Korean Medicinal Herb Extracts Inhibit Melanin Formation in Clone M-3 Mouse Melanocyte Cell Lines

  • In order to search for anti -melanin formation agents from Korean medicinal herbs, we selected 21 Korean medicinal herbs, based on a review of Korean traditional medicine books and the recommendations of Korean traditional medical doctors. We tested for inhibition effect of melanin pigmentation of Clone M-3 mouse melanocyte cell lines when we treated the extracts of 21 medicinal herbs in the mouse melanocyte cell lines, respectively. Among 21 medicinal herb extracts, 5 extracts showed a inhibition effect of melanin formation. The sample Phaseolus radiatus L, Cordyceps militaris, Pinellia ternata, Phellinus linteus and Citrus junos Tanaka showed a significantly little formation of melanin pigments compared with control groups. Especially extract of Citrus junos Tanaka was more potent than the others. These results suggest that extract of Korean Citrus junos may represents an excellent candidate for inhibition of melanin pigmentation at in vitro level.

Inhibition Effects of Toxic Solvent Mixture in Catalytic Oxidation Process (유독성 유기용매의 촉매산화공정에서 혼합조성에 따른 간섭효과)

  • The selective catalytic oxidation of toxic aromatic solvents (benzene, toluene, ethylbenzene, and styrene) and their mixtures were studied on a $Pt/{\;}{\gamma}-Al_2O_3$ catalyst at temperature ranging from $160~350^{\circ}C$. The deep conversion of aromatic solvents was increased as the inlet concentration was decreased and the reaction temperature was increased. The reactivity increases in order benzene > toluene > ethylbenzene > styrene. In mixture, remarkable effects on reaction rate and selectivity have been evidence ; the strongest inhibition effect is shown by styrene and increase in a reverse order with respect to that of reactivity. The inhibition effect was increased in order styrene > ethylbenzene > toluzene > benzene. This trend is due to the competition adsorption between the two or three reactants on the oxidized catalyst. Also, the deep conversion change of benzene was a small in tertiary mixtures(including of benzene and styrene) comparing with conversion characteristics of binary mixture with styrene. This result was due to small concentration of styrene. which had very strong inhibition effect.

The inhibitive effect of erythritol on growth and acidogenic ability of Streptococcus mutans (에리스리톨의 Streptococcus mutans에 대한 성장력과 산생성능의 억제효과)

  • The purpose of this study was to closely examine the inhibitive effect of erythritol on growth and acidogenic ability of Streptococcus mutans. As expected, the growth of S. mutans was comparably increased with the addition of sucrose. However, xylitol and erythritol remarkably reduced the growth of S. mutans. Growth inhibition was detected at more than 5% of erythritol although xylitol showed growth inhibition effect at all concentrations tested. Growth inhibition effect was monitored with the combination of same concentration of erythritol and other carbohydrates. Combination of 5% or 10% erythritol with xylitol showed effective growth inhibition. Addition of 2.5%, 5%, or 10% erythritol with sorbitol also showed growth inhibition. From these results, erythritol showed potency of growth inhibition of S. mutans, which is involved in dental caries, and was confirmed to be an excellent sugar substitute, which has effect on preventing caries.

  • To study antimicrobial activities of Impatiens balsamina, its leaf and stem was extracted by methanol and water. The extracts were tested their microbial inhibition activities against several microorganisms, staphylococcus aureus KCTC 1928, Escherichia coli KCTC 1116, Klebsiella pneumoniae KCTC 2690, Trichophyton mentagrophytes KCTC 6316. The methanol extract and water extract of the leaf and stem didn't show the inhibition effect on E. coli but showed the inhibition effect against the ohter 3 species of the microorganisms tested. The extract of leaf showed higher inhibition than the extract of stem. And the methanol extract showed higher inhibition than the water extract.

Regulation of NAD+- Specific Isocitrate Dehydrogenase from Pythium ultimum

  • The $NAD^+$-specific activity of a dual coenzyme-specific isocitrate dehydrogenase (IDH; EC 1.1.1.41) from the primitive fungus Pythium ultimum was investigated to elucidate the regulatory factors that may influence the intracellular distribution of carbon and the availability of intermediates, e.g. citrate, for fatty acid synthesis. Inhibition of $NAD^+$-IDH activity by diphospho- and triphosphonucleotides (ATP, ADP, and GTP) reflected the sensitivity of this enzyme to cellular energy charge even though monophosphonucleotides (AMP and GMP) had little effect on activity. NADPH, but not NADH, substantially inhibited $NAD^+$-IDH activity, showing noncompetitive inhibition with isocitrate. Oxalacetate and ${\alpha}$-ketoglutarate showed competitive inhibition with isocitrate, while citrate and cis-aconitate showed mixed-noncompetitive inhibition with isocitrate. Inhibition by these substances ranged from 29 to 46% at 10 mM. The inhibitory effect of oxalacetate was increased synergistically by glyoxylate, which alone caused 31% uncompetitive inhibition at 10 mM, and a mixture of the two substances at 1 mM each showed 98% inhibition of $NAD^+$-IDH activity. The regulation of $NAD^+$-IDH in Pythium ultimum seems to be a complex process involving mitochondrial metabolites. The addition of glyoxylate (3 mM) and oxalacetate (3 mM) to the culture medium resulted in the production of 49% more lipid by P. ultimum.

The Experimental Study on Antifungal effects of mixture of Sophorae Subprostratae Radix, Acoti Radix, and Hibisci Syriaci Cortex by the Three Types of Extraction on three species of the dermatophytes. (다양한 山豆根·川烏·木槿皮 혼합물 용액이 足部白癬菌에 미치는 영향)

  • This experimental study was performed to investigate the antifungal effects of mixture of Sophorac Subprostratae Radix, Aconti Radix, and Hibisci Syriaci Cortex by the three types of extraction in the dermatophytes such as Trichophyton rubrum, Trichophyton mentagrophytes, and Epidermophyton floccosum. The following results were obtained by using Minimum Inhibition Concentration(MIC) and Inhibition Zone. 1. MIC on Trichophyton mentagrophytes in Sample S-2, Sample S-3, and Sample-Y were 10${\mu}l$, respectively, showing the highest antifungal effect. 2. MIC on Epidermophyton floccosum in Sample S-2, Sample S-3, and Sample-Y were 10㎕ respectively, showing the highest antifungal effect. 3. MIC on Trichophyton rubrum in Sample S-3, Sample-Y were 10${\mu}l$ respectively, showing the highest antifungal effect. 4. The size of inhibition zone on Trichophyton mentagrophytes for Sample S-3 were 22.3mm in 10${\mu}l$, showing the highest antifungal effect. 5. The size of inhibition zone on Epidermophyton floccosum for Sample S-3 was 34.3mm in 10${\mu}l$, showing the highest antifungal effect. 6. The size of inhibition zone on Trichophyton rubrum for Sample-Y was 26.5mm in 10${\mu}l$, showing the highest antifungal effect. Whereas. in extractions, that of Sample S-3 was 12.5mm in 10${\mu}l$, showing the highest antifungal effect. 7. From the above results, Sample S-3 on Trichophyton mentagrophytes, Epidermophyton floccosum and Sample-Y on Trichophyton rubrum showed the highest antifungal effects. This results suggests that vinegar extraction excels other extraction method using water or ethanol to measure the antifungal effect on dermatophytes. In addition, the result shows that the extract of herbal medicines can be used to tinea pedis and if further study is performed, the use of the extract of herbal medicines will be valuable and beneficial in the clinical medicines. The effect of wrinkle care and whitening by Rhus verniciflua Stokes (건칠의 항주름 효과와 미백효과 연구)

  • Objectives : Recently, the demands for the effective and safe depigmentating and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The aim of this study is to search new materials from the traditional herbal medicines which inhibit the aging process of skin in vitro.Methods : Human dermis cell (HS68) was used to test the effect of Rhus verniciflua Stokes (RVS). 80% ethanol or water extracts were screened for their inhibitory activities against elastase. Elastase inhibition effect was tested by microplate reader instrument. And MMP-1 suppression effect of RVS was tested by western blot. These cells were investigated the viability by MTS assay. And also the inhibition effect of tyrosinase by RVS was tested.Results : RVS (final concentrstion 1 ㎎/㎖ appeared over 30% of inhibition of elastase activity. So we are investigated anti wrinkle effects of Rhus verniciflua Stokes look through MMP-1 inhibition activity, also Extracts of RVS showed higher anti-tyrosinase activity than arbutin as final concentration 1 mg/ml. These results suggest that herbal medicines could be strong potential sources of inhibition of anti-aging and whitening effects for the skin.Colclusions : RVS was the best suppressor candidate of elastase activity among other control oriental drugs. It was found that RVS did not have toxicity to cells. We found proper concentration of RVS to treat in HS68 culture and investigated not only wrinkle care effect but also whitening effect of RVS.

Antioxidative Properties of Amaranth Cauline Leaf and Suppressive Effect against CT-26 Cell Proliferation of the Sausage Containing the Leaf

  • The study investigated antioxidative properties and rectal cancer cell inhibition effect of amaranth (Amaranthus cruentus L.) cauline leaves (ACL) to produce the sausage with ACL powder (ACLP). Antioxidative effects of ACLP prepared with different stem lengths (10-45 cm) were evaluated through DPPH, ABTS, reducing power, total phenol, and total flavonoid. Inhibition effect on rectal cancer cells growth was also examined with CT-26 cell. To determine appropriate ACL amounts in sausage formula, response surface methodology was used. The sausages without ACL (control) and the sausage with ACL (ACLP sausage) were the subjected to the examinations of antioxidation, growth inhibition on CT-26, and physicochemical properties (pH and water content). ACLP made from the leaf with 15 cm length stem generally showed the highest antioxidative effect through results of DPPH, ABTS, reducing power, total phenol, and total flavonoid. ACLP also showed inhibition effect on the proliferation of CT-26, depending on concentration of ACLP. The surface response model showed that 4.87 g of ACLP was optimized amount for sausage production. Physicochemical properties between optimized ACLP and control sausages were not significantly different. Higher antioxidative effect of optimized ACLP sausage extract was observed (p<0.05) in antioxidation tests than control sausage extract except for DPPH. Cell viability of CT-26 cells were higher (p<0.05) in ACLP than in control sausage extracts. These results indicate that ACLP has functional effects on antioxidation activity and growth inhibition on CT-26 cell, and thus, it should be useful as a supplement in sausage, which may some effect as ACLP itself.

The Comparative Study of the Effects of Fructificatio Inonoti Obliqui Aqueous Extract according to the Extraction Temperature(II) -Anti-oxidativy Activity, anti inflammatory effect and cancer cell multiplication inhibition effect- (차가버섯 물 추출물의 추출온도에 따른 효능 비교 연구(II) -항산화 효능, 소염 및 항암 효과 연구-)

  • Objectives : The present study purposed to compare the antioxidant effect, anti inflammatory effect and cancer cell multiplication inhibition effect of Fructificatio Inonoti Obliqui aqueous extract according to extraction temperature. Methods : We medicated animal models, which had experimental oxidation, with Fructificatio Inonoti Obliqui total extract and $50^{\circ}C$ low temperature leachate, and performed hematological analysis and blood chemical analysis with measuring SOD, GSH, catalase, NO and MDA content in the liver. In addition, we made comparative observation of anti inflammatory effect and anti-cancer effect. Results : Compared to the control group, both the group medicated with Fructificatio Inonoti Obliqui total extract and with $50^{\circ}C$ low-temperature leachate were found to decrease the number of thrombocytes in blood plasma and NO content while to increase SOD activity and catalase activity significantly. Both groups also showed anti-inflammatory effect against THP-1 cells and a multiplication inhibition effect against liver cancer cells and stomach cancer cells significantly. Conclusions : Both Fructificatio Inonoti Obliqui total extract and Fructificatio Inonoti Obliqui $50^{\circ}C$ low-temperature leachate have significant antioxidant effect, anti inflammatory effect and anti cancer effect.

The Effect of Salviae Radix on Oxidat-Inhibition of Phosphate Uptake in Renal Proximal Tubular Cells (단삼약침액(丹蔘藥鍼液)이 신장(腎臟) 근위세뇨관세포(近位細尿管細胞)에서 산화제(酸化劑)에 의한 인산(燐酸)의 이동억제(移動抑制)에 미치는 영향(影響))

  • This study was undertaken to determine if Salviae Radix (SR) exerts protective effect against oxidant-induced inhibition of phosphate uptake in renal proximal tubular cells. Membrane transport function and cell death were evaluated by measuring phosphate uptake and trypan blue exclusion, respectively, in opossum kidney (OK) cells, an established proximal tubular cell line. $H_2O_2$ was used as a model oxidant. $H_2O_2$ inhibited the phosphate uptake in a dose-dependent manner over the concentration range of 0.1-0.5 mM. Similar fashion was observed in cell death. However, the phosphate uptake was more vulnerable to $H_2O_2$ than cell death, suggesting that $H_2O_2$-induced inhibition of phosphate uptake is not totally attributed to cell death. Decreasedphosphate uptake was associated with ATP depletion and inhibition of $Na^+$-pump activity as determined by direct inhibition of $N^+-K^+$-ATPase activity. When cells were treated with $H_2O_2$ in the presence of 0.05% SR, the inhibition of phosphate uptake and cell death induced by $H_2O_2$ was significantly attenuated. SR restored ATP depletion and decreased $Na^+-K^+$-ATPase activity, and this is likely responsible for the protective effect of SR on decreased phosphate uptake. The protective effect of SR was similar to the $H_2O_2$ scavenger catalase. SR reacts directly with $H_2O_2$ to reduce the effective concentration of the oxidant. The iron chelator deferoxamine prevented the inhibition of phosphate uptake and cell death induced by $H_2O_2$, suggesting that $H_2O_2$-induced cell injury is resulted from an iron-dependent mechanism. These results indicate that SR exerts the protective effect against $H_2O_2$-induced inhibition of phosphate uptake by reacting directly with $H_2O_2$ like the $H_2O_2$scavenger enzyme catalase, in OK cells. However, the underlying mechanism remains to be explored.
