Anticariogenic Effects and Inhibition of Glucosyltransferase Activity of Chrysanthemun indicum L. Extracts (감국(Chrysanthemun indicum L.) 추출물의 항충치효과와 Glucosyltransferase 저해활성 탐색)

  • The anticariogenic effects and inhibition of glucosyltransferase activity of Chrysanthemun indicum L. extracts were investigated against 5 strains of microorganisms. The ethanol extracts of Chrysanthemun indicum L. showed the growth inhibition effects on the Streptococcus mutans and oral bacteria. The inhibition rate of glucosyltransferase activity of Chrysanthemun indicum L. extracts showed 78.4 to 92.3% range. The extracts were analysed by using solvent fractionation, silica gel adsorption chromatography, Sephadex LH-20 column chromatography, TLC, HPLC techniques. Three components whose molecular weights ranged from 200 to 400 were confirmed to have the anticariogenic activity.

Effect of Inhibition on Glucosultransferase and Antimicrobial Activity of Polyphenol Fraction of Gallnut and Red Grape Husk (오배자와 적포도과피 폴리페놀 분획물의 항균성 및 Glucosyltransferase 저해효과)

  • The antimicrobial and glucosyltransferase(GTase) inhibition activity were searched for 30 species of various folk drugs and by products of food industry. Among them, two species, gallnut and red grape husk water extracts, were selected for the powerful antimicrobial and GTase inhibition activity. The polyphenol fractions of gallnut and red grape husk were showed very greater antimicrobial activity on both Gram(+) and (-), B. subtilis and E. coli. The minimum antimicrobial activity of gallnut polyphenol fraction were 1.0mg for B. subtilis and 3,0mg for E. coli. Red grape husk was 2.0mg for B. subtilis and 3.0mg for E coli. The polyphenol fractions of gall nut and red grape husk were showed powerful GTase inhibition activity. The concentrations of these fractions for 80% inhibition of GTase activity were 1.08$\times$10$\^$-3/mg/㎖ and 1.08$\times$10$\^$-2/mg/㎖, respectively. The most abundant compound in these fraction seems to be polyphenol derivatives. From these results, we think that the gallnut and the red grape husk polyphenol fraction had more antimicrobial and anti-plaque activities than artificial synthetic preservatives as an economic point of view.

  • We investigated the inhibition effects of teeth decalcification and glucosyltransferase(GTase) activity on the Juniperus rigida S. et Z. Teeth decalcifications by Streptococcus mutans were respectively inhibited to 70.13, 74.93% on methanol and n hexane fractions. In the inhibition test of GTase activities by solvent fractions of J. rigida, they were respectively inhibited to 86.6, 89.5% to a similar degree. And in the result to identify GTase produced by S. mutans with SDS PAGE, the band near 65KD estimated as GTase did not show in the lanes of methanol, n hexane and chloroform fractions.

  • In the course of our studies, novel flavan-3-ol structure isolated from Theobroma cacao L. husk was established by the thiolysis, desulfurization and spectroscopic method. The structure was identified for cinnamtannin A-2 containing the tetrameric epicatechin and molecular weight was [1153] by FAB-MS negative ion. The inhibitory effect on the glucosyltransferase activity was investigated. Cinnamtannin A-2 showed complete inhibition at 0.03 mM and inhibited on the glucosyltransferase noncompetitively. The hydroxyl group of flavan-3-ol was supposed to be the essential element for inhibition on the glucosyltransferase.

  • Dental caries is one of the most common oral diseases in the world. Glucosyltransferase (Gtase) of Streptococcus mutans (S. mutans) plays an important role in the develo pment of dental caries. For the purpose to develop anti- caries, we examined the effect of metabolites isolated from Streptomyces sp. M-20 on Gtase and the growth of S. mutans. Streptomyces sp. M-20 isolated from Mongolian soil showed 95~96% sequence homology with that of Streptomyces lin- colnensis. The metabolites of Streptomyces sp. M-20 were partially purified by extraction with ethyl acetate, silica gel column chromatography and preparative TLC. Partially purified metabolite, red colored component (MR-20) in ethyl acetate fraction showed potent antibacterial activitiy against S. mutans and inhibitory activity against Gtase purified from S. mutans, while another isolated yellow component (MY-20) showed no activity against S. mutans. The inhibitory activity of MR-20 against Gtase was confirmed by activity staining on SDS-polyacrylamide gel electrophoresis. The concentration of MR-20 for 50% inhibition $(IC_{50})$ against Gtase activity was $60{\mu}g/ml$. These results suggest that MR-20 can be developed for antibacterial agent and anticaries.

  • In the course of studies for anti-plaque agents, novel procyanidin structure isolated from Artocarpus heterophyllus folium was established by thiolysis and spectroscopic analysis. The chemical structure was identified for $(-)-epiafzelecin-(4{\beta}{\rightarrow}8)-afzelecin-(4{\alpha}-8)-catechin$ containing the trimeric flavan-3-ols and molecular weight was 833[M-H] by FAB-MS negative ion method. The inhibitory effect on the glucosyltransferase activity was investigated, novel compound showed complete inhibition at 1.0 mM and inhibited on the glucosyltransferase noncompetitively.

Anticariogenic Activity and Glucosyltransferase Inhibition of Phenolic Compounds (페놀성 화합물의 항충치활성 및 Glucosyltransferase 억제효과)

  • Fourteen phenolic compounds(benzoic acid, p-hydroxybenzoic acid, protocatechuic acid, vanillic acid, syringic acid, gallic acid, caffeic acid, ferulic acid, (+)-catechin, quercetin, rutin, catechol, chlorogenic acid and L-ascorbic acid) were examined for their effects on the anticarigenic activity. Among tested samples, catechol was significantly inhibited the S. mutans, exhibiting an clear zone 18.5-19.5mm by 10 mg/disc level. The minimal inhibition concentration(MIC) of the phenolic compounds for Streptococcus mutans, M1 and M2 strain were determined as 2,000 ppm, whereas catechol was 1,000 ppm. The activity of glucosyltransferase(GTase) was significantly inhibited by catechol, at 10 ppm(58.7%), 50 ppm(60.7%) and 100 ppm(88.4%) and 500 ppm(89.6%), respectively. Among them, catechol showed most significant anticariogenic activity as well as inhibition of GTase activity.

Isolation of 3-Galloylprocyanidin B3, a Glucosyltransferase Inhibitor from the Korean Green Tea Leaves

  • In the course of surveying the anti-plaque agents for dental caries prevention, the extract of Korean green tea leaves (KGTL) was tested for inhibitory activity against Streptococcus mutans adhering to glass surfaces in the presence of crude glucosyltransferase (GTase). The extracts of KGTL showed a positive inhibitory activity against GTase. The active compound was purified through Sephadex LH-20 and MCI gel CHP-20P columns. A positive reaction was shown in the anisaldehyde-$H_2SO_4$ test, which confirmed the condensed tannin. The inhibitory compound was identified as 3-galloylprocyanidin $B_3$ through IR, negative FAB-mass, and $^{1}H$-NMR spectroscopic analyses. Acetone extract and 3- galloylprocyanidin $B_3$ of KGTL showed inhibitory effect against GTase. The percent of inhibition was determinated to be 71.84% (P<0.01) with 10 mM 3-galloylprocyanidin B3. The 3-galloylprocyanidin $B_3$, which possessed a galloyl, showed a higher inhibitory activity against glucosyltransferase than monomeric (+)-catechin and procyanidine $B_3$ which had no galloyl group.

  • For glucosyltransferase(GTase) inhibitors, two flavan-3-ols were isolated from Theobroma cacao beam husk. They showed positive reaction with $anisaldehyde-H_2SO_4$ solution, $FeCl_3$ to be confirmed as dimeric flavan-3-ols on TLC and were identified as procyanidin B-1 [(-)-epicatechin-$(4{\beta}{\rightarrow}8)$-catechin] and procyanidin B-3 [(+)-catechin-$(4{\beta}{\rightarrow}8)$-catechin by spectroscopic analysis. Their inhibitory effect on glucosyltranaferase activity was also investigated and procyanidin B-1 showed 50% inhibition at 0.3 mM. They inhibited on the glucosyltransferasa noncompetitively and dimeric flavan-3-ol containing (-)-epicatechin had higer inhibitory activity.

Isolation and Identification of Anticariotic Compound from Sophora flavescens Ait. (고삼으로부터 항우식활성 물질의 분리)

  • The purpose of this study is to investigate anticariotic activity of the ethyl acetate soluble extract of Sophora flavescens Ait. for the prevention of dental caries and glucosyltransferase activity caused by Streptococcus mutans. The fraction 5-4-3 showed strong growth inhibition activity against Streptococcus mutans (MIC, 3.13 $\mu\textrm{g}$/ml). The glucosyltransferase activity of the active fraction 5-4-3 inhibited the formation of glucan and showed 77% of the antiproliferative effect at 100 $\mu\textrm{g}$/ml (P<0.05). Two flavanones, (2S)-2'-methoxy kurarinone (1) and (+)-kurarinone (2), were isolated from the active fraction 5-4-3 of the ethyl acetate soluble extract of S. flavescens Ait. Their structures were elucidated using spectroscopic methods.
