adherence of HL-60 cells, it was reextracted with ethyl acetate, hexane, and ethyl ether and chromatographed three times to isolate asadisulphide. HL-60 cells were grouped into untreated control, TPA-treated, asadisulphide-teated and TPA+asadisulphide-treated groups, and structural changes of these cells were observed using light microscope, scanning electron microscope and transmission electron microscope to examine the inhibitory effects of asadisulfide on the TPA-induced adherence of HL-60 cells. Light microscopic observations showed that asadisulphide has inhibitory effects on the cell aggregation, extention of cytoplasmic processes and inhibition of substrate adhesion of HL-60 cells. Using scanning and transmission electron microscope, it was observed that cell surfaces and several ultrastructures of TPA-treated HL-60 cell were different from control group, while there were no remarkable differences between asadisulphide-treated and TPA+asadisulphide-treated group. These results could suggest that asadisulphide has the inhibitory effects on the TPA-induced structural changes of HL-60 cells.

Paraquat 유독성에 대한 Flavonoid류의 독성경감효과 (Scavenging Effects of Flavonoids on Paraquat Induced Toxicity)

  • To investigate and evaluated the scavenging and antioxidative effects of various flavonoids on paraquat induced toxicity, in vivo and vitro tests of eight flavonoids (catechin, epocatechin, flavone, chrysin, apigenin, quercetin, morin and biochanin A) were carried out. The generation of reactive oxygen substances(ROS) in PMS-NADH system $H_2O_2$ induced hemolysis and lipidperoxidation to blood, NADPH dependent lipidperoxidation to liver and lung microsome by paraquat were studied.The results are summerized as follows; 1) In the concentration ranges from 3.3 to 9.8$\mu$M of catechin,epicatechin, quercetin and biochanin A removed the 50% of DPPH radical scavenging effects. 2) In the concentration ranges from 0.60 to 1.86 mM of catechin, epicatechin, quercetin and biochanin A showed the inhibitory and antioxidative activity on superoxide anion which gernerated in PMA-NADH system. 3) In the concentration ranges from 0.12 to 0.49mM of catechin, epicatechin, quercetin and biochanin A showed the inhibitory and antioxidative activity on H202 which generated in PMA-NADH system. 4) In the concentration ranges from 0.6 x10$^{-5}$ to 6.3 x 10$^{-5}$mM of catechin, epicatechin, flavone, chrysin, quercetin and morin showed the inhibitory and antioxidative activity on $H_2O_2$ induced hemolysis to blood 5) All flavonoids tested exhibited inhibitory and antioxidative effects on paraquat induced liver and tung microsomal lipidperoxidation. Therefore, all flavonoids evaluated showed the useful compounds for scavenger and antioxidant on paraquat induced toxicity.

인진쑥 추출물의 병원성 세균에 대한 생체내 증식 억제 효과 (Inhibitory Effects of Artemisia capillaris Extract on the Pathogenic Bacteria in Mice)

  • Artemisia capillaris THUNB is a perennial herb that belongs to the family Compositae spp and the most common plant among the various herbal folk remedies used in treatment of abdominal pain, hepatitis, chronic liver disease, jaundice and coughing in Korea. This experiment was conducted to investigate the inhibitory effects of orally administrated Artemisia capillaris extracts on the pathogenic bacteria in 200 ICR mice. The experimental groups showed inhibitory effects on the bacteria in $1{\sim}3$ days after inoculation. After 21 days of inoculation, no viable bacterial cells appeared in the feces of both experimental groups while they did appear in the control group. The results of these studies indicate Artemisia capillaris extract exhibited excellent antimicrobial and inhibitory effects on the food poisoning pathogenic bacteria; S. enteritidis, E. coli O157:H7, L. monocytogenes and S. aureus.

Suppressive Effects of a Truncated Inhibitor K562 Protein-Derived Peptide on Two Pro-inflammatory Cytokines, IL-17 and TNF-α

  • Inhibitor K562 (IK) protein was first isolated from the culture medium of K562 cells, a leukemia cell line, and is an inhibitory regulator of interferon-γ-induced major histocompatibility complex class II expression. Recently, exogenous truncated IK (tIK) protein showed potential as a therapeutic agent for inflammation-related diseases. In this study, we designed a novel putative anti-inflammatory peptide derived from tIK protein based on homology modeling of the human interleukin-10 (hIL-10) structure, and investigated whether the peptide exerted inhibitory effects against pro-inflammatory cytokines such as IL-17 and tumor necrosis factor-α (TNF-α). The peptide contains key residues involved in binding hIL-10 to the IL-10 receptor, and exerted strong inhibitory effects on IL-17 (43.8%) and TNF-α (50.7%). In addition, we used circular dichroism spectroscopy to confirm that the peptide is usually present in a random coil configuration in aqueous solution. In terms of toxicity, the peptide was found to be biologically safe. The mechanisms by which the short peptide derived from human tIK protein exerts inhibitory effects against IL-17 and TNF-α should be explored further. We also evaluated the feasibility of using this novel peptide in skincare products.

방풍통성산(防風通聖散)이 고지방사료(高脂肪飼料) 식이(食餌)로 유발(誘發)된 비만(肥滿)생쥐에서 비만유전자(肥滿遺傳子) 및 관련인자(關聯因子)에 미치는 영향(影響) (The Inhibitory Effects of Bangpuangtongseoung-san on the Obese gene and Obese Inhibitory about Obese-mouse induced by High Fat Diet)

  • Objectives : In this study, the aim was to investigate the inhibitory effects of Bangpungtong-seoung-san on the obese gene and obese inhibitory about obese-mouse induced by high fat diet. Methods : In order to investigate the effects of Bangpuangtongseoung-san(BPTS) on the obese gene and obese inhibitory, C57BL/6 mice were induced by high fat diet. C57BL/6 mice were divided into three groups(normal, high fat diet with control, high fat diet with BPTS extract) and fed for 13weeks. Results : 1. The change of body weight and the final increase of body weight were decreased significantly. 2. The amount of the adipocyte in body weight was decreased significantly. 3. In primary adipocytes, ${\beta}3AR$ gene expression was increased significantly and leptin gene expression was decreased in $100{\mu}g/ml$ density significantly. 4. In primary adipocytes, the amount of TNF-${\alpha}$ was decreased in $100{\mu}g/ml$ density significantly and the amount of leptin was decreased but did not show significance. 5. In adipocytes tissue, the expression of leptin was decreased and the expression of ${\beta}3AR$ was increased. Conclusions : These results suggest that BPTS may inhibit the expression of the obese gene mad obese inhibitory about obese-mouse induced by high fat diet.

Inhibitory Effects of Brown Algae Extracts on Histamine Production in Mackerel Muscle via Inhibition of Growth and Histidine Decarboxylase Activity of Morganella morganii

  • This study was performed to investigate the inhibitory effects of brown algae extracts on histamine production in mackerel muscle. First, antimicrobial activities of brown algae extracts against Morganella morganii were investigated using a disk diffusion method. An ethanol extract of Ecklonia cava (ECEE) exhibited strong antimicrobial activity. The minimum inhibitory concentration (MIC) of ECEE was 2 mg/ml. Furthermore, the brown algae extracts were examined for their ability to inhibit crude histidine decarboxylase (HDC) of M. morganii. The ethanol extract of Eisenia bicyclis (EBEE) and ECEE exhibited significant inhibitory activities (19.82% and 33.79%, respectively) at a concentration of 1 mg/ml. To obtain the phlorotannin dieckol, ECEE and EBEE were subjected to liquid-liquid extraction, silica gel column chromatography, and HPLC. Dieckol exhibited substantial inhibitory activity with an $IC_{50}$ value of 0.61 mg/ml, and exhibited competitive inhibition. These extracts were also tested on mackerel muscle. The viable cell counts and histamine production in mackerel muscle inoculated with M. morganii treated with ${\geq}2.5 $ MIC of ECEE (weight basis) were highly inhibited compared with the untreated sample. Furthermore, treatment of crude HD-Cinoculated mackerel muscle with 0.5% ECEE and 0.5% EBEE (weight basis), which exhibited excellent inhibitory activities against crude HDC, reduced the overall histamine production by 46.29% and 56.89%, respectively, compared with the untreated sample. Thus, these inhibitory effects of ECEE and EBEE should be helpful in enhancing the safety of mackerel by suppressing histamine production in this fish species.
